Evaluation of squeeze-induced somnolence in neonatal foals.

OBJECTIVE: To test the hypothesis that application of a rope restraint device would result in behavioral, electroencephalographic, and humoral changes consistent with sleep and analgesia in neonatal foals. ANIMALS: 8 healthy neonatal foals. PROCEDURES: Following acclimatization to experimental conditions, each foal underwent a series of assessments before and during or at the end of a period of restraint via application of a restraint device (soft linen rope). Assessments included measurements of heart and respiratory rates, rectal temperature, and circulating β-endorphin and steroid hormone

Pharmacogenetic treatments for drug addiction: alcohol and opiates.

AIMS: Psychiatric pharmacogenetics involves the use of genetic tests that can predict the effectiveness of treatments for individual patients with mental illness such as drug dependence. This review aims to cover these developments in the pharmacotherapy of alcohol and opiates, two addictive drugs for which we have the majority of our FDA approved pharmacotherapies. METHODS: We conducted a literature review using Medline searching terms related to these two drugs and their pharmacotherapies crossed with related genetic studies. RESULTS: Alcohol's physiological and subjective effects are asso

Role of opioid antagonists in the treatment of women with glucoregulation abnormalities.

Beta-endorphin were detected in the endocrine pancreas and seem able to influence insulin and glucagon release. Hence, endogenous opioids could have a role in glucoregulation and in the pathogenesis of obesity beyond the previously detected effects on appetite. Metabolic abnormalities, such as hyperinsulinemia, insulin-resistance and obesity, are common features of polycystic ovary syndrome (PCOS), and seem to have a pathogenetic role in this disorder. A link between opioids and PCOS-related hyperinsulinism is suggested by the finding of altered central opioid tone and elevated beta-endorphins…

Enhancement of natural immunity seen after voluntary exercise in rats. Role of central opioid receptors.

Chronic voluntary exercise in wheels for 5 weeks in spontaneously hypertensive rats (SHR) augments in vivo natural killer (NK) cell cytotoxicity. Endogenous beta-endorphin is increased in cerebrospinal fluid after voluntary exercise in rats and we have recently shown that beta-endorphin administered i.c.v. augments NK cell mediated cytotoxicity in vivo in a similar way as chronic voluntary exercise. We have now further investigated the involvement of central opioid systems in the exercise-induced augmentation in natural immunity. Exercise consisted of voluntary running in wheels for 5 weeks. I

Changes in the levels of several endogenous opioid peptides in dog cerebrospinal fluid following morphine administration.

The cisterna magna of dogs anesthetized with sodium Surital was fitted with a cannula, and cerebrospinal fluid (CSF) was withdrawn before (control) and one hour after the s.c. injection of 10 mg/kg of morphine sulfate (morphine). The CSF from control and morphine-treated dogs was purified initially by gel filtration. Each fraction was submitted to opiate bioassay procedures, followed by high performance liquid chromatography (HPLC) purification on a mu-Bondapak C18 column. Two of the CSF fractions from HPLC purification showed greater opiate-like activity after morphine treatment than that in

Transient hypoalgesia under physical exercise--relation to silent ischaemia and implications for cardiac rehabilitation.

A series of studies with humans as well as experiments carried out on animals could show that physical exercise leads to temporary hypoalgesia. Reduced sensitivity to pain is not only demonstrable after long-distance exercise (such as marathon run) but also after intensive physical exercise on a laboratory ergometer. Pain threshold elevation is most pronounced during maximal exertion, but hypoalgesia remains present also after exercise is stopped demonstrating that a systemic analgetic effect is induced by the exercise process. Pre-exercise pain threshold level is returned to approximately 60

Evidence for altered opioid activity in patients with cancer.

Endogenous opioid peptides have been shown to be involved in the regulation of tumour growth. At present, however, no data are available about the secretion of opioid peptides in cancer patients. To draw some preliminary conclusions on opioid brain function in human neoplasms, we evaluated hypophyseal hormone responses to the administration of a met-enkephalin analogue, FK 33-824. The study included 14 patients affected by early or advanced neoplastic disease, 12 healthy subjects and 7 patients with a chronic medical illness other than cancer. FK 33-824 was given intravenously at a dose of 0.3

Restraint stress decreases afternoon plasma prolactin levels in female rats. Influence of neural antagonists and agonists on restraint-induced changes in plasma prolactin and corticosterone.

Female Sprague-Dawley rats were ovariectomized, given estrogen, and blood samples were obtained via an atrial catheter in the afternoon during the prolactin (PRL) surge. Restraint stress applied at 16.00 h and continued for 3 h resulted in marked decrease in plasma prolactin (PRL) and an increase in plasma corticosterone (B). The neural mechanism(s) involved in the plasma PRL decrease to restraint stress in the afternoon were examined using neural agonists and antagonists. The administration of pimozide, a dopamine antagonist, increased plasma PRL and completely prevented the restraint-induced

Corticotropin-releasing factor (ovine) and vasopressin exert a synergistic effect on adrenocorticotropin release in man.

