Cellular concentrations of glutamine synthetase in murine organs.

Glutamine synthetase (GS) is the only enzyme that can synthesize glutamine, but it also functions to detoxify glutamate and ammonia. Organs with high cellular concentrations of GS appear to function primarily to remove glutamate or ammonia, whereas those with a low cellular concentration appear to primarily produce glutamine. To validate this apparent dichotomy and to clarify its regulation, we determined the GS concentrations in 18 organs of the mouse. There was a >100-fold difference in GS mRNA, protein, and enzyme-activity levels among organs, whereas there was only a 20-fold difference in

Neurobehavioral deficits, oxidative dysregulation, and inflammatory responses in male Swiss mice following sub-acute exposure to ethanol extract of Leiotrametes lactinea Berk.

BACKGROUND: Leiotrametes lactinea (Berk.) Welti & Courtec. a mushroom reported to possess antimicrobial, antioxidant, antitumor and analgesic activities. However, there is limited information on its safety profile in the central nervous system. This study thus evaluates the neurotoxic effect of ethanol extract of Leiotrametes lactinea (EELL) in male Swiss mice following 30 days oral exposure. METHOD: Twenty-four (24) Swiss male mice were divided into 4 groups (n = 6) and orally administered EELL for 30 days at doses of 50, 100, and 200 mg/kg; the last group served as the healthy control. Synthesis, biological evaluation and in silico studies of novel propargyl-tethered isatin hydrazones as monoamine oxidase inhibitors for Parkinson's disease.

Neurodegenerative disorders such as Parkinson's disease are closely associated with dysregulated activity of monoamine oxidase-B (MAO-B), which leads to dopamine depletion and oxidative stress. Despite the availability of numerous monoamine oxidase (MAO) inhibitors on the market, their irreversibility and associated side effects necessitate the development of more effective and reversible MAO-B inhibitors. In this study, a series of twenty-one indole-based derivatives (PSH1-PSH21), collectively designated as PSH, was synthesised and evaluated for inhibitory activity against MAO isoforms. Most

Extension of cytokines' role in Behcet's disease associated peripheral neuropathy.

BACKGROUND/OBJECTIVE: Behçet's disease (BD) is a chronic multisystem inflammatory disorder with diverse neurological manifestations. While central nervous system involvement is well recognized, peripheral neuropathy remains an underdiagnosed and poorly understood complication. Chemokines, particularly C-C motif chemokine ligand 21 (CCL21), play a critical role in immune-mediated neuroinflammation and have been implicated in several autoimmune diseases. This study aimed to investigate the association between selected CCL21 gene polymorphisms and BD-associated peripheral neuropathy, and to asses

Organization of sensorimotor activity in anterior cruciate ligament reconstructed individuals: an fMRI conjunction analysis.

INTRODUCTION: Anterior cruciate ligament reconstruction (ACLR) is characterized by persistent involved limb functional deficits that persist for years despite rehabilitation. Previous research provides evidence of both peripheral and central nervous system adaptations following ACLR. However, no study has compared functional organization of the brain for involved limb motor control relative to the uninvolved limb and healthy controls. The purpose of this study was to examine sensorimotor cortex and cerebellar functional activity overlap and non-overlap during a knee motor control task between

Atomoxetine promotes incentive value of modafinil and sensitizes exploratory behavior.

Substance dependence is a disorder that alters the functioning of the nervous system due to frequent abuse of drugs. The role of dopamine in the addictive effect of psychostimulants is well known; however, the involvement of the noradrenergic system is still unclear and poorly understood, though drugs like cocaine and amphetamines are known to exert significant activity on this system. The drug modafinil (MOD) has no proven addictive effect. It promotes wakefulness by acting mainly on the dopaminergic system and, to a lesser degree, the noradrenergic (NOR) system. Atomoxetine (ATX) is a non-st

Plasma and Urinary Levels of Nerve Growth Factor Are Elevated in Primary Hypertension.

Nerve growth factor (NGF) is the main neurotrophic factor that can control sympathetic nerve innervation and sympathetic neural activity in cardiovascular organs. Although NGF overproduction and its influences on the sympathetic nervous system have been shown in hypertensive animals, NGF status and its association with sympathetic nerve activity have not yet been explored in human hypertension. In the present study, therefore, plasma and urinary levels of NGF and those of catecholamines (i.e., indices for NGF status and sympathoadrenal activity, respectively) were compared between 83 untreated

Somatization, but not depression, is characterized by disorders in the tryptophan catabolite (TRYCAT) pathway, indicating increased indoleamine 2,3-dioxygenase and lowered kynurenine aminotransferase activity.

BACKGROUND: Reduced plasma tryptophan occurs in depression and somatization. Induction of indoleamine 2,3-dioxygenase (IDO) with consequent synthesis of tryptophan catabolites (TRYCATs) and lowered tryptophan are associated with the onset of depression in the puerperium and during interferon-alpha treatment. Depression is accompanied by lowered kynurenic acid, a neuroprotectant, or increased kynurenine, a neurotoxic TRYCAT. AIMS AND METHODS: To examine plasma tryptophan; kynurenine; kynurenic acid; the kynurenine / tryptophan (KY/TRP) ratio, indicating IDO activity; and the kynurenine / kynur

Phosphorylation of Pyruvate Dehydrogenase as a Marker of Neuronal Inactivity in the Enteric Nervous System.

BACKGROUND: The enteric nervous system (ENS) regulates essential gut functions through interactions between neurons and glial cells. While tools for studying neuronal activation are well-established, methods for tracking neuronal inactivation remain underdeveloped. Phosphorylated pyruvate dehydrogenase (pPDH) has emerged as a marker of neuronal inactivity in the brain. This study investigates whether pPDH can similarly serve as a reliable marker of neuronal inactivity in the ENS. METHODS: Whole-mount preparations of the colonic myenteric plexus from mice were treated with veratridine (neurona

Recent Advances in Medicinal Plant Research for Mitigating Aluminium Toxicity.

