Assessment of active fibrotic remodeling in patients with idiopathic pulmonary fibrosis (IPF)/progressive pulmonary fibrosis (PPF) using dual-time-point [68Ga]Ga-FAPI-46-PET/CT.

BACKGROUND: Progressive fibrosing interstitial lung diseases (ILD) continue to have a poor prognosis. To date, there is no imaging method that can reliably detect fibrotic remodeling processes or distinguish between active and dormant fibrotic changes in the lungs. The novel PET/CT with fibroblast activation protein inhibitor (FAPI) targets activated fibroblasts. The aim of this study was to evaluate the feasibility of [68Ga]Ga-FAPI-46 PET/CT to detect and quantify fibroblast-related pulmonary activity, and to explore the association between this activity and pulmonary function, as well as the

Stable expression of protective protein/cathepsin A-green fluorescent protein fusion genes in a fibroblastic cell line from a galactosialidosis patient. Model system for revealing the intracellular transport of normal and mutated lysosomal enzymes.

Fibroblastic cell lines derived from a galactosialidosis patient, stably expressing the chimaeric green fluorescent protein variant (EGFP) gene fused to the wild-type and mutant human lysosomal protective protein/cathepsin A (PPCA) cDNA, were first established as a model system for revealing the sorting and processing of lysosomal enzymes and for investigating the molecular bases of their deficiencies. In the cell line expressing the wild-type PPCA-EGFP chimaera gene (EGFP-PPwild), an 81 kDa form (27 kDa EGFP fused to the C-terminus of the 54 kDa PPCA precursor) was produced, then processed in

The role of HIF-1α in nicotine-induced root and bone resorption during orthodontic tooth movement.

BACKGROUND: In orthodontic tooth movement (OTM), pseudo-inflammatory processes occur that are similar to those of nicotine-induced periodontitis. Previous studies have shown that nicotine accelerates OTM, but induces periodontal bone loss and dental root resorption via synergistically increased osteoclastogenesis. This study aimed to investigate the role of hypoxia-inducible factor 1 alpha (HIF-1α) in nicotine-induced osteoclastogenesis during OTM. MATERIALS/METHODS: Male Fischer-344 rats were treated with l-Nicotine (1.89 mg/kg/day s.c., N = 10) or NaCl solution (N = 10). After a week of pre

Coding GNAS mutations leading to hormone resistance impair in vitro agonist- and cholera toxin-induced adenosine cyclic 3',5'-monophosphate formation mediated by human XLalphas.

Most loss of function mutations of GNAS identified in different forms of pseudohypoparathyroidism disrupt not only the stimulatory G protein alpha-subunit (Gsalpha), but also its paternally expressed variant, XLalphas. However, the possibility that XLalphas deficiency contributes to disease pathogenesis has remained unexplored. We therefore examined the signaling property of human XLalphas and the effects of one novel (XLalphas(H704P) or Gsalpha(H362P)) and two previously described (XLalphas(DelI724) and XLalphas(Y733X) or Gsalpha(DelI382) and Gsalpha(Y391X), respectively) GNAS mutations on ei

Cells retrovirally transfected with fibroblast growth factor-2 isoforms exhibit altered adenylate cyclase activity and G-protein functionality.

Basic fibroblast growth factor (FGF-2) is synthesized as different molecular mass isoforms all lacking the signal-peptide sequence. The high molecular-mass isoforms (21-24 kDa) possess a signal sequence directing their nuclear translocation. The role of each isoform is still poorly understood, however, modifications in intracellular signalling pathways could explain some effects of these peptides. In order to evaluate the role of FGF-2 isoforms on the adenylate cyclase (AC) signalling pathway, we retrovirally infected a rat pancreatic cell line (AR4-2J) with point-mutated FGF-2 cDNAs, allowing

Radiofrequency ablation of atrial flutter with 1:1 accessory pathway conduction improved cardiac function in a patent with PRKAG2 Cardiomyopathy. Insights with 68Ga-FAPI PET/CT imaging.

