Effects of High- and Low-Fat Meals on the Bioavailability and Pharmacokinetics of Votoplam, a HTT Gene Splicing Modifier.

Votoplam is a novel, orally bioavailable, small molecule HTT gene splicing modifier that is being developed for the treatment of Huntington's disease. This was a single dose, open-label, two-period, crossover food effect study that evaluated the effect of high- and low-fat meals on 20 mg votoplam in healthy participants. There was a washout of 21 days between the two periods. Twenty-six participants completed the study. There were minimal changes in the bioavailability and pharmacokinetics of votoplam following administration of a single dose of 20 mg votoplam when taken with low-fat and high-

[Effect of homocysteine on gluconeogenesis in mice].

OBJECTIVE: To investigate the expressions of glucose-6-phosphatase (G6Pase) and phosphoenolpyruvate carboxykinase (PEPCK) in the liver of mice with hyperhomocysteinemia (HHcy) and explore the mechanism of gluconeogenesis induced by homocysteine. METHODS: Fifty mice were randomly divided into normal control group (n=25) and HHcy group (n=25) and fed with normal food and food supplemented with 1.5% methionine, respectively. After 3 months of feeding, the fasting blood glucose and insulin levels were determined, and HOMA insulin resistance index (HOMA-IR) was calculated. The expressions of G6Pas

[Nutrition and health--infections caused by food].

The annual incidence of food-borne infections in the Netherlands is estimated to be 250,000 or more; registration, however, is lacking. Meat, poultry, milk and eggs are contaminated primarily by intestinal animal commensals (Salmonella, Campylobacter, E. coli O157:H7, Yersinia enterocolitica) or secondarily by animals, humans and the environment during processing (typhoid fever, Shigella, Listeria, Clostridium, hepatitis A virus, Norwalk virus, parasites). The guidelines for the prevention of contamination are insufficient. Intensity of production and the economic importance of fast, large-sca

Hypergastrinemia in rheumatoid arthritis.

Fasting hypergastrinemia was noted in 13-41% of 82 of outpatients with definite or classical rheumatoid arthritis. No differences were noted between the normogastrinemic and the hypergastrinemic subjects in their age, sex, duration of rheumatoid disease, serological status, or medications. The food-stimulated gastrin response was 117 mg/L, or 216% above the basal levels, and the integrated gastrin response was increased to the level previously reported in patients with duodenal or gastric ulcer disease. These results suggest that hypergastrinemia is a feature of patients with rheumatoid arthri

Impact of Different Durations of Fasting on Intestinal Autophagy and Serum Metabolome in Broiler Chicken.

Fasting-induced autophagy in the intestine is beneficial for body health. This study was designed to explore the relationship between the host metabolism and intestinal autophagy. Broilers were randomly assigned into 48 cages. At 0 (CT), 12 (FH12), 24 (FH24), 36 (FH36), 48(FH48), and 72 h (FH72) before 09:00 a.m. on day 25, eight cages of birds were randomly allotted to each fasting time point using completely random design, and their food was removed. At 09:00 a.m. on day 25, the blood and jejunum were sampled for serum metabolome and autophagy gene analyses, respectively. The results showed

Fasting before contrast-enhanced CT and the incidence of acute adverse reactions: a single-center randomized clinical trial.

OBJECTIVES: To evaluate the effect of eliminating the traditional preparatory fasting policy before contrast-enhanced CT on acute adverse reactions and to identify potential risk factors in a Spanish population sample, since many European patients still experience this unnecessary measure in clinical practice. METHODS: Outpatients who underwent non-emergency CT to either 6 h of solid food fasting (control group) or an unrestricted consumption of solids (intervention group). Adverse reactions during contrast media administration and up to 30 min afterward were recorded and their incidence was

Impact of nutritional factors on the pharmacokinetics of antipsychotics - a systematic review.

INTRODUCTION: Nutritional habits and specific dietary components can significantly alter the pharmacokinetics (PK) of antipsychotics, potentially leading in worst cases to therapeutic failure or increased toxicity. Despite its clinical importance, a comprehensive systematic overview of these interactions remains a gap in current psychiatric research. This systematic review aims to evaluate how food intake or specific nutrients influence the PK parameters of antipsychotic medications. METHODS: A systematic search was conducted across Web of Science, PubMed, Cochrane Database, and Scopus. Out o

Dietary Patterns and Nutritional Status in Bariatric Surgery Candidates-A Cross-Sectional Study.

BACKGROUND/OBJECTIVES: Observing a patient's dietary behaviour before bariatric surgery may help to predict their diet (and indirectly, the rate of weight loss) after the procedure. Consequently, the aim of this study was to identify dietary patterns (DPs) in bariatric surgery candidates, as well as to assess the relationship between DPs, degree of obesity, and body composition. METHODS: The participants were comprised of 117 bariatric surgery candidates. Data concerning their diet, lifestyle, and socioeconomic status was collected using the KomPAN® questionnaire. The following three DPs were

Grocery Shopping and Eating Behaviors Differ Between Persons With and Without a Spinal Cord Injury.

