In schistosomiasis, a parasitic disease caused by helminths, the parasite eggs induce a T helper 2 cell (T(H)2) response in the host. Here, the specific role of human monocyte-derived dendritic cells (DCs) in initiation and polarization of the egg-specific T cell responses was examined. We demonstrate that immature DCs (iDCs) pulsed with schistosome soluble egg antigens (SEA) do not show an increase in expression of co-stimulatory molecules or cytokines, indicating that no conventional maturation was induced. The ability of SEA to affect the Toll-like receptor (TLR) induced maturation of iDCs

The Fragile-X mental retardation syndrome is the leading form of inherited mental retardation. Dendritic analysis in a mouse model (FraX) found abnormal pruning in somatosensory cortex. To further characterize dendritic abnormalities and assess their occurrence in other brain regions, we examined mitral cells in FraX mice olfactory bulbs. FraX mice exhibited dendritic abnormalities consistent with somatosensory cortex, suggesting that deficient pruning is found in multiple brain regions.

BACKGROUND: In fast firing, parvalbumin (PV)-expressing (Pvalb) interneurons, PV acts as an intracellular Ca2+ signal modulator with slow-on … wild-type neurons. The increase in dendrite length and branching, as well as thickness of proximal dendrites of selected PV-/- Pvalb neurons is likely the result of the observed increased density and length of mitochondria in these PV-/- Pvalb neuron dendrites. The increased branching and soma size directly linked to the absence of PV is assumed to contrib …

Both endocytic uptake and viral fusion can lead to human immunodeficiency virus type 1 (HIV-1) transfer to CD4+ lymphocytes, either through directional regurgitation (infectious transfer in trans [I-IT]) or through de novo viral production in dendritic cells (DCs) resulting in a second-phase transfer to CD4+ lymphocytes (infectious second-phase transfer [I-SPT]). We have evaluated in immature monocyte-derived DCs both pathways of transfer with regard to their susceptibilities to being blocked by potential microbicidal compounds, including cyanovirin (CNV); the plant lectins Hippeastrum hybrid

Analysis of serial ultrathin sections of the human aortic intima detected a new cell yet to be described in the literature. These cells, which we have designated Vascular Dendritic Cells, appeared in contact with each other and with other intimal cells. Vascular dendiritic cells are characterised by ultrastructural features similar to those of dendritic cells, including a well developed smooth endoplasmic reticulum and the presence of several processes which were 3-5 or more times in excess of the size of the cell body. In areas of the normal aorta resistant to atherosclerosis, vascular dendri

Clonal histiocytopathy involving non-Langerhans mature, monocyte-derived dendritic cells is a poorly characterized cutaneous manifestation of chronic myeloproliferative diseases. We describe the clinical and histopathological features of a well-differentiated histiocytopathy composed of mature, CD1a-negative, S100-negative, langerin-negative monocyte-derived dendritic cells, proposing it as a potential dermatologic marker for acute and/or chronic myeloproliferative diseases. This case series includes two cases and a review of eight others from the Weill Cornell Medicine Dermatopathology databa

BACKGROUD: Atopic dermatitis (AD) is a chronic, inflammatory skin disease characterized by intense pruritus and xerosis. Dendritic cells (DC) play an essential role in tissue inflammation in atopic dermatitis (AD) skin, especially the inflammatory epidermal dendritic cells (IDEC), a particular subset of myeloid dendritic cells (mDC). The aim of the present study was to assess the phenotype and function of mDC and circulating IDEC-like in peripheral blood mononuclear cells (PBMC) of adults with AD.
METHODS: We selected 21 AD patients and 21 non-AD controls, age and gender matched. Expressions

Aqueous zinc(Zn)-metal batteries (AZMBs) have attracted significant attention owing to their high safety, economy, and potential applications in stationary energy storage systems. However, AZMBs still suffer from low reversibility and short lifespan of the Zn anode, resulting from the instability of the Zn/electrolyte interface. Herein, a hybrid interface combining a solid electrolyte interphase (SEI) and an electrostatic shield layer was introduced into the AZMBs through a simple electrolyte-regulating strategy. This hybrid interface simultaneously establishes a Zn phosphate (Zn-P-O) based SE

