Preclinical Models to Study the Molecular Pathophysiology of Meniere's Disease: A Pathway to Gene Therapy.

Background: Meniere's disease (MD) is a set of rare disorders that affects >4 million people worldwide. Individuals with MD suffer from episodes of vertigo associated with fluctuating sensorineural hearing loss and tinnitus. Hearing loss can involve one or both ears. Over 10% of the reported cases are observed in families, suggesting its significant genetic contribution. The condition is polygenic with >20 genes, and several patterns of inheritance have been reported, including autosomal dominant, autosomal recessive, and digenic inheritance across multiple MD families. Preclinical research us

Protective CTL-dependent immunity and enhanced immunopathology in mice immunized by particle bombardment with DNA encoding an internal virion protein.

Anti-viral CTL were induced in vitro using a particle bombardment device or "gene-gun" to deliver plasmid DNA encoding the nucleoprotein of the lymphocytic choriomeningitis virus (LCMV). Using this plasmid we were able to study T cell-mediated immunity in the absence of a neutralizing Ab response. Upon a single DNA immunization, a nearly 2 log10 reduction in splenic viral titers was observed 3 days after LCMV infection. After two or three immunizations a greater than 3 log10 inhibition of viral titers in the spleen was observed, with most animals having no detectable virus. C57BL/6 mice immuni

Batch potency testing of inactivated erysipelas vaccines by ELISA--development, validation and implementation.

Inactivated erysipelas vaccines are widely used to protect pigs against erysipelas disease caused by the bacterium Erysipelothrix (E.) rhusiopathiae. Quality control tests for this vaccine are laid down in the European Pharmacopoeia (Ph.Eur.) Monograph No. 64. A laboratory animal model using a vaccination-challenge procedure is currently required as batch potency test. More than 10 years ago we initiated the first studies to develop an alternative ELISA potency model to replace this regulatory challenge test in mice. A short retrospective outline of the various steps from the development of th

Evaluation of the Use of Sub-Immunodominant Antigens of Babesia bovis with Flagellin C Adjuvant in Subunit Vaccine Development.

Bovine babesiosis caused by the tick-borne apicomplexan parasite Babesia bovis remains a threat for cattle worldwide, and new vaccines are needed. We propose using immune-subdominant (ISD) antigens as alternative vaccine candidates. We first determined that RAP-1 NT and RRA are subdominant antigens using recombinant antigens in ELISAs against sera from B. bovis-protected cattle. Protected animals demonstrated high antibody responses against the known immunodominant rRAP-1 CT antigen, but significantly lower levels against the rRAP-1 NT and rRRA antigens. Next, a group of cattle (n = 6) was vac

A multivariate statistical and data mining analysis of spatial memory-related behaviour following bilateral vestibular loss in the rat.

Vestibular dysfunction in animals and humans is associated with a variety of cognitive and anxiety disorders, and it has been difficult to determine how the different symptoms may be related to one another. The aim of this study was to determine the extent to which the spatial memory deficits that occur following bilateral vestibular deafferentation (BVD) in rats can be attributed to other behavioural symptoms. Spatial memory was measured using a spatial T maze alternation task (STM), while locomotor activity and anxiety were measured using open field, elevated plus and T mazes, respectively.

Adaptations in the walking pattern of spinal cord injured rats.

Walking ability is a measure of recovery used in many studies that test experimental strategies to treat injuries or diseases of the central nervous system (CNS) in animal models. A common measure in the rat animal model of thoracic spinal cord injury (SCI) is visual inspection and scoring of hind limb activity, which allows the documentation of movements associated with the recovery of locomotor function. In this study, we expand on previously documented visible changes in the locomotor pattern following SCI. The spontaneous recovery of locomotion in rats with thoracic SCIs of variable extent

The demonstration of tissue clonality by X-linked enzyme histochemistry.

We report here the first demonstration in normal, as opposed to chimaeric mice, of the clonal architecture of the small intestine, oesophagus, breast, and thyroid, using polymorphism of the X-linked enzyme glucose-6-phosphate dehydrogenase (G6PD, EC 1.1.1.49) as a clonal marker. Reproducible results using X-linked histochemistry were found to depend upon the simultaneous study of tissue from normal mice, mice homozygous for abnormal enzyme levels, and heterozygous mice. In tissues from homozygous animals where levels of enzyme activity normally vary, the use of a physiological stimulus to redu

Non-steroidal anti-inflammatory drug gastropathy: causes and treatment.

Aspirin and non-aspirin non-steroidal anti-inflammatory drugs (NSAIDs) almost invariably cause acute gastroduodenal injury and probably account for approximately 12,000 ulcer bleeding episodes and 1200 deaths per annum in the United Kingdom. Clinically significant intestinal toxicity is also recognized but less clearly defined. The main risk factors for NSAID-related peptic ulcer complications are age, past history, use of higher risk individual NSAIDs, drug dose, concurrent use of warfarin or corticosteroids. The underlying reason for NSAID use and Helicobacter pylori status is not clearly as

Bioreactors, scaffolds and microcarriers and in vitro meat production-current obstacles and potential solutions.

In vitro meat production presents a potential viable alternative for meat consumption, which could provide the consumer with a product indistinguishable from the original, with very similar nutritional and culinary values. Indeed, the alternative products currently accessible often lack comparable nutritional value or culinary attributes to their animal-derived counterparts. This creates challenges for their global acceptance, particularly in countries where meat consumption holds cultural significance. However, while cultured meat research has been progressing rapidly in recent years, some si

Blood collection from the sublingual vein in mice and hamsters: a suitable alternative to retrobulbar technique that provides large volumes and minimizes tissue damage.