BACKGROUND: Spinal cord injury (SCI) heightens the susceptibility to neurogenic obesity, which is driven by alterations in energy metabolism and suboptimal dietary habits after injury. Those with SCI may adjust their grocery shopping and eating behaviors, notably influencing overall health outcomes. OBJECTIVES: To assess differences in grocery shopping and eating behaviors between persons with and without (controls) SCI. METHODS: In an online, cross-sectional study, adults with (n = 207) and without (n = 52) SCI completed a one-time survey that assessed grocery shopping behavior (shopping fr

Feeding behaviour after injection of α-adrenergic receptor agonists into the median raphe nucleus of food-deprived rats.

This study investigated the participation of median raphe nucleus (MnR) α1-adrenergic receptors in the control of feeding behaviour. The α1-adrenergic agonist phenylephrine (PHE) and α2-adrenergic agonist clonidine (CLON) (at equimolar doses of 0, 6 and 20 nmol) were injected into the MnR of: a) rats submitted to overnight fasting (18 h); or b) rats maintained with 15 g of lab chow/day for 7 days. Immediately after the drug injections, the animals were placed in the feeding chamber and feeding and non-ingestive behaviours such as grooming, rearing, resting, sniffing and locomotion were recorde

Nanoplasmonic sensor for foodborne pathogens detection. Towards development of ISO-SERS methodology for taxonomic affiliation of Campylobacter spp.

According to EU summary report on zoonoses, zoonotic agents and food-borne outbreaks in 2017, Campylobacter was the most commonly reported gastrointestinal bacterial pathogen in humans in the EU. Unfortunately, the standard methods for the detection of thermotolerant Campylobacter spp. in foods are time-consuming. Additionally, the qualified staff is obligatory. For this reason, new methods of pathogens detection are needed. The present work demonstrates that surface-enhanced Raman scattering (SERS) is a reliable and fast method for detection of Campylobacter spp. in food samples. The proposed

The National Cardiothoracic Centre, Accra Ghana: proceedings of the second International Update Course in Cardiology - improving the coverage of cardiology services.

Many developing countries now face the growing phenomenon of the double burden of disease. Most are still grappling with infectious diseases resulting from poor environmental sanitation and lack of access to good drinking water like malaria, cholera, and enteric fever. At the same time changes in diet and lifestyle in general in these countries is resulting in increasing numbers of people with obesity, sedentary life styles, increased salt intake from fast foods, increased smoking and consumption of alcohol and fizzy drinks, hypertension and diabetes. To increase the scope and depth of cardiov…

Changes in brain tissue and behavior patterns induced by single short-term fasting in mice.

In humans, emaciation from long-term dietary deficiencies, such as anorexia, reportedly increases physical activity and brain atrophy. However, the effects of single short-term fasting on brain tissue or behavioral activity patterns remain unclear. To clarify the impact of malnutrition on brain function, we conducted a single short-term fasting study as an anorexia model using male adult mice and determined if changes occurred in migratory behavior as an expression of brain function and in brain tissue structure. Sixteen-week-old C57BL/6J male mice were divided into either the fasted group or

Experience with DPP-4 inhibitors in the management of patients with type 2 diabetes fasting during Ramadan.

A large proportion of Muslim patients with type 2 diabetes mellitus (T2DM) elect to fast during the holy month of Ramadan. For these patients hypo- and hyperglycemia constitute two major complications associated with the profound changes in food pattern during the Ramadan fast, and efficacious treatment options with a low risk of hypoglycemia are therefore needed to manage their T2DM as effectively and safely as possible. Dipeptidyl peptidase-4 (DPP-4) inhibitors modulate insulin and glucagon secretion in a glucose-dependent manner, and consequently a low propensity of hypoglycemia has consist

[Anorexia nervosa: An addiction? Application of the addiction model to eating disorders. A narrative commented review].

OBJECTIVES: Addiction offers a framework for the understanding of eating disorders, particularly those characterized by hyperphagia, with growing interest in food addiction. However, the application of the addiction model to anorexia nervosa remains more controversial. In this commented narrative review, we examine and discuss the addictive features of anorexia nervosa. METHODS: Commented narrative review of the literature. RESULTS: Anorexia nervosa could be the consequence of the loss of control of several objects of positive reinforcement: food restriction, physical hyperactivity, and food

Applying network analysis to explore family and peer influence on diet and health in households of Singaporean young adults: findings from an online network survey.

OBJECTIVE: The complexity and nuance of how social networks shape dietary behaviours and health dynamics remain underexplored, particularly in collectivist societies where family and peer relationships strongly impact health. This study applies social network analysis to examine these dynamics in Singapore. DESIGN: An online household survey of young adults (age 21-35) and family (21+) assessed the consumption of healthy food groups (fruit, vegetable intake), unhealthy food groups (fast food, snack consumption) and social network characteristics (interaction frequency, emotional closeness, sh

Effects of A One-week Fasting Therapy in Patients with Type-2 Diabetes Mellitus and Metabolic Syndrome - A Randomized Controlled Explorative Study.

There is increasing experimental evidence for beneficial effects of calorie restriction and intermittent fasting in type 2 diabetes mellitus (T2DM). In humans, prolonged fasting is established as a health-promoting complementary treatment in Europe and claimed to improve metabolism by a complex hormetic response. We aimed to investigate effects of a one-week fasting period compared to usual care in T2DM by means of a pilot trial. Patients with manifest T2DM medically treated with oral hypoglycemic agents and/or insulin were randomly assigned to a 7-day fasting program followed by dietary advic
