A Mendelian randomization study to reveal gut-disc axis: causal associations between gut microbiota with intervertebral disc diseases.

PURPOSE: Emerging evidence suggests a link between gut microbiota and intervertebral disc diseases (IDDs); however, the causal relationships remain unclear. This study aimed to evaluate the causal effects of gut microbiota on the risk of cervical disc disorders (CDD), other intervertebral disc disorders (OIDD), pyogenic intervertebral disc infections, and discitis, shedding light on the potential "gut-disc axis". METHODS: Genetic variation data for 202 gut microbiota taxa were obtained from the Dutch Microbiome Project, and disease outcome data were sourced from the FinnGen consortium. A Mend

HLA-B*27 and Ankylosing Spondylitis: 50 Years of Insights and Discoveries.

PURPOSE OF REVIEW: To commemorate the 50th anniversary of the groundbreaking discovery of a remarkably strong association between HLA-B*27 and ankylosing spondylitis (AS). RECENT FINDINGS: In addition to HLA-B*27, more than 116 other recognized genetic risk variants have been identified, while epigenetic factors largely remain unexplored in this context. Among patients with AS who carry the HLA-B*27 gene, clonally expanded CD8 + T cells can be found in their bloodstream and within inflamed tissues. Moreover, the α and β chain motifs of these T-cell receptors demonstrate a distinct affinity fo

MALT lymphoma: A paradigm of NF-κB dysregulation.

Extranodal marginal zone lymphoma of mucosa-associated lymphoid tissue (MALT lymphoma) invariably arises from a background of chronic microbial infection and/or autoimmune disorder at diverse mucosal sites. The prolonged chronic infection and/or autoimmunity generate active immune and inflammatory responses that provide a setting for evolution and development of autoreactive B-cells, their expansion and eventual malignant transformation following acquisition of genetic changes. The immune responses also play a critical role in sustaining the growth and survival of the transformed cells as show

Substitutive Hospital at Home for older persons: effects on costs.

Emerging evidence suggests that gut microbiota is associated with functional gastrointestinal disorders (FGIDs). However, findings regarding microbial alterations in FGIDs have been inconsistent across observational studies, and the direction and causality of these associations remain unclear. We conducted a bidirectional two-sample Mendelian randomization (MR) study to investigate potential causal associations between gut microbiota and two common FGIDs, irritable bowel syndrome (IBS) and functional dyspepsia (FD). Genome-wide association study (GWAS) summary statistics of gut microbiota, IBS

HLA-B27 expression modulates gram-negative bacterial invasion into transfected L cells.

The mechanism by which HLA-B27 confers genetic susceptibility to the seronegative spondyloarthropathies ankylosing spondylitis, Reiter's syndrome, and reactive arthritis, is not well understood. The current concept of an extraarticular bacterial infection functioning as the triggering event in a genetically susceptible host suggests the possibility of direct microbial-MHC interaction. We have addressed the role of HLA-B27 in microbial-host cell interaction by examining invasion by putatively arthritogenic gram-negative bacteria. Target cells used were murine L cells transfected with HLA-B27, H

Molecular mechanisms of Polyendocrine Metabolic Ovarian Syndrome and identification of novel druggable targets for drug discovery and repositioning.

Polyendocrine Metabolic Ovarian Syndrome (PMOS) is a heterogeneous disorder that mainly affects women of reproductive age. Genetic predisposition, insulin resistance, androgen excess, inflammation, mitochondrial dysfunction, and changes in gut microbial composition were notable contributors to the prognosis of PMOS. Several genetic variations, including mutations, polymorphisms, and variations in gene regulation, have been identified that predispose individuals to PMOS. A complex network involving metabolic, hormonal, and inflammatory pathways plays a major role in disease progression. To iden

Genetic risk, dysbiosis, and treatment stratification using host genome and gut microbiome in inflammatory bowel disease.

