Fine tuning of DNA repair in transcribed genes: mechanisms, prevalence and consequences.

Cells fine-tune their DNA repair, selecting some regions of the genome in preference to others. In the paradigm case, excision of UV-induced pyrimidine dimers in mammalian cells, repair is concentrated in transcribed genes, especially in the transcribed strand. This is due both to chromatin structure being looser in transcribing domains, allowing more rapid repair, and to repair enzymes being coupled to RNA polymerases stalled at damage sites; possibly other factors are also involved. Some repair-defective diseases may involve repair-transcription coupling: three candidate genes have been sugg

Human xeroderma pigmentosum group D gene encodes a DNA helicase.

Xeroderma pigmentosum (XP), a genetically heterogeneous human disease, results from a defect in nucleotide excision repair of ultraviolet-damaged DNA. XP patients are extremely sensitive to sunlight and suffer from a high incidence of skin cancers. Cell fusion studies have identified seven XP complementation groups, A-G. Group D is of particular interest as mutations in this gene can also cause Cockayne's syndrome and trichothiodystrophy. The XPD gene was initially named ERCC2 (excision repair cross complementing) as it was cloned using human DNA to complement the ultraviolet sensitivity of a …

The molecular basis of xeroderma pigmentosum.

BACKGROUND: Xeroderma pigmentosum is an extremely rare, autosomal recessive disease characterized by a more than 1000-fold increase in nonmelanoma skin cancer. Individuals with this disease can be divided into eight complementation groups: A-G and V for variant. Each one represents a different genetic defect in DNA repair. OBJECTIVE: To review the molecular basis of xeroderma pigmentosum. RESULTS: Deficiencies in various gene products in the nucleotide excision repair pathway cause xeroderma pigmentosum in complementation groups A-G. The molecular basis of the variant group remains to be elu…

Thymidylate synthase inhibitory drugs induce p53-dependent pathways differently.

Thymidylate synthase (TS) is a key enzyme in thymidylate biosynthesis and an established target of chemotherapeutics such as 5-fluoro-2'-deoxyuridine (5FdUR) and raltitrexed (RTX). Inhibition of TS disrupts the dUTP:dTTP balance, leading to uracil misincorporation, triggering futile base excision repair cycles, DNA strand breaks, and ultimately cell death. Interestingly, when the main uracil-DNA repair pathway is inhibited, treatment with TS-inhibitory drugs still leads to cell death. Beyond its catalytic role, TS also binds RNA, autoregulating its own translation and interacting with transcri…

Popliteal artery pseudoaneurysm caused by distal femoral osteochondroma in a 13-year-old boy.

A 13-year-old boy presented with 10-day left distal thigh swelling and pain after running. Examination revealed a 7.0 × 9.0 cm firm popliteal mass, absent popliteal pulse, and weak dorsalis pedis pulse. Computed tomography angiography demonstrated a 69 × 91 mm pseudoaneurysm with multiple osteochondromas. Open repair included sac evacuation, primary repair of a 1.0 cm arterial defect with interrupted 6-0 polypropylene sutures, and excision of the offending osteochondroma with burr smoothing. The dorsalis pedis pulse was restored immediately, and recovery was uneventful. Osteochondroma is a rar

The transcription-repair coupling factor CSA is required for efficient repair only during the elongation stages of RNA polymerase II transcription.

The known nucleotide excision repair (NER) defects of xeroderma pigmentosum (XP) and Cockayne syndrome (CS) cells can be exploited to analyze mechanisms of repair of UV-induced cyclobutane pyrimidine dimers (CPDs) at nucleotide (nt.) resolution. The two gene products of the CS complementation groups (CSA and CSB) have been implicated in the preferential repair of the transcribed strand of human genes. We had previously described very efficient repair of CPDs at sequences near the transcription initiation site of the human JUN gene in normal fibroblasts. Here, we have analyzed repair in a CSA f…

Transcription-coupled repair: tangled up in convoluted repair.

Significant progress has been made in understanding the mechanism of transcription-coupled nucleotide excision repair (TC-NER); however, numerous aspects remain elusive, including TC-NER regulation, lesion-specific and cell type-specific complex composition, structural insights, and lesion removal dynamics in living cells. This review summarizes and discusses recent advancements in TC-NER, focusing on newly identified interactors, mechanistic insights from cryo-electron microscopy (Cryo-EM) studies and live cell imaging, and the contribution of post-translational modifications (PTMs), such as …

Transanal minimally invasive surgery (TAMIS): applications beyond local excision.

Transanal minimally invasive surgery (TAMIS) is a new technique for the local excision of rectal neoplasia. This platform employs ordinary laparoscopic instruments to achieve high-quality local excision. The TAMIS platform, however, is quite versatile. Described here are applications of TAMIS beyond local excision, ranging from repair of a rectourethral fistula to reverse proctectomy.

The DNA repair endonuclease XPG binds to proliferating cell nuclear antigen (PCNA) and shares sequence elements with the PCNA-binding regions of FEN-1 and cyclin-dependent kinase inhibitor p21.

