Systematic analysis of the basic/helix-loop-helix (bHLH) transcription factor family in pummelo (Citrus grandis) and identification of the key members involved in the response to iron deficiency.

BACKGROUND: Iron (Fe) deficiency is a common problem in citrus production. As the second largest superfamily of transcription factors (TFs), the basic/helix-loop-helix (bHLH) proteins have been shown to participate in the regulation of Fe homeostasis and a series of other biological and developmental processes in plants. However, this family of members in citrus and their functions in citrus Fe deficiency are still largely unknown. RESULTS: In this study, we identified a total of 128 CgbHLHs from pummelo (Citrus grandis) genome that were classified into 18 subfamilies by phylogenetic comparis

Characterization of Rice NADPH oxidase genes and their expression under various environmental conditions.

Plasma membrane NADPH oxidases (Noxs) are key producers of reactive oxygen species under both normal and stress conditions in plants. We demonstrate that at least eleven genes in the genome of rice (Oryza sativa L.) were predicted to encode Nox proteins, including nine genes (OsNox1-9) that encode typical Noxs and two that encode ancient Nox forms (ferric reduction oxidase 1 and 7, OsFRO1 and OsFRO7). Phylogenetic analysis divided the Noxs from nine plant species into six subfamilies, with rice Nox genes distributed among subfamilies I to V. Gene expression analysis using semi-quantitative RT-

Expression profiling of two stress-inducible genes encoding for miraculin-like proteins in citrus plants under insect infestation or salinity stress.

The expression of two genes, namely Mir1 and Mir3 and the abundance of their encoded proteins, the putative miraculin-like proteins, MLP1 and MLP3, showing similarity to the Kunitz family of protease inhibitors, were monitored in the leaves of the citrus variety, 'Clementine' after Tetranychus urticae infestation and elicitor treatments, or in the leaves of three other diploid citrus: 'Willow leaf', 'Cleopatra' mandarins and 'Trifoliate' orange, as well as their respective doubled diploids and the allotetraploid somatic hybrid 'FLHORAG1' under salt stress. RT-PCR and 2-DE indicated that Mir1 a

Resveratrol and nitric oxide synergistically enhance resistance against B. cinerea in tomato fruit by regulating phytohormones.

Resveratrol (RVT), a plant antitoxin, plays an important role in plant resistance against pathogens. While nitric oxide (NO) as an essential signaling factor in disease resistance enhancement is well documented, the potential molecular interplay RVT and NO in postharvest tomato fruit defense against Botrytis cinerea (B.cinerea) still needs exploration. In this study, exogenous RVT reduced gray mold caused by B.cinerea in tomato fruit, with 20 μM being the most effective. Tomato fruit were treated with 20 μM RVT enhanced resistance against B.cinerea, as indicated by reduced symptoms of disease

[Nonexpressor of pathogenesis-related genes 1 (NPR1): a key node of plant disease resistance signalling network].

The NPRI (nonexpressor of pathogenesis-related genes (1) gene, firstly cloned in Arabidopsis thaliana, is a key gene involved in regulation of plant disease resistance. It plays a pivotal role not only in systemic acquired resistance (SAR) and induced systemic resistance (ISR), but also in basic resistance and resistance (R) gene-dependent resistance. NPR1 monomerization induced by enhanced reducing condition after oxidative burst, and the accumulation of NPR1 monomers in the nuclei, are required and enough for expression of PR (pathogenesis-related) genes and SAR. NPR1 regulates PR gene expre

MicroRNA396a-5p and -3p induce tomato disease susceptibility by suppressing target genes and upregulating salicylic acid.

Plants have evolved a variety of mechanisms to perceive and resist the assault of pathogens. The biotrophs, necrotrophs and hemibiotrophs are types of plant pathogens that activate diverse salicylic acid (SA) and jasmonic acid (JA) signaling pathways. In this study we showed that the expressions of miR396a-5p and -3p in Solanum lycopersicum (S. lycopersicum) were both down-regulated after infection by hemibiotroph Phytophthora infestans (P. infestans) and necrotroph Botrytis cinerea (B. cinerea) infection. Overexpression of miR396a-5p and -3p in transgenic tomato enhanced the susceptibility of

The immune repressor BIR1 contributes to antiviral defense and undergoes transcriptional and post-transcriptional regulation during viral infections.

