GRP78: A possible relationship of COVID-19 and the mucormycosis; in silico perspective.

Mucormycosis is a severe fungal infection reported in many cancer survivors, diabetic and immune-suppressed patients during organ transplants. A vast spark in the reported COVID-19 cases is noticed in India during the second wave in May 2021, when Mucormycosis is declared an epidemic. Despite being a rare disease, the mortality rate associated with Mucormycosis is more than 40%. Spore coat proteins (CotH) are essential proteins in many pathogenic bacteria and fungi. CotH3 was reported as the vital protein required for fungal virulence in Mucormycosis. We previously reported the involvement of

Acidic Mammalian Chitinase Negatively Affects Immune Responses during Acute and Chronic Aspergillus fumigatus Exposure.

Chitin is a polysaccharide that provides structure and rigidity to the cell walls of fungi and insects. Mammals possess multiple chitinases, which function to degrade chitin, thereby supporting a role for chitinases in immune defense. However, chitin degradation has been implicated in the pathogenesis of asthma. Here, we determined the impact of acidic mammalian chitinase (AMCase) (Chia) deficiency on host defense during acute exposure to the fungal pathogen Aspergillus fumigatus as well as its contribution to A. fumigatus-associated allergic asthma. We demonstrate that chitin in the fungal ce

Resorcylic Acid Lactones and Isocoumarin Derivatives from the Marine-Associated Polar Fungus Penicillium sp. OUCMDZ-4014.

Marine-associated polar fungi are promising sources of structurally diverse natural products. To investigate polyketide metabolites from the Antarctic moss-derived fungus Penicillium sp. OUCMDZ-4014, HSQC NMR combined with DeepSAT analysis was used to prioritize fractions enriched in cyclic and aromatic polyketides, and chromane-like metabolites. Chromatographic separation yielded ten resorcylic acid lactone-derived aromatic polyketides, including RAL macrolides, ring-opened esters, and isocoumarins, among which compounds 4-7 were new. Their planar structures were established by HRESIMS and 1D

Primary Cutaneous Cryptococcosis Mimicking Vasculitis in a Patient with Rheumatoid Arthritis.

Opportunistic infections caused by Cryptococcus spp. are well-documented among people with HIV. However, the incidence of cryptococcosis has also recently risen among other immunocompromised patients. Here, we reported a rare case of primary cutaneous cryptococcosis in a 73-year-old man who had been taking corticosteroids long-term for rheumatoid arthritis (RA). Despite receiving antimicrobial treatment, he developed painful, non-healing ulceronecrotic lesions accompanied by haemorrhagic bullae on his right forearm and upper arm. Initially, it was thought that the patient had RA-associated vas

α-Glucan-type exopolysaccharides with varied linkage patterns: Mitigating post-prandial glucose spike and prolonging the glycemic response.

Microbial exopolysaccharides (EPSs) are traditionally known as prebiotics that foster colon health by serving as microbiota nutrients, while remaining undigested in the small intestine. However, recent findings suggest that α-glucan structures in EPS, with their varied α-linkage types, can be hydrolyzed by mammalian α-glucosidases at differing rates. This study explores α-glucan-type EPSs, including dextran, alternan, and reuteran, assessing their digestive properties both in vitro and in vivo. Notably, while fungal amyloglucosidase - a common in vitro tool for carbohydrate digestibility analy

Neutrophil swarms containing myeloid-derived suppressor cells are crucial for limiting oral mucosal infection by C. albicans.

Oral mucosal colonization by C. albicans (Ca) is benign in healthy people but progresses to deeper infection known as oropharyngeal candidiasis (OPC) that may become disseminated when combined with immunosuppression. Cortisone-induced immunosuppression is a well-known risk factor for OPC, however the mechanism by which it permits infection is poorly understood. Neutrophils are the primary early sentinels preventing invasive fungal growth, and here we identify that in vivo neutrophil functional complexes known as swarms are crucial for preventing Ca invasion which are disrupted by cortisone. Equilibria of complexes in the aqueous cobalt(II)-N-(2-hydroxybenzyl)phenylalanine system and their biological activity compared to analogous Schiff base structures.

UNLABELLED: Due to their excellent prospects in biological applications, Schiff bases and their complexes are a source of continuing interest. The present study examines the formation of four cobalt(II) complexes with the reduced Schiff base N-(2-hydroxybenzyl)phenylalanine (PhAlaSal) in alkaline aqueous solution by pH-metry. UV-Vis and ESI-MS studies confirmed the model of proposed species. Kinetic analysis indicated that the single- and bi-ligand cobalt(II) complexes transitioned from octahedral to tetrahedral structures. The Schiff base and its complexes detected under physiological pH were

Clathrin- and Arp2/3-independent endocytosis in the fungal pathogen Candida albicans.