OBJECTIVES: Inflammatory bowel diseases (IBD), comprised of Crohn's disease (CD) and ulcerative colitis (UC), are characterized by a complex pathophysiology that is thought to result from an aberrant immune response to a dysbiotic luminal microbiota in genetically susceptible individuals. New technologies support the joint assessment of host-microbiome interaction. METHODS: Using whole genome sequencing and shotgun metagenomics, we studied the clinical features, host genome, and stool microbial metagenome of 85 IBD patients, and compared the results to 146 control individuals. Genetic risk sc

The interaction between host genetics and the microbiome in the pathogenesis of spondyloarthropathies.

PURPOSE OF REVIEW: The intestinal microbiome is increasingly implicated in the pathogenesis of ankylosing spondylitis, reactive arthritis, and other diseases collectively known as the spondyloarthropathies (SpAs). In common with other complex inflammatory diseases, SpAs have both a strong genetic and environmental component. Recent genetic studies have highlighted host pathways that may intersect the host-microbiota interaction and offer novel paradigms to understand the pathophysiology of these diseases. RECENT FINDINGS: Genetic association studies have identified genes such as RUNX3, PTPEN2

Heart of the matter-infection and xenotransplantation.

In this clinicopathological conference, invited experts discussed a previously published case of a patient with nonischemic cardiomyopathy who underwent heart transplantation from a genetically modified pig source animal. His complex course included detection of porcine cytomegalovirus by plasma microbial cell-free DNA and eventual xenograft failure. The objectives of the session included discussion of selection of immunosuppressive regimens and prophylactic antimicrobials for human xenograft recipients, description of infectious disease risk assessment and mitigation in potential xenograft do

Inhalant and Additional Mucosal-Related Environmental Risks for Rheumatoid Arthritis.

Rheumatoid arthritis (RA) occurs as the result of a complex interplay of environmental factors in a genetically susceptible individual. There is considerable evidence that the lungs may serve as an initial site of tolerance loss in the generation of RA-related autoimmunity, and several environmental inhalant exposures and lung diseases have been associated with RA risk. There is additional evidence that immune and microbial dysregulation of other mucosal sites, including the oral and gastrointestinal mucosa, may contribute to the development of RA. Epidemiologic evidence linking mucosal exposu

Histologic Review of Sarcoidosis in a Neck Lymph Node.

Sarcoidosis is a multisystem granulomatous disease of unknown etiology. It may occur at any age, but is most commonly seen in young to middle age adults. Sarcoidosis remains more common in women regardless of geographic or racial boundaries. Although the etiology remains unclear, the most common explanation is that sarcoidosis is a disease of immunologic dysregulation triggered by an as yet unidentified environmental or microbial antigen in genetically susceptible persons. We review a case of sarcoidosis with was initially diagnosed in a neck lymph node that was removed for evaluation of metas

Avian paramyxovirus type 1-associated severe pneumonia in humans: Molecular characterization and zoonotic transmission risk.

BACKGROUND: Avian paramyxovirus serotype 1 (APMV-1, Newcastle disease virus) is a major poultry pathogen. Human infections are rare and typically self-limiting, but its potential to cause severe respiratory disease and the mechanisms underlying cross-species transmission remain understudied. METHODS: We analyzed a 65-year-old male with severe pneumonia who had contact with sick backyard feeder chickens. Immunocompetence was evaluated via routine blood tests and serum immunoglobulin levels. mNGS identified 40 APMV-1 sequence reads (50.6% microbial abundance) covering 10.78% of the genome. APMV

Decoding Biothreats With FT-ICR-MS: Metabotyping of Bacillus cereus Spores Through Untargeted Metabolomics.

Genome sequencing is the gold standard for microbial identification. However, it fails to distinguish closely related species with overlapping genetic features. This is the case of Bacillus cereus and Bacillus cytotoxicus, two members of the B. cereus group with distinct pathogenic profiles. While B. cereus, a well-known opportunistic pathogen involved in intestinal and extraintestinal infections, produces the tissue-destructive exoenzymes hemolysin BL (HBL), nonhemolytic enterotoxin (NHE), and cytotoxin K (CytK-2) as well as the plasmid-encoded emetic toxin cereulide, the thermotolerant speci

Decoding fungal communication networks: molecular signaling, genetic regulation, and ecological implications.

