Gut microbiota-testis axis: FMT improves systemic and testicular micro-environment to increase semen quality in type 1 diabetes.

BACKGROUND: Clinical data suggest that male reproductive dysfunction especially infertility is a critical issue for type 1 diabetic patient (T1D) because most of them are at the reproductive age. Gut dysbiosis is involved in T1D related male infertility. However, the improved gut microbiota can be used to boost spermatogenesis and male fertility in T1D remains incompletely understood. METHODS: T1D was established in ICR (CD1) mice with streptozotocin. Alginate oligosaccharide (AOS) improved gut microbiota (fecal microbiota transplantation (FMT) from AOS improved gut microbiota; A10-FMT) was t

Large Neutral Amino Acid Therapy Increases Tyrosine Levels in Adult Patients with Phenylketonuria: A Long-Term Study.

The standard treatment for phenylketonuria (PKU) is a lifelong low-phenylalanine (Phe) diet, supplemented with Phe-free protein substitutes; however, adult patients often show poor adherence to therapy. Alternative treatment options include the use of large neutral amino acids (LNAA). The aim of this study was to determine the Phe, tyrosine (Tyr), and Phe/Tyr ratio in a cohort of sub-optimally controlled adult patients with classical PKU treated with a new LNAA formulation. Twelve patients received a Phe-restricted diet plus a slow-release LNAA product taken three times per day, at a dose of 1

Pharmacokinetic optimisation in the treatment of gastro-oesophageal reflux disease.

Gastro-oesophageal reflux disease (GORD) is a very common disorder of upper gastro-intestinal motility, differing widely in severity and prognosis. Medical therapy of GORD has involved antacids, alginates, prokinetic agents and antisecretory compounds, primarily H2 receptor antagonists and proton pump inhibitors. Knowledge of the pharmacokinetics of these compounds is important, to optimise the therapeutic benefit in each patient. GORD patients are often elderly and pharmacokinetics are move variable in this group. Furthermore, they often suffer from other diseases needing medical therapy and

Tailored therapy guided by multichannel intraluminal impedance pH monitoring for refractory non-erosive reflux disease.

A relevant percentage of non-erosive reflux disease (NERD) is refractory to proton pump inhibitors (PPIs) treatment. Multichannel intraluminal impedance pH (MII-pH) monitoring should give useful pathophysiological information about refractoriness. Therefore, our aim was to assess whether this technique could be useful to guide a 'tailored' therapy in refractory NERD. We retrospectively recruited NERD patients undergoing MII-pH monitoring for unsuccessful treatment. All patients had undergone upper endoscopy, and those with erosive esophagitis were excluded. No patient received PPI during MII-p

Spatiotemporal Targeting Randle Cycle and Immune Checkpoint for Potent Antitumor Therapy.

Tumor metabolic reprogramming plays a crucial role in cancer progression and therapeutic resistance. The competitive and compensatory relationship between glucose and lipid metabolism-known as the Randle cycle-poses a major challenge to single-pathway metabolic inhibition strategies. In this study, we developed a glucose oxidase-based nanogel (GOX-NG) system using catechol-functionalized alginate, which exhibits enhanced tumor penetration, prolonged retention, and sustained glucose depletion in the tumor microenvironment. When combined with etomoxir (ETX), a fatty acid oxidation (FAO) inhibito

Dynamically responsive hydrogel with mechanical stimulation enhances diabetic wound healing via activation of Piezo1-mediated efferocytosis.

Correcting the disordered metabolism and achieving dynamic, comprehensive management of chronic diabetic wounds remains a significant challenge. This study presents a double-network dynamic hydrogel exhibiting long-term anti-inflammatory, antioxidant properties, and tunable mechanical strength. The hydrogel is primarily composed of modified chitosan, hyaluronic acid, sodium alginate, and ZnO2/Fe3+ nanoparticles. The incorporated ZnO2/Fe3+ nanoparticles enable microenvironmental regulation by responding to H+ or reactive oxygen species (ROS), while the released Fe3+ ions drive hydrogel network

Microneedle drug delivery system based on hyaluronic acid for improving therapeutic efficiency of hypertrophic scars.

Hypertrophic scar (HS) is a disease with excessive skin fibrosis and collagen disorder, which is generally caused by abnormal wound repair process after burn and trauma. Although intralesional injection of 5-fluorouracil (5-Fu) has been used in clinical treatment of HS, the patients' compliance of injection treatment is poor. In this study, a double-layer dissolution microneedle (MN) containing asiaticoside (AS) and 5-Fu was designed for the treatment of HS. Biological macromolecules materials affected the formability, drug release, and hardness of MNs. Therefore, several types of biomacromole

A Semisynthetic Oligomannuronic Acid-Based Glycoconjugate Vaccine against Pseudomonas aeruginosa.

Pseudomonas aeruginosa is one of the leading causes of nosocomial infections and has become increasingly resistant to multiple antibiotics. However, development of novel classes of antibacterial agents against multidrug-resistant P. aeruginosa is extremely difficult. Herein we develop a semisynthetic oligomannuronic acid-based glycoconjugate vaccine that confers broad protection against infections of both mucoid and nonmucoid strains of P. aeruginosa. The well-defined glycoconjugate vaccine formulated with Freund's adjuvant (FA) employing a highly conserved antigen elicited a strong and specif

A Sustainable Solution to Skin Diseases: Ecofriendly Transdermal Patches.