Proliferating cell nuclear antigen (PCNA) is a DNA polymerase accessory factor that is required for DNA replication during S phase of the cell cycle and for resynthesis during nucleotide excision repair of damaged DNA. PCNA binds to flap endonuclease 1 (FEN-1), a structure-specific endonuclease involved in DNA replication. Here we report the direct physical interaction of PCNA with xeroderma pigmentosum (XP) G, a structure-specific repair endonuclease that is homologous to FEN-1. We have identified a 28-amino acid region of human FEN-1 (residues 328-355) and a 29-amino acid region of human XPG…

Correction of xeroderma pigmentosum complementation group D mutant cell phenotypes by chromosome and gene transfer: involvement of the human ERCC2 DNA repair gene.

Cultured cells from individuals afflicted with the genetically heterogeneous autosomal recessive disorder xeroderma pigmentosum (XP) exhibit sensitivity to UV radiation and defective nucleotide excision repair. Complementation of these mutant phenotypes after the introduction of single human chromosomes from repair-proficient cells into XP cells has provided a means of mapping the genes involved in this disease. We now report the phenotypic correction of XP cells from genetic complementation group D (XP-D) by a single human chromosome designated Tneo. Detailed molecular characterization of Tne…

[Hereditary diseases with primary and secondary DNA repair defects].

Two basic ways of DNA repair are discussed. The first one is nucleotide excision repair after exposure to UV light regarded on using examples of various eukaryotic cells, detailed description of human diseases related to genetic defects of this process (xeroderma pigmentosum, Cockayne's syndrome, and trychotiodystrophy). Contemporary ideas on genetics of excision repair are presented. Relevant genes and their mutations are shown. The second one is a process of double strand breaks repair regarded as the basic type of lethal damage, provoked by ionizing radiation. This process was first describ…

DNA damage and repair in translational oncology: an overview.

Unknown to early investigators, DNA damage and repair has been a major focus of anticancer therapy from the beginning of clinical oncology. … Some of our most useful clinical agents depend on causing DNA damage that is repaired by nucleotide excision repair. X-irradiation induces damage that is mostly repaired by base excision repair and double-strand break repair. We are now learning how to modulate select DNA repair pathways to benefit patients with breast cancer and other malignancies.

Repair-induced DNA double strand breaks after ultraviolet-light and either aphidocolin or 1-beta-D-arabinofuranosylcytosine/hydroxyurea.

We recently showed that the DNA repair processes occurring after u.v. light induce double strand breaks (DSBs) in the DNA of normal human fibroblast (IMR-90) but not of xeroderma pigmentosum group A cells. We call these lesions 'repair-induced double strand breaks' (RDSBs), since they only occur in excision repair competent cells. In this work, we asked whether these RDSBs occur in IMR-90 cells at low u.v. doses and whether the RDSBs are themselves repairable. We reasoned that holding open the excision-repair induced gaps by inhibiting nucleotide polymerization after u.v. light with hydroxyure…

Xeroderma pigmentosum group F caused by a defect in a structure-specific DNA repair endonuclease.

Nucleotide excision repair, which is defective in xeroderma pigmentosum (XP), involves incision of a DNA strand on each side of a lesion. We isolated a human gene homologous to yeast Rad1 and found that it corrects the repair defects of XP group F as well as rodent groups 4 and 11. Causative mutations and strongly reduced levels of encoded protein were identified in XP-F patients. The XPF protein was purified from mammalian cells in a tight complex with ERCC1. This complex is a structure-specific endonuclease responsible for the 5' incision during repair. These results demonstrate that the XPF…

DNA repair gene polymorphisms and prostate cancer risk in South Australia--results of a pilot study.

OBJECTIVE: Single nucleotide polymorphisms (SNPs) in DNA repair genes may impact on DNA damage, and cancer risk. To elucidate the role of SNPs in DNA repair genes in prostate cancer (PC) we conducted a case-control study comprising of 118 Caucasian men affected with late onset PC and 132 age-matched healthy controls from South Australia. METHODS AND MATERIALS: We examined the association between PC risk with nonsynonymous SNPs (nsSNPs) in 5 genes involved in 3 DNA-repair pathways: (1) base excision repair (BER): hOGG1 C1245G (Ser326Cys) and XRCC1 G28152A (Arg399Gln); (2) nucleotide excision r…

The endogenous nuclease sensitivity of repaired DNA in human fibroblasts.

The limited DNA excision repair that occurs in the chromatin of UV-irradiated growth arrested cells isolated from a xeroderma pigmentosum (XP) complementation group C patient is clustered in localized regions. The repaired DNA was found to be more sensitive to nicking by endogenous nucleases than the bulk of the DNA. The extra-sensitivity does not change with increasing amounts of DNA damage or repair activity in the locally-repaired regions and is retained through a 24-h chase period. We suggest that these results are due to the occurrence of DNA repair limited to pre-existing, non-transient …
