Induced pluripotent stem cells-derived myeloid-derived suppressor cells regulate the CD8+ T cell response.

Myeloid-derived suppressor cells (MDSCs) are markedly increased in cancer patients and tumor-bearing mice and promote tumor growth and survival by inhibiting host innate and adaptive immunity. In this study, we generated and characterized MDSCs from murine-induced pluripotent stem cells (iPSCs). The iPSCs were co-cultured with OP9 cells, stimulated with GM-CSF, and became morphologically heterologous under co-culturing with hepatic stellate cells. Allogeneic and OVA-specific antigen stimulation demonstrated that iPS-MDSCs have a T-cell regulatory function. Furthermore, a popliteal lymph node a

Japanese encephalitis virus replicon-based vaccine expressing enterovirus-71 epitope confers dual protection from lethal challenges.

BACKGROUND: To construct safer recombinant flavivirus vaccine, we exploited Japanese encephalitis virus (JEV) replicon-based platform to generate single-round infectious particles (SRIPs) that expressed heterologous neutralizing epitope SP70 derived from enterovirus-71 (EV71). Such pseudo-infectious virus particles, named SRIP-SP70, although are not genuine viable viruses, closely mimic live virus infection to elicit immune responses within one round of viral life cycle. RESULTS: We found that, besides gaining of full protection to thwart JEV lethal challenge, female outbred ICR mice, when we

Validation of quantitative ELISAS for measuring anti-Yersinia pestis F1 and V antibody concentrations in nonhuman primate sera.

This study systematically validated two quantitative enzyme-linked immunosorbent assays (ELISAs) for determining Yersinia pestis anti-F1 or anti-V IgG concentration in cynomolgus macaque sera. The results demonstrated that these ELISAs are reliable, reproducible, and suitable for their intended use to measure both anti-F1 and anti-V IgG in monkey sera following vaccination with a heterologous recombinant fusion F1-V protein (rF1-V). Statistical analysis demonstrated assay precision, accuracy, specificity, linearity/dilutional linearity, and robustness for both assays. The quantitative ranges o

Workshop on in vitro neutralization of Chlamydia trachomatis: summary of proceedings.

A task force evaluated an in vitro antibody-mediated chlamydial neutralization assay for its utility as a method to assess functional correlates of antibody responses to Chlamydia trachomatis. Two monoclonal antibodies that recognize different major outer membrane protein (MOMP) epitopes for a C. trachomatis serovar B strain exhibit good in vitro neutralizing activity, with a maximum of 90% neutralization. Calculations based on the 50% neutralization point indicated that 100% neutralization could theoretically be achieved when only 10% of the MOMP molecules bound antibody. Monoclonal antibodie

Babesia bovis: isolation of a protective antigen by using monoclonal antibodies.

Three distinct monoclonal antibody-producing hybridomas have been produced against a partly purified protective fraction of Babesia bovis. All three stain the parasite or infected erythrocytes or both in precise and different manners when fluorescent-antibody techniques are used. The relevant antigens for each monoclonal antibody were isolated by immunoadsorption, their native molecular weights being 1.3 X 10(6), 180 X 10(3), and 44 X 10(3). Each antigen reacted in serological assays with homologous and heterologous bovine antisera to B. bovis. Susceptible splenectomized calves were immunized

Oral immunization against pox. Studies on fowl pox as a model.

Oral vaccination against fowl pox is both effective and harmless. A suitable vaccine strain is the attenuated HP-1 strain in the 200th to 400th tissue culture passage. For optimal immunization virus of the 200th to 270th passage level should be applied twice at an interval of 3 to 4 weeks. Vaccination dose should contain 10(7.0)TCID50. Chickens may be effectively immunized already at 5 days. Immunity is against both homologous and heterologous virus and proves equally resistant to oral, cutaneous and intravenous challenge. Protection of mucous membranes of the upper respiratory and digestive t

Challenges and Strategies for Developing Recombinant Vaccines against Leptospirosis: Role of Expression Platforms and Adjuvants in Achieving Protective Efficacy.

The first leptospiral recombinant vaccine was developed in the late 1990s. Since then, progress in the fields of reverse vaccinology (RV) and structural vaccinology (SV) has significantly improved the identification of novel surface-exposed and conserved vaccine targets. However, developing recombinant vaccines for leptospirosis faces various challenges, including selecting the ideal expression platform or delivery system, assessing immunogenicity, selecting adjuvants, establishing vaccine formulation, demonstrating protective efficacy against lethal disease in homologous challenge, achieving

Critical role of CD4+ T cells and IFNγ signaling in antibody-mediated resistance to Zika virus infection.

