Water consumption contributes to maternal PFAS exposure: From source to metabolic perturbations.

Per- and polyfluoroalkyl substances (PFAS) are widespread persistent pollutants and pose a risk to human health. However, the transfer efficiencies (TEs) of PFAS from source water into maternal serum, and its metabolic pathways linking PFAS exposure and human disease are remain unclear. Here, we present an integrative study combining multi-water and serum PFAS analysis, lipidomics, and machine learning to decipher TEs and PFAS-metabolism interactions. Analyzing 30 PFAS across source water (N = 18), tap water (N = 18), water during drinking water treatment process (DWTP, N = 12), and serum of p

Ecosurveillance reveals subtle metabolic effects on the non-native cane toad (Rhinella marina) from low levels of accumulated environmental per- and polyfluoroalkyl substances.

Per- and polyfluoroalkyl substances (PFAS) are synthetic chemicals that persist in the environment, bioaccumulate, and may have toxic effects. This study used non-native cane toads (Rhinella marina) to examine PFAS and metal accumulation and impacts in large terrestrial amphibians from urban and peri-urban areas. We quantified 38 PFAS compounds and 36 environmental and legacy metal(loid)s in 52 adult cane toad livers collected from six locations around Southeast Queensland, Australia, along a known PFAS gradient. Associations among PFAS, metal(loid) concentrations, and whole-organism metrics w

Carbon footprint of Pars plana vitrectomies in rhegmatogenous retinal detachment in Germany between 2013 and 2023 - A cohort study of fluorinated gas emissions.

PURPOSE: Climate change, largely driven by greenhouse gas emissions, is a global threat. The healthcare sector contributes significantly - 4.4% of global and 5.2% of Germany's CO2 emissions. Rhegmatogenous retinal detachment (RRD) is a common ophthalmic emergency requiring surgical intervention to prevent blindness. Fluorinated gases (FG) such as SF6, C2F6, and C3F8 are routinely used in pars plana vitrectomy (ppV) due to their expansive properties, but they have a strong greenhouse effect. With an European Union-wide ban under consideration, this study assesses the associated emissions from p

Perfluorobutanesulfonate Interfering with the Intestinal Remodeling During Lithobates catesbeiana Metamorphosis via the Hypothalamic-Pituitary-Thyroid Axis.

The intestinal remodeling during amphibian metamorphosis is essential for adapting to various ecological niches of aquatic and terrestrial habitats. However, whether and how the widespread contaminant, perfluorobutanesulfonate (PFBS) affects intestinal remodeling remains unknown. In this study, tadpoles (Lithobates catesbeianus) at the G26 stage were exposed to environmentally relevant concentrations of PFBS (0, 1, 3, and 10 μg/L) until the end of metamorphosis. PFBS exposure resulted in reduced thyroid follicular glia; down-regulation of gene transcripts related to thyroid hormone synthesis;

Non-destructive surface sampling of PFAS exposure on firefighter protective gears: Potential implications for occupational exposure.

Per- and polyfluoroalkyl substances (PFAS) are a group of anthropogenic pollutants that pose significant global environmental and health challenges. While PFAS exposure occurs through various pathways, occupational exposure among firefighters is particularly concerning. This study introduces a non-invasive, and non-destructive sample collection protocol using textile wipes to assess PFAS on firefighter safety gear and potential health issues. Dampened polypropylene fabrics were used to swab 12 firefighter turnout gear sets, including jacket shoulders, pant shins, inside and outside of SCBA mas

Does exposure to zinc, methomyl, and perfluorooctanoic acid alter feeding behavior on MUG® in the freshwater amphipod Gammarus fossarum?

Feeding rate alteration is one of the first observed responses when animals are exposed to toxic stress and is recognized as a relevant tool for studying chemical compounds toxicity. However, food substrates that are currently used for ecotoxicity tests are not always easily available compared with referenced products. Using the European freshwater amphipod Gammarus fossarum, we here propose a standardized food substrate fabricated with referenced ingredients: the MUG® (meal unit for gammarid) for ecotoxicity tests. To investigate the suitability of using MUG to study behavioral response of am

Adsorption of perfluorooctanoic acid from aqueous matrices onto chitosan-modified magnetic biochar: Response surface methodology-based modeling, performance, and mechanism.

Perfluorooctanoic acid (PFOA) removal has gained significant attention due to its environmental stability and potential toxicity. This study aims to synthesize a chitosan-modified magnetic biochar (CS_MBC) for efficient PFOA removal from aqueous solutions. Various CS loading ratios (0.25:1, 0.5:1, and 1:1) were explored to determine the optimal adsorbent, with preliminary experiments exhibiting superior performance of CS1_MBC. To explore the impact of various experimental conditions (pH, adsorbent dose, time, and initial PFOA concentrations) on PFOA removal and optimize these parameters, centr

Per- and polyfluoroalkyl substances (PFAS) in placental compartments: Histopathological and toxicological data integration in an Italian cohort.

Per- and polyfluoroalkyl substances (PFAS) are synthetic environmental contaminants with widespread industrial and consumer applications, characterized by strong chemical stability and environmental persistence. Recent studies have highlighted placental permeability to PFAS, though evidence of direct histopathological impairment remains limited. This study aimed to investigate potential associations between PFAS exposure and histopathological abnormalities in placental samples. A total of 23 at-term pregnant women were recruited from two hospitals in Italy as part of a multicenter study. Place

Twenty-eight-day perfluorooctanoate exposure does not affect immune cell populations in naïve mice.

