Aqueous flare and retinal capillary changes in eyes with diabetic retinopathy.

We examined 112 eyes of 112 adult-onset diabetic patients by fundus fluorescein angiography and with the laser flare cell meter (LFCM) in order to evaluate the possible correlation between retinal capillary changes and aqueous flare. Diabetic eyes were divided into the following groups: minor background diabetic retinopathy (MDR), nonproliferative diabetic retinopathy with predominant capillary occlusion (ONP), nonproliferative diabetic retinopathy with predominant capillary dilation and exudation (ENP) and proliferative diabetic retinopathy (PDR). 108 age-matched eyes of 108 normal subjects w

Muscle biopsy findings in systemic capillary leak syndrome.

Two patients with systemic capillary leak syndrome (SCLS) were followed up clinically for 5 and 2 years, respectively. Muscle biopsy was performed 1 week after the end of an acute crisis in one patient, and after 3 years in the other. In both cases, muscle capillary basement membranes were extremely thick--more than 15 times thicker than normal. Capillary basement membrane enlargement appears to be a permanent lesion, probably limited to muscle vessels. This finding has not been previously reported in SCLS, and would appear to indicate a relationship with the pathogenesis and severity of the c

The compensation of post-traumatic oedema in the rabbit at different ages.

1. The compensation of fluid loss from the circulation after an LD(50) period of bilateral hind-limb ischaemia in a thermoneutral environment has been studied in the rabbit from birth until young adulthood.2. The fluid loss from the circulation was calculated from the increase in the great vessel haematocrit value as the red cell volume and F(cells) were unchanged after limb ischaemia. This ;apparent' fluid loss was compared with the ;actual' fluid loss calculated from the increase in water content of the injured limbs.3. The 0- to 1-day-old rabbit was better able to restore fluid, and protein

Valproic Acid-Induced Autistic-Like Behavior Is Accompanied by Intestinal Damage Driving Changes in Gut Permeability in a Sex-Dependent Way in Rats.

A significant proportion of individuals with autism spectrum disorder (ASD) experience gastrointestinal disturbances exacerbating behavioral symptoms. Therefore, this study investigated alterations in the intestinal mucosa that influence intestinal permeability in rats exposed to valproic acid (VPA) in utero, as well as the sexual differences in it. After assessing social behavior, intestinal permeability was assessed using FITC-dextran, and vascular permeability was evaluated through Evans blue dye tests in the intestine and blood-brain barrier (BBB) of male and female offspring. Furthermore,

Pathogenesis and significance of nonprimary focal and segmental glomerulosclerosis.

Injury of the glomerular microvasculature by nonimmunologic processes is often the underlying mechanism of progressive deterioration of renal function in patients with a variety of renal disorders. The structural hallmark of this injury is focal and segmental glomerulosclerosis, often accompanied by entrapment of hyalin. Although such lesions are quite characteristic for diseases that primarily affect the glomerular podocyte, similar damage occurs in association with functional and structural adaptive changes that develop as a consequence of a significant loss of functioning nephrons or other

RAGE/NF-κB signaling mediates paraquat-induced alveolar-capillary barrier injury in a human alveolar microphysiological system.

Paraquat (PQ), a highly toxic herbicide that causes fatal pulmonary injury, has no specific clinical antidotes, mainly due to incomplete understanding of its pathogenic mechanisms. Inhalational exposure to PQ is the primary route of toxicity among agricultural workers. However, conventional experimental models fail to accurately replicate the physiological characteristics of the alveolar-capillary barrier and the air-liquid interface (ALI). In this study, we developed an Alveolar Microphysiological System (AMS) that co-cultures alveolar epithelial cells (EPCs) and pulmonary microvascular endot

Fluid resuscitation with human albumin or hydroxyethyl starch--are there differences in the healing of experimental intestinal anastomoses?

OBJECTIVES: Restoration of the macro- and microcirculation is important for the healing of gastrointestinal anastomoses. Colloids and crystalloids are widely used for blood volume therapy. We evaluated the effects of human albumin, hydroxyethyl starch (HES) 130/0.4 and saline on the microcirculation and on wound healing in colon anastomoses in rats. MATERIAL AND METHODS: Male Wistar rats received a colonic end-to-end anastomosis. The animals were randomized into three groups and a single 3-ml dose of either 20% human albumin, 6% HES 130/0.4 or 0.9% saline was applied intravenously. Six, 24, 4

Urinary excretions of lipocalin-type prostaglandin D synthase predict renal injury in type-2 diabetes: a cross-sectional and prospective multicentre study.

BACKGROUND: Urinary excretions of lipocalin-type prostaglandin D synthase/beta-trace (L-PGDS) probably reflect the increased permeability of injured glomerular capillary walls of the kidney. We tested the hypothesis in cross-sectional and prospective studies that urinary L-PGDS excretions predict renal injury in type-2 diabetes. METHODS: (1) In the cross-sectional studies, we evaluated whether urinary L-PGDS excretions were able to predict renal diseases in a pooled population including 793 healthy subjects and 200 patients with various forms of renal diseases. (2) We determined the cut-off p

Hypoxia increases the proliferation of brain capillary endothelial cells via upregulation of TMEM16A Ca2+-activated Cl- channels.