Protective adaptive immunity to Zika virus (ZIKV) has been mainly attributed to cytotoxic CD8+ T cells and neutralizing antibodies, while the participation of CD4+ T cells in resistance has remained largely uncharacterized. Here, we show a neutralizing antibody response, dependent on CD4+ T cells and IFNγ signaling, which we detected during the first week of infection and is associated with reduced viral load in the brain, prevention of rapid disease onset and survival. We demonstrate participation of these components in the resistance to ZIKV during primary infection and in murine adoptive tr

Boosting BCG to protect against TB.

Given that TB still constitutes a tremendous public health problem at the start of the 21st Century, it may come as a surprise that Bacillus Calmette-Guérin (BCG), developed nearly 100 years ago, is today still the only vaccine available against TB. Owing to its limited efficiency in controlling TB, much effort has been deployed to develop new, improved vaccines, with initial preclinical models showing encouraging results. However, since most individuals worldwide have been vaccinated with BCG, new vaccine developments have to be placed in that context. Consequently, several approaches explore

Trained immunity-based mucosal immunotherapies for preventing respiratory infections: a translational, evidence-graded appraisal of platforms, populations, and the protection-immunopathology balance.

Conventional respiratory vaccines protect through antigen-specific immunity, leaving broad gaps against antigenically diverse and emerging pathogens and inducing little protection at the airway surface itself. Trained immunity, the durable epigenetic and metabolic reprogramming of innate immune cells and their bone marrow progenitors, offers antigen-agnostic, heterologous protection and can be induced directly at the respiratory mucosa, providing a mechanistic rationale for a new class of prophylactic immunotherapies. Here we appraise this field from a clinical-translational, evidence-graded s

Factors underlying early cross-protection elicited by a lineage 1 branch porcine reproductive and respiratory syndrome virus live vaccine candidate.

Porcine reproductive and respiratory syndrome virus (PRRSV) is a major swine pathogen that causes considerable economic losses globally. Previous studies have demonstrated that the L1 branch PRRSV SD-R vaccine candidate offers effective clinical protection against multiple PRRSV sublineages, including L1.8 (NADC30-like), L1.5 (NADC34-like), and L8.7 (HP-PRRSV and HP-like PRRSV). However, the mechanisms underlying cross-protection in SD-R remain to be fully elucidated. Based on three different PRRSV-MLV vaccines (CH-1R, HuN4-F112, and SD-R), we systematically evaluated the humoral and cellular

Nonspecific protection of heat-inactivated Mycobacterium bovis against Salmonella Choleraesuis infection in pigs.

Trained immunity is the capacity of innate immune cells to produce an improved response against a secondary infection after a previous unrelated infection. Salmonellosis represents a public health issue and affects the pig farming industry. In general, vaccination against salmonellosis is still facing problems regarding the control of distinct serovars. Therefore, we hypothesized that an immunostimulant based on heat inactivated Mycobacterium bovis (HIMB) could have an immune training effect in pigs challenged with Salmonella enterica serovar Choleraesuis (S. Choleraesuis) and decided to explo

Leishmania major p27 gene knockout as a novel live attenuated vaccine candidate: Protective immunity and efficacy evaluation against cutaneous and visceral leishmaniasis in BALB/c mice.

Leishmaniasis is a growing health problem in many parts of the world and efforts to find vaccine against the disease are a public health priority. Live attenuated vaccines are the gold standard for protection against intracellular pathogens such as Leishmania spp. Defined genetic alteration of the Leishmania genome can be achieved using a gene-targeted disruption strategy that allows for the selection of parasites lacking genes essential for long-term survival and virulence. Previously, we demonstrated that genetically modified live attenuated Leishmania major, lacking the p27gene (Lmp27-/-) i

Comparative evaluation of the protective efficacy of two formulations of a recombinant Chlamydia abortus subunit candidate vaccine in a mouse model.

