Prognostic value of cell proliferation markers, tumour suppressor proteins and cell adhesion molecules in primary squamous cell carcinoma of the larynx and hypopharynx.

In an attempt to identify molecular prognostic markers, a series of laryngeal and hypopharyngeal carcinomas was examined for PCNA, Ki67, p27(Kip1), p53, E-cadherin and CD44 by immunohistochemistry and for DNA content by flow cytometry. No correlation was found between E-cadherin, CD44, p53 or DNA ploidy and the clinicopathological data. The fraction of cancer cells immunolabelled for p27(Kip1) correlated with tumour differentiation, but not with lymph node metastasis. In contrast, the PCNA, Ki67 and S-phase fractions of cancer cells were significantly higher in tumours with lymph node metastas

Molecular subtyping of endometrial carcinoma cell lines uncovers subtype-specific targetable vulnerabilities.

Endometrial carcinoma (EC), the most common gynecologic cancer type in developed countries, encompasses four molecular subtypes (POLEmut, MMRd, p53abn, and NSMP) that have prognostic values and guide treatment decisions. Additionally, dual loss of ARID1A and ARID1B (referred to as ARID1A/B) characterizes a significant portion of dedifferentiated/undifferentiated EC (DD/UDEC), a rare but highly aggressive subtype of EC. To advance the translational research for ECs, we analyzed the genomic features of a panel of 39 EC cell lines, leading to the identification of cell lines representing each of

Significant response to toripalimab plus axitinib for metastatic chromophobe renal cell carcinoma with sarcomatoid differentiation: a case report and literature review.

Chromophobe renal cell carcinoma (chRCC) is a rare subtype of renal cell carcinoma (RCC). Sarcomatoid differentiation is considered a result of dedifferentiation of the primary tumor. The coexistence of both components (chromophobe and sarcomatoid) in a single renal tumor has been infrequently reported. Currently, only isolated cases of metastatic sarcomatoid chRCC treated with immune checkpoint inhibitors (ICIs) or targeted therapies have been documented. Here, we present the first case of metastatic sarcomatoid chRCC treated with toripalimab plus axitinib and provide a review of the relevant

Comparison of calcium-binding proteins expressed in cultured hNT neurons and hNT neurons transplanted into the rat striatum.

An alternative source of cells for neural transplantation and brain repair that has many characteristics of immature neurons is the hNT neuron, derived from an embryonal human teratocarcinoma (NTera2) cell line that is terminally differentiated in vitro with retinoic acid. The majority of hNT neurons are GABAergic in cell culture. We have determined the calcium-binding protein (CBP) phenotypes of hNT neurons for three CBPs, calretinin (CR), calbindin D-28K (CB), and parvalbumin (PV), in cell culture and after transplantation into the rat striatum. In cell culture, 95% of all cell profiles were

Inflammaging impairs peripheral nerve maintenance and regeneration.

The regenerative capacity of peripheral nerves declines during aging, contributing to the development of neuropathies, limiting organism function. Changes in Schwann cells prompt failures in instructing maintenance and regeneration of aging nerves; molecular mechanisms of which have yet to be delineated. Here, we identified an altered inflammatory environment leading to a defective Schwann cell response, as an underlying mechanism of impaired nerve regeneration during aging. Chronic inflammation was detected in intact uninjured old nerves, characterized by increased macrophage infiltration and

Wallerian degeneration and axonal regeneration after sciatic nerve crush are altered in ICAM-1-deficient mice.

The intercellular cell adhesion molecule-1 (ICAM-1) has been implicated in the recruitment of immune cells during inflammatory processes. Previous studies investigating its involvement in the process of Wallerian degeneration and focusing on its potential role in macrophage recruitement have come to controversial conclusions. To examine whether Wallerian degeneration is altered in the absence of ICAM-1, we have analyzed changes in the expression of axonal and Schwann cell markers following sciatic nerve crush in wildtype and ICAM-1-deficient mice. We report that the lack of ICAM-1 leads to imp

A paired comparison between glioblastoma "stem cells" and differentiated cells.