The blood-brain barrier (BBB) is mainly formed by brain capillary endothelial cells (BCECs) and is exposed to hypoxic environments under pathological conditions. The effects of hypoxia on the expression and activity of Ca2+-activated Cl- (ClCa) channels, TMEM16A, were examined in bovine brain endothelial t-BBEC117 cells and mouse BCECs. The expression of TMEM16A was upregulated by hypoxia. Whole-cell ClCa currents increased under hypoxia. Hypoxia also increased cell proliferation and trans-endothelial permeability, which were attenuated by ClCa channel blockers or TMEM16A siRNA. These findings

Systemic subclinical lymphedema due to obesity as the cause of clinical lymphedema: A new concept.

On evaluating patients with lymphedema, the authors found that obesity is associated with generalized edema of all extremities and the trunk with increased intracellular and extracellular fluids, thus suggesting a new concept of lymphedema that the authors have termed systemic subclinical lymphedema. Animal studies show that obesity and its progression lead to changes in the lymphatic system and microcirculation with alterations in lymphatic motility, inflammatory processes, capillary permeability and immune response. Systemic subclinical lymphedema is diagnosed when above normal fluid levels …

Abrupt onset of muscle dysfunction after treatment for Grave's disease: a case report.

Myopathy is a known complication of hypothyroidism, commonly characterized by an elevation in Creatine Kinase (CPK) due to increase capillary permeability proportional to the hypothyroid state. Thyroid hormone is important for the expression of fast myofibrillar proteins in the muscle. In hypothyroidism the expression of these proteins are deficient and there is an increase accumulation of slow myofibrillar proteins. A rapid or abrupt descend in thyroid hormones caused by radioiodine therapy after prolonged hyperthyroidism can lead to local hypothyroid state within the muscle tissue, resulting…

Localized bullous pemphigoid in a patient with primary lymphoedema tarda.

We report a case of localized bullous pemphigoid (BP) in a woman patient with primary lymphoedema tarda. There is only one previous case reported of localized pemphigoid in an area of lymphoedema, this being of the cicatricial variant. Slow circulation in the lymphatic vessels, increased capillary permeability with preferential localization of antibodies in the area, and potential cleavage of the epidermal junction due to increased hydrostatic pressure leading to autoimmunity, have all been advocated as possible pathogenic mechanisms. Nevertheless, we consider that the mechanism by which local…

Cochlear vascular changes in response to loud noise.

The findings of several authors suggest that alterations in cochlear vascular physiology may contribute to noise-induced hearing loss. Although the influence of noise on cochlear microcirculation is controversial, recent investigations have identified a number of microvascular alterations during noise exposure. Researchers, using intravital microscopy, a technique which offers the advantage of continuous in vivo observation of cochlear lateral wall vessels, have observed alterations in red blood cell velocity and capillary vasoconstriction within the cochlea during exposure to noise. These alt

Adult respiratory distress syndrome.

The Adult Respiratory Distress Syndrome (ARDS) is a fulminant form of respiratory failure affecting many seriously ill patients. The early manifestations of ARDS are caused by increased permeability of the alveolo-capillary barrier leading to pulmonary edema, stiff lungs, and a large right-to-left intrapulmonary shunt. Polymorphonuclear leukocytes (PMNS) are involved in the pathogenesis of most ARDS, and multiple PMN mechanisms can effect pulmonary injury; interactions between PMN adherence, proteolytic enzyme release, and oxygen radical production are emphasized. ARDS therapy remains largely

Noncardiac pulmonary edema precipitated by tracheal intubation in patients with inhalation injury.

Ten patients with body surface burn and clinical evidence of inhalation injury developed transient, reversible pulmonary edema within 5 min after endotracheal intubation. Hemodynamic studies within 1 hr after intubation revealed normal cardiac output and pulmonary artery wedge pressure (WP). Additionally, in the latter 4 patients, protein concentration of edema fluid (EF) aspirated from the trachea was 58-104% of plasma (P) total protein. These findings suggest that altered capillary permeability was responsible for this transient pulmonary edema. It is postulated that glottic generated expira…

[Mechanisms of acute pulmonary edema during meningococcal shock in children (author's transl)].

Meningococcic shocks may induce fatal acute pulmonary edemas. Ten children, who died in this condition, have been compared to 19 others, who during the treatment of the shock, developed right heart failure, without acute pulmonary edema (8 deaths, 11 survivals). The three groups of children could not be differentiated regarding their clinical status on admission, during the shock phase, or the treatments administered. Composition, volume and speed of administration of fluids were similar in the three groups. An increase in pulmonary capillary permeability could have occurred, and lead to the d…