UNLABELLED: Clathrin-mediated endocytosis (CME) is conserved among eukaryotes and has been extensively analyzed at a molecular level. Here, we present an analysis of CME in the human fungal pathogen Candida albicans that shows the same modular structure as those in other fungi and mammalian cells. Intriguingly, C. albicans is perfectly viable in the absence of Arp2/3, an essential component of CME in other systems. In C. albicans, Arp2/3 function remains essential for CME as all 15 proteins tested that participate in CME, including clathrin, lose their characteristic dynamics observed in wild-

Design, Synthesis, and Bioactivity Evaluation of Novel Indene Amino Acid Derivatives as Succinate Dehydrogenase Inhibitors.

Succinate dehydrogenase inhibitors (SDHIs) represent one of the three predominant fungicide categories in contemporary agricultural markets, garnering an increasing level of research interest. Building upon our prior work utilizing aminocyclobutanecarboxylic acid as a linker, we designed and synthesized a novel series of indene amino acid derivatives to optimize hydrophobic interactions with the SDH enzyme. These derivatives demonstrated potent in vitro antifungal activity against Rhizoctonia solani, Botrytis cinerea, and Sclerotinia sclerotiorum, with compound i19 exhibiting efficacy comparab

Disrupted Salivary Fungal Community in Patients With Oral Lichen Planus.

INTRODUCTION AND AIMS: Oral lichen planus (OLP), a chronic inflammatory oral mucosal disorder with malignant potential, has been associated with oral microbial dysbiosis. While bacterial community alterations in OLP are well-documented, the fungal community architecture and ecological dynamics, particularly within salivary microbiota, remain poorly characterised. METHODS: Saliva samples were collected from 30 participants and stratified into 3 cohorts: 10 healthy controls (HC), 10 reticular OLP (R-OLP) patients and 10 erosive OLP (E-OLP) patients. The fungal community profile was assessed usi

Glycolysis-mediated energy metabolism mechanisms: Synergistic antifungal activity of (±)-methylagrimonolide against drug-resistant Candida albicans.

Drug-resistant C. albicans infection is difficult to treat, and many studies have shown that energy metabolism disorder caused by glycolysis pathway can be an important research field of antifungal drugs. In present work, in vitro and in vivo antifungal activities of agrimonolide-based compounds combined with FLC. The results indicated that (±)-methylagrimonolide (1) combined with FLC showed good synergistic inhibition against FLC-resistant ATCC14053-FR with MIC50 of 8 μM and FICI of 0.08. In addition, the combined drugs could also improve the survival status and pathological damage of differe

3D-QSAR-Guided Molecule Design and Protein-Ligand Interaction Studies for Discovery of Succinate Dehydrogenase Inhibitors.

Succinate dehydrogenase (SDH) is an ideal fungicidal target. Here, a three-dimensional quantitative structure-activity relationship (3D-QSAR) was established based on the data of 74 compounds with activity against Rhizoctonia solani. By integrating our previously reported 3D-QSAR findings with the current model, a series of pyrazole-containing compounds were rationally designed and synthesized. Bioassay results demonstrated that B5, with an EC50 of 0.002 μg/mL against R. solani, outperformed fluxapyroxad (0.01 μg/mL). Enzyme activity assays revealed that the IC50 of B5 was 0.12 μM, significant

The structure of a C. neoformans polysaccharide motif recognized by protective antibodies: A combined NMR and MD study.

Cryptococcus neoformans is a fungal pathogen responsible for cryptococcosis and cryptococcal meningitis. The C. neoformans' capsular polysaccharide and its shed exopolysaccharide function both as key virulence factors and to protect the fungal cell from phagocytosis. Currently, a glycoconjugate of these polysaccharides is being explored as a vaccine to protect against C. neoformans infection. In this study, NOE and J-coupling values from NMR experiments were consistent with a converged structure of the synthetic decasaccharide, GXM10-Ac3, calculated from MD simulations. GXM10-Ac3 was designed

[Pyogenic infectious complications in recipients with allogenic kidneys: clinical and bacteriological aspects].

The bacteriological study of wound discharge indicated no changes in the structure of the microflora in the allogenic kidneys of recipients throughout the follow-up. Among microbes isolated there were prevalent gram-positive microbes whose proportion has slightly increased in the past year, with Staphylococcus, mainly epidermal Staphylococcus, which contributes to the etiology. At the same time, studies of wound discharge in the past years showed that the incidence of mixed infections had increased from 4.3 in 1989-1991 to 15.6% in 1994-1997. Bacteriological urinalysis found no great differenc
