INTRODUCTION: Resident and exudate macrophages play an important role in the development of liver cirrhosis. Ionized calcium binding adaptor molecule 1(+) (Iba1(+)) and galectin-3(+) (Gal-3(+)) macrophages regulate liver fibrosis probably through pro-inflammatory and pro-fibrotic factors. Macrophages show polarized functions in liver fibrosis; however, M1-/M2-polarization of Iba1(+) and Gal-3(+) macrophages remains obscured. This study investigated the M1-/M2-polarized properties of Iba1(+) and Gal-3(+) macrophages in chemical-induced liver cirrhosis.
MATERIALS AND METHODS: Cirrhosis was indu

OBJECTIVE: Juvenile dermatomyositis (DM) is a systemic autoimmune disorder of unknown immunopathogenesis in which the immune system targets the microvasculature of skeletal muscles, skin, and other organs. The current mainstay of therapy is a steroid regimen in combination with other immunosuppressive treatments. To date, no validated markers for monitoring disease activity have been identified, which hampers personalized treatment. This study was undertaken to identify a panel of proteins specifically related to active disease in juvenile DM.
METHODS: We performed a multiplex immunoassay for

The non-integrin laminin receptor (LAMR1/RPSA) and galectin-3 (Gal-3) are multi-functional host molecules with roles in diverse pathological processes, particularly of infectious or oncogenic origins. Using bimolecular fluorescence complementation and confocal imaging, we demonstrate that the two proteins homo- and heterodimerize, and that each isotype forms a distinct cell surface population. We present evidence that the 37 kDa form of LAMR1 (37LRP) is the precursor of the previously described 67 kDa laminin receptor (67LR), whereas the heterodimer represents an entity that is distinct from t

The eye is an immune-privileged organ, and corneal transplantation is therefore one of the most successful organ transplantation. The immunosuppressive intraocular microenvironment is known as one of the mechanisms underlying immune privilege in the eye. T-cell immunoglobulin and mucin domain (Tim)-3 is a regulatory molecule for T-cell function, and galectin (Gal)-9 is a Tim-3 ligand. We investigated the role of this pathway in establishing the immune-privileged status of corneal allografts in mice. Gal-9 is constitutively expressed on the corneal epithelium, endothelium and iris-ciliary body

Galectin-1 (Gal-1) is important in immune function and muscle regeneration, but its expression and localization in adult tissues and primary leukocytes remain unclear. To address this, we generated a specific monoclonal antibody against Gal-1, termed alphahGal-1, and defined a sequential peptide epitope that it recognizes, which is preserved in human and porcine Gal-1, but not in murine Gal-1. Using alphahGal-1, we found that Gal-1 is expressed in a wide range of porcine tissues, including striated muscle, liver, lung, brain, kidney, spleen, and intestine. In most types of cells, Gal-1 exhibit

BACKGROUND: Galectin-1 (gal-1), a member of the mammalian beta-galactoside-binding proteins, binds to cell surface glycoproteins (Mucin-1) on trophoblast cells. Although it has been demonstrated that gal-1 induces cell differentiation processes in these cells, no information on its signal transduction processes is available so far. As tyrosine phosphorylation is a major mechanism that controls multiple biological processes including cell differentiation, survival and proliferation, the aim of this study was to examine which human receptor tyrosine kinases (RTKs) were phosphorylated in trophobl

Controlling chronic immunoinflammatory diseases such as lesions in the eye caused by infection with HSV represents a therapeutic challenge. Since CD4(+) T cells are the primary orchestrators of lesions, targeting activated CD4(+) T cell subsets and increasing the representation of cells that express regulatory function would be a logical therapeutic approach. We show that this outcome can be achieved by therapy, systemic or local, with the lectin family member galectin-9. This molecule, which is a natural product of many cell types, acts as a ligand to the inhibitory molecule TIM-3 (T cell Ig

Shiga toxins (Stx) 1 and 2 are responsible for intestinal and systemic sequelae of infection by enterohemorrhagic Escherichia coli (EHEC). However, the mechanisms through which enterocytes are damaged remain unclear. While secondary damage from ischemia and inflammation are postulated mechanisms for all intestinal effects, little evidence excludes roles for more primary toxin effects on intestinal epithelial cells. We now document direct pathologic effects of Stx on intestinal epithelial cells. We study a well-characterized rabbit model of EHEC infection, intestinal tissue and stool samples fr

Galectin is an animal lectin that has high affinity to beta-galactoside of glycoconjugates. In the present study, cellular expression of galectin subtypes in the urinary system of adult mice was examined by in situ hybridization and immunohistochemistry. The major subtype expressed in the murine urinary system was galectin-3, which was expressed continuously from the kidney to the distal end of the urethra. The renal cortex expressed galectin-3 more intensely than the medulla. Renal galectin-3 immunoreactivity was strongest in the cortical collecting ducts, where principal cells were the sole

