Polymyositis occurring during alpha-interferon treatment for malignant melanoma: a case report and review of the literature.

Polymyositis is a systemic autoimmune disorder characterised by proximal muscle weakness which most frequently involves the limbs, neck and trunk. Alpha interferon is an antiviral molecule with well-known immunomodulatory and antiproliferative effects, but its use is often associated with a variety of side effects, in particular autoimmune phenomena. We report the occurrence of polymyositis during treatment with alpha-interferon in a patient affected by malignant melanoma. Indirect evidence suggests that in this case the patient's myositis was not linked to her neoplasia, since the melanoma wa

Both human alpha/beta and gamma interferons upregulate the expression of CD48 cell surface molecules.

We have established a cDNA library from interferon (IFN)-treated human lymphoblastoid Daudi cells and made use of differential screening to search for yet unidentified IFN-regulated genes. In the course of these studies, we have isolated a human cDNA coding for the glycosyl-phosphatidylinositol-linked (GPI) membrane glycoprotein CD48 (TCT-1, Blast-1). Various studies demonstrated that the murine CD48 is the predominant counterreceptor for the mouse CD2 and is involved in the regulation of T cell activation. Since the murine CD48 is functionally homologous to the human CD2 ligand LFA-3 (CD48),

Development of autoimmune thyroid diseases during long-term treatment of hematological malignancies with alpha-interferons.

In recent years recombinant alpha-interferons (alpha-IFNs) have been widely used in the treatment of several hematological malignancies. Prolonged courses of IFN have been shown to induce autoantibodies and to support the exacerbation or even the development of autoimmune diseases. In this report we describe the development of symptomatic autoimmune thyroid diseases in 4 (7.4%) out of 54 patients in chronic treatment with recombinant alpha-IFNs in our department. Two patients developed a disease resembling Hashimoto's thyroiditis after 17 and 49 months of continuous IFN treatment, while the ot

Clinical and histologic predictors of response to interferon-alpha in patients with chronic hepatitis C viral infection.

To evaluate if any pretreatment characteristics of patients with chronic hepatitis C (HCV) can be used to predict response to the current recommended dose (3 million units three times a week) and higher doses of interferon-alpha (IFN), we retrospectively assessed the response of 37 patients with HCV who were treated with IFN. Sixteen patients (43%) responded to the standard dose of IFN with normalization of ALT. Weight and liver histology were found to be significant factors for response. The responders weighed significantly less than nonresponders (161.8 +/- 35.5 lb versus 200.3 +/- 45.4 lb, …

Disappearance of metastatic renal cell carcinoma to the base of the tongue after systemic administration of interferon-alpha.

We describe a 41-year-old Japanese man with metastases of the lung, bone, lymph nodes and the base of the tongue 34 months after right radical nephrectomy. One month after systemic administration of human lymphoblastoid interferon (IFN), the metastasis to the base of the tongue completely disappeared, while other metastases increased both in size and number. Though the true effect of IFN-alpha on the metastatic renal cell carcinoma to the base of the tongue is still unknown, our case indicates that the tongue is a possible sensitive site to IFN-alpha therapy.

A clinical trial of recombinant bovine interferon alpha 1 for the control of bovine respiratory disease in calves.

The effects of recombinant bovine interferon alpha 1 (rBoIFN alpha 1) on the incidence of bovine respiratory disease (BRD) were examined in 60 young male cattle. Each group of clinically healthy 30 male cattle was injected intramuscularly with either rBoIFN alpha 1 (5 mg/head) or physiological saline 5 times in 6 months. They were examined daily for the clinical symptoms of respiratory distress. The clinical signs of respiratory distress in rBoIFN alpha 1-treated cattle were significantly less severe than those of the placebo group. The incidence rate, relapse rate and mean sick days of respir

[Clinical study on the effect of natural alpha-interferon (HLBI) in the treatment of adult T-cell leukemia].

The phase II trial of natural interferon-alpha (HLBI) in treatment of adult T-cell leukemia was carried out as a cooperative study. Of the 24 cases which could be evaluated, 3 cases in crisis type and 5 cases in chronic type with lymphadenopathy and/or skin infiltration achieved PR, giving a response rate of 33.3%. The anti-tumor effect of HLBI for skin lesion could be assessed in 16 cases with skin infiltration, giving a response rate of 50.0% (5 CR and 3 PR) and demonstrating a high efficacy. Of the 31 eligible patients, side effects were recognised in 27 (87.1%). Major subjective and object…

Homozygous deletion of the alpha- and beta 1-interferon genes in human leukemia and derived cell lines.

