Phenylalanine hydroxylase deficiency caused by a single base substitution in an exon of the human phenylalanine hydroxylase gene.

A novel restriction fragment length polymorphism in the phenylalanine hydroxylase (PAH) locus generated by the restriction endonuclease MspI was observed in a German phenylketonuria (PKU) patient. Molecular cloning and DNA sequence analyses revealed that the MspI polymorphism was created by a T to C transition in exon 9 of the human PAH gene, which also resulted in the conversion of a leucine codon to a proline codon. The effect of the amino acid substitution was investigated by creating a corresponding mutation in a full-length human PAH cDNA by site-directed mutagenesis followed by expressio

A 19-base pair deletion in the pro-alpha 2(I) gene of type I procollagen that causes in-frame RNA splicing from exon 10 to exon 12 in a proband with atypical osteogenesis imperfecta and in his asymptomatic mother.

Previous observations established that fibroblasts from a proband with atypical osteogenesis imperfecta synthesized about equal amounts of normal pro-alpha 2(I) chains and shortened pro-alpha 2(I) chains of type I procollagen. The pro-alpha 2(I) chains were shortened because of an in-frame deletion of most or all of the 18 amino acids encoded by exon 11 of the pro-alpha 2(I) gene. Here it was demonstrated that one of the proband's alleles for the pro-alpha 2(I) gene contained a 19-base pair deletion at the junction of intervening sequence 10 and exon 11 that produced an RNA splicing defect. Mutation-Dependent Conformational Dynamics and Dual-Mode Afatinib Binding in EGFR Exon 19 Variants.

Exon 19 mutations in the epidermal growth factor receptor (EGFR) kinase domain, including the common ΔE746-A750 deletion and the less frequent L747P substitution, are clinically important drivers of non-small cell lung cancer (NSCLC). Although these mutations occur within the β3-αC region, how they alter EGFR conformational dynamics and influence the response to the covalent inhibitor afatinib remains incompletely understood. Here, we investigated mutation-dependent changes in EGFR conformational ensembles, dual-mode of afatinib binding, and Cys797-mediated covalent reaction energetics in wild

SF3B3-dependent inclusion of STOX1 exon 3 facilitates invasion and tumor progression in colorectal cancer.

Alternative splicing dysregulation contributes to colorectal cancer (CRC) progression, yet the roles of specific splicing factors, such as SF3B3, remain poorly defined. To identify key dysregulated splicing factors, RNA sequencing was performed comparing high-metastatic (SW620) and low-metastatic (SW480) CRC cell lines. Notably, SW480 and SW620 are isogenic cell lines derived from the primary tumor and lymph node metastasis of the same CRC patient, respectively. Their consistent genetic background minimizes confounding factors, enabling reliable detection of metastasis-associated molecular alt

Evaluation for the Efficacy of Exon-Skipping Therapy Using In Vivo Assessment of Skeletal Muscle Function and Fragility.

Splice-switching oligonucleotide (SSO) therapy is expected to be a promising treatment for muscular diseases such as Duchenne muscular dystrophy (DMD). Preclinical studies using animal models of muscle diseases are essential for the development of SSO therapy. In addition to the histological and biochemical analyses, an in vivo muscle torque measurement using supramaximal electrical stimulation has become a standard protocol for the evaluation of drug efficacy. Nevertheless, in order to accurately measure the torque production in animal models, there are some critical points that should be bro

The Enigma of Big Tau Exon 4a: Genomic Architecture, Biophysical Identity, and Unique Evolutionary Mechanisms.

The microtubule-associated protein tau, encoded by the MAPT gene, serves as a major component of the neuronal cytoskeleton, facilitating the assembly, stabilization, and spatial organization of microtubules. Much of the work on tau has focused on the low-molecular-weight (LMW) isoforms abundantly expressed in the central nervous system (CNS) and their pathological aggregation in tauopathies. However, a different variant known as "Big tau", present in the peripheral nervous system (PNS) and selective CNS regions has distinct structural and functional properties and offers a unique perspective o

Acidosis promotes exon skipping through sequestering SR-rich splicing factors in nuclear speckles.

Deregulation of alternative splicing has long been associated with aggressive cancer phenotypes characterized by acute or chronic acidosis. Recently, it was hypothesized that this deregulation results from the sequestration of SR-rich splicing factors in nuclear speckles. However, this hypothesis lacked experimental evidence. Here, we demonstrate that, within the pH range consistent with acidosis, phosphorylated SR-rich fragments of splicing factors undergo a phase transition in minimal and multicomponent models of nuclear speckles. This acidification effect resembles the effect of SR dephosph

THRB splice site variants lead to exon 4 skipping and TRβ1 gain-of-function syndrome.

BACKGROUND: Heterozygous c.283+1G>A and c.283G>A variants in the THRB gene, encoding for thyroid hormone receptor (TR)β1 and β2, lead to autosomal dominant macular dystrophy (ADMD). We report the detailed clinical characterization of two first-degree relatives with ADMD, heterozygous for THRB c.283+1G>A, and an unrelated ADMD patient with a novel variant, c.283G>C. The genomic and molecular consequences of both variants were studied. METHODS: gDNA and mRNA were obtained from leukocytes. Clinical characterization included biochemistry, bone density and body composition, ECG, echocardiography,

Comprehensive characterization of MET exon 14 skipping mutations in non-small cell lung cancer.

