Effects of naloxone on dopamine release in the nigrostriatal system.

Simultaneously with a systemic injection of naloxone (NAL), the effects of the nigral application of the d(+)-amphetamine (AMPH) or of right forepaw stimulation on the release of [3H]dopamine [( 3H]DA) in the two caudate nuclei (CN) and the two substantia nigrae (SN) were examined in halothane-anesthetized cats. These experiments were carried out using four push-pull cannulae implanted bilaterally in the CN and SN and continuously supplied with [3H]tyrosine [( 3H]Tyr), the metabolic precursor of dopamine. Nigral AMPH application (1 muM) produced a local increase of the [3H]DA in spite of the N

Involvement of the cholinergic mechanism in depression of the caudate spindle.

Involvement of the cholinergic and catecholaminergic mechanisms in the caudate spindle recorded from the anterior and posterior sigmoid gyri was examined in cats. Physostigmine (0.01 to 0.1 mg/kg i.v.) abolished the appearance of the caudate spindle. These inhibitory effects were antagonized by the administration of atropine (0.25 to 1 mg/kg i.v.). The caudate spindle was inhibited by high frequency stimulation of the mesencephalic reticular formation: This inhibitory effect was antagonized by atropine. On the other hand, L-DOPA (25 to 50 mg/kg, i.v.), L-DOPA + MAO inhibitor and methamphetamin

Efferent connections of the prelimbic (area 32) and the infralimbic (area 25) cortices: an anterograde tracing study in the cat.

The projections from the caudal part of the medial frontal cortex, encompassing the prelimbic area (PL) and the infralimbic area (IL) (Brodm … l formation. Another prominent output of PL concerns projections to an extensive medial part of the caudate nucleus and the ventral striatum, whereas fibers from IL only distribute most ventrally in the striatum. In the amygdaloid complex, fibers from PL were found to reach the basolateral, basomedial, and central nuclei, and fibers from IL to distribute to the m …

Antidepressant-like Effects of Medial Forebrain Bundle Deep Brain Stimulation in Rats are not Associated With Accumbens Dopamine Release.

BACKGROUND: Medial forebrain bundle (MFB) deep brain stimulation (DBS) is currently being investigated in patients with treatment-resistant depression. Striking features of this therapy are the large number of patients who respond to treatment and the rapid nature of the antidepressant response. OBJECTIVE: To study antidepressant-like behavioral responses, changes in regional brain activity, and monoamine release in rats receiving MFB DBS. METHODS: Antidepressant-like effects of MFB stimulation at 100 μA, 90 μs and either 130 Hz or 20 Hz were characterized in the forced swim test (FST). Chan

Afferent projections to the cingulate cortex in albino rats: a study with a retrograde labeling method using fluoro-gold.

We studied the neuronal populations that project their axons to the cingulate cortex in albino rats using the retrograde fluorescent dye of 4% Fluoro-Gold injected into the anterior, middle and posterior portions of the cingulate cortex. The result showed that the following ipsilateral structures are sending fibers to these three portions: the prefrontal cortex, frontoparietal motor cortex, indusium griseum, dorsal endopiriform nucleus, lateral part of medial mammillary nucleus, nuclei of diagonal band of Broca, anterior pretectum, anterior part of caudate-putamen, hippocampal formation, anter

Connections of the anterior ectosylvian visual area (AEV).

We have previously described a visual area situated in the cortex surrounding the deep infolding of the anterior ectosylvian sulcus of the c … f AEV being related to more ventral portions of the LPm and LM-Sg complex. AEV also projects to the caudate nucleus, the putamen, the lateral amygdaloid nucleus, the superior colliculus, and the pontine nuclei. It is concluded that AEV is a visual association area which functionally relates the visual with both the motor and the limbic system and that it might pl …

An Unusual Case of Hypersomnolence.

A young woman presented with acute-onset hypersomnia. Due to the acute onset, the possibility of secondary cause was considered. Magnetic resonance imaging of the brain was showing involvement of deep gray structures surrounding the third ventricle such as thalamus, hypothalamus, and caudate nucleus with true diffusion restriction. Differentials of vasculitic infarct or inflammatory cause were considered. Among inflammatory causes, neuromyelitis optica spectrum disorder (NMOSD) was considered due to predominant diencephalic involvement. Serum aquaporin-4 immunoglobulin G was positive. No other…

Cerebello-Parietal Functional Connectivity in Amnestic Mild Cognitive Impairment.

The role of the cerebellum in amnestic mild cognitive impairment (aMCI), typically a prodromal stage of Alzheimer's disease, is not fully understood. We studied the lobule-specific cerebello-cerebral connectivity in 15 cognitively normal and 16 aMCI using resting-state functional MRI. Our analysis revealed weaker connectivity between the cognitive cerebellar lobules and parietal lobe in aMCI. However, stronger connectivity was observed in the cognitive cerebellar lobules with certain brain regions, including the precuneus cortex, posterior cingulate gyrus, and caudate nucleus in participants w…

Rare case of duplicated middle cerebral artery and acute ischaemic stroke managed successfully with mechanical thrombectomy.

