Bacterial lipopolysaccharide forms aggregates with apolipoproteins in male and female rat brains after ethanol binges.

Alcohol binge drinking allows the translocation of bacterial lipopolysaccharide (LPS) from the gut to the blood, which activates the peripheral immune system with consequences in neuroinflammation. A possible access/direct signaling of LPS to/in the brain has not yet been described under alcohol abuse conditions. Apolipoproteins are compounds altered by alcohol with high affinity to LPS which may be involved in its transport to the brain or in its elimination. Here, we explored the expression of small components of LPS, in its free form or bound to apolipoproteins, in the brain of female and m…

Effect of Regional COVID-19 Lockdowns on Alcohol Use and Smoking in Two Chilean University Communities: A Difference-In-Difference Analysis.

INTRODUCTION: Effects of lockdowns on alcohol use and smoking remain unclear. By exploiting the variation in regional COVID-19 lockdowns in two Chilean regions, we examined their effect on alcohol use and smoking in two university communities. METHODS: We conducted a quasi-experimental study in university communities in two Chilean regions, Coquimbo and Araucanía. Participants were a longitudinal cohort (n = 896) assessed seven times from July 2020 ( t 1 ) to April 2021 ( t 7 ). Outcomes were weekly grams of alcohol use, heavy episodic drinking (HED) and cigarettes smoked per day. RESU

A randomized controlled trial of a brief intervention to reduce alcohol use among female sex workers in Mombasa, Kenya.

OBJECTIVE: We assessed whether a brief alcohol intervention would lead to reduced alcohol use and sexually transmitted infection (STI)/HIV i … status at enrollment. RESULTS: There was a statistically significant reduction in alcohol use and binge drinking at 6 and 12 months, with intervention participants reporting less than one third of the odds of higher levels of drinking than the control group. The intervention did not impact laboratory-confirmed STI/HIV incidence, self-reported condom use, or se …

Association between the risk of alcohol use and unprotected sex in adolescents in a city in the southern region of Brazil.

This paper aims to analyze alcohol abuse and unprotected sex and the association between them in students in a city in the southern region of Brazil. This is a cross-sectional study using a quantitative approach with 590 secondary school students from two public schools. Regarding alcohol use, 14% scored from eight to 40 in The Alcohol Use Disorders Identification Test (AUDIT), which means, at least, a risky drinking behavior, with higher rate among boys. Moreover, 31.1% indulged themselves in binge drinking, which means drinking six or more doses according to AUDIT; and even among those who a…

Hyperbilirubinaemia and haemolytic anaemia in acute alcoholic hepatitis: there's oil in them thar veins.

A Caucasian woman in her late 30s was evaluated after a period of binge drinking and found to have hyperbilirubinaemia for which she was referred for consideration of cholecystectomy. After exclusion of other possibilities, Zieve's syndrome was diagnosed. This is a condition of hyperbilirubinaemia, Coombs' negative haemolytic anaemia and hyperlipidaemia associated with alcoholism. Abstinence from alcohol remains the only known effective treatment, and appreciation of the entity can prevent unnecessary biliary procedures. The patient improved with supportive measures and was discharged in stabl…

Physical training mitigates alveolar bone and blood enzymatic antioxidants defense impairment induced by binge ethanol consumption in rats.

We aimed to investigate the effectiveness of physical training as a protective strategy to mitigate alveolar bone damage and blood antioxidant defense caused by ethanol (EtOH) consumption in a binge-drinking pattern. Male Wistar rats aged approximately 90 days were divided into four groups: control, training, EtOH, and training + EtOH. The physical training protocol was conducted on a treadmill for four consecutive weeks, while the animals in the EtOH group were administered EtOH via orogastric gavage for three consecutive days each week, following the binge drink pattern. After the training p

Assessing the utility of the neurosteroid zuranolone to modify alcohol-related behaviours.

Alcohol use is a leading risk factor for premature mortality, yet effective pharmacotherapies remain limited. Neurosteroids, such as allopregnanolone, modulate γ-aminobutyric acid type A (GABAA) receptors and influence alcohol-related behaviours. Zuranolone, an orally bioavailable synthetic analogue of allopregnanolone recently approved for postpartum depression, represents a potential candidate for therapeutic repurposing in alcohol use disorder (AUD). Here, we assessed the effects of acute and daily zuranolone on alcohol-related behaviours in a preclinical binge drinking model, comparing out…

Adolescent ethanol exposure reprograms HMGB1 responsivity to adult ethanol challenge in males.

Adolescent intermittent ethanol (AIE)-a model of human adolescent binge drinking- produces lasting changes in ethanol responsivity, including increased adult ethanol consumption and behavioral tolerance. However, the neuroimmune and neuroplastic mechanisms underlying these adaptations are unclear. Therefore, we used a model of human adolescent binge drinking (AIE; 5 g/kg/day ethanol or water, i.g., 2-day on/2-day off cycle from postnatal day (PND) 25-54) to assess disruptions to immune, neuronal, and endocrine responsivity to an acute ethanol challenge (4 g/kg) in adulthood (PND 105-110) in ma…

Sex differences in the neuroimmune response following heavy, binge-like alcohol exposure in adolescent rats.

Adolescents who consume alcohol show a high prevalence of binge drinking, which has been linked to brain damage and neuroimmune reactions that increase risk for developing an alcohol use disorder (AUD). Adolescent female drinking patterns have surpassed males, yet little is known about damaging effects of alcohol in females. Known sex differences in neuroimmune reactivity, specifically microglial reactivity, suggest that the female brain will differ from males. Therefore, we examined indicators of neuroimmune activation and neurodegeneration following 2 days of heavy, bingelike exposure in ado…
