Emerging approaches of wound healing in experimental models of high-grade oral mucositis induced by anticancer therapy.

Clinical guidelines for oral mucositis (OM) still consist in palliative care. Herein, we summarize cellular and molecular mechanisms of OM ulceration in response to chemical therapies in animal models. We discuss evidenced anti-inflammatory and anti-oxidant drugs which have not been ever used for OM, such as synthetic peptides as well as cell therapy with mesenchymal stem cells; amniotic membranes, mucoadhesive polymers loaded with anti-inflammatory agents and natural or synthetic electrospun. These approaches have been promising to allow the production of drug-loaded membranes, scaffolds for

[Advances in the study of vincristine: an anticancer ingredient from Catharanthus roseus].

Vincristine is a dimer-indo-alkaloid which is extracted from the leaves of Catharanthus roseus. It is effective to treat acute lymphocytic cell leukemia, Hodgkin disease and non-Hodgkin disease clinically. But the severe side effects, such as neurotoxic and tissue damage, limit its application. In this paper, we summarize physical, chemical, pharmacological and pharmacokinetical properties of VCR and advances in decreasing its side effects. In clinic, association with other medication is adopted. In pharmaceutics, people adopt some new methods and technology such as conjugation with the antibo

Transplantation of encapsulated mitochondria alleviates dysfunction in mitochondrial and Parkinson's disease models.

Mitochondrial transplantation holds significant potential for the treatment of mitochondrial diseases. However, how to efficiently deliver exogenous mitochondria to somatic cells or tissues remains unresolved. We present a mitochondrial transplantation approach to deliver mitochondria into the cells and tissues of mice and monkeys with high efficiency, based on encapsulating mitochondria with vesicles derived from the plasma membrane of erythrocytes. Treatment with encapsulated mitochondria complemented the loss, deletion, or mutation of mitochondrial DNA, thereby rescuing the associated bioen

Development of Degradable Silylated Castor Oil-Based Microparticles for the Sustained Release of a Lipophilic Anti-Cancer Drug.

Background/Objectives: Hybrid microparticles (HMP) based on silylated vegetable oils have recently emerged as promising carriers for the sustained delivery of poorly water-soluble drugs, offering a controlled release without a burst effect and excellent biocompatibility. However, their limited degradability and the acidic conditions required for the sol-gel encapsulation process restrict their biomedical applicability, particularly for acid-sensitive active compounds. The aim of the present study was to develop new degradable vegetable oil-based HMP with improved drug-solubilization capacity w

A Polarity-Sensitive Dicyanoisophorone Based Fluorescent Probe for Targeted Human Serum Albumin Detection in Blood and Urine With Cellular Imaging.

Protein biomarkers serve as a crucial prognostic indicator for life-threatening disorders in the era of personalized medicine. In this context, we developed a polarity-sensitive fluorescent probe, DQDM, for the sensitive detection and quantification of human serum albumin (HSA). It operates through an intramolecular charge-transfer (ICT) mechanism showing weak fluorescence in ~100% PBS. Notably, upon binding to HSA, a substantial (~40-fold) fluorescence enhancement at ~650 nm was observed, which arises from its encapsulation within the low-polarity, hydrophobic binding cavity of the protein re

Chitosan-polysaccharide composite carriers under in vitro digestion: Classification, gastrointestinal behavior, and bioaccessibility implications-A review.

Chitosan-polysaccharide composites should be evaluated as gastrointestinally responsive structures rather than as generic encapsulation matrices. We organize food-relevant systems into four architectures: interfacial multilayers, hydrogel and microgel networks, particulate carriers, and hierarchical hybrids. Within this framework, we examine how material attributes, carrier structure, cargo properties, and digestion protocols govern protection, structural reorganization, release, bioaccessibility, and probiotic viability. The evidence shows that high retention, encapsulation efficiency, or muc

Antioxidant Ascorbic Acid Influences Cell-mediated Alterations in Localized Hydrogel Crosslinking and Affects Neo-Cartilage Connectivity in Hydrolytically Degradable Hydrogels.

