[Reversible "experimental" human arterial hypertension after fortuitous ligation of an isolated polar renal artery of aortic origin].

During the course of an aorto-femoral bypass in a normotensive young man, an inferior polar artery arising low on the aorta, and supplying the right kidney, was ligated. The limited ischaemia (demonstrated by later arteriography) of the parenchyma at the pole of the kidney was responsible, during the following weeks, for severe hypertension with a raised level of circulating plasma renin, but one that was rapidly treatable. This finding, which was fortuitous but could be ranked as "experimental", demonstrates the important role of abnormal polar arteries and of the renal parenchyma in human ar

Experimental immune complex disease of the lung. The pathogenesis of a laboratory model resembling certain human interstitial lung diseases.

Membranous and/or proliferative pneumonitis, similar in certain features to human interstitial pneumonitis, developed in rabbits making hyperactive antibody response to daily injections of bovine serum albumin (BSA) administered in multiple large doses sufficient to maintain the state of relative antigen-antibody equivalence. The pulmonary lesions were associated with deposition in alveolar capillary walls and interstitium of antigen, host globulin and complement, presumably in immune complexes. In some rabbits chronic interstitial pneumonitis, characterized by thickening of alveolar capillary

Necessary, Legendary and Detrimental Components of Human Colorectal Organoid Culture Medium: Raising Awareness to Reduce Experimental Bugs.

The creation of a specific culture medium for colorectal organoids in 2011 heralded a new era in human primary cultures by enabling the indefinite expansion of normal and pathological epithelial organoids. The original formula has been used ever since, with only minor, lab-specific modifications. The goal of culturing organoids from different tissues has relied on saving and propagating the pluripotent stem cell. The "magic bullet" and all its subsequent derivatives have pursued this goal. Consequently, agonist and antagonist signals are chronically activated in the organoid medium, forcing or

Recombinant human Thymosin β4 ameliorates experimental colitis and intestinal fibrosis through suppression of mineralocorticoid receptor signaling.

Inflammatory bowel disease (IBD) is a chronic inflammatory disorder characterized by epithelial barrier disruption, persistent mucosal inflammation, and progressive intestinal fibrosis, for which effective therapeutic options remain limited. Thymosin β4 (Tβ4) is a highly conserved endogenous peptide with established roles in tissue repair and immune regulation, but its contribution to IBD pathogenesis has not been fully elucidated. Here, we found that TMSB4X, the gene encoding Tβ4, was downregulated in colonic tissues of IBD patients. To investigate its functional significance, we generated Tm

Humans choose trunk lean angles consistent with minimal metabolic costs-an integrative experimental and musculoskeletal simulation approach.

Despite the substantial mass and moment of inertia of the trunk, its influence on minimizing metabolic cost in human locomotion is poorly understood. We show that self-selected trunk leaning in humans aligns with minimized metabolic costs during running. Utilizing an integrative experimental and musculoskeletal simulation approach, we reveal how trunk leaning redistributes joint moments and metabolic demands. We report estimates of muscle-specific changes in metabolic demands and contraction conditions for key muscles, including the hamstrings, uniarticular hip extensors and plantar flexors, r

An integrated experimental-computational electromagnetic-thermal framework for predicting bioheat response of the human head to wearable wireless devices.

The increasing use of wearable wireless devices operating in close proximity to the human body has raised the need for reliable assessment of electromagnetic exposure and associated thermal effects in biological tissue. In particular, wireless earbuds produce both electromagnetic energy absorption and device self-heating as potential sources of localized temperature elevation near the head. This study presents an integrated experimental-computational-electromagnetic-thermal framework for predicting the bioheat response of the human head. The approach combines infrared thermography measurements

Experimental investigation on developing human-centric AEB based on drivers' collision-avoidance capability in safety-critical scenarios.

Advanced Driving Assistance Systems (ADAS) can mitigate traffic accidents by issuing warnings or executing automated interventions in human-vehicle shared driving systems. However, their adaptability to human drivers remains limited, leading to inadequate coordination, increased collision risk, and potentially severe occupant injuries in safety-critical scenarios. This limitation stems from insufficient integration of drivers' inherent collision-avoidance behaviors into system design. Existing research lacks quantitative methods for assessing how drivers exhibit varying collision-avoidance cap

Exploring potential toxicological pathways of triclosan in human ovarian dysfunction via integrated network toxicology and experimental validation.

Triclosan (TCS), a compound ubiquitous in personal care products, is now a prevalent environmental contaminant across various ecosystems and has been detected in human tissue samples. Despite its prevalence and strong association with female infertility, the mechanisms by which TCS induces ovarian toxicity have not been thoroughly studied. This study employed an integrated approach combining network toxicology, molecular docking, transcriptomics, and cell experiments to systematically investigate the molecular mechanisms linking TCS to two core ovarian dysfunction diseases: polycystic ovary sy

Mitochondrial Proteostasis Links Diabetes and Sarcopenia: Cross-Scale Convergence from Experimental Models to Human Multi-Omics.