Aluminium is the third common metal of the earth, yet, it is physiologically non-essential and its higher levels on regular exposure can be harmful to the human body, affecting many vital organ systems. Environmental aluminium contamination and toxicity thereof is regarded as a 'silent killer' to the mankind as it is associated with the precipitation of several serious health complications chiefly involving nervous, blood and musculoskeletal systems. Various medicinal plants have been reported to possess aluminium toxicity ameliorative effects. A compilation of research published over the past

Periodic limb movements in Parkinson's disease: a critical review of pathophysiology and a framework for clinical management.

Periodic limb movements (PLM) refer to periodic episodes of repetitive and stereotypical limb movements, predominantly affecting the lower extremities. The prevalence of PLM is estimated at 17.6% to 86.7% in patients with Parkinson's disease (PD), which is much higher than 4% to 11% in the general population. PD is a chronic neurodegenerative movement disorder characterized by both motor and non-motor symptoms (NMS), and PLM represents one of the common non-motor manifestations. PLM in PD has garnered increasing attention due to its high prevalence, association with reduced sleep quality, elev

The Cardiac Orthostatic Stress Response as a Function of Internalizing Psychopathology and Sex in Typically Developing Youth-An Observational Study.

Stress-induced physiological changes offer insights into autonomic nervous system (ANS) function beyond resting cardiac activity, relevant to both physical and mental illnesses. Limited research exists on heart rate (HR) reactivity in children and adolescents, particularly in the context of internalizing psychopathology. In a study of n = 48 typically developing adolescents (mean age 16.31 years, SD = 0.51), HR and vagal-mediated heart rate variability (vmHRV) were assessed by electrocardiography during an orthostatic challenge, which served as a standardized physiological stressor. EEG Microstate as a Marker of Adolescent Idiopathic Scoliosis.

The pathophysiology of Adolescent Idiopathic Scoliosis (AIS) is not yet fully understood, but multifactorial hypotheses have been proposed that include defective central nervous system (CNS) control of posture, biomechanics, and body schema alterations. To deepen CNS control of posture in AIS, electroencephalographic (EEG) activity during a simple balance task in adolescents with and without AIS was parsed into EEG microstates. Microstates are quasi-stable spatial distributions of the electric potential of the brain that last tens of milliseconds. The spatial distribution of the EEG characteri

Palmitoylation of hIFITM1 inhibits JEV infection and contributes to BBB stabilization.

Human brain microvascular endothelial cells (hBMECs) are the main component cells of the blood-brain barrier (BBB) and play a crucial role in responding to viral infections to prevent the central nervous system (CNS) from viral invasion. Interferon-inducible transmembrane protein 1 (IFITM1) is a multifunctional membrane protein downstream of type-I interferon. In this study, we discovered that hIFITM1 expression was highly upregulated in hBMECs during Japanese encephalitis virus (JEV) infection. Depletion of hIFITM1 with CRISPR/Cas9 in hBMECs enhanced JEV replication, while overexpression of h

A case report of sinoatrial arrest caused by temporal lobe epilepsy in subclinical glioblastoma.

BACKGROUND: Atrial fibrillation with symptomatic bradycardia, higher grade atrioventricular block, and sinus node disease are all common indications for permanent pacemaker implantation. The most frequent causes of sinus node disease treated with pacemaker implantation involve degenerative structural changes of the sinus node; less often, extrinsic causes (such as damage due to myocardial infarction or heightened parasympathetic nervous system activity) lead to pacemaker implantation. CASE PRESENTATION: A 50-year-old patient with syncope and documented sinoatrial arrest was referred. Neurolog

The effect of early burn injury on sensitivity to future painful stimuli in dairy heifers.

Animals that experience painful procedures as neonates are more sensitive to pain later in life. We evaluated whether disbudding with a heated iron at 3 (n = 12), 35 (n = 9), or 56 (n = 20) d of age affected heifers' pain responses to vaccine injections at 11 mo of age. Heifers responded to the injection procedure with struggling and changes in eye temperature and heart rate variability compared to a sham procedure the day before, and still had a heightened response 6 d later, regardless of disbudding age. However, some heart rate variability indices suggested increased sympathetic dominance i

Sleep Disturbance in Bipolar Disorder: Neuroglia and Circadian Rhythms.

The worldwide prevalence of sleep disorders is approximately 50%, with an even higher occurrence in a psychiatric population. Bipolar disorder (BD) is a severe mental illness characterized by shifts in mood and activity. The BD syndrome also involves heterogeneous symptomatology, including cognitive dysfunctions and impairments of the autonomic nervous system. Sleep abnormalities are frequently associated with BD and are often a good predictor of a mood swing. Preservation of stable sleep-wake cycles is therefore a key to the maintenance of stability in BD, indicating the crucial role of circa

The Psychopharmacological Revolution in the USSR: Schizophrenia Treatment and the Thaw in Soviet Psychiatry, 1954-64.

Twentieth-century psychiatry was transformed in the 1950s and 1960s by the introduction of powerful psychopharmaceuticals, particularly Chlo … nabled them to successfully challenge the Stalin-era dogma in their field (Ivan Pavlov's 'theory of higher nervous activity'). Unlike in the West, however, the new psychopharmaceuticals did not lead to deinstitutionalisation. I argue that the new drugs did not disrupt the existing Soviet system because, unlike the system in the West, the Soviets were already dedicated, a …
