Unraveling the aroma-producing mechanism of the xerophilic fungus Xerochrysium coryli via integrated transcriptomic and volatile metabolomic analysis.

To investigate the aroma-producing mechanism of the tobacco-derived fungus Xerochrysium coryli, this study employed an integrated approach combining volatile metabolomics and transcriptomics. A total of 61 volatile metabolites were identified, and 17,269 genes were annotated. By integrating metabolite dynamics, differential gene expression, and pathway enrichment, we constructed a "gene-metabolite-pathway" association network. Significantly associated aroma-producing pathways included: aromatic amino acid conversion (associated with aromatic compounds such as phenylacetaldehyde and 2-phenyleth

Hos2-mediated KatG1 decrotonylation orchestrates ROS scavenging in rice blast infection.

In plant-pathogen interaction, plants trigger reactive oxygen species (ROS) bursts to defend against pathogen invasion, while pathogens evolved antioxidant systems to scavenge ROS for infection. Our study reveals a regulatory mechanism of ROS adaptation and scavenging in the rice blast fungus Magnaporthe oryzae. We found that KatG1, a bifunctional catalase-peroxidase, acts as a ROS-responsive regulatory enzyme. During infection, the deacetylase Hos2 mediates the decrotonylation of KatG1 at K422, initiating the ROS clearance process. Decrotonylated KatG1 depolymerizes into monomers, interacts w

Comparative genomics of Cadophora luteo-olivacea reveals a divergent lineage, conserved functional repertoires, and strain-level phenotypic variation.

Cadophora luteo-olivacea has been reported from different hosts and environments and is associated with grapevine trunk diseases, yet the extent to which isolates differ in genome composition, functional potential, and pathogenicity remains poorly understood. Here, we performed a comparative genomic analysis of 12 C. luteo-olivacea isolates recovered from grapevine, almond, apple, Crocus bulbs, soil, air, wastewater, and deep-sea sediment. Genome assemblies were highly complete (BUSCO >99%) and ranged from 46.94 to 50.70 Mbp. Pairwise average nucleotide identity (ANI) revealed a cohesive 11-st

The hijacking of a wheat Exo70-like protein by a fungal pathogen triggers disease.

Tan spot is an economically important fungal disease of wheat that occurs globally. In this necrotrophic pathosystem, pathogen effectors are recognized by wheat genes in an inverse gene-for-gene manner, leading to the hijacking of host defense mechanisms and ultimately disease susceptibility. Here, we identified the tan spot susceptibility gene Tsc2 through positional cloning, mutagenesis, and transgenic complementation. Tsc2 encodes a diverged form of the exocyst subunit Exo70FX15 that localizes to the nucleus and cytoplasm and does not interact directly with the fungal effector. A 21‑nucleot

Structurally diverse secondary metabolites from the dung-inhabiting fungus Botryotrichum murorum.

A combination of confidence-aware MS/MS-based metabolomics and conventional chemical screening approaches was employed to investigate the metabolome of Botryotrichum murorum DSM 113281, isolated from tortoise dung. The metabolomic survey revealed a high degree of chemical diversity; however, low production rates necessitated large-scale solid-state cultivation to purify the major constituents. This approach led to the discovery of tortoisellide A (1), which features a rare polyketide γ-lactol scaffold, an unprecedented thio-substituted analogue 2 of the 14-membered bis-macrolactone antibiotic

Unveiling the hidden virome of Sclerotinia sclerotiorum: New tools and discoveries in mycovirus detection.

Mycoviruses are widespread in fungi and can influence fungal physiology, pathogenicity, and ecological interactions. In the plant pathogenic fungus Sclerotinia sclerotiorum, increasing attention has been directed toward understanding the diversity and functional roles of its associated viruses. This review synthesizes current knowledge on mycovirus detection methods and summarizes the diversity of viruses reported in S. sclerotiorum. Early detection strategies based on double-stranded RNA profiling, virion purification, and RT-PCR laid the foundation for mycovirus discovery but were limited in

Rapid detection of cultivated truffle fungi (Tuber) from truffles and ectomycorrhizal root tips using loop-mediated isothermal amplification.

UNLABELLED: Truffle fungi, such as Tuber melanosporum and T. aestivum, are prized luxury food products that have been historically cultivated in Mediterranean regions of Europe. Recent advances in truffle cultivation have led to the establishment of productive truffle orchards in North America, South America, China, and Australia. Despite these advances, a consistent issue has been the introduction and accidental cultivation of non-target truffle species. Non-target truffle species (e.g., T. indicum and T. brumale) often morphologically resemble target species (e.g., T. melanosporum) and may u

A novel Cryphonectria species (Cryphonectriaceae, Diaporthales) discovered on Eucalyptus, including insights into the mating biology of the genus.

The Cryphonectriaceae (Diaporthales, Ascomycota) accommodates numerous important fungal pathogens that cause branch and stem canker diseases on woody plants worldwide. These include, for example, Cryphonectria (Cry.) parasitica, the causal agent of chestnut blight, which caused the near eradication of the American chestnut (Castanea dentata) in North America. The profound ecological and economic impact of Cry. parasitica has prompted considerable research interest in the diversity, biology, and pathogenicity of other species in the Cryphonectriaceae globally. During a 2023 disease survey in No…
