Optical genome mapping enhances cytogenetic analysis in recurrent miscarriage: confirmation of a suspected (1;10) chromosomal translocation.

BACKGROUND: Optical genome mapping (OGM) is a next-generation cytogenetic technique that may be beneficial for detecting subtle structural chromosomal alterations that can go unnoticed with conventional studies in couples with recurrent pregnancy loss. CASE PRESENTATION: We report the case of a couple referred to our assisted reproduction unit due to a history of recurrent pregnancy loss. Initially, conventional cytogenetic studies were performed to identify a possible genetic cause. To this end, the karyotypes of both members of the couple were determined. The fetal tissue from the third mis

[Cytogenetical and molecular analyses in a horse with SRY-negative sex reversal].

Numerical and structural aberrations of chromosomes may cause malformations, embryonal losses and reduced or missing fertility. In male horses the fertility is rather well controlled through their semen evaluation. For mares there are no mandatory regulations which specify their use in a breeding programme. Therefore, mares with chromosomal aberrations, which exclude reproduction success may be chosen for breeding. The present case describes a horse, offically registered as a female, which was presented for a breeding exam. On this occasion, small and inactive ovaries were diagnosed. Although

Cytogenetical findings of recurrent meningiomas. A study of 10 tumors.

Cytogenetic analyses of 10 cases of recurrent meningiomas growing in culture between 1-10 days are reported, of which seven showed benign morphology, one atypical, and two, malignant features. Normal karyotypes with nonclonal alterations were found in three cases, one case with only monosomy 22, and complex karyotypes in the remaining six. Four cases were hypodiploid, one pseudodiploid, and one hyperdiploid. The chromosomes most often involved in structural rearrangements were 1, 7, and 14 and the losses were chromosomes 7, 10, 14, 15, 18, and 22. Ring chromosome, dicentrics, double minutes, a

Chromosome analysis of human spermatozoa stored in vitro.

Two-hundred-sixty-five chromosome spreads from control human sperm samples capacitated in TEST-yolk buffer at 4 degrees C and 232 chromosome spreads from sperm samples incubated in vitro in Biggers-Whitten-Whittingham (BWW) for 24 h at room temperature prior to capacitation, were studied after fusion of sperm cells with zona-free hamster eggs. Sperm cells were provided by two volunteer donors. The results indicate an increase in chromosome structural abnormalities after in-vitro incubation of the spermatozoa from 1.8 to 7.7% in donor no. 1 and from 4.5 to 12.5% in donor no. 7. Overall, structu

Effect of cryopreservation on the frequency of chromosomal abnormalities and sex ratio in human sperm.

The effects of cryopreservation on the frequency and type of chromosomal abnormalities in human sperm were investigated. Employing a technique that enables direct visualization of human sperm chromosomes following in vitro penetration of hamster oocytes, sperm samples from 10 normal men were examined before and after freezing in liquid nitrogen. A total of 1,960 sperm karyotypes were analyzed, 1,132 before freezing and 828 after freezing. There was no significant difference in the frequency of structural chromosomal anomalies (10.5% prefreeze vs. 8.5% postfreeze), but there was a significant d

Mosaicism or pseudomosaicism: the problem of hypermodal cells in amniotic fluid cell culture.

A series of 2029 consecutive amniotic fluid specimens studied for prenatal genetic diagnosis were reviewed and reassessed so as to evaluate the frequency and clinical significance of hypermodal cells in amniotic fluid cell cultures. Hypermodal cells were defined as those with more than 46 chromosomes, and were characterized by an additional structurally normal or structurally abnormal chromosome. Of 2029 specimens, 47 (2.31 per cent) contained a total of 167 hypermodal cells. True fetal mosaicism was detected in three cases (0.14 per cent). All had hypermodal cells in more than one culture fla

Effect of Aerobic Physical Exercise in an Animal Model of Duchenne Muscular Dystrophy.

Duchenne muscular dystrophy (DMD) is a condition caused by an amendment to the X chromosome, inherited as a recessive trait, and affects 1:3500 live births, especially males. Low-intensity exercise is known to decrease certain parameters associated with muscular degeneration in animal models of progressive muscular dystrophies. In the present study, 28-day-old male mdx and wild-type (wild) mice were used. The animals were subjected to a low-intensity physical exercise protocol for 8 weeks. It was found that this protocol was able to reduce oxidative stress in muscle tissue and in most of the C

Immunohistochemical analysis of presenilin-1 expression in the mouse brain.

At least 22 different mutations associated with early-onset familial Alzheimer's disease (AD) in various kindreds have been reported to occur in a recently identified gene on chromosome 14, presenilin 1 (PS-1) (Sherrington et al. (1995) Nature 375, 754-760 [1] and reviewed by Van Broeckhoven (1995) Nat. Genet. 11, 230-231 [2]). In order to study the localization of PS-1 in the brain, we raised a polyclonal antiserum specific to a fragment of the predicted protein sequence of PS-1. PS-1 immunostaining was found intracellularly, in the perikaria of discrete cells, mostly neurons, appearing as th

[Cytogenetic aspects of diffuse scleroderma : structural anomalies and sister chromatid exchanges (author's transl)].

Structural chromosome anomalies (1 477 cells examined) and sister chromatid exchanges after two replication cycles with BrdU (771 cells studied) were evaluated in 12 patients with diffuse scleroderma and having received no recent or important irradiation. The increase of structural anomalies, chromatidic as well as chromosomal, is always low, inconstant and cannot be considered as having a diagnostic value. Increase of sister chromatid exchanges could be a more sensitive method of investigation. In particular, it is not influenced by low doses of diagnostic X-rays.

NUT Midline Carcinoma of the Nasal Cavity.