PRKAG2 syndrome is an autosomal dominant cardiac glycogenosis caused by mutations in the PRKAG2 gene, clinically characterized by the triad of ventricular hypertrophy, conduction disease, and ventricular pre-excitation, along with a high propensity for supraventricular arrhythmias. This report describes a young male carrier of the R302Q mutation who developed wide QRS tachycardia due to atrial flutter with 1:1 conduction via an accessory pathway. Electrophysiological study revealed atrial flutter activation in a counterclockwise direction around the tricuspid annulus, coexisting with a left-si

Therapeutic effects of serum extracellular vesicles in liver fibrosis.

The lack of approved therapies for hepatic fibrosis seriously limits medical management of patients with chronic liver disease. Since extracellular vesicles (EVs) function as conduits for intercellular molecular transfer, we investigated if EVs from healthy individuals have anti-fibrotic properties. Hepatic fibrogenesis or fibrosis in carbon tetrachloride (CCl4)- or thioacetic acid-induced liver injury models in male or female mice were suppressed by serum EVs from normal mice (EVN) but not from fibrotic mice (EVF). CCl4-treated mice undergoing EVN therapy also exhibited reduced levels of hepa

SQSTM1/p62 and PPARGC1A/PGC-1alpha at the interface of autophagy and vascular senescence.

UNLABELLED: Defective macroautophagy/autophagy and mitochondrial dysfunction are known to stimulate senescence. The mitochondrial regulator PPARGC1A (peroxisome proliferator activated receptor gamma, coactivator 1 alpha) regulates mitochondrial biogenesis, reducing senescence of vascular smooth muscle cells (VSMCs); however, it is unknown whether autophagy mediates PPARGC1A-protective effects on senescence. Using ppargc1a-/- VSMCs, we identified the autophagy receptor SQSTM1/p62 (sequestosome 1) as a major regulator of autophagy and senescence of VSMCs. Abnormal autophagosomes were observed in

Metformin exhibits gender specific impact on telomere dynamics by enhancing RAP1 expression in type-2 diabetes mellitus.

Telomere length serves as a critical biomarker of ageing, and diabetes is associated with shorter telomeres. This study aims to investigate the association and underlying molecular mechanism between telomere attrition rate in healthy individuals and patients with type 2 diabetes who consume the anti-diabetic drug metformin. Leukocyte telomere length was measured using the telomere restriction fragment assay in 111 healthy individuals and in 73 individuals with type 2 diabetes who were consuming metformin. Telomere length-regulating mRNA and protein expression studies were performed. The BJ fib

Structural basis for oxygen degradation domain selectivity of the HIF prolyl hydroxylases.

The response to hypoxia in animals involves the expression of multiple genes regulated by the αβ-hypoxia-inducible transcription factors (HIFs). The hypoxia-sensing mechanism involves oxygen limited hydroxylation of prolyl residues in the N- and C-terminal oxygen-dependent degradation domains (NODD and CODD) of HIFα isoforms, as catalysed by prolyl hydroxylases (PHD 1-3). Prolyl hydroxylation promotes binding of HIFα to the von Hippel-Lindau protein (VHL)-elongin B/C complex, thus signalling for proteosomal degradation of HIFα. We reveal that certain PHD2 variants linked to familial erythrocyt

PEG4-Modified FAP-CAIX Heterodimeric Radioligand for Precision Imaging and Radiotherapy.

Tumor heterogeneity severely limits the efficacy of single-targeted radiopharmaceuticals. FAP and CAIX are important biomarkers overexpressed in tumors and the tumor microenvironment. We designed a PEG4-modified heterodimeric radioligand, FC, for dual targeting of FAP and CAIX. FC showed high binding affinity toward FAP (IC50 = 18.2 ± 0.23 nM). [68Ga]Ga-FC exhibited significantly higher tumor uptake than monomeric tracers in FAP- or CAIX-positive tumor models at 120 min postinjection. PEG4 modification markedly enhanced hydrophilicity, tumor retention, and in vivo stability. Mirco-SPECT imagin

Structural Optimization of Fibroblast Activation Protein Inhibitors Through Zwitterionic and PEG Modification Strategy: Impact on Pharmacokinetics and Tumor Imaging.