Skin is the largest epithelial surface of the human body, with a surface area of 2 m2 for the average adult human. Being an external organ, it is susceptible to more than 3000 potential skin diseases, including injury, inflammation, microbial and viral infections, and skin cancer. Due to its nature, it offers a large accessible site for administrating several medications against these diseases. The dermal and transdermal delivery of such medications are often ensured by utilizing dermal/transdermal patches or microneedles made of biocompatible and biodegradable materials. These tools provide c

Organic matter affects phosphorus recovery during vivianite crystallization.

Vivianite crystallization is a promising route for phosphorus (P) recovery from P-rich wastewater. However, organic matter (OM) in wastewater may influence vivianite formation. In this study, the effects of four representative OMs, glucose, bovine serum albumin (BSA), humic acid (HA) and sodium alginate (SA), on P recovery by vivianite were investigated. The results showed that P recovery efficiency was inhibited by HA and SA, declining by 3.7% and 12.1% under HA (100 mg/L) and SA (800 mg/L), respectively. BSA, HA and SA affected the aggregated form of vivianite crystals. Vivianite particle si

HPV Lesions and Other Issues in the Oral Cavity Treatment and Removal without Pain.

Due to different oral and dental conditions, oral mucosa lesions such as those caused by the human papilloma virus and temporomandibular joint pathologies often have to be treated by surgical, ablative or extractive procedures. The treatment and control of pain and inflammation during these procedures is essential to guarantee the patient's well-being. For the foregoing reason, a hydrogel based on sodium alginate and hyaluronic acid containing 2% of ketorolac tromethamine has been developed. We characterized it physically, mechanically and morphologically. The rheological results suggest that

Hydrogel-assisted neuroregeneration approaches towards brain injury therapy: A state-of-the-art review.

Recent years have witnessed the development of an enormous variety of hydrogel-based systems for neuroregeneration. Formed from hydrophilic polymers and comprised of up to 90% of water, these three-dimensional networks are promising tools for brain tissue regeneration. They can assist structural and functional restoration of damaged tissues by providing mechanical support and navigating cell fate. Hydrogels also show the potential for brain injury therapy due to their broadly tunable physical, chemical, and biological properties. Hydrogel polymers, which have been extensively implemented in re

Biomolecules for Removal of Heavy Metal.

BACKGROUND: Patents reveal that heavy metals are natural constituents of the earth's crust, but some heavy metals like cadmium, lead, mercury, arsenic etc. are injurious to living organisms at higher concentration. Nowadays, anthropogenic activities have altered geochemical cycles and biochemical balance of heavy metals. Biomolecules are used nowadays for removal of heavy metals compared to other synthetic biosorbents due to their environmental friendly nature and cost effectiveness. OBJECTIVE: The goal of this work is to identify the role of biomolecules like polysaccharides, polypeptid

Construction of anticoagulant poly (lactic acid) films via surface covalent graft of heparin-carrying microcapsules.

Heparin has been widely investigated in medicine and biomedical applications as an anticoagulant. This paper described a covalent modification of poly (lactic acid) (PLA) films with heparin-carrying microcapsules. The heparin-carrying microcapsules were achieved by layer-by-layer (LbL) self-assembly technology by which poly (allylamine hydrochloride) (PAH) and heparin were alternately coated on the Ca(2+)-cross-linked alginate hydrogel. The microcapsules were characterized by scanning electron microscopy (SEM), zeta potential analysis and Fourier transform infrared spectroscopy (FTIR), and the

Pomelo-derived exosomes suppress vesicular stomatitis virus infection through GSK3β dependent regulation of innate immune signaling.

RNA virus infection triggers robust innate immune activation and inflammatory injury. Using vesicular stomatitis virus (VSV) as a model, we investigated the antiviral and immunomodulatory effects of pomelo-derived extracellular vesicles (EVPomelo). Among multiple plant vesicles screened, EVPomelo exhibited the strongest anti-VSV activity in vitro and in vivo. LC-MS analysis identified penicillic acid (PA) as an important endogenous bioactive component. Mechanistically, EVPomelo suppressed viral replication and reduced IFN-β and IL-6 production, accompanied by the inhibition of the signal trans

A pH-responsive hydrogel facilitates osteogenesis by coordinating immunomodulation and ROS scavenging.

Diabetic bone defects suffer from impaired self-repair due to pathological microenvironments characterized by excessive reactive oxygen species (ROS) accumulation and immune dysregulation. To address this challenge, we developed a multifunctional composite hydrogel (Gel/ZR) by integrating resveratrol (RSV)-loaded ZIF-8 nanoparticles into an oxidized alginate-gelatin self-healing matrix. This system leverages the pH-responsive degradation of ZIF-8 to enable targeted release of RSV and Zn2+ in acidic bone defect sites. Comprehensive characterization confirmed the hydrogel's injectability, self-h

Metal-ion-coordinated silk fibroin nanogels for enhanced oral chemotherapy of colorectal cancer.

The efficacy of oral chemotherapy for colorectal cancer (CRC) is hampered by poor drug stability and absorption in the upper gastrointestinal tract, as well as inadequate targeting efficiency at tumor sites. To address these issues, we proposed a simple and biocompatible nanogel, which was co-assembled from chemotherapeutic drug 5-fluorouracil (5-FU), regenerated silk fibroin (SF) as a natural protein carrier, and metal ions (Ni2+/Cu2+). The obtained nanogel system exploited the coordination interactions among 5-FU, amino acid residues, and metal ions to form a multifunctional oral nano-drug s
