The Role of Viruses in Cellular Transformation and Cancer.

BACKGROUND: Viral infections are established contributors to oncogenesis, leading to significant public health challenges. This systematic review aims to synthesize current knowledge on the mechanisms of viral cellular transformation and their association with various cancers. RECENT FINDINGS: Studies reveal key mechanisms of oncogenesis, including direct viral integration into the host genome, expression of viral oncogenes, and indirect pathways such as chronic inflammation and immune evasion. Notably, Human Papillomavirus (HPV) was linked predominantly to cervical and oropharyngeal cancers,

New approaches for cryptococcosis treatment and prevention-a systematic review.

BACKGROUND: Cryptococcosis remains a life-threatening fungal infection with high mortality despite current antifungal therapy. The toxicity, limited accessibility, poor tissue penetration, and increasing resistance associated with conventional antifungals highlight the need for innovative preventive and therapeutic strategies. This systematic review summarises recent advances in cryptococcosis management and identifies priorities for future research and clinical translation. METHODS: PubMed, Embase, Web of Science, Cochrane Library, and ClinicalTrials.gov were systematically searched for stud

Antiviral Resistance and Phage Counter Adaptation to Antibiotic-Resistant Extraintestinal Pathogenic Escherichia coli.

Extraintestinal pathogenic Escherichia coli (ExPEC), often multidrug resistant (MDR), is a leading cause of urinary tract and systemic infections. The crisis of emergent MDR pathogens has led some to propose bacteriophages as a therapeutic. However, bacterial resistance to phage is a concerning issue that threatens to undermine phage therapy. Here, we demonstrate that E. coli sequence type 131, a circulating pandemic strain of ExPEC, rapidly develops resistance to a well-studied and therapeutically active phage (ϕHP3). Whole-genome sequencing of the resisters revealed truncations in genes invo

PROTACs in Antivirals: Current Advancements and Future Perspectives.

Proteolysis-targeting chimera (PROTAC) technology has demonstrated remarkable progress in tumor therapy, attributed to its unique capability of catalytically degrading "undruggable" targets. In the context of the ongoing global health threat posed by the Coronavirus Disease 2019 (COVID-19) pandemic, the application scope of PROTAC technology has been gradually extended to the field of antiviral research. Unlike traditional small molecule inhibitors, PROTAC employs an "event-driven" mechanism to achieve ubiquitination-mediated degradation of target proteins. This approach holds great promise in

IL-28B As a Predictor of Sustained Virologic Response in Patients with Chronic Hepatitis C Virus Infection.

Genome-wide association studies have recently identified single nucleotide polymorphisms in proximity to the interleukin-28B (IL-28B) gene that can predict sustained virologic response (SVR) in patients with chronic hepatitis C virus (HCV) infection who are undergoing therapy with pegylated interferon (IFN) a and ribavirin. IL-28B encodes IFN-λ3, a type III IFN involved in host antiviral immunity. Favorable variants of the 2 most widely studied IL-28B polymorphisms, rs1 2979860 and rs8099917, are strong pretreatment predictors of early viral clearance and SVR in patients with genotype 1 HCV in

Phage therapy for intestinal infections: efficacy, challenges, and future directions in translational research.

Phage therapy has emerged as a promising alternative to conventional antibiotics for combating intestinal bacterial infections, especially in the era of rising antimicrobial resistance. Despite its therapeutic potential, the clinical translation of phage therapy remains hindered by limited large-scale trial data and incomplete mechanistic understanding. This review systematically evaluates the efficacy of phage therapy in animal models of intestinal diseases, encompassing bacterial infection-induced diarrhea (e.g., cholera, typhoid fever), bacterial enteritis, and sepsis. By synthesizing evide

Prospects of molecularly-targeted therapies for cervical cancer treatment.

Cervical cancer is the third most common cancer among women worldwide and is responsible for 275.000 deaths each year. The development of cervical cancer has been linked to cell cycle disturbances caused by persistent expression of high- risk HPV oncoproteins (E5, E6 and E7), which modulate the expression of host genes and cellular microRNAs. An estimated 5 million women throughout the world are currently infected by HPV and several of them will develop invasive cervical cancer. Despite failures in conventional screening tests, approved therapies have no direct effect on HPV infection. In view

Epstein-Barr virus associated diseases: Immune alterations and targeted approaches.

Epstein-Barr virus (EBV) is a ubiquitous human γ-herpesvirus associated with a wide spectrum of diseases, ranging from autoimmune diseases to malignancies. Recognized as the first human oncogenic virus, multifaceted pathogenesis of EBV-associated diseases has been extensively explored. However, recent evidence suggests that EBV modulates immune cell function to influence the initiation and progression of associated diseases. Expressing a diverse repertoire of viral RNAs and proteins, EBV not only maintains latency with multiple immune evasion mechanisms, but also modulates host cell biology an

Vaccination-enabled immune readiness for checkpoint blockade.

Immune checkpoint blockade has transformed cancer therapy by demonstrating that durable tumour control can be achieved through immune modulation rather than direct cytotoxicity. However, primary resistance remains the dominant clinical outcome across solid tumours, reflecting a fundamental limitation of checkpoint inhibitors: they do not initiate antitumour immunity, but amplify immune responses that are already underway. Here, we advance immune readiness as a unifying framework to explain primary resistance to immune checkpoint inhibitors. Immune readiness is defined as a dynamic and programm

Preclinical efficacy and safety assessment of engineered regulatory T cells for treatment of IPEX and other autoimmune disorders.

