Dissociation and reassociation of oligomeric viral glycoprotein subunits in the endoplasmic reticulum.

The vesicular stomatitis virus (VSV) glycoprotein (G) forms noncovalently linked trimers in the endoplasmic reticulum (ER) prior to transport to the cell surface. Here we examined the formation of heterotrimers between wild-type and mutant subunits that were retained in the ER by C-terminal retention signals. When G protein was coexpressed with mutant subunits that formed trimers at the wild-type rate and were transported from the ER at the wild-type rate, heterotrimers were readily detected. In contrast, when G protein was coexpressed with mutant subunits that formed trimers at the wild-type

Volatile organic compounds from the salt-tolerant Glutamicibacter halophytocola KLBMP 5180 enhance salt stress tolerance in tomato seedlings by regulating antioxidant defense, ion homeostasis, and auxin, jasmonic acid/ethylene signaling pathways.

The application of salt-tolerant plant growth-promoting rhizobacteria (PGPR) represents a promising strategy to alleviate salt stress in crops. However, the mechanisms by which volatile organic compounds (VOCs) from actinomycetes mitigate salinity stress remain unclear. In this study, exposure to VOCs from the halotolerant actinomycete Glutamicibacter halophytocola KLBMP 5180 significantly promotes the growth of tomato seedlings under salt stress, as evidenced by increased fresh weight, lateral root number, and chlorophyll content. Physiological analyses show that VOCs exposure reduces oxidati

MeMLO12 Impairs Cassava Resistance to Xanthomonas axonopodis pv. manihotis by Repressing Reactive Oxygen Species, Callose Deposition, and Defense Hormone Pathways.

Cassava (Manihot esculenta Crantz) is a vital food and energy crop in tropical regions, yet its yield is significantly threatened by cassava bacterial blight, caused by Xanthomonas axonopodis pv. manihotis (Xpm). Mildew resistance locus O (MLO) genes are known negative regulators of plant immunity, but their roles in cassava disease susceptibility remain unclear. In this study, we identified the MeMLO12 gene from cassava and investigated its function in response to Xpm infection. Subcellular localization revealed that MeMLO12 is specifically localized in the nucleus. Functional analysis via tr

MLN4924 enhances RSL3-induced ferroptosis sensitivity in glioblastoma via inhibiting the STAT3/GPX4 axis.

Glioblastoma (GBM) is one of the most aggressive and treatment-resistant tumors of the central nervous system, demanding innovative therapeutic strategies. Ferroptosis, an iron-driven lipid peroxidation-dependent cell death, has emerged as a potential means to overcome resistance, with glutathione peroxidase 4 (GPX4) as its key suppressor. MLN4924 (pevonedistat), a first-in-class inhibitor of neddylation currently in clinical trials, has shown strong antitumor activity across multiple malignancies. However, its role in ferroptosis regulation and the underlying molecular mechanisms remain poorl

Receptor-like kinases BIR1 and BIR3 modulate antiviral resistance by different mechanisms.

BRASSINOSTEROID INSENSITIVE1-ASSOCIATED RECEPTOR KINASE 1 (BAK1)-INTERACTING RECEPTOR-LIKE KINASE (BIR) proteins are negative regulators of cell death and defense against microbes in Arabidopsis thaliana. Here, we demonstrate that the members of the BIR family function as negative regulators of antiviral resistance in Arabidopsis. We show that during tobacco rattle virus (TRV) infection, BIR1 and BIR3 gene expression is antagonistically regulated by salicylic acid and jasmonic acid signaling pathways. BIR1 and BIR3 negatively regulate TRV resistance via distinct mechanisms. Our data indicate t

The Magnaporthe oryzae effector MoBys1 suppresses rice immunity by targeting OsCAD2 to manipulate host jasmonate and lignin metabolism.

Rice blast disease caused by Magnaporthe oryzae poses a severe threat to rice production. To counteract M. oryzae, plants synthesize jasmonate (JA) and lignin, two primary defense-related metabolites, to initiate defense programs. However, the mechanism through which M. oryzae modulates JA- and lignin-mediated plant immunity remains unclear. In this study, a novel M. oryzae effector, MoBys1, was identified as being involved in pathogenesis. Knockout of MoBys1 in M. oryzae significantly reduced its infection ability. Conversely, overexpression of MoBys1 in rice impaired the rice defense respons

CAM5, WRKY53, and TGA5 regulate defense gene expression mediated by the volatile organic compound ethyl vinyl ketone in arabidopsis.

Plants produce ethyl vinyl ketone (evk) in response to biotic stress; however, the downstream defense responses triggered by evk remain unclear. Our research utilizing reverse-transcription quantitative PCR showed that evk treatment antagonized the effects of salicylic acid (SA) and jasmonic acid (JA) and that CALMODULIN 5 (CAM5) positively regulated WRKY53 expression, which, in turn, positively regulated the expression of SA-related genes. Additionally, the GST-pulldown assay, yeast two-hybrid assays, and luciferase complementation test all suggested that CAM5 and WRKY53 physically interacted

Inhibition of NEDDylation enhances the cytostatic effect of Rohinitib on chronic lymphocytic leukemia cells.

Chronic lymphocytic leukemia (CLL) is an incurable hematologic neoplasm that primarily affects elderly individuals. Its treatment consumes a significant amount of clinical resources, a demand expected to grow due to increasing life expectancy in our society. Two characteristics of B-CLL lymphocytes that have been scarcely explored as therapeutic targets are their high RNA translation and increased ubiquitin-like post-translational modifications (UBL-PTM). Previous studies have analyzed the effects of inhibiting these two processes in B-CLL cells separately, yielding promising results. Here we

Ectopic and transient expression of VvDIR4 gene in Arabidopsis and grapes enhances resistance to anthracnose via affecting hormone signaling pathways and lignin production.