Cancer stem cells (CSC) have been postulated to be responsible for the key features of a malignancy and its maintenances, as well as therapy resistance, while differentiated cells are believed to make up the rapidly growing tumour bulk. It is therefore important to understand the characteristics of those two distinct cell populations in order to devise treatment strategies which effectively target both cohorts, in particular with respect to cancers, such as glioblastoma. Glioblastoma is the most common primary brain tumour in adults, with a mean patient survival of 12-15 months. Importantly, t

Reverse differentiation as a gene filtering tool in genome expression profiling of adipogenesis for fat marker gene selection and their analysis.

BACKGROUND: During mesenchymal stem cell (MSC) conversion into adipocytes, the adipogenic cocktail consisting of insulin, dexamethasone, indomethacin and 3-isobutyl-1-methylxanthine not only induces adipogenic-specific but also genes for non-adipogenic processes. Therefore, not all significantly expressed genes represent adipogenic-specific marker genes. So, our aim was to filter only adipogenic-specific out of all expressed genes. We hypothesize that exclusively adipogenic-specific genes change their expression during adipogenesis, and reverse during dedifferentiation. Thus, MSC were adipogen

Dedifferentiated liposarcoma presenting with ascites as the initial clinical manifestation: a case report and literature review.

Dedifferentiated liposarcoma (DDLPS) is a high-grade soft tissue sarcoma predominantly arising in the retroperitoneum, characterized by an insidious clinical course. This study reports a 63-year-old female patient who presented with abdominal distension and decreased appetite, without other systemic complaints. Malignant tumor cells were identified in both ascitic smears and cell blocks. Combined with immunohistochemical (IHC) staining, the findings were consistent with sarcoma cells. Fluorescence in situ hybridization (FISH) analysis of the tissue biopsy revealed clustered amplification of mo

Glioblastoma and resistance to radiotherapy: role of cancer stem cells subpopulations, hypoxia and therapeutic strategies.

Glioblastoma is the most aggressive primary brain tumor in adults, characterized by rapid progression, resistance to therapy, and inevitable recurrence. Despite standard treatment-surgical resection, X-ray radiotherapy, and temozolomide chemotherapy-prognosis remains poor. Growing evidence indicates that glioma stem cells (GSCs) and hypoxia drive this resistance and recurrence. This review examines distinct GSC subtypes: mesenchymal GSCs, the most aggressive and invasive; proneural GSCs, which are more radiosensitive but highly proliferative and contribute to recurrence; and slow-cycling GSCs,

PKR activation-induced mitochondrial dysfunction in HIV-transgenic mice with nephropathy.

HIV disease remains prevalent in the USA and chronic kidney disease remains a major cause of morbidity in HIV-1-positive patients. Host double-stranded RNA (dsRNA)-activated protein kinase (PKR) is a sensor for viral dsRNA, including HIV-1. We show that PKR inhibition by compound C16 ameliorates the HIV-associated nephropathy (HIVAN) kidney phenotype in the Tg26 transgenic mouse model, with reversal of mitochondrial dysfunction. Combined analysis of single-nucleus RNA-seq and bulk RNA-seq data revealed that oxidative phosphorylation was one of the most downregulated pathways and identified sig

Ceramide from sphingomyelin hydrolysis induces neuronal differentiation, whereas de novo ceramide synthesis and sphingomyelin hydrolysis initiate apoptosis after NGF withdrawal in PC12 Cells.

BACKGROUND: Ceramide, important for both neuronal differentiation and dedifferentiation, resides in several membranes, is synthesized in the endoplasmic reticulum, mitochondrial, and nuclear membranes, and can be further processed into glycosphingolipids or sphingomyelin. Ceramide may also be generated by hydrolysis of sphingomyelin by neutral or acidic sphingomyelinases in lysosomes and other membranes. Here we asked whether the differing functions of ceramide derived from different origins. METHODS: We added NGF to PC12 cells and to TrkA cells. These latter overexpress NGF receptors and are

Histopathology of gastric carcinoids: a survey of 42 cases.