Photopolymerizable and degradable poly(ethylene glycol) (PEG) hydrogels are a promising platform to deliver chondrocytes for cartilage tissue engineering. Previous studies reported that cells are exposed to and react with free-radicals produced during encapsulation, creating a pericellular region of reduced crosslink density measured by the distance Rd. This study tests the hypothesis that increasing Rd improves spatial elaboration of deposited extracellular matrix (ECM) (i.e., micro-tissue) in a degrading hydrogel. Chondrocytes pre-treated or not with the antioxidant ascorbic acid atlow (1 nM

Intracellular Delivery of Full-Length Antibodies via Poly-l-lysine-Coated PEG-PLGA Polymersomes Enables Noninvasive Pulmonary Immunotherapy.

The intracellular delivery of full-length antibodies offers substantial therapeutic potential but remains limited by poor cellular uptake, extracellular degradation, and inefficient encapsulation strategies. Here, we report a noninvasive, scalable, and biocompatible nanocarrier platform based on poly-l-lysine (PLL)-coated polyethylene glycol-block-poly(lactic-co-glycolic acid) (PEG-PLGA) polymersomes for efficient intracellular antibody delivery. Coating polymersomes with 30 kDa ε-poly-l-lysine increased antibody encapsulation efficiency to ∼ 80%. It enabled precise modulation of surface charg

Resolving the mRNA Encapsulation-Release Trade-off via Compensatory Forces in Engineered Ionizable Lipids.

A critical challenge in lipid nanoparticle (LNP) delivery of messenger RNA (mRNA) is the inherent trade-off between stable encapsulation and efficient intracellular release. Here, this challenge is addressed through rationally engineering compensatory forces between mRNA and LNP, leveraging short-range intermolecular interactions (van der Waals, hydrogen bonding) to dynamically balance long-range Coulombic binding. Guided by a computational-experimental framework, a "contact number" metric is developed to decipher mRNA and LNP binding hierarchies, enabling the strategic incorporation of short-

Biotin-modified starch nanoparticles for resveratrol delivery: Encapsulation mechanism and synergistic efficacy in ulcerative colitis therapy.

Resveratrol (RES) is a promising anti-inflammatory polyphenol for treating ulcerative colitis (UC), but its efficacy is constrained by poor solubility and colon-targeting efficiency. Biotin, as an active ligand, has been widely used to functionalize nanocarriers to enhance intestinal cell permeability, thereby increasing the absorption of bioactive components. Biotin modification enlarged the helical cavity of starch into a V7-type structure, creating a more favorable hydrophobic microenvironment for the encapsulation of guest molecules. The mass ratio of resveratrol to starch was 1:10 to 1:2,

Stingless bee propolis-loaded alginate/zinc nanogels: Enhanced propolis bioavailability and effects on melanoma cells migration.

Bioactive compounds with low aqueous solubility often exhibit limited bioavailability, which restricts their therapeutic potential. This study explores the antimelanoma activity of the dichloromethane propolis extract (DPE), a terpene-rich fraction from the stingless bee Melipona quadrifasciata and enhances its solubility via encapsulation in zinc-crosslinked alginate nanogels (pNG) through ionic gelation technique. Unlike its well-studied polar counterpart, the non-polar and semi-polar fraction remains largely unexplored, offering new biological opportunities to treat diseases such as melanom

Development of nanostructured lipid carriers as a promising tool for methotrexate delivery: physicochemical and in vitro evaluation.

The aim of the present study is to fabricate the stable nanostructured lipid carriers (NLCs) using biocompatible excipients for the encapsulation of Methotrexate (MTX), a chemotherapeutic agent for breast cancer treatment. MTX has restricted clinical applications owing to its low solubility, non-specific targeting and adverse side effects. Glyceryl Monostearate (GMS) and Miglyol 812 (MI1) were chosen as solid and liquid lipids, respectively, for the fabrication of NLCs, and the influence of variation of solid and liquid composition was investigated. The prepared NLCs exhibited long-term stabil

5-Fluorouracil drug delivery system based on bacterial nanocellulose for colorectal cancer treatment: Mathematical and in vitro evaluation.

Colorectal cancer (CRC) is the third most common worldwide. Its treatment includes adjuvant chemotherapy with 5-fluorouracil (5FU) administered intravenously. 5FU is an antineoplastic drug of the fluoropyrimidines group, widely used in the treatment of solid tumors, mainly CRC. Nevertheless, it causes several adverse effects and poor effectiveness due to its short half-life. This work aimed to employ bacterial nanocellulose (BNC) as an encapsulation material for the oral administration of 5FU. First, the adsorption phenomena were analyzed by isotherms, thermodynamic parameters, and kinetic mod
