Separation of bioconjugated quantum dots using capillary electrophoresis.

Capillary electrophoresis (CE) with laser-induced fluorescence (LIF) detection was used to separate different bioconjugated CdSe/ZnS quantum dots (QDs). The QD nanocrystals studied were conjugated to the biomolecules streptavidin, biotin, and immunoglobulin G. The bioconjugated QDs showed different electrophoretic mobilities, which appear to depend upon the biomolecule that is attached to the QD and the buffer solution used. The use of a polymeric additive into the CE run buffer improved the resolution of the bioconjugates. Under CE conditions, the interaction between QD bioconjugates containi

Nail fold capillary observation in replanted severed fingers.

Observation of circulation in replanted severed fingers often relies on the clinical experience of the doctor. This article discusses nail fold capillary loops (NFCL) observation in the monitoring of the perfusion and circumfluence of replanted digits. In our study, 30 patients with a total of 39 replanted digits were selected. The WX-9A900 Microcirculation Checkout System (MCS) was adopted to perform evaluation of circulation postoperatively. The digits were monitored by clinical examination and MCS at 5- to 7-h intervals for 24 h and then at 2-days intervals for 14 days. Our results showed t

Stepping out of the flow: capillary extravasation in cancer metastasis.

In order for cancer cells to successfully colonize a metastatic site, they must detach from the primary tumor using extracellular matrix-degrading proteases, intravasate and survive in the circulation, evade the immune response, and extravasate the vasculature to invade the target tissue parenchyma, where metastatic foci are established. Though many of the steps of metastasis are widely studied, the precise cellular interactions and molecular alterations associated with extravasation are unknown, and further study is needed to elucidate the mechanisms inherent to this process. Capillary electrophoretic studies on the photogenotoxic potential of pharmaceutical substances.

Drug-induced phototoxic skin responses, including photoirritation, photoallergy and photogenotoxicity, are identified as adverse reactions. In this study, we attempted to develop effective analytical tools to predict the photogenotoxic potential of pharmaceutical substances with the use of pBR322 DNA, a plasmid DNA. pBR322 DNA was irradiated with simulated solar light in the presence of photosensitizers, and its structural conversion was assessed by agarose gel electrophoresis (AGE), transmission electron microscopy (TEM) and capillary gel electrophoresis (CGE). The generation of reactive oxyg

Characterization of quantum dots using capillary zone electrophoresis.

Commercially available quantum dots (QDs) were characterized using CE. The CE instruments were laboratory-built, each being capable of both electrokinetic and hydrodynamic injection. Modes of detection include UV absorption and LIF. The CE-LIF system was further modified to handle microliter sample volumes during injection. Sodium phosphate (5-25 mM, pH 7.5-11) was found to be a good buffer electrolyte. Sodium mercaptoproprionate CdTe/CdS (ADS620) QDs and carboxylic acid CdSe/ZnS (T2-Evitag) QDs yielded high separation efficiencies of N = 1.5x10(6) plates at t(M) = 10 min and N = 1.0x10(5) pla

Disseminated lobular capillary hemangioma: two case reports.

Lobular capillary hemangioma, also known as pyogenic granuloma, is a common, solitary, benign neoplasm of the skin and mucous membranes. The etiology of this lesion remains unknown. Lobular capillary hemangioma can present rarely in a disseminated form, usually associated with other disorders. Two patients, aged 17 and 33, were admitted to the dermatology clinic with disseminated lobular capillary hemangiomas which appeared suddenly. There was no history of trauma in either case. The histopathology of the lesions was consistent with lobular capillary hemangioma. No associated disorders were fo

Determination of small phosphorus-containing compounds by capillary electrophoresis.

The review focuses on the analysis of small phosphorus-containing compounds by capillary electrophoresis (CE) with different detection modes including UV absorption, laser-induced fluorescence, conductometry, amperometry, atomic spectrometry, and mass spectrometry. Determinations of phosphates, organophosphate, and chemical warfare agents in environmental samples such as water, soil and grains are emphasized. To achieve better sensitivity, high-resolving power, and reproducibility, the optimum separation conditions for various analytes (samples) are different. We compare the merits and demerit

Alveolar capillary dysplasia with congenital misalignment of pulmonary vessels.

Alveolar capillary dysplasia with misalignment of pulmonary vessels is an uncommon congenital cause of persistent pulmonary hypertension of the newborn. It is universally fatal, and diagnosis is entirely dependent upon surgical lung biopsy. We present a case of alveolar capillary dysplasia with misalignment of pulmonary vessels occurring in a full-term neonate, emphasizing that early involvement of the thoracic surgeon for a histological diagnosis allows expensive and ineffective treatments to be avoided.

Intravenous pyogenic granuloma or intravenous lobular capillary hemangioma.