Blood examination is a key element in studies of laboratory animals. In rodents, retrobulbar venous plexus puncture is a commonly used method for obtaining a blood sample. Although this technique yields large volumes of blood, the disadvantage is that it can lead to severe tissue damage. The aim of the present study was to develop the puncture of V. sublingualis as a suitable alternative technique for drawing blood in mice and other rodents. In rats, this method has been established for collecting large blood volumes. During the first part of the study, the sublingual bleeding technique was de

Influence of a garlic-based additive on the performance parameters and intestinal morphology of replacement gilts in Ukraine under commercial conditions.

BACKGROUND AND AIM: The global ban on antibiotic growth promoters (AGPs) in livestock production has increased the search for safe and effective natural alternatives that can maintain productivity and support animal health. Phytogenic feed additives from medicinal plants are gaining more attention because of their antimicrobial, antioxidant, and immunomodulatory properties. Garlic (Allium sativum) and caraway (Carum carvi) contain bioactive compounds that can enhance digestive function and intestinal health in monogastric animals. Replacement gilts are a critical group in pig production, as th

Transcranial electrical stimulation as a therapeutic strategy for Alzheimer's disease: Current uses and challenges.

Alzheimer's disease (AD) is an age-related neurodegenerative disorder for which there are currently rarely effective drug treatments available to halt or slow down its progression. With the aging of the world population, AD as the primary cause of dementia, is rapidly becoming one of the most expensive, lethal, and burdening diseases of this century. In recent years, the new method used to treat nervous system diseases including AD is transcranial electrical stimulation (tES) with non-invasive and for regulating the flexibility of neural circuits operation and behaviors. The rationale of tES f

Peromyscus spp. Deer Mice as Rodent Model of Acute Leptospirosis.

Leptospirosis is a global zoonotic disease affecting humans, wildlife, companion, and domestic animals. Incidental hosts can contract the disease directly or indirectly from asymptomatic reservoir hosts, most commonly small rodents. The Golden Syrian hamster is recognized as the dominant rodent model for acute leptospirosis because the animals are susceptible to many serovars and are used to maintain laboratory strains and test bacterin vaccine efficacy. However, hamsters are primarily used in survival-based studies, and investigations into host immune response and disease pathogenesis are lim

Recombinant vesicular stomatitis virus glycoprotein carrying a foot-and-mouth disease virus epitope as a vaccine candidate.

Foot-and-mouth disease (FMD) is one of the most highly contagious animal diseases. In an effort to overcome the drawbacks of the currently used inactivated foot-and-mouth disease virus vaccine, a novel recombinant protein carrying foot-and-mouth disease virus VP1 GH loop epitope linked to vesicular stomatitis virus glycoprotein was expressed in a baculovirus system. Its antigenicity was confirmed with ELISA using monoclonal antibody against foot-and-mouth disease virus. Twice immunizations one month apart in field pigs resulted in a significant antibody increase compared to the glutathione S-t

Development of a new haemostatic dissecting forceps utilizing controlled heat as an energy source.

We tested a prototype of a new thermal surgical system in animal experiments. This device utilizes controlled heat as an energy source and seals and divides small- to medium-size vessels. The forceps we used in the current study are shaped like dissecting forceps used in conventional open surgery, and their grippers can open bilaterally. Heating elements are built into a gripper. The temperature adjustment is controlled by monitoring the electric resistance. Since the new device utilizes no ultrasonic energy, unfavorable phenomena such as cavitations or mist production are not observed. In a p

[Stapotomy with ultrasound? Animal experiment study of possible side effects].

Stapedectomy by ultrasound has been proposed as an alternative method for perforating the stapes footplate in surgery for otosclerosis. Possible functional adverse effects of ultrasound perforation of the otic capsule in guinea pigs were investigated in the present study by means of vestibular evoked potentials (VsEP). VsEP were elicited by pulsed linear accelerations applied to the animal's head following surgical removal of the middle ear, following intense ultrasound drilling around the otic capsule and after drilling a small hole in the anterior bony wall of the vestibule. All manipulation

Techniques for the Assessment of In Vitro and In Vivo Antifungal Combinations.

Systemic fungal infections are associated with high mortality rates despite adequate treatment. Moreover, acquired resistance to antifungals is increasing, which further complicates the therapeutic management. One strategy to overcome antifungal resistance is to use antifungal combinations. In vitro, several techniques are used to assess drug interactions, such as the broth microdilution checkerboard, agar-diffusion methods, and time-kill curves. Currently, the most widely used technique is the checkerboard method. The aim of all these techniques is to determine if the interaction between anti

Intravenous Lipid Emulsions in Infants: Is Balanced Better?

Parenteral nutrition (PN) is frequently required by extremely preterm infants due to gastrointestinal immaturity and complications of prematurity. Parenteral nutrition-associated cholestasis (PNAC) and intestinal failure-associated liver disease (IFALD) are common complications of prolonged PN. Plant-based intravenous lipid emulsions, containing proinflammatory omega-6 fatty acids and phytosterols, may contribute to these conditions as well as other comorbidities such as bronchopulmonary dysplasia and retinopathy of prematurity. Intravenous lipid emulsions containing animal-based fats, such as

Characterization and Testing the Efficiency of Acinetobacter baumannii Phage vB-GEC_Ab-M-G7 as an Antibacterial Agent.

Acinetobacter baumannii is a gram-negative, non-motile bacterium that, due to its multidrug resistance, has become a major nosocomial pathogen. The increasing number of multidrug resistant (MDR) strains has renewed interest in phage therapy. The aim of our study was to assess the effectiveness of phage administration in Acinetobacter baumannii wound infections in an animal model to demonstrate phage therapy as non-toxic, safe and alternative antibacterial remedy. Using classical methods for the study of bacteriophage properties, we characterized phage vB-GEC_Ab-M-G7 as a dsDNA myovirus with a
