Crosstalk between astrocytes and microglia results in increased degradation of α-synuclein and amyloid-β aggregates.

BACKGROUND: Alzheimer's disease (AD) and Parkinson's disease (PD) are characterized by brain accumulation of aggregated amyloid-beta (Aβ) and alpha-synuclein (αSYN), respectively. In order to develop effective therapies, it is crucial to understand how the Aβ/αSYN aggregates can be cleared. Compelling data indicate that neuroinflammatory cells, including astrocytes and microglia, play a central role in the pathogenesis of AD and PD. However, how the interplay between the two cell types affects their clearing capacity and consequently the disease progression remains unclear. METHODS: The aim o

Glycerol-Based Soft Elastomeric Degradable Biomaterials with Tunable Physicochemical and Mechanical Properties.

The widespread clinical adoption of implantable devices has driven growing interest in biodegradable alternatives that eliminate the need for secondary removal surgeries. Among these, glycerol-based thermosets have emerged as promising biocompatible materials. Compared with established glycerol-based thermosets, incorporating shorter-chain aliphatic dicarboxylic acids into the glycerol backbone may enhance hydrophilicity and potentially accelerate hydrolytic degradation, while varying the curing conditions enables tailorable properties for biomedical applications requiring temporary structural

An integrated machine learning and chemical space network approach for the design of potent epigenetic HDAC6 inhibitors for targeting neurological disorders.

Histone deacetylase 6 (HDAC6) is increasingly recognized as a key regulator of cytoskeletal dynamics and intracellular transport in both neurodevelopmental and neurodegenerative disorders. Despite the large number of HDAC6 inhibitors reported, only a few have shown effective in vivo activity in neurological disease models. In this context, machine-learning (ML) approaches offer a powerful strategy for extracting chemical, physical, and biological features from a large and complex dataset of 4307 HDAC6 inhibitors. The current research aimed to develop high-quality ML-based classification framew

Chimeric enediynes as potent antivirals against coronaviruses by targeting the conserved free-fatty-acid binding site on the spike protein.

Conventional targeted protein degradation (TPD) strategies operate by hijacking intracellular proteolytic machinery, limiting their therapeutic scope. Here, we present protein-radical-oxidation targeting enediyne chimera (PROTEC), a TPD platform that can be located exclusively outside of host cells and degrades pathogen proteins via a radical-based oxidative mechanism. The maleimide-fused enediyne core undergoes cycloaromatization under physiological conditions to generate reactive radical species. Strategic incorporation of sulfate-based hydrophilicity-adjusting groups (HAGs) restricts PROTEC

A Novel Biosensor for Ferrous Iron Developed via CoBiSe: A Computational Method for Rapid Biosensor Design.

Genetically encoded biosensors enable the monitoring of metabolite dynamics in living organisms. We present CoBiSe, a computational biosensor design approach using Constraint Network Analysis to identify optimal insertion sites for reporter modules in molecular recognition elements (MREs). Applied to the iron-binding protein DtxR from Corynebacterium glutamicum, CoBiSe identified a flexible connective loop (residues 138-150) for inserting the reporter module, resulting in IronSenseR, a novel ratiometric biosensor for ferrous iron (Fe2+). IronSenseR demonstrates high specificity for Fe2+ with d

Isolation of Proteins and Extracellular Vesicles Secreted by Pollen of Actinidia chinensis L. During In Vitro Germination.

Extracellular vesicles (EVs) are membrane-bound particles secreted into the extracellular space by cells of any living kingdom. Plant-derived EVs (PDEVs) in particular are regarded as promising nutraceutical and drug carriers, but their purification is hindered by the contamination from intracellular and intramural vesicles, which is inherently associated with the existing PDEVs isolation protocols. In the last decade, pollen has been proven to release EVs, called pollensomes, during in vitro germination. When secreted in a liquid culture medium, pollensomes can be easily isolated with minimal

Intracellular enhancement technique for gadoxetic acid-enhanced hepatobiliary-phase magnetic resonance imaging: evaluation of hepatic function.

PURPOSE: To investigate the utility of intracellular enhancement (ICE) technique which suppresses signals from the extracellular space for the evaluation of hepatic function on gadoxetic acid-enhanced hepatobiliary-phase (HBP) images. METHODS: We subjected 67 patients with suspected neoplastic hepatic lesions to gadoxetic acid-enhanced HBP imaging with and without ICE [i-HBP, conventional-HBP (c-HBP)]. A radiologist calculated the liver/spleen contrast (LSC) [LSC = signal intensity (SI) of liver/SI of spleen]. Receiver-operating analysis was used to evaluate the diagnostic value of the LSC on

Electron cryotomography of intact motile cilia defines the basal body to axoneme transition.

Cells use motile cilia to generate force in the extracellular space. The structure of a cilium can be classified into three subdomains: the intracellular basal body (BB) that templates cilium formation, the extracellular axoneme that generates force, and the transition zone (TZ) that bridges them. While the BB is composed of triplet microtubules (TMTs), the axoneme is composed of doublet microtubules (DMTs), meaning the cilium must convert between different microtubule geometries. Here, we performed electron cryotomography to define this conversion, and our reconstructions reveal identifying s

Dephosphorylated rather than hyperphosphorylated Tau triggers a pro-inflammatory profile in microglia through the p38 MAPK pathway.

