Natural Anti-Infective Pulmonary Proteins: In Vivo Cooperative Action of Surfactant Protein SP-A and the Lung Antimicrobial Peptide SP-BN.

The anionic antimicrobial peptide SP-B(N), derived from the N-terminal saposin-like domain of the surfactant protein (SP)-B proprotein, and SP-A are lung anti-infective proteins. SP-A-deficient mice are more susceptible than wild-type mice to lung infections, and bacterial killing is enhanced in transgenic mice overexpressing SP-B(N). Despite their potential anti-infective action, in vitro studies indicate that several microorganisms are resistant to SP-A and SP-B(N). In this study, we test the hypothesis that these proteins act synergistically or cooperatively to strengthen each other's micro

A combined genetic and phenotypic marker approach enables precise detection of hypervirulent Klebsiella pneumoniae and reveals associated traits of capsule overproduction and tellurite resistance.

We integrated genetic and phenotypic analyses to characterize carbapenem-resistant hypervirulent K. pneumoniae (CRhvKp) and liver abscess-associated K. pneumoniae (LAKp) isolates, aiming to identify potential biomarkers for the accurate detection of hvKp. A total of 660 carbapenem-resistant K. pneumoniae (CRKp) and 69 LAKp isolates were screened for five hvKp-associated biomarkers (peg-344, iroB, iucA, prmpA, prmpA2) and the string test. Among LAKp isolates, 49.3% were positive for all five biomarkers and the string test (5 genes-string test positive), while 39.1% carried all markers but were

The Capsular Polysaccharide Obstructs Wall Teichoic Acid Functions in Staphylococcus aureus.

BACKGROUND: The cell envelope of Staphylococcus aureus contains 2 major secondary cell wall glycopolymers: capsular polysaccharide (CP) and wall teichoic acid (WTA). Both CP and WTA are attached to the cell wall and play distinct roles in S. aureus colonization, pathogenesis, and bacterial evasion of host immune defenses. We aimed to investigate whether CP interferes with WTA-mediated properties. METHODS: Strains with natural heterogeneous expression of CP, strains with homogeneous high CP expression, and CP-deficient strains were compared regarding WTA-dependent phage binding, cell adhesion,

Bacterial and host factors affecting acquisition of Streptococcus pneumoniae in a murine model.

Initial acquisition is a critical step in the establishment of colonization that in turn enables transmission to new hosts and potentially leads to invasive disease. Here, we studied host and bacterial factors affecting acquisition, as distinct from those affecting colonization density, using Streptococcus pneumoniae (Spn) in a mouse model. Acquisition was quantified using nasal inocula of limiting size to determine the proportion of hosts infected, as well as the median infectious dose. Infant mice (<7 days of age) were highly susceptible to acquisition (ID50 < 30 CFU) compared with adults (>

Proteomic identification of Acanthamoeba castellanii excretory-secretory proteins associated with Klebsiella pneumoniae capsule enlargement: a candidate role for C-type lysozyme.

BACKGROUND: Acanthamoeba castellanii and Klebsiella pneumoniae are environmental commensals that co-exist in aquatic habitats. Our previous studies demonstrated that A. castellanii genotype T4 (AcT4) induces capsule enlargement and hypermucoviscosity in K. pneumoniae through small excretory-secretory proteins (ESPs; <10 kDa) that activate the Rcs two-component system, upregulating capsule biosynthesis genes independently of virulence plasmids. However, the molecular identity of these active ESPs remained undefined. RESULTS: To identify candidate proteins, we performed label-free quantitative

The unique Legionella longbeachae capsule favors intracellular replication and immune evasion.

Legionella longbeachae and Legionella pneumophila are the most common causative agents of Legionnaires' disease. While the clinical manifestations caused by both species are similar, species-specific differences exist in environmental niches, disease epidemiology, and genomic content. One such difference is the presence of a genomic locus predicted to encode a capsule. Here, we show that L. longbeachae indeed expresses a capsule in post-exponential growth phase as evidenced by electron microscopy analyses, and that capsule expression is abrogated when deleting a capsule transporter gene. Surface charge of Streptococcus pneumoniae predicts serotype distribution.

Streptococcus pneumoniae (pneumococcus) frequently colonizes the human nasopharynx and is an important cause of pneumonia, meningitis, sinusitis, and otitis media. The outer cell surface of pneumococcus may assume various degrees of negative charge depending on the polysaccharide capsule, of which more than 90 serotypes have been identified. The negative charge of capsular polysaccharides has been proposed to electrostatically repel pneumococci from phagocytic cells, and avoidance of phagocytosis correlates with higher carriage prevalence. We hypothesized that the surface charge of pneumococcu

Induction of labyrinthitis ossificans after pneumococcal meningitis: an animal model.

Newly formed disorganized bone fills the open spaces within the otic capsule in various pathologic conditions, resulting in labyrinthitis ossificans. The pathologic mechanisms of this disease remain poorly understood. To better study the sequence of events and contributing mechanisms involved in labyrinthitis ossificans, an animal model was developed. Three groups of Mongolian gerbils received either an intralabyrinthine injection of normal saline solution (group 1) or Streptococcus pneumoniae polysaccharide capsule antigens (groups 2 and 3). The temporal bones were harvested after 3 months an

Evaluation of a passive microcapsule agglutination test for the screening of human leptospirosis.