BIR1 is a receptor-like kinase that functions as a negative regulator of basal immunity and cell death in Arabidopsis. Using Arabidopsis thaliana and Tobacco rattle virus (TRV), we investigate the antiviral role of BIR1, the molecular mechanisms of BIR1 gene expression regulation during viral infections, and the effects of BIR1 overexpression on plant immunity and development. We found that SA acts as a signal molecule for BIR1 activation during infection. Inactivating mutations of BIR1 in the bir1-1 mutant cause strong antiviral resistance independently of constitutive cell death or SA defens

Gene expression analysis of rice seedling under potassium deprivation reveals major changes in metabolism and signaling components.

Plant nutrition is one of the important areas for improving the yield and quality in crops as well as non-crop plants. Potassium is an essential plant nutrient and is required in abundance for their proper growth and development. Potassium deficiency directly affects the plant growth and hence crop yield and production. Recently, potassium-dependent transcriptomic analysis has been performed in the model plant Arabidopsis, however in cereals and crop plants; such a transcriptome analysis has not been undertaken till date. In rice, the molecular mechanism for the regulation of potassium starvat

Molecular characterization of a miraculin-like gene differentially expressed during coffee development and coffee leaf miner infestation.

The characterization of a coffee gene encoding a protein similar to miraculin-like proteins, which are members of the plant Kunitz serine trypsin inhibitor (STI) family of proteinase inhibitors (PIs), is described. PIs are important proteins in plant defence against insects and in the regulation of proteolysis during plant development. This gene has high identity with the Richadella dulcifica taste-modifying protein miraculin and with the tomato protein LeMir; and was named as CoMir (Coffea miraculin). Structural protein modelling indicated that CoMir had structural similarities with the Kunit

miR160a-ARF4 module regulates the biosynthesis of Panax notoginseng saponins.

Panax notoginseng is an economically vital medicinal plant, prized for its bioactive P. notoginseng saponins (PNS), which possess significant therapeutic effects in cardiovascular and cerebrovascular diseases. Despite the elucidation of the core enzymatic steps in PNS biosynthesis, the posttranscriptional mechanisms that coordinate these metabolic fluxes remain largely obscure. This study explored the Pn-miR160a-ARF4 module as a pivotal regulation mode in the biosynthesis of PNS. Under methyl jasmonate stimulation, Pn-miR160a was significantly upregulated, subsequently targeting and cleaving t

Over-expression of rice R1-type MYB transcription factor confers different abiotic stress tolerance in transgenic Arabidopsis.

Among various abiotic stresses, water deficit hit the first in the list followed by heavy metal stresses as a serious environmental growth-limiting factor that restricts the global crop yield. Molecular approaches will help us to trace key regulators which are involved in stress-related phenomenon to enhance crop productivity. The present study functionally characterized one of the key regulators, OsMYB-R1 in Arabidopsis. Phylogenetic analyses indicated that OsMYB-R1 had a close relationship with Sorghum bicolour and Zea mays. Ectopic expression of OsMYB-R1 in Arabidopsis resulted in improved

Phytomelatonin as a central molecule in plant disease resistance.

Melatonin is an essential phytohormone in the regulation of many plant processes, including during plant development and in response to stress. Pathogen infections cause serious damage to plants and reduce agricultural production. Recent studies indicate that melatonin plays important roles in alleviating bacterial, fungal, and viral diseases in plants and post-harvest fruits. Herein, we summarize information related to the effects of melatonin on plant disease resistance. Melatonin, reactive oxygen species, and reactive nitrogen species form a complex loop in plant-pathogen interaction to reg

Genome-Wide Identification, Gene Structure and Expression Analysis of the MADS-Box Gene Family Indicate Their Function in the Development of Tobacco (Nicotiana tabacum L.).

MADS-box genes play a pivotal role in various processes, including floral and seed development, controlling flowering time, regulation of fruits ripening, and respond to abiotic and biotic stressors in planta. Tobacco (Nicotiana tabacum) has been widely used as a model plant for analyzing the gene function, however, there has been less information on the regulation of flowering, and the associated genes. In the present study, a total of 168 NtMADS-box genes were identified from tobacco, and their phylogenetic relationship, chromosome locations, and gene structures were further analyzed. NtMADS

AtATM3 is involved in heavy metal resistance in Arabidopsis.