The loss of bands p21-22 from one chromosome 9 homologue as a consequence of a deletion of the short arm [del(9p)], unbalanced translocation, or monosomy 9 is frequently observed in the malignant cells of patients with lymphoid neoplasias, including acute lymphoblastic leukemia and non-Hodgkin lymphoma. The alpha- and beta 1-interferon genes have been assigned to this chromosome region (9p21-22). We now present evidence of the homozygous deletion of the interferon genes in neoplastic hematopoietic cell lines and primary leukemia cells in the presence or absence of chromosomal deletions that ar

Interferon-alpha-2C in the treatment of advanced hairy cell leukaemia. Results of a phase II trial.

The efficacy of recombinant interferon-alpha 2 in the treatment of advanced hairy cell leukaemia is investigated in 31 patients. Preliminary results show a complete and partial response rate of 69%. An optimal dose study utilizing maximal stimulation of an interferon-dependent pathway was carried out in 5 patients. The optimal dose of the order of 5 X 10(5) IU/day is effective and without side-effects. Interferon treatment did not enhance natural killer cell activity and therefore a direct mode of action upon tumour cells is proposed.

Association of visceral fat and plasmacytoid dendritic cell-derived interferon alpha with SARS-CoV-2 infection.

Type I interferon (IFN-I: IFNα/β) production by plasmacytoid dendritic cells (pDCs) is critical for controlling viral infections. Visceral fat obesity is associated with severe outcomes in viral infections, but its effect on pDC-mediated IFN-I production in the context of SARS-CoV-2 infection remains unclear. This cross-sectional study evaluated cytokine production by blood pDCs in response to TLR7/8 stimulation and analyzed its association with visceral fat area (VFA). IFN-I production was exclusively observed in pDCs following TLR7/8 stimulation. The proportion of IFNα-positive pDCs (pDC-IFN

Oral interferon alpha treatment significantly reduces halitosis and Porphyromonas gulae growth in dogs with periodontal disease.

Periodontal disease (PD) is one of the most common infectious conditions in dogs, associated with plaque accumulation, gingival inflammation, halitosis, and tooth loss. Halitosis is primarily caused by volatile sulfur compounds (VSCs) generated by anaerobic bacteria, including Porphyromonas gulae (P. gulae), a pathogen closely linked to PD progression. This study examined the effects of oral interferon alpha (IFN-α) on halitosis and bacterial activity in dogs with PD. Thirty-two dogs with moderate-to-severe PD were enrolled and allocated to untreated controls (n = 10) or IFN-α treatment (n = 2

Multimodal characterization of the responsiveness of eight hepatitis D virus genotype isolates to interferon-alpha treatment.

UNLABELLED: Chronic hepatitis delta virus (HDV) infection causes the most severe form of viral hepatitis. Although humans produce 12 subtypes of interferon-alpha (IFN-α), IFN-α2a has been the only commonly used treatment against HDV. Previously, we characterized eight HDV genotype isolates with varying replication kinetics. Herein, we systematically investigated the antiviral efficacy of IFN-α2a and other IFN-α subtypes against HDV genotypes 1-8 during de novo infection, cell mitosis, and in quiescent cells. Our findings revealed that IFN-α2a exhibits potent but varied efficiency against HDV 1…

Short-term pegylated interferon alpha in chronic HBV patients with ultra-low HBsAg: a retrospective study.

BACKGROUND AND AIMS: Hepatitis B surface antigen (HBsAg) clearance-defined as an HBsAg level below the lower limit of detection-is critical for the functional cure of chronic hepatitis B (CHB). This study evaluated the efficacy of short-term pegylated interferon alpha (Peg-IFNα) therapy in achieving HBsAg clearance in CHB patients with ultra-low HBsAg levels (<50 IU/ml). METHODS: A total of 378 CHB patients with HBsAg levels <50 IU/ml were enrolled, including 206 nucleos(t)ide analogue (NUC)-treated patients and 172 inactive HBsAg carriers (IHCs). The NUC-treated cohort was divided into 83 pa

Results of Response-Guided Therapy with Pegylated Interferon Alpha 2a in Chronic Hepatitis B and D.

Pegylated interferon alpha 2a continues to be used for the treatment of chronic hepatitis D. The reported on-treatment virologic response varies between 17 and 47%, with relapses in more than 50% of these patients. No stopping rules have been defined, and the duration of the treatment is not clearly established, but it should be between 48 and 96 weeks. In total, 76 patients with compensated liver disease treated with peg-interferon according to the Romanian National protocol for the treatment of hepatitis D were retrospectively included. The duration of treatment was up to 96 weeks, with the