An unselected series of 42 gastric carcinoids has been reviewed. Clinically the tumours simulated common gastric lesions including ulcer, polyp and carcinoma. No endocrine symptoms were identified. The tumours were most frequent in the body of the stomach and in 25% in that site were multiple. Morphologically most tumours when classified according to Soga (1974) demonstrated a mixed growth pattern. Six tumours displayed an atypical morphology (type D): they were larger and metastasized more frequently than the rest of the tumours. Six tumours contained a few scattered argentaffinic cells but t

Ramulus Mori (Sangzhi) alkaloids enhance pancreatic β-Cell maturation and function by targeting the CD81/endoplasmic reticulum stress pathway.

BACKGROUND: β-cell dedifferentiation and impaired maturation are crucial contributors to β-cell dysfunction in type 2 diabetes mellitus (T2DM), yet targeted therapies remain limited. OBJECTIVE: To investigate the protective effects of Ramulus Mori (Sangzhi) Alkaloids (SZ-A) on β-cell function and maturation in T2DM. METHODS: A T2DM mouse model was induced by high-fat diet and streptozotocin, followed by SZ-A treatment. Single-cell RNA sequencing (scRNA-seq) was used to profile β-cell subpopulations, identifying Cd81 as a marker of β-cell immaturity. Cd81-overexpressing diabetic mice, as well…

Peripheral T cell lymphoma: immunologic and cell-kinetic observations associated with morphological progression.

Peripheral T cell lymphomas (PTCLs) form a morphologically heterogeneous group of non-Hodgkin's lymphomas that are generally considered to have immunophenotypes associated with mature T cells, usually those of helper T cells. We now describe and correlate the clinical, morphological, immunologic, and cell-kinetic findings based on the evaluation of eight tissue samples obtained at various times from a 13-year-old girl with PTCL. The early morphological expressions of this patient's PTCL were those of diffuse mixed-cell lymphoma and focal large-cell lymphoma (LCL) evolving from the histologic p

Undifferentiated pleomorphic sarcoma of the small intestine presenting with intussusception in a patient with prior well-differentiated liposarcoma.

An 88-year-old man presented with abdominal pain and vomiting. Contrast-enhanced computed tomography revealed a target sign, suggesting intussusception caused by a small-bowel tumor. Laparoscopic exploration was performed, followed by manual reduction using the Hutchinson technique and segmental small-bowel resection. Histopathological examination revealed a 3-cm submucosal tumor composed of pleomorphic tumor cells. Immunohistochemistry showed no specific lineage differentiation, and staining for MDM2 and CDK4 was negative. Because of his history of well-differentiated liposarcoma, metastatic

Updates in Peripheral Nerve Sheath and Adipocytic Tumors.

Certain areas in peripheral nerve sheath and adipocytic neoplasm diagnosis historically were characterized by a lack of clarity and consensus regarding terminology, diagnostic criteria, and prognosis. Recent work has led to important clarifications in diagnostic criteria and/or outcome data for neurofibromas with atypia and MDM2 amplification-negative adipocytic tumors. However, these clarifications remain incompletely recognized by many pathologists, in part due to the paucity of these neoplasms. This paper reviews the history and recent advances in diagnostic criteria, terminology, and under

The Pathogenesis of Primary Biliary Cholangitis: A Comprehensive Review.

Primary biliary cholangitis (PBC) is a chronic cholestatic liver disease characterized by autoimmune destruction of small to medium size intrahepatic bile ducts. The etiology of PBC remains unknown and pathogenesis features immune-mediated biliary injury, alongside the consequences of chronic cholestasis. PBC is strongly associated with the loss of immune tolerance against mitochondrial antigens and the subsequent presence of an articulated immunologic response that involves both humoral and cellular responses. Both environmental factors and genetic variants increase PBC susceptibility. Biliar

Myocardin: A novel player in atherosclerosis.

Myocardin (MYOCD) the most important coactivator of serum response factor (SRF), plays a critical role specifically in the development of cardiac myocytes and vascular smooth muscle cells (VSMCs). Binding of Myocardin to the SRF on the CArG box-containing target genes can transcriptionally activate a variety of downstream muscle-specific genes, such as Sm22α, Acta2, Myh11, and several other signaling pathways. Myocardin expression represents a contractile and differentiated SMC phenotype. Loss of Myocardin, however, represents a synthetic and dedifferentiated phenotype, a hallmark in atheroscl