FOXP3 is an essential transcription factor driving lineage commitment and function of regulatory T cells (Tregs). We previously reported a proof-of-concept study describing engineered Tregs (EngTregs) generated using homology-directed repair-based editing of the FOXP3 gene in CD4+ T cells, resulting in constitutive, high-level endogenous FOXP3 expression leading to acquisition of a Treg phenotype and robust in vitro and in vivo suppressive function. Here, we expand this gene-editing strategy to integrate a functional FOXP3 cDNA to enable EngTreg therapy for immune dysregulation, poly-endocrino

COVID-19-Associated Fungal Co-Infections: Pathogenesis, Diagnostics, Biomarkers, and Therapeutic Strategies.

From a health perspective, the 21st century has witnessed the emergence and global impact of several major viral diseases, most notably coronavirus disease 2019 (COVID-19) caused by SARS-CoV-2, which has been associated with a substantial burden of secondary fungal infections. Patients suffering from COVID-19 illnesses frequently developed fungal co-infections, which can worsen clinical outcomes and complicate therapeutic efforts. Hospitalized individuals with viral infections are particularly susceptible to invasive fungal pathogens, including Aspergillus, Candida, and Mucorales species. The

Aeromonas hydrophila Necrotizing Soft-Tissue Infection With Acute-on-Chronic Calcaneal Osteomyelitis Following Freshwater Exposure in a Patient With Poorly Controlled Diabetes Mellitus.

Aeromonas hydrophila is a gram-negative bacterium found in freshwater environments that can cause rapidly progressive necrotizing soft-tissue infections (NSTIs), particularly in patients with predisposing host factors such as diabetes mellitus. The organism rarely affects deeper structures such as bone, which can make diagnosis and treatment more difficult. We report the case of a 68-year-old man with poorly controlled type 2 diabetes mellitus, heart failure with reduced ejection fraction, and prior calcaneal trauma who developed an NSTI with acute-on-chronic calcaneal osteomyelitis after stag

Alternative therapies against Mycobacterium abscessus infections.

BACKGROUND: Mycobacterium abscessus (Mab) is considered as the most pathogenic rapid-growing mycobacteria in humans, causing pulmonary and extra-pulmonary diseases, especially in patients with cystic fibrosis. Mab shows intrinsic and acquired resistance to many drugs, leaving limited treatment options that lead to a generally poor prognosis. The standard therapeutic regimen last for more than 6 months and consists of a drug cocktail that ideally includes a macrolide and amikacin. Yet, toxicity and efficacy are suboptimal due also to the high toxicity. There is a need to introduce innovative an

Resistance to systemic immune checkpoint inhibition in the peritoneal niche.

Immune checkpoint inhibition (ICI) is an established therapeutic option for patients with deficient mismatch repair or high levels of microsatellite instability tumors. Yet, response to ICI among this group is varied, with nearly one-third of patients exhibiting primary resistance. Initial efforts in studying mechanisms of resistance to ICI have focused on intrinsic tumor factors. Host factors such as metastatic niches have unique biological properties that may mediate resistance to ICI but have been less studied date. Metformin is associated with favorable outcomes in patients with COVID-19 and type 2 diabetes mellitus.

Coronavirus disease 2019 (COVID-19) is a new pandemic the entire world is facing since December of 2019. Several risk factors are identified in developing severe disease and one of which is preexisting type 2 diabetes mellitus. Metformin is known to have host-directed anti-viral and anti-inflammatory properties. However, whether these effects offer lower mortality remains unclear. In this retrospective study, we aim to address whether metformin use prior to admission decreases mortality in patients with COVID-19 and pre-existing type 2 diabetes mellitus. A total of 1356 hospitalized patients w

Interactions of coronaviruses with ACE2, angiotensin II, and RAS inhibitors-lessons from available evidence and insights into COVID-19.

The rapid spread of a novel coronavirus, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has led to an ongoing pandemic of coronavirus disease 2019 (COVID-19). Recently, angiotensin-converting enzyme 2 (ACE2) has been shown to be a functional receptor for SARS-CoV-2 to enter host target cells. Given that angiotensin receptor blockers (ARBs) and an ACE inhibitor (ACEI) upregulated ACE2 expression in animal studies, the concern might arise regarding whether ARBs and ACEIs would increase the morbidity and mortality of COVID-19. On the other hand, animal data suggested a potential pr

The Artemisinin-Derived Autofluorescent Compound BG95 Exerts Strong Anticytomegaloviral Activity Based on a Mitochondrial Targeting Mechanism.

Human cytomegalovirus (HCMV) is a major human pathogen associated with severe pathology. Current options of antiviral therapy only partly satisfy the needs of a well-tolerated long-term treatment/prophylaxis free from drug-induced viral resistance. Recently, we reported the strong antiviral properties in vitro and in vivo of the broad-spectrum anti-infective drug artesunate and its optimized derivatives. NF-κB signaling was described as a targeting mechanism and additional target proteins have recently been identified. Here, we analyzed the autofluorescent hybrid compound BG95, which could be