Fibroblast activation protein (FAP), highly overexpressed in cancer-associated fibroblasts (CAFs), is crucial in tumor pathogenesis and progression, making it an important target for diagnosis and therapy. This study presents the design of a series of FAP inhibitors (FAPIs) derived from UAMC-1110 derivative, modified with zwitterions and polyethylene glycol (PEG). The novel 68Ga-labeled tracers show improved pharmacokinetics compared to 68Ga-FAPI-04. Small animal positron emission tomography/computed tomography (micro-PET/CT) on U87MG tumor-bearing nude mice revealed that 68Ga-FAPI-BN-1, incor

[18F]FAPI PET/CT outperforms [18F]FDG in distinguishing nodal immune flare from lymph node metastases during immunotherapy for lung cancer.

PURPOSE: Nodal immune flare (NIF) is an immune-related adverse event affecting lymph nodes after lung cancer immunotherapy. Differentiating NIF from lymph node metastasis using [18F] fluorodeoxyglucose ([18F]FDG) is challenging. This study aimed to evaluate whether [18F] fibroblast activation protein inhibitor ([18F]FAPI), a novel tracer targeting fibroblast activation protein (FAP), can distinguish NIF from lymph node metastasis after immunotherapy. METHODS: We retrospectively enrolled 589 lung cancer patients who received immunotherapy and subsequently underwent restaging by [18F]FDG PET/CT

mRNA-laden LNP-enabled in situ CAR-macrophage alleviates liver fibrosis via inhibiting activated HSCs and modulating the immune microenvironment.

Liver fibrosis, marked by an abnormal buildup of extracellular matrix (ECM), poses a major health threat. Myofibroblasts, predominantly derived from hepatic stellate cells (HSCs) and portal fibroblasts, are the primary drivers of ECM synthesis. Fibroblast activation protein (FAP), highly expressed by activated HSCs, is a pivotal player in the pathogenesis of liver fibrosis. Delineating the mechanisms underlying HSC activation and devising strategies to curb their hyperactivity are paramount for the management and prevention of liver fibrosis. In this study, we explored a pioneering therapeutic

FAP as a prognostic biomarker and radiomics-based predictor of angiogenesis-associated recurrence in Adamantinomatous craniopharyngioma.

BACKGROUND: Adamantinomatous craniopharyngioma (ACP) is a histologically benign yet clinically aggressive intracranial tumor characterized by high recurrence rates. Fibroblast activation protein (FAP), a marker of cancer-associated fibroblasts (CAFs), is implicated in tumor progression and microenvironment remodeling. This study evaluates the prognostic significance of FAP and develops a radiomics-based model for non-invasive preoperative risk stratification. METHODS: Immunohistochemical analysis was performed on 54 ACP cases to assess FAP expression levels. Transcriptomic data from 110 ACP c

Functional Comparison of Molluscum Contagiosum Virus vFLIP MC159 with Murine Cytomegalovirus M36/vICA and M45/vIRA Proteins.

UNLABELLED: Molluscum contagiosum virus (MCV) gene MC159 encodes a viral FLICE inhibitory protein (vFLIP) that inhibits caspase-8-mediated apoptosis. The MC159 protein was also reported to inhibit programmed necrosis (necroptosis) and modulate NF-κB activation by interacting with RIP1 and NEMO. The importance of MC159 during MCV infection has remained unknown, as there is no system for propagation and genetic manipulation of this virus. Here we investigated the functions of MC159 during viral infection using murine cytomegalovirus (MCMV) as a surrogate virus. MC159 was inserted into the MCMV g

Association between autoimmune disease and neurodevelopmental disorder: a Mendelian randomization analysis.

INTRODUCTION: Neurodevelopmental disorders such as attention deficit and disruptive behaviour disorders (ADHD), autism spectrum disorder (ASD), and schizophrenia have been increasingly prevalent recently. Previous research has demonstrated that inflammatory activity from autoimmune diseases is involved in neurological diseases. However, some studies question the association between inflammatory activities and neurodevelopmental disorders. Herein, we attempt to clarify this relationship using Mendelian randomization (MR) analysis. METHODS: We used systemic lupus erythematosus (SLE), rheumatoid