AtATM3, an ATP-binding cassette transporter of Arabidopsis (Arabidopsis thaliana), is a mitochondrial protein involved in the biogenesis of iron-sulfur clusters and iron homeostasis in plants. Our gene expression analysis showed that AtATM3 is up-regulated in roots of plants treated with cadmium [Cd(II)] or lead (II); hence, we investigated whether this gene is involved in heavy metal tolerance. We found that AtATM3-overexpressing plants were enhanced in resistance to Cd, whereas atatm3 mutant plants were more sensitive to Cd than their wild-type controls. Moreover, atatm3 mutant plants expres

CRISPR-Cas9-mediated mutagenesis of the SlSRM1-like gene leads to abnormal leaf development in tomatoes.

BACKGROUND: Leaves, which are the most important organs of plants, can not only fix carbon sources through photosynthesis, but also absorb nutrients through transpiration. Leaf development directly determines the growth, flowering and fruiting of plants. There are many factors that affect leaf development, such as the growth environment, gene expression, and hormone synthesis. In this study, tomatoes were used to study the role of the transcription factor Solanum lycopersicum salt-related MYB1-like (SlSRM1-like) in the development of tomato leaves. RESULTS: Loss-of-function of the SlSRM1-like

Genome-Wide Identification of NLP Family Genes in Cultivated Strawberry (Fragaria × ananassa Duch.) and Analysis of Their Expression Under Heat and Botrytis cinerea Stresses.

Nodule inception (NIN)-like proteins (NLPs) are plant-specific transcription factors regulating nutrient absorption, growth, and stress tolerance; however, their roles in stress responses remain largely uncharacterized. Cultivated strawberry (Fragaria × ananassa 'Camarosa') serves as an ideal model for dissecting the evolution and function of NLP genes. In this study, 37 FaNLP genes were identified genome-wide. Phylogenetic analysis classified them into three subfamilies, which are evenly distributed across seven chromosomes. Divergent exon-intron structures and conserved motif compositions su

Soybean gene GmMLP34 regulates Arabidopsis negative response to high temperature stress.

The functions of major latex proteins (MLPs) in plant defense and stress responses have been widely documented; however, their roles in HT stress response in soybeans have not been elucidated. This study investigated the role of GmMLP34, a member of the major latex protein (MLP) family, in the response of soybeans to HT stress. Transcriptome analysis of HT-resistant (JD21) and HT-sensitive (HD14) soybean leaves under HT stress (43.40 ± 1.70 °C) and field conditions revealed differential expression of GmMLP34. Further examination across different HT-resistant varieties showed that GmMLP34 was d

PmLBD3 links auxin and brassinosteroid signalling pathways on dwarfism in Prunus mume.

BACKGROUND: Grafting with dwarf rootstock is an efficient method to control plant height in fruit production. However, the molecular mechanism remains unclear. Our previous study showed that plants with Prunus mume (mume) rootstock exhibited a considerable reduction in plant height, internode length, and number of nodes compared with Prunus persica (peach) rootstock. The present study aimed to investigate the mechanism behind the regulation of plant height by mume rootstocks through transcriptomic and metabolomic analyses with two grafting combinations, 'Longyan/Mume' and 'Longyan/Peach'. Examining Short-Term Responses to a Long-Term Problem: RNA-Seq Analyses of Iron Deficiency Chlorosis Tolerant Soybean.

Iron deficiency chlorosis (IDC) is a global crop production problem, significantly impacting yield. However, most IDC studies have focused on model species, not agronomically important crops. Soybean is the second largest crop grown in the United States, yet the calcareous soils across most of the upper U.S. Midwest limit soybean growth and profitability. To understand early soybean iron stress responses, we conducted whole genome expression analyses (RNA-sequencing) of leaf and root tissue from the iron efficient soybean (Glycine max) cultivar Clark, at 30, 60 and 120 min after transfer to ir

The primary module in Norway spruce defence signalling against H. annosum s.l. seems to be jasmonate-mediated signalling without antagonism of salicylate-mediated signalling.

A key tree species for the forest industry in Europe is Norway spruce [Picea abies (L.) Karst.]. One of its major diseases is stem and butt rot caused by Heterobasidion parviporum (Fr.) Niemelä & Korhonen, which causes extensive revenue losses every year. In this study, we investigated the parallel induction of Norway spruce genes presumably associated with salicylic acid- and jasmonic acid/ethylene-mediated signalling pathways previously observed in response to H. parviporum. Relative gene expression levels in bark samples of genes involved in the salicylic acid- and jasmonic acid/ethylene-me
