Sleep serves a fundamental role in memory consolidation, but this can be influenced by physiological context. For instance, satiated Drosophila require sleep for appetitive memory consolidation, but starved flies that must forage for food switch to sleep-independent memory. As ethanol intoxication alters behaviors such as sleep and memory, we asked how it affects the role of sleep in memory consolidation. Here, we demonstrate that acute ethanol inverts the canonical link between sleep and memory in Drosophila. Ethanol selectively impairs memory consolidation in satiated flies by causing a swit

Eating behavior stands as a fundamental determinant of animal survival and growth, intricately regulated by an amalgamation of internal and external stimuli. Coordinated movements of facial muscles and the mandible orchestrate prey capture and food processing, propelled by the allure of taste and rewarding food properties. Conversely, satiation, pain, aversion, negative emotion or perceived threats can precipitate the cessation or avoidance of eating activities. In recent years, the parasubthalamic nucleus (PSTN), located in the lateral hypothalamic area, has emerged as a focal point in feedin

Observational and experimental studies indicate a remarkably consistent association between rapid growth and weight gain during infancy and higher risks for obesity in later childhood and adult life. This association appears to be equally relevant to breastfed and formula milk-fed infants, and infants small for gestational age and with normal birth weight. The type of infant milk feeding, energy intake, and milk nutrient composition are important determinants of infant growth and weight gain. There is also accumulating evidence that genetic factors related to adult obesity susceptibility act i

The immune system is proposed as the key to understanding the etiology and treatment of psychosocial disease. There is a dense communication network between the immune system and the central nervous system (CNS). Immune cell cytokines, via direct action on the CNS, induce fever, alter sleep, pain perception and pituitary hormone release and reduce appetite and activity in animals. Interleukin-2 and interferon given to humans result in global behavioral and cognitive pathology. Activation of the immune system by pathogens produces global cognitive and behavioral pathology also. Recently, contro

Pediatric obesity is a chronic, multifactorial neuroendocrine disease in which disrupted gut-brain signaling, hypothalamic appetite regulation, adipose tissue dysfunction, and altered inter-organ communication sustain positive energy balance. Incretin-based therapies translate this biology into mechanism-based treatment, yet their developmental implications remain incompletely defined. This narrative review integrates evidence on GLP-1 and GIP receptor signaling with pediatric trial data and clinical implementation, with particular attention to developmental modifiers of gut-brain signaling an

OBJECTIVE: Alexander disease (AxD) is a severe neurodegenerative disorder caused by gain-of-function mutations in the gene for GFAP, which lead to protein aggregation and a primary astrocytopathy. Symptoms vary, but failure to thrive (FTT) and frequent emesis are common and cause significant morbidity. Here we investigate GDF15, a member of the TGFβ superfamily, which regulates energy balance and appetite, as a potential mediator of FTT in AxD.
METHODS: In this study, we use the Gfap+/R237H rat model (R237H), in which pups fail to gain weight after weaning and become frail and impaired as the

BACKGROUND: Bulimia nervosa (BN) involves recurrent episodes of uncontrolled eating followed by compensatory behaviors. Stress is implicated in BN, affecting the hypothalamic-pituitary-adrenal (HPA) axis and ghrelin, a key appetite-regulating hormone. Studies report conflicting HPA axis findings in BN, necessitating further investigation.
OBJECTIVE: To examine the impact of acute stress on cortisol and serum ghrelin and eating disorder symptoms in women with BN and healthy controls (HC).
METHODS: Participants underwent a socially evaluated cold pressor test (CPT) and control condition (quiet

Dementia, with loss of memory, cognitive abilities, and independent daily functioning, is increasing worldwide, related to an aging population. Currently, there is no curative treatment for dementia. Treatment of the frequently occurring behavioral and psychological symptoms of dementia (BPSD) is partially effective and associated with significant side effects. Cannabinoids are lipophilic molecules acting on the CB1 end CB2 receptors, essential for main biological processes such as sleep, appetite, memory, and pain. Cannabinoids might have a positive impact on amyloid formation in Alzheimer's

This study investigated trends within a custom Sports Nutrition Assessment for Consultation (SNAC) survey designed to identify nutrition-related risk factors among post-operative lower-extremity youth athletes. Athletes aged 8-18 years who completed the SNAC at a sports medicine institution after lower-extremity surgery were reviewed for associations between SNAC questions and age/sex differences. Of 477 patients (15.0 ± 2.0 years; 47.8% female), 319 (66.9%) answered 'yes' to at least one question and were identified for a consult, though 216 (64.3%) declined. The most frequent questions to pr