Fungal communication networks regulate essential biological processes, enabling fungi to adapt to environmental changes, coordinate development, and establish interactions within microbial communities. These networks are mediated by diverse signaling molecules, including volatile organic compounds (VOCs), peptide signaling molecules, and quorum-sensing molecules, which facilitate intra- and interspecies communication. The intricate regulation of these signals occurs through specialized signal transduction pathways such as G-protein-coupled receptors (GPCRs) and two-component regulatory systems

Evolutionary Systems Biology Identifies Genetic Trade-offs in Rice Defense against Aboveground and Belowground Attackers.

Like other plants, wild and domesticated rice species (Oryza nivara, O. rufipogon, and O. sativa) evolve in environments with various biotic and abiotic stresses that fluctuate in intensity through space and time. Microbial pathogens and invertebrate herbivores such as plant-parasitic nematodes and caterpillars show geographical and temporal variation in activity patterns and may respond differently to certain plant-defensive mechanisms. As such, plant interactions with multiple community members may result in conflicting selection pressures on genetic polymorphisms. Here, through assays with

Dietary fat, bile acid metabolism and colorectal cancer.

Colorectal cancer (CRC) risk is predominantly driven by environmental factors, in particular diet. A high intake of dietary fat has been implicated as a risk factor inducing the formation of pre-neoplastic lesions (e.g., adenomatous polyps) and/or exacerbating colonic tumorigenesis. Recent data attributed the tumor-promoting activity of high-fat diets to their effects on gut microbiota composition and metabolism, in particular with regard to bile acids. Bile acids are synthesized in the liver in response to dietary fat and facilitate lipid absorption in the small intestine. The majority of bil

A Proposal for Formation of Archaean Stromatolites before the Advent of Oxygenic Photosynthesis.

Stromatolites are solid, laminar structures of biological origin. Living examples are sparsely distributed and formed by cyanobacteria, which are oxygenic phototrophs. However, stromatolites were abundant between 3.4 and 2.4 Gyr, prior to the advent of cyanobacteria and oxygenic photosynthesis. Here I propose that many Archaean stromatolites were seeded at points of efflux of hydrogen sulfide from hydrothermal fields into shallow water, while their laminar composition arose from alternating modes of strictly anoxygenic photosynthetic metabolism. These changes were a redox regulatory response o

Maternal diet and genetics shape the human milk metabolome.

Human milk contains a diverse array of metabolites that contribute to infant nutrition, immune development, and microbial colonization. The maternal factors shaping the milk metabolome, and the relative contribution of genetics or diet vs. other factors, remain poorly understood. Here, we profiled 458 milk metabolites in 349 one-month postpartum human milk samples and integrated metabolomic data with maternal diet, clinical, transcriptomic, and genomic measurements. Maternal diet was broadly associated with milk metabolite composition, with significant correlations identified between dietary f

Bibliometric analysis of human microbiota-associated animal model (2005-2025).

BACKGROUND: The research on human microbiota-associated (HMA) animal models is an important tool for studying the human microbiome and holds great potential for elucidating disease mechanisms and microbe-based therapeutic interactions. However, a systematic bibliometric assessment of this field has been limited. METHODS: This study employed bibliometric methods, retrieving relevant publications published between 2005 and 2025 from the Web of Science Core Collection, Scopus, and PubMed, and visualizing the data with VOSviewer and CiteSpace. RESULTS: The analysis revealed a continuous upward t

The oral-vascular axis: immune mechanisms linking periodontal dysbiosis to systemic vascular pathology.

Periodontitis is among the most prevalent chronic inflammatory diseases worldwide and may affect vascular health beyond the oral cavity. Framed within the concept of an oral-vascular axis, this review synthesizes clinical and mechanistic evidence linking periodontal disease with atherosclerotic cardiovascular disease (ASCVD). Epidemiological studies and meta analyses consistently associate periodontitis with higher risks of coronary heart disease (CHD), stroke, and cardiovascular mortality, with modest but reproducible effect sizes that persist after adjustment for traditional risk factors.