A man in his early 80s presented with acute onset aphasia and right-sided weakness with an NIH Stroke Scale (NIHSS) of 17. He was last seen normal 9 hours before the presentation. MRI of the brain showed acute infarcts in the left caudate, lentiform nucleus and corona radiata. MRI angiogram (MRA) revealed an occluded left main middle cerebral artery (MCA) and an associated ipsilateral patent duplicated middle cerebral artery (DMCA). Mechanical thrombectomy (MT) was performed, and he was discharged with an NIHSS of 8. In this report, we review and discuss the challenges during the intervention

Pearls & Oy-sters: Huntington Disease Presenting as Primary Progressive Aphasia: A Case of Semantics.

We present a case of semantic variant primary progressive aphasia as the presenting feature in a patient with Huntington disease (HD). The patient initially developed progressive language impairment including impaired naming and object knowledge and single-word comprehension and then developed chorea and behavioral changes. An MRI of the brain showed left anterior temporal lobe and hippocampal atrophy. A neurologic FDG PET/CT showed reduced metabolism in the head of the left caudate nucleus. Huntingtin gene testing revealed an expansion of 39 CAG repeats in 1 allele. This case outlines the sub…

[Immunohistochemical and ultrastructural studies of subependymal giant cell astrocytomas in children with tuberous sclerosis].

Ten cases of subependymal giant cell astrocytoma associated with sclerosis tuberosa were reevaluated in order to assess their phenotyping and biologic features. All tumours were multifocal, located within lateral ventricles, often overlying the head of the caudate nucleus or protruding into the third ventricle. The phenotype of SGCA disclosed a complex pattern: giant cells were GFAP positive, some of them were stained with antibodies against neurofilament and NSE. Ultrastructurally, the cells of SGCA contained frequent dense bodies, numerous intermediate filaments and microtubules. Biologicall…

Increased concentrations of immunoreactive cathepsin D in supraoptic nucleus of the Brattleboro rat.

The distribution of cathepsin D, an aspartyl endopeptidase, was measured in selected, discrete nuclei of the forebrain of the Brattleboro rat by means of microdissection and radioimmunoassay. The results indicate that cathepsin D is widely distributed, but in varying amounts among nuclear groups in this region of the brain. High concentrations were detected in the supraoptic and paraventricular nuclei. In studies of the vasopressin-deficient Brattleboro rat, an increased content of cathepsin D in the supraoptic nucleus was observed compared to the heterozygous control. No differences were dete

Tyrosine hydroxylase and tryptophan hydroxylase activities and its cofactor biopterin level in brain regions of the rolling mouse: The influence of thyrotropin releasing hormone.

Biopterin, the cofactor for tyrosine hydroxylase and tryptophan hydroxylase, was decreased in caudate nucleus, hypothalamus and cerebellum of the rolling mouse. Though there were not significant differences of tyrosine hydroxylase and tryptophan hydroxylase activities between the rolling and normal control mouse in the hypothalamus, the rolling showed significant increase of biopterin concentration and tyrosine hydroxylase activity after administration of thyrotropin releasing hormone (TRH). These results suggest that ataxic gait of the rolling mouse may be partly due to some abnormalities of …

The regional distribution of the nerve-stimulating phosphopeptide (nerveside, substance B) in the central nervous system.

1. The nerve-stimulating phosphopeptide, nerveside, is present mainly in the cerebrum and brain stem, while the cerebellum contains only a trace amount of it.2. Nerveside is uniformly distributed over all parts of the cerebral cortex.3. The cerebral cortex contains the greatest concentration of nerveside. If the nerveside activity of 1 g cerebral cortex is expressed as 100 then the relative activities of the other parts of the C.N.S. are hippocampus 41, caudate nucleus 21, diencephalon (without hypothalamus) 24, midbrain 19, pons 7, medulla 12 and cerebellum 6.4. The nerveside activity of the …

Extensive cortical damage in a case of Creutzfeldt-Jacob disease: clinicoradiological correlations.

MRI demonstrated extensive cortical involvement in a patient with pathologically proven Creutzfeldt-Jacob disease. The whole brain was atrophic; some of the supratentorial cortex, putamen and caudate nucleus gave high signal on T2-weighted images; the changes were more extensive on diffusion-weighted images (DWI). Comparison of the history, and the sites of atrophy and signal change suggested that the latter predominates in regions with long-lasting damage and prevalent gliosis, while high signal on DWI indicate current neuronal loss. This case widens the range of MRI findings in patients with…

[Hemichorea caused by striatal infarct in a young type 1 diabetic patient].

Movement disorders such as chorea and ballism rarely occur in diabetes mellitus. We report the case of 26-year-old man with a 13-year-history of type 1 diabetes mellitus. He presented with a right side hemichorea. Brain CT-scan and MRI showed an infarction of the head of the caudate nucleus and the anterior part of the putamen. Presence of microangiopathy affecting retina, kidneys and peripheral nerves suggest a similar involvement of the lenticulo-striatal arteries. Hemichorea and hemiballism usually occur in older patients presenting type 2 diabetes mellitus. Non-ketotic hyperglycaemia is th…

Neuropathology in glutaric acidaemia type 1.

The neuropathology in three cases of glutaric acidaemia type 1 is presented. All three showed extensive neuronal loss in the caudate nucleus and the putamen, with only small numbers of large neurons surviving. The globus pallidus showed moderate shrinkage and gliosis but no conspicuous decreases in neurons. Severe spongiform change was seen in many regions, involving predominantly white matter. These features are very similar to some cases described previously in familial striatal degeneration in childhood, but are different from other cases. If found in a brain at autopsy, they should lead to…