BACKGROUND: DIR (Dirigent) proteins play important roles in the biosynthesis of lignin and lignans and are involved in various processes such as plant growth, development, and stress responses. However, there is less information about VvDIR proteins in grapevine (Vitis vinifera L). RESULTS: In this study, we used bioinformatics methods to identify members of the DIR gene family in grapevine and identified 18 VvDIR genes in grapevine. These genes were classified into 5 subfamilies based on phylogenetic analysis. In promoter analysis, various plant hormones, stress, and light-responsive cis-ele

Nitrogen forms and metabolism affect plant defence to foliar and root pathogens in tomato.

Nitrogen (N) influences a myriad of physiological processes while its effects on plant defences and the underlying mechanisms are largely unknown. Here, the interaction between tomato and pathogens was examined under four N regimes (sole NO3- or mixed NO3- /NH4+ of total 1 and 7 mM N, denoting low and high N regimes, respectively) followed by inoculation with two bacterial pathogens, Pseudomonas syringae and Ralstonia solanacearum. Tomato immunity against both pathogens was generally higher under low N as well as NO3- as the sole N source. The disease susceptibility was reduced by silencing N

Investigating Proteome and Transcriptome Defense Response of Apples Induced by Yarrowia lipolytica.

A better understanding of the mode of action of postharvest biocontrol agents on fruit surfaces is critical for the advancement of successful implementation of postharvest biocontrol products. This is due to the increasing importance of biological control of postharvest diseases over chemical and other control methods. However, most of the mechanisms involved in biological control remain unknown and need to be explored. Yarrowia lipolytica significantly inhibited blue mold decay of apples caused by Penicillium expansum. The findings also demonstrated that Y. lipolytica stimulated the activitie

The green peach aphid Myzus persicae perform better on pre-infested Chinese cabbage Brassica pekinensis by enhancing host plant nutritional quality.

The green peach aphid, Myzus persicae Sulzer, is a notorious pest on vegetables, which often aggregates in high densities on crop leaves. In this study, we investigated whether M. persicae could suppress the resistance level of Chinese cabbage Brassica pekinensis. M. persicae performed better in terms of weight gain (~33% increase) and population growth (~110% increase) when feeding on previously infested (pre-infested) Chinese cabbage compared with those on non-infested plants. However, when given a choice, 64% of the aphids preferred to settle on non-infested leaves, while 29% of aphids chos

Constitutive expression of the poplar WRKY transcription factor PtoWRKY60 enhances resistance to Dothiorella gregaria Sacc. in transgenic plants.

WRKY proteins are involved in various physiological processes in plants, especially in coping with diverse biotic and abiotic stresses. However, limited information is available on the roles of specific WRKY transcription factors in poplar defense. In this study, we reported the characterization of PtoWRKY60, a Group IIa WRKY member, from Populus tomentosa Carr. The gene expression profile of PtoWRKY60 in various tissues showed that it significantly accumulated in old leaves. Phylogenetic analyses revealed that PtoWRKY60 had a close relationship with AtWRKY18, AtWRKY40 and AtWRKY60. PtoWRKY60

MAPK-dependent JA and SA signalling in Nicotiana attenuata affects plant growth and fitness during competition with conspecifics.

BACKGROUND: Induced defense responses to herbivores are generally believed to have evolved as cost-saving strategies that defer the fitness costs of defense metabolism until these defenses are needed. The fitness costs of jasmonate (JA)-mediated defenses have been well documented. Those of the early signaling units mediating induced resistance to herbivores have yet to be examined. Early signaling components that mediate herbivore-induced defense responses in Nicotiana attenuata, have been well characterized and here we examine their growth and fitness costs during competition with conspecific

Interaction of plant cell signaling molecules, salicylic acid and jasmonic acid, with the mitochondria of Helicoverpa armigera.

The cotton bollworm, Helicoverpa armigera is a polyphagous pest in Asia, Africa, and the Mediterranean Europe. Salicylic acid (SA) and jasmonic acid (JA) are the cell signaling molecules produced in response to insect attack in plants. The effect of these signaling molecules was investigated on the oxidative phosphorylation and oxidative stress of H. armigera. SA significantly inhibited the state III and state IV respiration, respiratory control index (RCI), respiratory complexes I and II, induced mitochondrial swelling, and cytochrome c release in vitro. Under in vivo conditions, SA induced s

Botrytis cinerea manipulates the antagonistic effects between immune pathways to promote disease development in tomato.

Plants have evolved sophisticated mechanisms to sense and respond to pathogen attacks. Resistance against necrotrophic pathogens generally requires the activation of the jasmonic acid (JA) signaling pathway, whereas the salicylic acid (SA) signaling pathway is mainly activated against biotrophic pathogens. SA can antagonize JA signaling and vice versa. Here, we report that the necrotrophic pathogen Botrytis cinerea exploits this antagonism as a strategy to cause disease development. We show that B. cinerea produces an exopolysaccharide, which acts as an elicitor of the SA pathway. In turn, the

Elicitor and Fusarium-induced expression of NPR1-like genes in banana.

The non-expressor of pathogenesis-related genes 1 (NPR1) is an essential positive regulator of salicylic acid (SA)-induced pathogenesis-related (PR) gene expression and systemic acquired resistance (SAR). Two novel full length NPR1-like genes; MNPR1A and MNPR1B, were isolated from banana by application of the PCR and rapid amplification of cDNA ends (RACE) techniques. The two identified MNPR1 sequences differed greatly in their expression profile using quantitative real time (qRT)-PCR following either elicitor or Fusarium oxysporum Schlecht f. sp. cubense (Smith) Snyd (Foc) treatment.