Galectin-3, a member of beta-galactoside-binding lectins, is expressed and secreted by a variety of cell types including human intestinal epithelial cells. The presence of anti-galectin-3 antibody in the sera of patients was analyzed by immunoblotting using recombinant human galectin-3. A substantially higher percentage of sera from Crohn's disease patients contained anti-galectin-3 IgG autoantibodies than from patients with ulcerative colitis, primary biliary cirrhosis, or autoimmune hepatitis and of apparently healthy control volunteers. In Crohn's disease patients the titer of autoantibodie

Carbohydrate-binding proteins, often called lectins, possess the unique ability to engage other macromolecules through recognition of carbohydrate structures. Among lectins, galectins have emerged as unique mammalian lectins that can possess high specificity for human carbohydrate-based blood group antigens. As such, galectins can differentiate red blood cells (RBCs) based on the blood group status of an individual. The ability of galectins to engage glycans can be studied by a wide variety of approaches. We describe herein the production of galectins and the characterization of galectin glyca…

Circulating monocyte-derived macrophages (MDMø) rapidly invade the brain after stroke, exerting both detrimental and beneficial effects. Elucidating mechanisms that mediate detrimental properties of MDMø may identify therapeutic strategies to divert MDMø from destructive phenotypes, while preserving their favorable effects. Toward this goal, the current study explores the function of Galectin-3 (GAL3) in MDMø and elucidates mechanisms whereby MDMø-derived GAL3 exacerbates stroke injury. In the acutely injured brain, GAL3 expression was upregulated primarily within MDMø. Global KO of GAL3 reduc

BACKGROUND: Intermittent hypoxia (IH), the pathophysiological hallmark of obstructive sleep apnea (OSA), promotes oxidative stress and cardiovascular remodeling. Galectin-3 (Gal-3) is a key mediator of hypoxia-associated inflammatory cardiac injury. ALKBH5, an m6A RNA demethylase detectable in plasma, has not been systematically characterized during hypoxic cardiovascular stress. This experimental study examined plasma ALKBH5 responses to graded IH and evaluated whether pharmacologic Gal-3 inhibition with modified citrus pectin (MCP) modulates circulating ALKBH5 levels and associated cardiac h

The TIM3 receptor acts as an immune checkpoint protein. Canine cancers exhibit higher pan-cancer penetrance of TIM3 compared to PD1, highlighting the potential of TIM3 as a compelling target in comparative immuno-oncology. We have used a highly diverse naïve canine scFv phage library to isolate antibodies to canine TIM3. Alternating rounds of biopanning were performed using either Fc or GST tagged canine TIM3 synthesized in mammalian cells. scFv sequences were identified using colony screening and next generation deep sequencing (NGS). The NGS protocol identified lower abundant frequency clone

Parkinson’s disease (PD) is a chronic and progressive neurodegenerative disorder, characterized by dopaminergic neuronal loss within the substantia nigra pars compacta (SNpc). The gradual progression of neuronal loss results in a significant decline in dopamine levels in the striatum, leading to several hallmarks of motor complications, including resting tremors, rigidity, postural instability, bradykinesia, and a variety of non-motor symptoms, like cognitive decline, mood disturbances, and autonomic dysfunction, reflecting the widespread neurochemical and structural involvement beyond the nig

Candida auris is an emerging multidrug-resistant fungal pathogen that uniquely colonizes the skin. The factors regulating C auris colonization are poorly understood. In this study, we examined the role of galectin-3 during C auris skin infection. We found that deletion of galectin-3 enhances the antifungal activity of neutrophils and increases resistance to C auris skin infection. Mechanistically, galectin-3 negatively regulates GLUT4 expression, glucose uptake, reactive oxygen species production, and fungal killing by neutrophils. Collectively, our findings uncover a detrimental role for gale

The protein galectin-3 is essential for cell migration, activation, and proliferation. It also functions as an inflammatory mediator and triggers caspase-3, which causes brain damage. The purpose of this study was to unveil the phytochemical richness of macadamia nut oil (MO) and assess its potential as a treatment for brain damage caused by potassium dichromate (PDC), as well as its impact on galectin-3/caspase-3 and Sitruin1. The active ingredients of MO were determined by thorough analyses using GC-MS, HPLC, and spectrophotometry. These included oleic acid (47.53%), palmitoleic acid (16.48%

Liver cirrhosis represents a major global health challenge with significant socioeconomic implications. Oxidative stress-mediated injury plays a pivotal role in driving fibrotic progression, with hepatic macrophages serving as a dominant source of reactive oxygen species (ROS). Consequently, targeted suppression of macrophage-derived ROS presents a promising therapeutic strategy. Single-cell RNA sequencing analysis identified Galectin-3 as positively correlated with macrophage oxidative stress and ROS production. Here, a dynamic DNA nano-antioxidant (DDN) is developed for hepatic macrophage-sp