Chlamydia abortus (C. abortus) is the causative agent of ovine enzootic abortion (OEA) and poses a zoonotic risk to pregnant women. Current live attenuated 1B vaccines are efficacious but cause disease in vaccinated animals and inactivated vaccines are only marginally protective. We tested the ability of a new C. abortus subunit vaccine candidate based on the conserved and immunogenic polymorphic membrane protein D (Pmp18D) formulated in CpG1826+FL (Fms-like tyrosine kinase 3 Ligand; Flt3L) or Vibrio cholerae ghosts (VCG) to induce innate and cross protective immunity against genital C. abortu

In vivo differences in the virulence, pathogenicity, and induced protective immunity of wboA mutants from genetically different parent Brucella spp.

To explore the effects of the genetic background on the characteristics of wboA gene deletion rough mutants generated from different parent Brucella sp. strains, we constructed the rough-mutant strains Brucella melitensis 16 M-MB6, B. abortus 2308-SB6, B. abortus S19-RB6, and B. melitensis NI-NB6 and evaluated their survival, pathogenicity, and induced protective immunity in mice and sheep. In mice, the survival times of the four mutants were very different in the virulence assay, from less than 6 weeks for B. abortus S19-RB6 to 11 weeks for B. abortus 2308-SB6 and B. melitensis NI-NB6. Live chimeric and inactivated Japanese encephalitis virus vaccines differ in their cross-protective values against Murray Valley encephalitis virus.

The Japanese encephalitis virus (JEV) serocomplex, which also includes Murray Valley encephalitis virus (MVEV), is a group of antigenically closely related, mosquito-borne flaviviruses that are responsible for severe encephalitic disease in humans. While vaccines against the prominent members of this serocomplex are available or under development, it is unlikely that they will be produced specifically against those viruses which cause less-frequent disease, such as MVEV. Here we have evaluated the cross-protective values of an inactivated JEV vaccine (JE-VAX) and a live chimeric JEV vaccine (C

Type 2 immune history trains lung macrophages for viral disease tolerance.

Severe respiratory viral disease varies widely among individuals and often reflects immunopathology rather than inadequate pathogen control, suggesting that previous immune history can prime the lungs towards disease tolerance. Here we show that nerve- and airway-associated macrophages (NAMs), a subset of interstitial macrophages, expand ephemerally after type 2 inflammation induced by Nippostrongylus brasiliensis. We therefore hypothesized that NAMs acquire epigenetically imprinted trained immunity and tested this using a heterologous challenge model in which mice that were previously infecte

Novel influenza vaccine M2SR protects against drifted H1N1 and H3N2 influenza virus challenge in ferrets with pre-existing immunity.

Current influenza vaccines do not provide effective protection against heterologous influenza viruses. The ability of the novel M2SR influenza vaccine to protect against drifted influenza viruses was evaluated in naïve ferrets and in ferrets with pre-existing immunity to influenza. In naïve ferrets, M2SR provided similar protection against drifted challenge viruses as the comparator vaccine, FluMist®. However, in ferrets with pre-existing immunity, M2SR provided superior protection than FluMist in two model systems. In the first model, ferrets were infected with influenza A H1N1pdm and influen

A multi-antigen chimeric PvCSP-MSP119 vaccine induces robust humoral responses against distinct stages of Plasmodium vivax.

Plasmodium vivax is the leading cause of malaria outside Africa, highlighting the need for an effective vaccine. However, the development of vaccines against this parasite remains challenging because of its complex life cycle and antigenic diversity. In this study, we evaluated vaccine formulations targeting both the pre-erythrocytic and erythrocytic stages of P. vivax using either individual antigens or a novel chimeric protein. The chimeric construct combined the C-terminal region and variant repeat domains of the P. vivax circumsporozoite protein (PvCSP) with the 19-kDa C-terminal fragment

The Interleukin-33-Group 2 Innate Lymphoid Cell Axis Represents a Potential Adjuvant Target To Increase the Cross-Protective Efficacy of Influenza Vaccine.

Interleukin-33 (IL-33) is a multifunctional cytokine that mediates type 2-dominated immune responses. In contrast, the role of IL-33 during viral vaccination, which often aims to induce type 1 immunity, has not been fully investigated. Here, we examined the effects of IL-33 on influenza vaccine responses. We found that intranasal coadministration of IL-33 with an inactivated influenza virus vaccine increases vaccine efficacy against influenza virus infection, not only with the homologous strain but also with heterologous strains, including the 2009 H1N1 influenza virus pandemic strain.