The effects of ovine CRF, lysine vasopressin (LVP), and their interrelationships, and rat hypothalamic extract (HME), on ACTH and beta-endorphin release by human pituitary tumor cells from two patients with Nelson's syndrome and one with Cushing's disease and on ACTH and cortisol secretion in vivo were studied. In cultured pituitary tumor cells, both LVP and CRF greatly stimulated ACTH and beta-endorphin release at maximally active concentrations of 0.1 microM and 10 nM, respectively. At these concentrations, the combination of the two substances had an additive or synergistic effect on hormon

Effects of pro-opiomelanocortin-derived peptides on plasma levels of glucagon, insulin and glucose.

Pro-opiomelanocortin (POMC) is a prohormone for several peptides including corticotropin, melanocyte stimulating hormones and beta-endorphin. POMC-derived peptides have been demonstrated in many tissues, including the hypothalamus and the endocrine pancreas, which play important roles in the control of plasma levels of glucagon, insulin and glucose. This article reviews the present knowledge concerning in vitro and in vivo effects of POMC-derived peptides on glucagon, insulin and glucose levels involving several possible mechanisms: direct effects on the endocrine pancreas (including endocrine

Response to low-dose pulsatile cortisol in Addison's disease with suspected corticotropinoma.

A 65 year old woman with long-standing Addison's disease treated with oral glucocorticoid and mineralocorticoid replacement had persistently high ACTH levels, inadequate suppression of ACTH on low-dose dexamethasone, sellar enlargement, and pigmentation, and thus resembled patients alleged to develop corticotropinomas while on oral replacement for adrenal insufficiency. Since animal studies suggested that rapid rises of corticosteroids within the physiologic range can inhibit ACTH release, we administered brief infusions of cortisol every three hours with total daily dose equal to her chronic

Arginine vasopressin deficient Brattleboro rats fail to develop tolerance to the hypothermic effects of ethanol.

We have tested the hypothesis that animals with reduced levels of arginine vasopressin (AVP) would show reduced tolerance to ethanol. Brattleboro rats either heterozygous or homozygous for the diabetes insipidus (DI) trait and normal Sprague-Dawley rats were exposed to ethanol vapor for 21 days. Two days later, tolerance was evaluated by monitoring body temperature reductions after intraperitoneal injection of 2 g/kg (20% w/v) ethanol. Under the same conditions of chronic ethanol exposure, Sprague-Dawley rats, but not Brattleboro rats, displayed tolerance to the hypothermic effects of intraper

Immuno-derived opioids as mediators of the immuno-enhancing and anti-stress action of melatonin.

The pineal neurohormone melatonin was shown to stimulate the release of opioid peptides from activated CD4+, T lymphocytes. These immuno-derived opioids or "lymphomorphins" crossreact with anti-beta-endorphin and anti-metenkephalin antisera, bind to opioid receptors in the thymus and are the mediators of the immunoenhancing and anti-stress action of endogenous and/or exogenous melatonin. These findings proved the existence of a novel immunoneuroendocrine physiological mechanism which may be related for the long range maintenance of the immune homeostasis in spite of the unavoidable stress and/

[Infantile apnea. The etiopathogenic aspects. I].

The paper reports on etiopathogenesis aspects of apnea, on the basis of the literature data. After showing the importance of the problem and the definitions accepted, the author presents the physiological and physiopathological framework of the breathing control. The paper deals with apnea and periodic breathing associated with evident diseases and procedures, and idiopathic apnea. The central, obstructive and mixed types of apnea and their characteristics are described. The final part of the paper dwells on other mechanisms involved in apnea: gastroesophageal flux, endorphines, increased seru

Bulimic vomiting alters pain tolerance and mood.

Bulimia, a disorder of episodic binging and purging, remains without a known etiology. A case report is presented of a patient who attributed bulimic episodes to efforts at inducing euphoria. Experimental pain tolerance was increased by bulimic vomiting, blocked by naloxone, but not by saline. Vomiting was also associated with falls in depression and anxiety. Plasma ACTH and cortisol, putative markers for beta-endorphin, also rose following vomiting. It is hypothesized that in some bulimics, the disorder arises by virtue of an addiction to one's own internally released endogenous opioid peptid

Interaction of amphetamine and naloxone in feeding behavior in guinea pigs.

Chronic naloxone treatment of schedule-fed guinea pigs caused an initial anorexia followed by a resumption of food and water intake, despite the continued presence of the narcotic antagonist. During this state of chronic naloxone treatment, amphetamine exerted a similar degree of anorexia to that in animals not pretreated with naloxone. In contrast, amphetamine tolerant guinea pigs exhibited supersensitivity to naloxone with respect to feeding behavior. Since chronic amphetamine exposure results in an increased level of beta-endorphin in the hypothalamus it is suggested that an enhanced endorp
