Antibodies to gliadin as a screening test for coeliac disease. A prospective study.

The diagnostic value of gliadin antibody determination using the fluorescent immunosorbent test was examined in a prospective study of 57 children with gastrointestinal disease. Antibodies to gliadin were found in all 20 patients with active coeliac disease, whereas 7 of these children (37%) had a normal xylose absorption test despite a flat small gut mucosa. Only 4 (14%) of 28 children with other gastrointestinal conditions had antibodies to gliadin, invariably in low titre. After at least 2 years on a gluten-free diet none of 9 children with coeliac disease in remission had demonstrable glia

Gut Morphological Structure-Microbial Characteristics in Elaphodus cephalophus: A Case Report.

The ruminant gastrointestinal tract hosts a complex microbial ecosystem vital for nutrient absorption, with each segment displaying distinct morphological and microbial compositional features compared to monogastric animals. While most studies rely on fecal samples, these fail to capture region-specific variations, limiting insight into microbe-physiology adaptations. In this study, we investigated Elaphodus cephalophus by measuring circular, longitudinal, and mucosal layer thickness across intestinal segments and by profiling bacterial and fungal communities via 16S and its rRNA sequencing. User-safe and efficient chitosan-gated porous carbon nanopesticides and nanoherbicides.

Nanopesticides are selected as one of ten chemical innovations that will change our world. Carboxylated porous carbon nanoparticles (PCNs) were used to encapsulate water-insoluble pesticides and subsequently capped with chitosan (CS) to prepare the CS-gated PCN (PCN@CS) nanopesticides for the controlled release of pesticides in response to acidic pH and elevated temperature with good fungicidal efficacy. To resolve the issue of gastrointestinal absorption of PQ upon ingestion of PQ formulation, it is an innovative strategy to select the carboxylated PCNs as the paraquat (PQ) nanocarriers to in…

An in vitro method for determining the bioaccessibility of pharmaceuticals in wildlife.

Wildlife can be exposed to human pharmaceuticals via prey that have accumulated the compounds from wastewater, surface water, sediment, and soil. One factor affecting internal absorption of pharmaceuticals is bioaccessibility, the proportion of the compound that enters solution in the gastrointestinal tract. Currently, the bioaccessibility of most pharmaceuticals in prey remains unknown for most wildlife species. The authors evaluated the potential of a 2-compartment in vitro gastrointestinal tract model to compare the bioaccessibility of the antidepressant fluoxetine from invertebrate prey fo

[Gastrointestinal dysfunction has important implications for plasma L-dopa concentrations in Parkinson's disease].

Gastrointestinal motility dysfunctions including anorexia, nausea, heartburn, bloating, etc. are common and frequent complication of Parkinson's disease (PD). Degeneration of enteric nerves system is supposed to be a pathogenesis of these symptoms. Impairment of gastric emptying (GE) leads to retardation of the drug delivery from stomach to jejunum, so that PD patients with GE impairment show the delayed elevation of plasma L-dopa concentration. Disturbance of L-dopa absorption will result in wearing-off and delayed-on, and these are called motor fluctuation. In our investigation, 69% of PD pa

Gastropathic sideropenia.

There has been an increasing awareness recently of subtle, non-bleeding gastrointestinal conditions that may result in abnormal iron absorption leading to iron-deficiency anaemia (IDA) in the absence of gastrointestinal symptoms. Thus, the importance of coeliac disease as a possible cause of IDA refractory to oral iron treatment, without other manifestations of malabsorption syndrome, is increasingly being recognized. In addition, Helicobacter pylori has been implicated in several recent studies as a cause of IDA refractory to oral iron treatment, and the anaemia responds favourably to H. pylo

Dietary chromium - forms and availabilities.

The chromium naturally occurring in plants eluted from Sephadex G-25 at approximately 2600 D. Total chromium was quantitated with flameless atomic absorption spectrophotometry. A plant ligand tagged with radioactive chromium both in vivo and in vitro migrated on Sephadex G-25 identically to the naturally occurring chromium compound. The molecular weight of the radioactively tagged chromium compound was 2600 daltons on Sephadex G-25. Similar complexes isolated from plant species were found attached to an organic ligand. The ligand appears to have 2 components, differing in composition by an ami

Anticholinergic suppression of fetal rabbit upper gastrointestinal motility.

OBJECTIVE: At birth the newborn digestive tract must assume the responsibility of assimilating nutrients for survival. Immature gastrointestinal motility in the neonate may result in impaired feeding and nutrition. Newborn gastrointestinal motility development requires the expression and functional maturation of gastrointestinal receptors. To explore the timing of fetal responses to gastrointestinal cholinergic motility agents, we assessed the effect of the anticholinergic agent atropine in the late-gestation rabbit fetus. METHODS: Seven pregnant New Zealand White rabbits were studied at day

Uptake of the colon carcinogen 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine by different segments of the rat gastrointestinal tract: its implication in colorectal carcinogenesis.

