Epigenetic Alterations Beyond CpG Islands in Periodontitis: In Silico Study of DNA Methylation Data.

OBJECTIVES: To classify differentially methylated CpG sites in periodontitis based on methylation shift direction and location relative to CpG islands. MATERIAL AND METHODS: We integrated DNA methylation data from 65 gingival samples (31 periodontitis, 34 healthy) from three GEO datasets. Preprocessing included normalization using R pipelines. Differential DNA methylation analysis was performed with the limma package. CpG sites (adjusted p < 0.05) were further evaluated using an exploratory threshold-crossing framework based on group-mean β-values (cutoff = 0.5) identifying Control low → Peri

Gender differences in psychophysically determined maximum acceptable weights and forces for industrial workers observed after twenty years.

PURPOSE: In the year 1991, manual materials handling guidelines were published by Liberty Mutual Research Institute for Safety. In these guidelines, maximum acceptable weights (MAWs) and forces (MAFs) for lifting, lowering, pushing, pulling, and carrying were derived from studies conducted in a 20 year span before the above publication date. The question is whether the present generation of workers has retained the same gender differences and absolute values in psychophysically determined MAWs and MAFs as those reflected in the guideline. METHODS: Twenty-four female industrial workers perform

Quantitative sensory testing of peripheral thresholds in patients with lifelong premature ejaculation: a case-controlled study.

INTRODUCTION: The main functional factors related to lifelong premature ejaculation (PE) etiology have been suggested to be penile hypersensitivity, greater cortical penile representation, and disturbance of central serotoninergic neurotransmission. AIMS: To quantitatively assess penile sensory thresholds in European Caucasian patients with lifelong PE using the Genito-Sensory Analyzer (GSA, Medoc, Ramat Yishai, Israel) as compared with those of an age-comparable sample of volunteers without any ejaculatory compliant. METHODS: Forty-two consecutive right-handed, fully potent patients with li

Air pressure-sensing ability of the middle ear-Investigation of sensing regions and appropriate measurement conditions.

OBJECTIVE: To study the association between ear diseases and the ability of the middle ear to sense air pressure changes, we elucidated the appropriate conditions for measurements and confirmed the pressure-sensing regions while quantitatively assessing the pressure-sensing ability of normal ears and of ears with traumatic perforation of the tympanic membrane. METHODS: In 318 normal ears (159 volunteers), an air pressure of 40daPas(-1) was loaded on to the external auditory canals to measure the minimum pressure sensed by subjects as a result of air pressure changes. The minimum pressure was

EMG analysis of the thenar muscles as a model for EMG-triggered larynx stimulation.

Paralysis of one or both sides of the larynx musculature compromises breathing and speech function. Currently there is no surgical remedy to restore adequate function of the larynx. A plausible alternative solution is triggered electrical stimulation of the paralysed larynx site using a laryngeal pacemaker. Triggering of the pacemaker succeeds via constant EMG measurement of the muscle activity of the healthy larynx side. The EMG data analysis described in this work is one possible approach for regulating pacemaker triggering. In this study we used EMG data from the thenar muscles as a model t

Trigeminal sensitization and desensitization in the nasal cavity: a study of cross interactions.

Chemical irritation in the human nasal cavity is poorly documented. In this field, an important issue concerns the differential responses produced by successive stimulation. Repeated identical chemical irritant stimuli can produce increases or decreases in responses (two phenomena known as self-sensitization or self-desensitization). In the same way, different molecules can interact and produce cross-sensitization or cross-desensitization. The aim of this study was to contribute to this question using two specific molecules, acetic acid (AA) and allyl isothiocyanate (AIC). As the self-sensitiz

Altered cortical excitability in obsessive-compulsive disorder.

OBJECTIVE: To assess cortical inhibitory and excitatory mechanisms in obsessive-compulsive disorder (OCD). BACKGROUND: Transcranial magnetic stimulation (TMS) studies have found decreased neuronal inhibition and a reduced cortical silent period in the primary motor area in Tourette's syndrome, focal dystonia, and other disorders believed to involve dysfunction of subcortical structures, including the basal ganglia. Dysfunction of the basal ganglia and linked regions also has been implicated in OCD, which has significant clinical and familial overlap with tic disorders. METHODS: We applied th

Are recognition deficits following occipital lobe TMS explained by raised detection thresholds?

It is known that transcranial magnetic stimulation (TMS) administered over the occipital pole suppresses recognition of visual objects. Our aim was to ascertain whether this suppression can be interpreted as a change in visual contrast threshold. Four subjects detected the orientation of an U-shaped hook flashed for 21 ms. Under control conditions, mean contrast threshold was found at 0.88 log units Weber contrast. Thresholds were raised if TMS was applied 40-200 ms after the visual stimulus. Maximum elevation was 1.67 log units under TMS at 120 ms stimulus onset asynchrony. This phenomenon ca

In vitro classical conditioning of abducens nerve discharge in turtles.

