The Opisthorchis viverrini genome provides insights into life in the bile duct.

Opisthorchiasis is a neglected, tropical disease caused by the carcinogenic Asian liver fluke, Opisthorchis viverrini. This hepatobiliary disease is linked to malignant cancer (cholangiocarcinoma, CCA) and affects millions of people in Asia. No vaccine is available, and only one drug (praziquantel) is used against the parasite. Little is known about O. viverrini biology and the diseases that it causes. Here we characterize the draft genome (634.5 Mb) and transcriptomes of O. viverrini, elucidate how this fluke survives in the hostile environment within the bile duct and show that metabolic pat

Evidences of increasing risk of dirofilarioses in southern Italy.

Given the spread of Aedes albopictus from northern to southern Italy, and the lack of updated data on Dirofilaria infections, this study was carried out to assess the infection risk for dogs and cats in Apulia region. During a 2-year study, 175 A. albopictus female specimens and samples of blood from 427 dogs (309 privately owned dogs and 118 shelter dogs) and 12 cats were collected. All blood samples were subjected to a modified Knott method, to a test for the detection of circulating Dirofilaria immitis antigen, and to a Dirofilaria species-specific real-time PCR for the simultaneous detecti

Assessment of a DNA vaccine encoding an anchored-glycosylphosphatidylinositol tegumental antigen complexed to protamine sulphate on immunoprotection against murine schistosomiasis.

Protamine sulphate/DNA complexes have been shown to protect DNA from DNase digestion in a lipid system for gene transfer. A DNA-based vaccine complexed to protamine sulphate was used to induce an immune response against Schistosoma mansoni anchored-glycosylphosphatidylinositol tegumental antigen in BALB/c mice. The protection elicited ranged from 33 to 44%. The spectrum of the elicited immune response induced by the vaccine formulation without protamine was characterized by a high level of IgG (IgG1> IgG2a). Protamine sulphate added to the DNA vaccine formulation retained the green fluorescent

Recent developments in research into the Cyathostominae and Anoplocephala perfoliata.

Intestinal helminths are an important cause of equine disease. Of these parasites, the Cyathostominae are the commonest group that infect horses. These nematodes consist of a complex tribe of 51 species, although individual horses tend to harbour 10 or so common species, in addition to a few rarer species. The Cyathostominae can be extremely pathogenic, and high levels of infection result in clinical symptoms ranging from chronic weight loss to colic, diarrhoea and death. As part of their life cycle, immature cyathostomins penetrate the large intestinal wall, where they can enter a state of in

Trematode diversity in southern Africa: metacercariae of the Clinostomidae and Cryptogonimidae in intermediate freshwater fish hosts.

The freshwater fish fauna of southern Africa is highly diverse; however, the magnitude of parasitic species they host is unevenly known. The region's documented adult trematode fish fauna is sparse, while the opposite is evident for intermediate trematode stages. Perceived difficulty in identification of underdeveloped stages lead to the exclusion of reporting metacercariae or lack either morphological or molecular data resulting in a depauperate comparative molecular data repository for species of the region and Africa as a whole. In an effort to address the morphological and molecular data v

The Multifaceted Roles of STAT6: Implications for Precision Immunotherapeutics.

STAT6, located on chromosome 12, encodes a transcription factor whose SH2 domain is central to its activation. While classically a Th2 mediator, aberrant STAT6 activation is observed in diverse malignancies. This review focuses on the molecular and functional landscape of STAT6, primarily in cancer and tumor immunology, while also surveying its emerging roles in allergy, infection, and autoimmunity to illustrate context-dependent signaling and identify areas for future investigation. In tumors, STAT6 fosters immunosuppression by polarizing macrophages toward M2 phenotypes and upregulates check

Paleoparasitological analysis on animal-origin coprolites from the Moche archaeological site Licapa II, Chicama Valley, Peru (450-900 AD).

Paleoparasitological research has allowed the recovery and identification of parasite structures like helminth eggs, protozoan oocysts, arthropod remains, as well as parasite ancient DNA molecules preserved in human and animal-origin archaeological material from Peruvian contexts. However, these studies are still scarce in the region despite its potential to trace parasite infections and diseases through history, and to help understand ecological relationships in the past. Here, we examined 36 coprolites from rodents and South American camelids recovered from a test pit, through light microsco

Diomedenema dinarctos n. sp. (Spiruromorpha: Desmidocercidae), a new species of lung nematode in Fiordland crested penguins (Eudyptes pachyrhynchus; Sphenisciformes), from South Island, New Zealand.

Fiordland crested penguins Eudyptes pachyrhynchus Gray are thick-billed, crested penguins endemic to New Zealand that breed in small colonies in inaccessible places. They are assessed as Vulnerable by the IUCN. This is the first report of helminth parasites from this penguin species. Herein a new species of Diomedenema is described (Spiruromorpha, Desmidocercidae) from the lungs of Fiordland crested penguins. Diomedenema dinarctos n. sp. has only two congeners and is closest morphologically to D. tavaresi. It differs from D. tavaresi in overall larger size, longer buccal cavity and oesophagus,

Molecular paleoparasitological hybridization approach as effective tool for diagnosing human intestinal parasites from scarce archaeological remains.

Paleoparasitology is the science that uses parasitological techniques for diagnosing parasitic diseases in the past. Advances in molecular biology brought new insights into this field allowing the study of archaeological material. However, due to technical limitations a proper diagnosis and confirmation of the presence of parasites is not always possible, especially in scarce and degraded archaeological remains. In this study, we developed a Molecular Paleoparasitological Hybridization (MPH) approach using ancient DNA (aDNA) hybridization to confirm and complement paleoparasitological diagnosi

Characterisation of fascioliasis lymnaeid intermediate hosts from Chile by DNA sequencing, with emphasis on Lymnaea viator and Galba truncatula.

In South America, Fasciola hepatica infection poses serious health problems in both humans and livestock. In Chile, the medical impact appears yearly stable and mainly concentrated in central regions, where the veterinary problem is highlighted by higher animal prevalences. Studies were undertaken by rDNA ITS-2 and ITS-1 and mtDNA cox1 sequencing to clarify the specific status of the lymnaeids, their geographical distribution and fascioliasis transmission capacity in Chile, by comparison with other American countries and continents. Results change the lymnaeid scenario known so far. The lymnae

Molecular evidence of Trichobilharzia franki Müller and Kimmig, 1994 (Digenea: Schistosomatidae) in Radix auricularia from Central Italy.

The molecular approach was used to identify the first focus of human cercarial dermatitis occurring during the spring-summer season in Vico Lake, a Natural Reserve Area in Central Italy. Sequences of 586 bp length of the partial region of 18S and ITS-1 ribosomal DNA from cercariae were obtained. They were compared with extant GenBank sequences of Trichobilharzia. Accordingly, the ocellate furcocercariae were identified as belonging to the species Trichobilharzia franki Müller and Kimmig, 1994. Sequences of the 18S (712 bp) and ITS-2 (218 bp) rDNA gene of snails belonging to the family Lymnaeid