2-Amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP) is a colon carcinogen in rats. In the present study the absorption of PhIP in the small and large intestine of Fischer 344 rats was determined, and the relevance of the differences in the degree of absorption of PhIP along the gastrointestinal axis for the PhIP-mediated colon carcinogenesis process is discussed. PhIP uptake was low in the different gut sections of Fischer 344 rats, the PhIP tissue levels varying in the following order: proximal jejunum>distal jejunum>proximal colon>distal colon=rectum. Furthermore, abcc2 was mainly express

In vitro digestion of milk proteins including intestinal brush border membrane peptidases. Transepithelial transport of resistant casein domains.

The use of enzymes from the brush border membrane (BBM) in simulated gastrointestinal digestion of milk proteins has been evaluated. With this purpose, the resistant sequences from casein and milk whey proteins after INFOGEST in vitro digestion with and without BBM have been analyzed by tandem mass spectrometry. The use of BBM revealed additional cleavages to those found with pancreatic enzymes, although the number of total identified peptides decreased due to the reduction of the peptide size. These new cleavages were mainly attributed to the activity of amino- and carboxy-peptidases, which w

The Role of Plasma Membrane Sodium/Hydrogen Exchangers in Gastrointestinal Functions: Proliferation and Differentiation, Fluid/Electrolyte Transport and Barrier Integrity.

The five plasma membrane Na+/H+ exchanger (NHE) isoforms in the gastrointestinal tract are characterized by distinct cellular localization, tissue distribution, inhibitor sensitivities, and physiological regulation. NHE1 (Slc9a1) is ubiquitously expressed along the gastrointestinal tract in the basolateral membrane of enterocytes, but so far, an exclusive role for NHE1 in enterocyte physiology has remained elusive. NHE2 (Slc9a2) and NHE8 (Slc9a8) are apically expressed isoforms with ubiquitous distribution along the colonic crypt axis. They are involved in pHi regulation of intestinal epitheli

ADCC-mediating capacity in children with cow's milk protein intolerance in relation to IgG subclass profile of serum antibodies to beta-lactoglobulin.

In a previous study sera from children with cow's milk protein intolerance (CMPI) exhibiting gastrointestinal symptoms were found to efficiently induce antibody-dependent cell-mediated cytotoxicity (ADCC) to beta-lactoglobulin-coated cells. In contrast, sera from children with coeliac disease showed a low ADCC-mediating capacity, despite high levels of IgG anti-beta-lactoglobulin antibodies. The study described here was undertaken to evaluate whether differences in IgG subclass profile of anti-beta-lactoglobulin antibodies could explain the observed variations in the ADCC-mediating capacity. Tract-wide transcriptomic and metabolomic profiling reveals gastrointestinal regionalization in Jiekarzi semi-fine wool sheep.

Jiekarzi semi-fine-wool sheep are indigenous to the Qinghai-Tibet Plateau, and may face substantial demands on digestive efficiency under low temperatures, hypoxia and seasonal forage limitation. However, the molecular organization of the gastrointestinal tract in this breed remains insufficiently characterized. Here, we profiled mucosal transcriptomes and digestive-fluid metabolomes from 10 gastrointestinal segments collected from six 12-month-old male Jiekarzi. The segments were grouped into four digestive regions: forestomach, true stomach, small intestine and large intestine. Transcriptome

Role of Focal Adhesion Kinase and NRF2 Pathways in Gastrointestinal Wound Healing.

The gastrointestinal epithelium forms a critical barrier that regulates nutrient absorption while preventing the translocation of harmful luminal contents. Disruption of this barrier initiates a coordinated wound healing response involving epithelial restitution, proliferation, and differentiation. Focal adhesion kinase (FAK) is central to this process, regulating focal adhesion (FA) turnover and cytoskeletal dynamics required for epithelial migration. Activation of FAK via phosphorylation at tyrosine 397 (Y397) promotes cell motility, proliferation, and survival, whereas loss of function impa

Induction of glucose intolerance by acute administration of rimonabant.

BACKGROUND/AIMS: Rimonabant is a cannabinoid CB1 receptor antagonist. Other CB1 antagonists have biphasic effects on blood glucose levels following acute administration. We therefore tested the effects of rimonabant on glucose tolerance following acute administration. METHODS: We tested the effects of oral and intracerebroventricular administration of rimonabant on blood glucose and gastrointestinal transit in mice following oral and intravenous glucose challenge. RESULTS: We found a dose-dependent increase in blood glucose from oral doses of rimonabant of 3 mg/kg and above. WIN55,212-2 (3 m

Lactose Intolerance in Adults: Biological Mechanism and Dietary Management.

Lactose intolerance related to primary or secondary lactase deficiency is characterized by abdominal pain and distension, borborygmi, flatus, and diarrhea induced by lactose in dairy products. The biological mechanism and lactose malabsorption is established and several investigations are available, including genetic, endoscopic and physiological tests. Lactose intolerance depends not only on the expression of lactase but also on the dose of lactose, intestinal flora, gastrointestinal motility, small intestinal bacterial overgrowth and sensitivity of the gastrointestinal tract to the generatio