In vitro classical conditioning of abducens nerve activity was performed using an isolated turtle brainstem-cerebellum preparation by direct stimulation of the cranial nerves. Using a delayed training procedure, the in vitro preparation was presented with paired stimuli consisting of a 1 sec train stimulus applied to the auditory nerve (CS), which immediately preceded a single shock US applied to the trigeminal nerve. Conditioned and unconditioned responses were recorded in the ipsilateral abducens nerve. Acquisition exhibited a positive slope of conditioned responding in 60% of the preparatio

Anesthetic blockade of the dorsolateral funiculus enhances evoked activity of spinal cord dorsal horn neurons.

1. A previous study of cat lumbar dorsal horn neurons found reduced responsiveness to A-fiber stimulation 1.5-12 h after thoracic dorsolateral funiculus (DLF) lesions. The present study was undertaken to determine whether this was due to the loss of descending activity or to factors specifically associated with injury by examining the response properties of dorsal horn cells before and during lidocaine blockade of the ipsilateral DLF. Electric shocks applied to the dorsal columns were used to search for dorsal horn cells. Noxious and nonnoxious cutaneous mechanical stimuli and graded electrica

Specific bronchial reactivity to toluene diisocyanate: dose-response relationship.

20 patients with toluene diisocyanate (TDI)-induced asthma were examined in order to assess their threshold of response to TDI during specific bronchial provocative tests (BPT). Specific bronchial hyperresponsiveness was evaluated by performing, on different days, specific BPT with increasing concentrations of TDI until a positive response was obtained; the threshold of response to TDI (low: 0.02-0.05 ppm; moderate: 0.1 ppm; high: 0.2-0.25 ppm) and the pattern of positive response were evaluated in comparison with some clinical features of the disease. The threshold of airway response to TDI w

Fiberoptic endoscopic evaluation of swallowing with sensory testing (FEESST) in healthy controls.

The purpose of this study was to introduce a new method of bedside assessment of both the motor and sensory components of swallowing called fiberoptic endoscopic evaluation of swallowing with sensory testing (FEESST). This approach combines the established bedside endoscopic swallowing evaluation with a more recently described technique that allows objective determination of laryngopharyngeal (LP) sensory discrimination thresholds by delivering air pulse stimuli to the mucosa innervated by the superior laryngeal nerve via a flexible endoscope. A prospective study was conducted of FEESST in 20

COVID-19 Incidence and Age Eligibility for Elementary School.

IMPORTANCE: Understanding the role of school attendance on transmission of SARS-CoV-2 among children is of importance for responding to future epidemics. Estimating discontinuities in outcomes by age of eligibility for school attendance has been used to examine associations between school attendance and a variety of outcomes, but has yet to be applied to describe associations between school attendance and communicable disease transmission. OBJECTIVE: To estimate the association between eligibility for elementary school and COVID-19 incidence. DESIGN, SETTING, AND PARTICIPANTS: This case seri

Using the Hounsfield Units Derived From Computed Tomography (CT) Scans to Differentiate Between the Subtypes of Allergic Fungal Rhinosinusitis: A Retrospective Study.

Background Allergic fungal sinusitis (AFS) is an inflammatory condition, often diagnosed using computed tomography (CT) scans, where Hounsfield units (HU) serve as a critical metric. However, the diagnostic process can be challenging due to the ambiguous patterns of sinus secretions. This study evaluates whether the HU measurements from preoperative CT scans can reliably differentiate between the subtypes of AFS and other chronic rhinosinusitis (CRS) entities by correlating these values with histopathological findings. Patients and methods A retrospective analysis was conducted on 120 patients

Printed MoS2 memristive nanosheet networks for spiking neurons with multi-order complexity.

Artificial neurons that reproduce the rich dynamical behaviour of biological spiking are essential for neuromorphic hardware and biohybrid interfaces, yet scalable solution-processed devices with physiologically relevant spiking characteristics remain elusive. Here we demonstrate aerosol-jet-printed memristive networks of MoS2 nanosheets that exhibit thermally activated filamentary switching and snap-back negative differential resistance, enabling volatile threshold switching in fully printed graphene/MoS2/graphene devices on flexible substrates. Mathematical modelling of the effects of public health educational campaigns on the transmission dynamics of Taenia solium Taeniasis.

Taenia solium Taeniasis is a zoonotic intestinal infection that poses a significant threat to public health and imposes a substantial economic burden on developing regions, particularly in Sub-Saharan Africa. This study formulates and rigorously analyses a deterministic mathematical model, comprised of a system of non-linear ordinary differential equations, to investigate the transmission dynamics of the parasite under the influence of public health education campaigns. Using the next-generation matrix method, we derive the effective reproduction number, R e d , as the fundamental threshold

Novel transcripts of EMT driving the malignant transformation of oral submucous fibrosis.

Oral submucous fibrosis (OSF) is a chronic, progressive, and fibrotic condition of the oral mucosa that carries an elevated risk of malignant transformation. We aimed to identify and validate novel genes associated with the regulation of epithelial-to-mesenchymal transition (EMT) in OSF. Genes regulating EMT were identified through differential gene expression analysis, using a LogFC threshold of -1 and + 1 and a padj value < 0.05, based on data from GEO datasets and the TCGA-HNSC datasets. The curated EMT genes were correlated with functional cancer states and subjected to clustering to ident