Microcapsules, absorbed with two mixed antigens--each composed of 3 sonicated leptospira serovars--were developed in the past as a microcapsule agglutination test (MCA-LS) for the screening of clinical leptospirosis. For this study, fifty serum samples, taken at an earlier and at a later stage of illness from 25 Italian in-patients with clinical symptoms of leptospirosis, were tested with both the MCA-LS one-dilution test and the microscopic agglutination (MA) test, the confirmatory test for leptospirosis, with 18 leptospira strains circulating in Italy. Compared with MA, MCA-LS showed a sensi

An outbreak of severe invasive meningococcal disease due to a capsular switched Neisseria meningitidis hypervirulent strain B:cc11.

OBJECTIVES: The aim was to investigate an outbreak of invasive meningococcal disease (IMD) in Southern Sardinia. METHODS: Epidemiological and microbiological investigations were performed. The latter included antimicrobial susceptibility testing and whole-genome sequencing (WGS). RESULTS: Seven individuals with severe IMD were found to be infected with serogroup B (MenB) Neisseria meningitidis in the first quarter of 2018. Five of the seven cases (five males; mean age 19 years; range 18-21 years; CFR 40%) were due to a unique strain B:P1.5-1,10-8:F3-6:ST-11(cc11), probably switched from the

Characterization of polysaccharide conformational epitopes by surface plasmon resonance.

SPR techniques can provide a wealth of insight into the nature of protein-carbohydrate interactions. Information not obtained readily by other methodologies can be gathered relatively quickly in a label-free manner with low sample consumption. This chapter focused on two applications in which SPR has been used to map conformational epitopes on bacterial polysaccharides recognized by protective antibodies. In one example, methods for demonstrating the conformational nature of the epitope recognized by an anti-GBS antibody were described. Dramatic epitopic stabilization at 2 repeating units with

Characterisation of isolates of Staphylococcus aureus from acute, chronic and subclinical mastitis in cows in Norway.

Eighty-six Staphylococcus aureus isolates from cases of bovine mastitis were characterised biochemically and with respect to serotype, multilocus enzyme electrophoresis genotypes, antibiotic sensitivity, and production of enterotoxins A through D (SEA-D) and toxic shock syndrome toxin-1 (TSST-1). The samples were obtained from 81 different cows from 79 Norwegian dairy herds in 10 different counties in southern Norway. There was an equal representation of isolates from cases of acute, chronic and subclinical mastitis. Multilocus enzyme electrophoresis using 13 genetic loci showed that 69 of 86

Clonal characterization of the human IgG antibody repertoire to Haemophilus influenzae type b polysaccharide. V. In vivo expression of individual antibody clones is dependent on Ig CH haplotypes and the categories of antigen.

Antibodies (Ab) to the polysaccharide capsule of Haemophilus influenzae type b (Hib-PS) provide protection against Haemophilus influenzae type b disease in children, and Hib-PS vaccines with different immunologic properties are widely used clinically. The repertoire of human anti-Hib-PS Ab induced by these vaccines is relatively restricted and can be divided into two types by the structure of the light chain V region. Ab using A2-V kappa II gene product, which account for the majority of anti-Hib-PS Ab response in most patients, show little somatic mutations. In contrast, non-Ab using A2-V kap

Childhood epiglottitis in recent years.

Fifty-five cases of acute epiglottitis were diagnosed and treated at the Children's Hospital of Philadelphia between 1985 and 1991. To evaluate the changing patterns of the disease over recent years, we compared the current results with those obtained from an earlier study reported at our institution in 1979. The overall annual incidence was higher in recent years than in the earlier study, with a sharp decline in new cases over the last two years. Thirty-six percent of the Hemophilus influenzae type B (HIB) infections were ampicillin-resistant in recent years, while no resistance had been fou

Prevalence of β-hemolytic groups C and F streptococci in patients with acute pharyngitis.

BACKGROUND: The roles of group C and F streptococci in causing endemic pharyngitis are still controversial, although group C streptococci are implicated in the outbreaks of pharyngitis and associated disorders. AIM: The aim of this study was to determine the prevalence and the role of these groups of β-hemolytic streptococci in acute pharyngitis with emphasis on the Streptococcus anginosus group. The antimicrobial susceptibility profile of these bacterial isolates and their ability to produce some virulence factors was also determined. MATERIALS AND METHODS: Throat swab specimens were collec

Cellular and molecular biology of Neisseria meningitidis colonization and invasive disease.

The human species is the only natural host of Neisseria meningitidis, an important cause of bacterial meningitis globally, and, despite its association with devastating diseases, N. meningitidis is a commensal organism found frequently in the respiratory tract of healthy individuals. To date, antibiotic resistance is relatively uncommon in N. meningitidis isolates but, due to the rapid onset of disease in susceptible hosts, the mortality rate remains approx. 10%. Additionally, patients who survive meningococcal disease often endure numerous debilitating sequelae. N. meningitidis strains are cl

Genomic epidemiology of Streptococcus pneumoniae reveals globally disseminated lineages and capsular switching in the Arabian Gulf region.

BACKGROUND: Streptococcus pneumoniae remains a major cause of pneumococcal disease globally despite widespread use of pneumococcal conjugate vaccines (PCVs). Genomic data from the Arabian Gulf remain limited, and ongoing changes in pneumococcal population structure may complicate serotype-based surveillance and vaccine impact assessment. METHODS: Whole-genome sequencing was performed on 250 S. pneumoniae isolates collected in the United Arab Emirates and Kuwait. Population structure was analysed using Global Pneumococcal Sequence Clusters (GPSCs). Serotypes and sequence types were determined,