Weight gain and metabolic disturbances are frequent in people treated with second generation antipsychotics (SGA). We aimed to investigate the effect of SGAs on eating behaviors, cognitions and emotions, as a possible contributor to this adverse effect. A systematic review and a meta-analysis were conducted following the Preferred Reporting Items for Systematic reviews and Meta-Analyses (PRISMA) guidelines. Original articles measuring outcomes relating to eating cognitions, behaviours and emotions, during treatment with SGAs were included in this review. A total of 92 papers with 11,274 partic

Anorexia-cachexia syndrome develops during the advanced stages of various chronic diseases in which patients exhibit a decreased food intake, weight loss, and muscle tissue wasting. For these patients, this syndrome is a critical problem leading to an increased rate of morbidity and mortality. The present pharmacological therapies for treating anorexia-cachexia have limited effectiveness. The Japanese herbal medicine rikkunshito is often prescribed for the treatment of anorexia and upper gastrointestinal (GI) disorders. Thus, rikkunshito is expected to be beneficial for the treatment of patien

Nausea and emesis are common side effects of gastrointestinal disease. Reports indicate that ghrelin and endocannabinoids, agents that stimulate appetite, also reduce emesis evoked by chemotherapy treatment, which suggests that stimulation of feeding inhibits the emetic system. In the following study we conducted a more direct test of this hypothesis by determining the impact of manipulating the motivation to eat on emesis, using food restriction and refeeding. Emesis was induced in musk shrews, a commonly used animal model for emesis research, using the cancer chemotherapy agent cisplatin (20

Anorexia nervosa (AN) is a debilitating and deadly disease characterized by low body mass index due to diminished food intake, and oftentimes concurrent hyperactivity. A high percentage of AN behavioral and metabolic phenotypes can be replicated in rodents given access to a voluntary running wheel and subject to food restriction, termed activity-based anorexia (ABA). Despite the well-documented bodyweight loss observed in AN human patients and ABA rodents, much less is understood regarding the neurobiological underpinnings of these maladaptive behaviors. Hunger-promoting hypothalamic agouti-re

The ability to exert self-control in the face of appetitive, alluring cues is a critical component of healthy development. The development of behavioral measures that use disease-relevant stimuli can greatly improve our understanding of cue-specific impairments in self-control. To produce such a tool relevant to the study of eating and weight disorders, we modified the traditional go/no-go task to include food and non-food targets. To confirm that performance on this new task was consistent with other go/no-go tasks, it was given to 147 healthy, normal weight volunteers between the ages of 5 a

The purpose of the study was to evaluate anabolic-androgenic steroid (AAS) abusing adults every 2 weeks with a comprehensive behavioral and clinical assessment battery. The study was conducted at the University of Pennsylvania Treatment Research Center; 10 subjects were enrolled and 7 completed the protocol. AASs and other drugs were obtained and self-administered by subjects through their usual mechanisms. On-study evaluations included medical, behavioral, and drug-use assessments. While a high incidence of mood disorders and substance abuse was found, few clinically relevant changes in physi

It has been hypothesized that brain development during adolescence perturbs reward processing in a way that may ultimately contribute to the risky decision making associated with this stage of life, particularly in young males. To investigate potential reward dysfunction during adolescence, Experiment 1 examined palatable fluid intake in rats as a function of age and sex. During a series of twice-weekly test sessions, non-food-deprived rats were given the opportunity to voluntarily consume a highly palatable sweetened condensed milk (SCM) solution. We found that adolescent male, but not female

Vaginocervical stimulation (VCS) induces the immediate-early gene product Fos in the ventromedial hypothalamus (VMH) of female rats. However, this induction is lower in ovariectomized rats that receive estradiol benzoate (EB) and progesterone (P) relative to an oil vehicle. We have observed that a substantial proportion of cells activated in the VMH by VCS stain for glutamate, and infusions of glutamate or its selective receptor agonists to the VMH inhibit both appetitive and consummatory sexual behaviors in females. This raises the possibility that VCS activates an inhibitory glutamate system

Psychotropic medications are an essential component of treating pediatric mental health disorders, and pediatricians are increasingly likely to prescribe them. Commonly used psychotropic medications include stimulants and nonstimulants used in the treatment of attention-deficit/hyperactivity disorder (ADHD); antidepressants used in the treatment of anxiety and depression; and antipsychotics indicated for use in autism, schizophrenia, mood disorders, severe impulsivity, and aggression. Stimulants are commonly associated with appetite suppression and initial insomnia and nonstimulants for ADHD a