Nuclear protein in testis (NUT) midline carcinoma (NMC) is a rare, aggressive, poorly differentiated form of squamous cell carcinoma caused by a chromosomal rearrangement of the NUT gene on chromosome 15. These tumors have a predilection for midline and paramidline structures of the upper aerodigestive tract and mediastinum and can affect patients across a broad age range, including children. In the current example, a 53 year old male presented with a mass originating in the left nasal cavity. The clinical, radiographic, and morphologic features of NMC are discussed.

Sex reversal in a child with duplication of sex reversing locus on the short arm of the X chromosome (Xp).

We report a child with a female phenotype possessing a karyotype of 46,XY,13p+. The child had female external genitalia, and manifested severe mental retardation, pulmonary atresia and multiple congenital abnormalities. Laparoscopy revealed the presence of streak gonads and Müllerian structures. Histological examination of the gonads showed ovarian-like stroma with immature seminiferous tubules. Chromosome and gene analyses demonstrated Xp11.23 (or 11.3)-pter duplication and an intact sex determinating factor of Y (SRY). The findings of this case suggest that duplication of Xp causes sex rever

Unilateral biorchism associated with contralateral rudimentary uterus.

A healthy 5-year-old boy presented with two palpable masses in the right hemiscrotum. An examination also showed a palpable mass in the left groin. During surgery, he was found to have right-sided biorchism and a left groin nodule attached to the left vas deferens and spermatic cord. The nodule was excised and histologic examination showed the presence of persistent Mullerian structures. A chromosome analysis showed the normal male karyotype. Both conditions have been previously reported, but this is the first report of both conditions occurring in the same patient.

Boundary sequences flanking the mouse tyrosinase locus ensure faithful pattern of gene expression.

Control of gene expression is dictated by cell-type specific regulatory sequences that physically organize the structure of chromatin, including promoters, enhancers and insulators. While promoters and enhancers convey cell-type specific activating signals, insulators prevent the cross-talk of regulatory elements within adjacent loci and safeguard the specificity of action of promoters and enhancers towards their targets in a tissue specific manner. Using the mouse tyrosinase (Tyr) locus as an experimental model, a gene whose mutations are associated with albinism, we described the chromatin s

Pericentric inversion of chromosome 5: a possible threat to male fertility?

Pericentric inversion of chromosome 5 (inv(5)(p12,q13] was diagnosed in two unrelated men with normal phenotype referred for infertility. The spermatogenic damage in these patients may be explained on the basis of the X-Y bivalent behaviour in cases of some balanced structural rearrangements of the chromosomes. The Authors propose a possible negative action of inv(5) in male gamete production and suggest a larger use of the chromosome analysis in the study of male infertility.

Improvement of high-resolution fluorescence in situ hybridisation mapping on chromosomes of Brassica oleracea var. capitata.

The low resolution of chromosome-based Fluorescence in situ hybridisation (FISH) mapping is primarily due to the structure of the plant cell wall and cytoplasm and the compactness of regular chromosomes, which represent a significant obstacle to FISH. In order to improve spatial resolution and signal detection sensitivity, we provide a reproducible method to generate high-quality extended chromosomes that are ~13 times as long as their pachytene counterparts. We demonstrate that proteinase K used in this procedure is crucial for stretching pachytene chromosomes of Brassica oleracea in the cont

Human Chromosomes: II. Preparation, Analysis and Diagnostic Implications of Abnormalities.

This presentation is designed to show the diagnostic implications of chromosomal abnormalities, and how in some cases chromosome analysis may be helpful in prognosis and counselling. Most males with Klinefelter's syndrome have chromatinpositive nuclei and an abnormal sex chromosome complex (usually XXY). In Turner's syndrome many such females have chromatin-negative nuclei and a deficient sex chromosome complex (usually XO). Multiple-X females have unusual chromatin patterns (two or three masses of sex chromatin) and abnormal sex chromosome complexes (XXX, XXXX, XO/XXX, etc.). One of the paren

Fusions near telomeres occur very early in the amplification of CAD genes in Syrian hamster cells.

Previous analyses by fluorescence in situ hybridization of structures present 20-30 cell generations after the primary events of mammalian gene amplification have shown that tens of megabases of DNA separate each copy of the selected gene in chromosomal arrays that contain up to 15 copies. Since these structures are very unstable, it is necessary to study amplified DNA as soon as possible after it has been formed to relate the structures observed to the primary mechanisms that generated them. Previously, new amplifications of the CAD gene were analyzed in colonies of 10(5) N-(phosphonoacetyl)-

The Types and Frequencies of X Chromosome Abnormalities in Women with Reproductive Problems.

INTRODUCTION: X chromosome architecture and integrity are essential for normal ovarian function. Both numerical and structural X chromosome abnormalities play an important role in female infertility. This study aimed to determine the types and frequency of X chromosome aberrations detected in women referred for cytogenetic investigation due to reproductive problems. METHODS: 2,936 women (average age: 37.5 years) were enrolled in the present study. Peripheral blood karyotyping was performed by conventional cytogenetic techniques. For each woman, 20 G-banded metaphases were studied and in case

Molecular cytogenetic characterization of pancreas cancer cell lines reveals high complexity chromosomal alterations.

Karyotype analysis can provide clues to significant genes involved in the genesis and growth of pancreas cancer. The genome of pancreas cancer is complex, and G-band analysis cannot resolve many of the karyotypic abnormalities seen. We studied the karyotypes of 15 recently established cell lines using molecular cytogenetic tools. Comparative genomic hybridization (CGH) analysis of all 15 lines identified genomic gains of 3q, 8q, 11q, 17q, and chromosome 20 in nine or more cell lines. CGH confirmed frequent loss of chromosome 18, 17p, 6q, and 8p. 14/15 cell lines demonstrated loss of chromosome