The functional specificity of tubulin isotypes has been demonstrated by various neurological diseases caused by an increasing number of mutations in tubulin isotypes. TUBA8 is specifically localized in cerebellar Purkinje cells, which exhibit the most elaborate dendritic trees in the central nervous system. However, the role and related molecular mechanism of TUBA8 in regulating neuronal dendritic morphology remain poorly understood. Here, we report that TUBA8 is required for neuronal dendrite development. As the most divergent member in α-tubulin isotypes, the expression of TUBA8 in Purkinje

Antigen presentation within solid tumours presents a significant clinical challenge for systemic immune activation. Although radiotherapy or chemotherapy can generate tumour antigens by destroying tumour cells, the insufficient number and impaired function of dendritic cells within solid tumours compromises effective antigen presentation. Here we present plant-derived nanoparticles coated with nanohydroxyapatite that promote dendritic cell activation and amplify the production of dendritic-cell-derived antigen-presenting vesicles in vivo through the modulation of intracellular immune conversio

Abnormal deposition of the microtubule-associated protein tau has long been associated with neurodegenerative diseases. While spine loss and neuronal death are hallmarks of tauopathy, how pathological tau affects synaptic activity in vivo and whether functional properties of individual synapses dictate the survival fate of dendritic spines remain elusive. Here we examined the visual response properties of dendrites and spines of layer 2/3 primary visual cortical neurons, using longitudinal two-photon calcium imaging in P301S mouse model of tauopathy. We found that neuronal outputs in tau mutan…

Dendritic cells (DCs) are essential immune system sentinels, playing a key role in activating naïve T cells against leukemic cells. They have an immunomodulatory function, promoting an immune response and, at the same time, regulating immune tolerance to avoid excessive reactions. Plasmacytoid dendritic cell (pDC) neoplasms are a complex group of disorders, and among the subtypes, acute myeloid leukemia (AML) with pDC dermatosis (AML-pDC) stands out. In this context, pDCs can play an immunomodulatory role by inhibiting or promoting immune responses and acting as tumor progenitors since, in som

Neurons are subject to extensive developmental regulation to ensure precise subtype-specific morphologies that are intimately tied to their function. Starburst amacrine cells (SACs) in the mammalian retina have a highly stereotyped, radially symmetric dendritic arbor that is essential for their role in direction-selective circuits in the retina. We show that PTEN, the primary negative regulator of the PI3K-AKT-mTOR pathway that is highly implicated in neurodevelopmental disorders, regulates SAC morphology in a cell-autonomous manner. Pten-deficient mouse SACs show a nearly twofold increase in

This study explored the immunomodulatory properties of the probiotic strains Lacticaseibacillus rhamnosus LR-32 and Lactobacillus acidophilus LA-5 by evaluating their effects on human dendritic cell (DC) responses. Immature DCs were obtained from human CD14⁺ monocytes and challenged with E. coli LPS and/or probiotic lactobacilli at a DC ratio of 1:10. Relative transcription of inflammatory mediators and pattern-recognition receptors was assessed by RT-qPCR. Cytokine production and surface marker expression were evaluated by ELISA and flow cytometry, respectively. DC function was further assess

Dendritic cells (DCs), the most proficient antigen-presenting cells, bridge innate and adaptive immunity and are critical for anti-cancer immune surveillance. Their function is precisely regulated by protein tyrosine kinases (PTKs), which integrate signals from external stimuli and internal cellular stress to control DC maturation, migration, and antigen presentation. This review systematically synthesizes current knowledge on PTK roles in DC-mediated anti-tumor immunity, with a focused analysis of their differential expression and function across human and mouse DC subsets-including conventio

Personalized vaccines provide the advantage of patient-specific antigen selection to optimize immune responses, a strategy extensively explored in oncology through neoantigen-targeted peptide, mRNA, and dendritic cell platforms. Peptide vaccines provide simplicity and stability though often elicit limited cytotoxic T-cell responses. What is more, mRNA vaccines lead to rapid, multiplexed neoantigen delivery, endogenous antigen processing and eventually improved immunogenic coverage. Dendritic cell-based vaccines have the potency to prime potent T-cells although this technology requires labor-in

Dendritic cells (DCs) comprise a heterogeneous population of specialized antigen-presenting cells that play central roles in the initiation and coordination of innate and adaptive immune responses. Recent advances in single-cell technologies and mechanistic studies have substantially expanded our understanding of DC subset heterogeneity, developmental trajectories, and functional specialization. Within the tumor microenvironment (TME), DCs are essential for the priming and maintenance of antitumor T cell immunity; however, they are also vulnerable to tumor-derived signals that drive dysfunctio