BACKGROUND: MET exon 14 skipping mutation (METΔex14) is a key driver event in non-small cell lung cancer (NSCLC) and can emerge as an acquired drug resistance mechanism to MET, EGFR or ALK inhibitors. The clinical and genomic features of METΔex14 in NSCLC require further characterization. METHODS: Our study included a total of 585 patients with METΔex14 + NSCLC, comprising 556 baseline samples, 53 samples from patients exhibiting resistance to MET inhibitors, and 16 samples from patients resistant to EGFR/ALK inhibitors. Genomic data from targeted next-generation sequencing (NGS) of tissue an

Reduced SRSF1 Expression Correlates with LCK Exon Skipping and Impaired Treg Induction in Immune Thrombocytopenia.

Immune thrombocytopenia (ITP) is an autoimmune disorder characterized by antibody-mediated platelet destruction and impaired regulatory T-cell (Treg) function, yet its molecular basis remains poorly defined. Here, we show that CD4+ naive T cells from patients with ITP exhibit diminished T-cell receptor (TCR) signaling and impaired in vitro Treg induction relative to healthy controls. RNA-sequencing revealed widespread alternative splicing dysregulation, most notably exon 8 skipping in LCK, a kinase central to TCR signaling and Treg differentiation. Using an LCK minigene combined with RNA pull-

RGR-opsin maintains cellular homeostasis by ameliorating the proteotoxicity of its exon-skipping splice variant.

PURPOSE: One of the alternative splicing variants of retinal G protein-coupled receptor (RGR), RGR-d, is recognized as a misfolded protein. With or without inhibition of the proteasome system, RGR-d has a cytotoxic effect on retinal pigment epithelium (RPE) cells. RGR-d mutant mice exhibit age-related macular degeneration-like pathologic changes. However, humans with RGR-d likely express both wild-type and splice variant forms. In this study, we investigate the protective influence of the co-expression of RGR and RGR-d protein in RPE cells. METHODS: ARPE-19 cells overexpressing both RGR and R

Novel frameshift variant in exon 7 of COL17A1 in a domestic shorthair kitten with junctional epidermolysis bullosa.

Junctional epidermolysis bullosa (JEB) is a congenital blistering skin disorder with clefting within the lamina lucida of the basement membrane zone. We describe the clinical and morphologic features of JEB in a 4-mo-old domestic shorthair kitten and identify the underlying genetic variant. The kitten was presented with blistering lesions affecting friction-prone areas of haired skin, mucocutaneous junctions, and oral mucosa. Histopathology revealed extensive subepidermal cleft formation in affected tissues. Periodic acid-Schiff (PAS) staining showed a thin, PAS-positive line along the dermal

Nonsense Mutation in USH2A Exon-13 Activates the Innate Immune Response in Müller Glial Cells.

Pathological USH2A mutations cause Usher syndrome type II, characterized by progressive retinitis pigmentosa and hearing and balance impairment. This study aims to investigate the cellular mechanisms underlying USH2A-related retinal degeneration using human induced pluripotent stem cell (hiPSC)-derived retinal organoids. The introduction of a homozygous nonsense mutation in the USH2A hotspot exon-13 resulted in normal photoreceptor development but loss of ciliary localization of usherin long form B and its interacting proteins, ADGRV1 and whirlin. Haemophilia A in a Female German Shepherd With Homozygosity for the FVIII p.C567Y Variant in Exon 11 of the F8 Gene.

Haemophilia A is an extremely rare disorder in females, as the causative F8 gene is located on the X chromosome. Female carriers, also known as 'conductors,' are typically heterozygous and therefore do not show clinical signs of the disease. However, in mild forms of haemophilia A, affected males may survive and mate. In such cases, mating a haemophilic male with a carrier female can result in female offspring who are homozygous for a pathogenic variant and consequently develop haemophilia A. In the present case, a female dog with a mild form of haemophilia A is described. Although it had a ma

A Noncanonical MET Exon 14 Splice-Site Variant in Pulmonary Sarcomatoid Carcinoma With Response to Capmatinib.

MET exon 14 skipping is an actionable oncogenic driver in non-small cell lung carcinoma (NSCLC); however, noncanonical splice-region variants are frequently classified as variants of unknown significance (VUS), which may result in missed therapeutic opportunities and highlight limitations in current DNA next-generation sequencing (NGS) reporting criteria. We report a patient with pulmonary sarcomatoid carcinoma harboring a noncanonical MET splice donor-proximal indel (c.3022_3028 + 13delinsA), initially interpreted as a VUS, who achieved a rapid and durable response to capmatinib. Exon Skipping by Ultrasound-Enhanced Delivery of Morpholino with Bubble Liposomes for Myotonic Dystrophy Model Mice.

Abnormal splicing of the chloride channel 1 (CLCN1) gene causes myotonic dystrophy type 1 (DM1). Therefore, controlling the alternative splicing process of this gene by antisense oligonucleotides can be a promising treatment for DM1. In this study, we describe an efficient phosphorodiamidate morpholino oligomer (PMO) delivery method by ultrasound-mediated Bubble liposomes, which is a known gene delivery tool with ultrasound exposure, to treat skeletal muscles in a DM1 mouse model, HSALR. Effective delivery of PMO using this technique can help control the alternative splicing of the Clcn1 gene