Perfluorooctanoate (C7F15CO2-, PFOA) is a perfluorinated compound widely used in the production of industrial products, such as non-stick cookware and firefighting foams. Although its production has been phased out in the United States, PFOA remains prevalent in the environment due to its persistence and resistance to degradation. As a result, trace levels of PFOA is detectable in water, soil, biota, and the general population. Environmental and toxicological concerns have emerged due to potential health risks associated with PFOA exposure, including effects on the immune system. Basic underly

Sub-acute exposure of male guppies (Poecilia reticulata) to environmentally relevant concentrations of PFOA and GenX induces significant changes in the testis transcriptome and reproductive traits.

Poly- and perfluoroalkyl substances (PFAS) are frequently detected in the environment and are linked to adverse reproductive health outcomes in humans. Although legacy PFAS have been phased out due to their toxicity, alternative PFAS are increasingly used despite the fact that information on their toxic effects on reproductive traits is particularly scarce. Here, we exposed male guppies (Poecilia reticulata) for a short period (21 days) to an environmentally realistic concentration (1 ppb) of PFOA, a legacy PFAS, and its replacement compound, GenX, to assess their impact on reproductive traits

Storms mobilize organophosphate esters, bisphenols, PFASs, and vehicle-derived contaminants to San Francisco Bay watersheds.

In urban to peri-urban watersheds such as those surrounding San Francisco Bay, stormwater runoff is a major pathway by which contaminants enter aquatic ecosystems. We evaluated the occurrence of 154 organic contaminants via liquid chromatography coupled to tandem mass spectrometry, including organophosphate esters (OPEs), bisphenols, per- and polyfluoroalkyl substances (PFASs), and a suite of novel urban stormwater tracers (SWCECs; i.e., vehicle-derived chemicals, pesticides, pharmaceuticals/personal care products, benzothiazoles/benzotriazoles). Dermal bioavailability of perfluoroalkyl substances using in vitro 3D human skin equivalent models.

Perfluoroalkyl substances (PFAS) have been identified in various products that come in contact with human skin, ranging from school uniforms to personal care products. Despite this, knowledge on human dermal uptake of PFAS is lacking. Thus, the human dermal absorption of 17 PFAS was assessed, for the first time, using in vitro 3D-human skin equivalent models exposed to 500 ng/cm2 PFAS dissolved in methanol over 24-36 h. The distribution of target PFAS is presented, based on three fractions: absorbed, un-absorbed, and retained within skin tissue (absorbable dose). Perfluoropentanoic acid (PFPeA

Perfluorooctane sulfonate (PFOS) and its selected analogs induce various cell death types in peripheral blood mononuclear cells.

The perfluoalkyl substance (PFASs) perfluorooctane sulfonate (PFOS) has been widely used in industry. However, PFOS is a persistent organic pollutant and has been gradually replaced by its short-chain analogs, perfluorohexane sulfonate (PFHxS) and perfluorobutane sulfonate (PFBS). PFASs are extremely persistent and are very frequently detected among the general population. The aim of the study was to determine the effect of selected PFASs on peripheral blood mononuclear cells (PBMCs) and the mechanisms of their action. PBMCs were exposed to PFOS, PFBS and PFHxS at concentrations ranging from 0

Tough, Antifreezing, and Piezoelectric Organohydrogel as a Flexible Wearable Sensor for Human-Machine Interaction.

Piezoelectric hydrogel sensors are becoming increasingly popular for wearable sensing applications due to their high sensitivity, self-powered performance, and simple preparation process. However, conventional piezoelectric hydrogels lack antifreezing properties and are thus confronted with the liability of rupture in low temperatures owing to the use of water as the dispersion medium. Herein, a kind of piezoelectric organohydrogel that integrates piezoelectricity, low-temperature tolerance, mechanical robustness, and stable electrical performance is reported by using poly(vinylidene fluoride)

Kynurenic Acid Plays a Protective Role in Hepatotoxicity Induced by HFPO-DA in Male Mice.

Following its introduction as an alternative to perfluorooctanoic acid, hexafluoropropylene oxide dimer acid (HFPO-DA) has been extensively detected in various environmental matrices. Despite this prevalence, limited information is available regarding its hepatotoxicity biomarkers. In this study, toxicokinetic simulations indicated that under repeated treatment, HFPO-DA in mice serum reached a steady state by the 4th day. To assess its subacute hepatic effects and identify potential biomarkers, mice were administered HFPO-DA orally at doses of 0, 0.1, 0.5, 2.5, 12.5, or 62.5 mg/kg/d for 7 d. R

All-Textile Piezoelectric Nanogenerator Based on 3D Knitted Fabric Electrode for Wearable Applications.

Flexible, air permeable and elastic self-powered sensors for human motion monitoring and assisted medical rehabilitation have recently become a hot research topic. However, most current piezoelectric sensors can not account for many characteristics. Addressing this challenge, an all-textile piezoelectric sensor (ATPS) based on 3D structured knitted fabric electrodes is reported. The ATPS consists of a piezoelectric element polyvinylidene fluoride nanofiber membrane, flexible knitted fabric electrodes, and an elastic self-adhesive bandage. Based on the flexible and efficient knitting technology

Impact of Environmental Exposures on Human Breast Milk Lipidome in Future Immune-Mediated Diseases.

The composition of human breast milk (HBM) exhibits significant variability both between individuals and within the same individual. While environmental factors are believed to play a role in this variation, their influence on breast milk composition remains inadequately understood. Herein, we investigate the impact of environmental factors on HBM lipid composition in a general population cohort. The study included mothers (All Babies In Southeast Sweden study) whose children later progressed to one or more immune-mediated diseases later in life: type 1 diabetes (n = 9), celiac disease (n = 24
