Identification and characterization of a novel p. S390C variant in TMC1 in an autosomal recessive family with non-syndromic hearing loss.

BACKGROUND: TMC1 variants cause autosomal dominant (DFNA36) and recessive (DFNB7/11) sensorineural hearing loss. However, the clinical significance of novel missense variants, particularly in East Asian populations, remains unclear. OBJECTIVE: Our study aims to clarify the pathogenicity of a novel variant p.S390C by clinical characterization, genetic and molecular model analysis in China. MATERIAL AND METHODS: Auditory features of the hearing loss were characterized in proband. Variant screening of 406 known deafness-associated genes was performed in proband by targeted next-generation seque

Abnormal white and gray matter functional connectivity is associated with cognitive dysfunction in presbycusis.

Presbycusis is characterized by high-frequency hearing loss and is closely associated with cognitive decline. Previous studies have observed functional reorganization of gray matter in presbycusis, but the information transmission between gray matter and white matter remains ill-defined. Using resting-state functional magnetic resonance imaging, we investigated differences in functional connectivity (GM-GM, WM-WM, and GM-WM) between 60 patients with presbycusis and 57 healthy controls. Subsequently, we examined the correlation between these connectivity differences with high-frequency hearing

Mood and Cognitive Disorders Following Hearing Loss: Impact of Hearing Aid Timing.

Background: Hearing loss is one of the most common yet often overlooked sensory deficits worldwide, with consequences extending well beyond auditory function. Mounting evidence highlights the complex interrelationships among hearing loss, cognitive decline, and psychosocial well-being. Neural mechanisms underlying this association include increased cognitive load, cortical reorganisation, and social isolation, which mediate the impact of auditory deprivation on the brain and mental health. Furthermore, hearing impairment is consistently associated with a higher risk of depression and anxiety,

Slc26a4-insufficiency causes fluctuating hearing loss and stria vascularis dysfunction.

SLC26A4 mutations can cause a distinctive hearing loss phenotype with sudden drops and fluctuation in patients. Existing Slc26a4 mutant mouse lines have a profound loss of hearing and vestibular function, with severe inner ear malformations that do not model this human phenotype. In this study, we generated Slc26a4-insufficient mice by manipulation of doxycycline administration to a transgenic mouse line in which all Slc26a4 expression was under the control of doxycycline. Doxycycline was administered from conception to embryonic day 17.5, and then it was discontinued. Auditory brainstem respo

[Active hearing implants in chronic otitis media].

In patients with inadequate hearing improvement after tympanoplasty and failure of conventional hearing aid fitting, active hearing implants provide an alternative treatment option. Active middle ear implants function as a vibromechanical bypass of the stiffness and damping effect of a poorly oscillating tympanic membrane and the (reconstructed) ossicular chain. The selection of the hearing system depends on the maximum output levels of the hearing system and the anatomical conditions in mostly multiply operated ears. The development of variable coupling elements for active middle ear implants

Audiological Findings in Hemodialysis Patients.

Hearing loss in end-stage chronic renal failure is more common than in the general population. The etiopathogenetic mechanisms reported included the use of ototoxic drugs, hypertension, fluid and electrolyte disorders associated with kidney failure, and factors related to hemodialysis itself, such as acute hypotension, reduced blood osmotic pressure, acute urea clearance, and the immunological reaction caused by dialyzer membranes. The aim of our study was to evaluate auditory function in patients with end-stage chronic renal failure, to study the prevalence of hearing loss and the factors tha

Age-related hearing loss and dementia risk across the lifespan: mechanisms, equity, and prevention.

Age-related hearing loss (ARHL) has emerged as a significant and potentially modifiable risk factor for neurodegenerative disorders, including Alzheimer's disease. A growing body of evidence links ARHL to structural and functional changes in the brain, with implications for cognitive decline and dementia onset. However, both ARHL and dementia are multifactorial conditions shaped not only by biological mechanisms but also by broader social determinants of health. Inequities in access to hearing care, underrepresentation of marginalised populations in research, and variability in intervention ou

Prestin as a context-dependent biomarker of outer hair cell stress: Integrating preclinical and clinical evidence.

The early detection of hearing loss remains a major clinical challenge. In many patients, once functional changes become evident on pure-tone audiometry, their recovery is limited. Outer hair cells (OHCs) are essential for cochlear amplification, and their injury is a common pathway in many forms of sensorineural hearing loss. However, reliable biomarkers that reflect hearing impairment in real-time are still lacking. Prestin, a motor protein located in the lateral membrane of OHCs, has been proposed as a potential marker of cochlear stress, but findings across studies vary, and its clinical s

The Antioxidant Effect of Rosmarinic Acid by Different Delivery Routes in the Animal Model of Noise-Induced Hearing Loss.

HYPOTHESIS: Trans-tympanic Rosmarinic Acid (RA), as compared with the systemic administration, protects against noise-induced auditory hair cell and hearing losses in rats in vivo. BACKGROUND: ROS production, lipoperoxidative damage, and an imbalance of antioxidant defences play a significant role in noise-induced hearing loss. Several molecules with antioxidant properties have been tested to restore redox homeostasis; however, drug delivery system represents a challenge for their effectiveness. In our model, acute and intense noise exposure induces hearing loss, hair cell death, and oxidativ

The impact of hearing aids on cognitive function and quality of life in patients with hearing impairment: A cross-sectional study.

OBJECTIVE: Age-related cognitive decline involves a complex set of factors. Among these factors, hearing loss is considered to have a significant impact, but the effect of hearing aid use remains unresolved. The purpose of this study was to evaluate the effects of hearing aid use by simultaneously assessing various factors not only cognitive function but also frailty, anxiety, depression, and quality of life (QOL) in patients with hearing loss. METHODS: The cross-sectional study at the Hearing Aid (HA) Center was conducted between 2020 and 2021. Initially, associations with cognitive function

Genetic studies identify known and novel variants for recessively inherited moderate to severe hearing loss in consanguineous families from Pakistan.

Variants of at least twenty-seven human genes are associated with stable or progressive nonsyndromic moderate to severe hearing loss inherited as a recessive condition. We ascertained 54 consanguineous families, predominantly from the Punjab province in Pakistan, with multiple individuals affected by moderate to severe or progressively profound hearing loss. After Sanger sequencing identified 4 families with GJB2 variants or a non-coding pathogenic variant in HGF, exome sequencing was carried out for selected samples from the remaining 50 families. Analyses revealed a total of 24 novel and 33

Cognitive decline and increased hippocampal p-tau expression in mice with hearing loss.

Hearing and cognition are commonly involved in both normal and pathological aging. Current clinical interest lies in whether peripheral hearing loss promotes cognitive decline. In our previous publication, the authors have shown a causal relationship between hearing and cognitive impairments in C57BL/6 mice. Here we extended the follow-up to 12 months to determine the long-term effects of hearing loss on cognition and to observe hippocampal p-tau and lipofuscin. One month old male mice were randomly allocated into two groups, the control (n = 12) and noise-induced hearing loss (NIHL) (n = 12).

Hearing and visual impairments.

Hearing impairments, with their resultant difficulties in communication function, represent a common chronic condition affecting one third of aged individuals. Loss of hearing ability pervades multiple domains of function in aged individuals, decreasing activity, increasing depressive symptoms, and confounding assessment of cognitive ability. Although they are not curable, hearing impairments are often remediable with appropriate audiologic evaluation and the prescription of amplification devices. The role of the geriatrician in managing hearing impairment is to recognize its presence, provide