Lobular capillary hemangioma is a vascular neoplasm that commonly occurs as a cutaneous tumor. When it involves the skin and mucosal surfaces, ulceration and suppuration may occur, hence the classic term of pyogenic granuloma. Intravenous pyogenic granuloma is a rare solitary form of lobular capillary hemangioma that usually occurs in the veins of the neck and upper extremities. We report the ultrasonographic and magnetic resonance imaging findings of a pyogenic intravenous granuloma localized in the right cephalic vein. The imaging and pathological findings and the differential diagnoses are

Pulmonary capillary hemangiomatosis arising in hereditary hemorrhagic telangiectasia.

Hereditary hemorrhagic telangiectasia (HHT), also known as Osler-Weber-Rendu syndrome, is an autosomal dominant inherited disease characterized by epistaxis, telangiectases, and visceral arteriovenous malformations that can lead to hemorrhage and other complications. We report the case of a 56-year-old female patient with HHT and pulmonary hypertension who died with intractable pulmonary and gastrointestinal bleeding. Autopsy revealed vascular malformations in the lungs, gastrointestinal tract, liver, spleen, and brain. Capillary proliferations in the pulmonary alveolar walls, characteristic o

Influence of peripheral arterial disease on capillary pressure in the foot.

BACKGROUND: Capillary perfusion and transmural pressure are delicately regulated by microvascular constriction mechanisms, which are activated upon a change in posture. Capillary flow is known to be disturbed in patients with severe peripheral arterial disease. To date, however, the influence of this disease on capillary pressure is unknown. METHODS: Capillary pressure in the nail fold of the hallux, ankle, and toe blood pressures were measured in the sitting and supine positions in 8 patients with intermittent claudication (F2), in 7 patients with rest pain and/or ischemic ulcers (F3-4), and

Cystic maculopathy with normal capillary permeability secondary to docetaxel.

BACKGROUND: Cystic maculopathy without leakage on fluorescein angiography has been described in cases of toxicity to niacin. We present a similar case of cystic maculopathy due to docetaxel (Taxotere) in the treatment of breast cancer. CASE REPORT: A 53-year-old Asian female presented with complaints of decreased vision soon after the initiation of treatment for breast cancer with docetaxel. A cystic maculopathy with a normal fluorescein angiogram was found on examination. Significant improvement in visual acuity along with resolution of the cystic maculopathy was found after changing to an a

Caspase 3 activation in nasal capillary in patients with epistaxis.

OBJECTIVE: We investigated whether an apoptosis of nasal microvessels contributes to probable mechanism of the onset of epistaxis. STUDY DESIGN AND SETTING: Nasal septal mucosa of Little's areas taken from patients without (n = 19) and with (n = 26) epistaxis were examined. Active caspase-3 in the mucosa was detected according to the methods of immunohistochemistry and Western blotting. On Western blot analysis of the homogenates of the mucosa, we also sought probable signaling factors after caspase-3 activation. RESULTS: Marked activation of caspase-3 was detected in the capillaries and its…

Capillary electrophoresis as an orthogonal technique in HPLC method validation.

High-performance liquid chromatography is usually used to assay the main compound and organic impurity content of drug substance and drug product during pharmaceutical development. A crucial validation parameter of these methods is specificity--the ability to unequivocally assess the analyte in the presence of component expected to be present. Typically, these include impurities, degradation products, and matrices. Besides adequate chromatographic separation with sufficient selectivity, additional 2- or 3-D spectroscopic or chromatographic tools are frequently necessary for this purpose. In ou

Group A streptococcal meningitis as a complication of an infected capillary haemangioma.

UNLABELLED: We report a case of group A streptococcal meningitis in an infant resulting from an infected capillary haemangioma. The child suffered significant morbidity including cerebral infarction, epilepsy, and developmental delay. Treatment of infected capillary haemangiomas remains controversial and inconsistent. CONCLUSION: Our experience of this infant, resulting in profound neurological morbidity suggests that group A Streptococcus can be a virulent organism in the young child and that capillary haemangiomas must be treated aggressively at the first sign of infection.

Detection of homocysteine by conventional and microchip capillary electrophoresis/electrochemistry.

A method based on capillary electrophoresis (CE) with electrochemical (EC) detection for the determination of both total homocysteine (tHcy) and protein-bound homocysteine (pbHcy) in plasma is described. Both end-column and off-column amperometric detection were investigated. Off-column detection resulted in a more sensitive assay for the determination of homocysteine (Hcy). The detection limit for homocysteine was 500 nM using off-column EC detection and the response was linear over the range 1-100 microM. Therefore, this assay is appropriate for the quantification of Hcy over the physiologic

Fetal capillary haemangioblastoma: an exceptional tumour. A review of the literature.

We report a case of a fetal haemangioblastoma located in the cerebellopontine angle. On prenatal ultrasonographic examination a hyperechogenic and heterogeneous mass with a major vascularization on colour Doppler imaging was observed. It increased progressively and laminated the cerebellum. A neoplastic tumour was suspected but its extent into the cerebral peduncle was unclear. Diagnosis was made at autopsy using histological, immunohistochemical and flow cytometric evaluation. Haemangioblastoma is an exceptional congenital tumour, which is either sporadic or integrated in von Hippel-Lindau di