Sarcopenia in type 2 diabetes mellitus is increasingly recognized as a mechanistic consequence of chronic metabolic stress rather than mere age-related comorbidity. This review synthesizes evidence demonstrating how insulin resistance, hyperglycemia, lipotoxicity, and inflammation converge on skeletal muscle mitochondrial proteostasis to drive progressive decline. We evaluate seven pathway modules-mitochondrial dynamics, mitophagy, biogenesis, oxidative phosphorylation, nicotinamide adenine dinucleotide (NAD+)/sirtuin (SIRT)-linked regulation, protein import, and the mitochondrial unfolded pro

Inhibition of human and rat placental 3β-hydroxysteroid dehydrogenases by gallates: Combined experimental, 3D-QSAR, and in silico analysis.

Gallates are extensively utilized as additives in food, pharmaceuticals, and cosmetics. Despite their widespread use, their impact on the endocrine system remains poorly understood. This study investigates the potential interaction of gallates with 3β-hydroxysteroid dehydrogenase (3β-HSD), a critical enzyme in placental progesterone synthesis. The inhibitory effects and structure-activity relationship (SAR) and 3D-QSAR of nine gallates on human (h3β-HSD1) and rat (r3β-HSD4) placental enzymes were evaluated. Results revealed that the inhibitory potency of gallates on h3β-HSD1 was dependent on t

Towards constructing a generalized structural 3D breathing human lung model based on experimental volumes, pressures, and strains.

Respiratory diseases represent a significant healthcare burden, as evidenced by the devastating impact of COVID-19. Biophysical models offer the possibility to anticipate system behavior and provide insights into physiological functions, advancements which are comparatively and notably nascent when it comes to pulmonary mechanics research. In this context, an Inverse Finite Element Analysis (IFEA) pipeline is developed to construct the first continuously ventilated three-dimensional structurally representative pulmonary model informed by both organ- and tissue-level breathing experiments from

Experimental model of unilateral ureteral obstruction reproduces key events of chronic kidney disease: From mice to humans.

Chronic kidney disease (CKD) is currently a serious global health problem, due to its high risk of progression, prevalence and mortality. It not only affects the kidneys but also causes multi-organ damage. Moreover, there is no effective pharmacological treatment, and the only available alternatives are dialysis or transplantation, both of which impose a significant financial burden on healthcare systems. Given these challenges, a better understanding of CKD is essential for early diagnosis, identifying the pathological events involved, and assessing is systemic consequences. To achieve this,

Joint Microbiota Suggests Articular Dysbiosis in Experimental Murine Spondyloarthritis and Histological Detection of Bacteria in Human SpA Joints.

BACKGROUND: Recent studies have provided evidence supporting the presence of a commensal joint microbiome; however, its role in the pathogenesis of spondyloarthritis (SpA) remains unclear. This study aimed to characterize the joint microbiome and assess its role in bacterial dissemination and systemic involvement. METHODS: DBA/1 mice with spontaneous arthritis (SpAD) and healthy BALB/c mice, as well as biopsies from SpA patients, were analyzed by histology (Gram staining and IHC), short-read next-generation sequencing of the 16S rRNA gene amplicons, and transcriptomics. Shared bacterial speci

In Vitro Experimental Approach for Studying Human Pulmonary Artery Smooth Muscle Cells and Endothelial Cells Proliferation and Migration.

Pulmonary arterial hypertension (PAH) is a severe vascular disease characterized by persistent precapillary pulmonary hypertension, leading to right heart failure and death. Despite intense research in the last decades, PAH remains an incurable disease with high morbidity and mortality. New directions and therapies to improve understanding and treatment of PAH are desperately needed. The pathological mechanisms leading to this fatal disorder remain mostly undetermined, although structural remodeling of the pulmonary vessels is known to be an early feature of PAH. Pulmonary vascular remodeling

Human cues in eHealth to promote lifestyle change: An experimental field study to examine adherence to self-help interventions.

eHealth lifestyle interventions without human support (self-help interventions) are generally less effective, as they suffer from lower adherence levels. To solve this, we investigated whether (1) using a text-based conversational agent (TCA) and applying human cues contribute to a working alliance with the TCA, and whether (2) adding human cues and establishing a positive working alliance increase intervention adherence. Participants (N = 121) followed a TCA-supported app-based physical activity intervention. We manipulated two types of human cues: visual (ie, message appearance) and relation