Tauopathies are a broad set of neurodegenerative dementias characterized by the aggregation of Tau protein. Activated microglia and elevated levels of pro-inflammatory molecules are also pathological hallmarks of tauopathies. In these diseases, intracellular Tau is secreted to the extracellular space, where it interacts with other cells, such as neurons and glia, promoting inflammation. However, the mechanism through which extracellular Tau triggers pro-inflammatory responses in microglia remains unknown. Primary microglia cultures were treated with extracellular Tau in its hyperphosphorylated

Molecular mechanisms of endothelial hyperpermeability: implications in inflammation.

Endothelial hyperpermeability is a significant problem in vascular inflammation associated with trauma, ischaemia-reperfusion injury, sepsis, adult respiratory distress syndrome, diabetes, thrombosis and cancer. An important mechanism underlying this process is increased paracellular leakage of plasma fluid and protein. Inflammatory stimuli such as histamine, thrombin, vascular endothelial growth factor and activated neutrophils can cause dissociation of cell-cell junctions between endothelial cells as well as cytoskeleton contraction, leading to a widened intercellular space that facilitates

Monoamine transporter as a target molecule for psychostimulants.

Methamphetamine (MAP), a drug of abuse known worldwide for its addictive effects and neurotoxicity, causes somatic and psychiatric disorders. MAP enters terminals/neurons via monoamine transporters, displaces both vesicular and intracellular monoamines, and facilitates the release of monoamines into the extraneuronal space through synaptic transport via the monoamine transporters. Chronic psychostimulant abusers exhibit psychotic features, including delusions and auditory hallucinations. The dopamine transporter (DAT) and the vesicular monoamine transporter 2 (VMAT2) play pivotal roles in the

R992C (p.R1192C) Substitution in collagen II alters the structure of mutant molecules and induces the unfolded protein response.

We investigated the molecular bases of spondyloepiphyseal dysplasia (SED) associated with the R992C (p.R1192C) substitution in collagen II. At the protein level, we analyzed the structure and integrity of mutant molecules, and at the cellular level, we specifically studied the effects of the presence of the R992C collagen II on the biological processes taking place in host cells. Our studies demonstrated that mutant collagen II molecules were characterized by altered electrophoretic mobility, relatively low thermostability, the presence of atypical disulfide bonds, and slow rates of secretion

Profiling of astrocyte properties in the hyperammonaemic brain: shedding new light on the pathophysiology of the brain damage in hyperammonaemia.

Acute hyperammonaemia (HA) causes cerebral oedema and severe brain damage in patients with urea cycle disorders (UCDs) or acute liver failure (ALF). Chronic HA is associated with developmental delay and intellectual disability in patients with UCDs and with neuropsychiatric symptoms in patients with chronic liver failure. Treatment often cannot prevent severe brain injury and neurological sequelae. The causes of the brain oedema in hyperammonaemic encephalopathy (HAE) have been subject of intense controversy among physicians and scientists working in this field. Currently favoured hypotheses a

Biochemistry of the elastic fibers in normal connective tissues and its alterations in diseases.

The elastic fibers present in various connective tissues of the body are responsible for physiologic elasticity of the organs. These fibers consist of 2 distinct components, elastin and the elastic fiber microfibrils. Controlled synthesis and balanced interaction of these 2 components are essential for normal fibrillogenesis. The intracellular biosynthesis of elastin by connective tissue cells, such as smooth muscle cells, involves assembly of the polypeptide chains on the membrane-bound ribosomes, hydroxylation of some prolyl residues to hydroxyproline, and secretion of the polypeptides packa

Mechanism of barbituric-acid protection against inhibition by alloxan of glucose-induced insulin release.

Isolated rat islets were maintained in a simple static incubation system and were exposed to alloxan for a period of five minutes. Alloxan inhibited subsequent glucose-induced insulin release in a dose-dependent manner at 37 degrees C., with 650 muM alloxan producing 94 per cent inhibition of insulin release. Barbituric acid, a compound structurally related to alloxan, provided complete protection (at 37 degrees C.) against this inhibition of insulin release when present during the alloxan exposure. At 23 degrees C., barbituric acid was shown to be absent from the intracellular space of the is…

Integrated Effects of Digestion Strategy and LC Gradient Architecture Define Host Cell Protein Detectability in CHO Null Harvests.

Accurate characterization of host cell proteins (HCPs) is critical for biologics development, yet analytical outcomes are strongly influenced by the digestion strategy and chromatographic design. Here, we systematically evaluated digestion parameters, including digestion mode, enzyme ratio, and incubation time, together with LC-MS gradient architecture using a CHO null harvest as a controlled model system to isolate workflow-dependent analytical effects from biological variability. Specifically, the study was designed to determine how these variables jointly influence HCP detection and the phy

A unified intracellular pH landscape with SITE-pHorin: a quantum-entanglement-enhanced pH probe.

An accurate map of intracellular organelle pH is crucial for comprehending cellular metabolism and organellar functions. However, a unified intracellular pH spectrum using a single probe is still lacking. Here, we developed a novel quantum entanglement-enhanced pH- sensitive probe called SITE-pHorin (single excitation and two emissions pH sensor protein), which features a wide pH-sensitive range and ratiometric quantitative measurement capabilities. We subsequently measured the pH of various organelles and their subcompartments, including mitochondrial subspaces, Golgi stacks, endoplasmic reti

Pathophysiology of dementias and implications for therapy.

Dementia is characterized clinically by progressive cognitive decline, often with impairment of memory. The pathology of dementias is either focal as with infarcts in Vascular Dementia or diffuse as typified by Alzheimer's disease. In many cases of Alzheimer's disease there is a mixture of focal infarcts and diffuse changes. Diffuse pathology in dementias comprises mainly intracellular and extracellular protein deposits. Intracellular inclusions are of tau protein (Alzheimer's disease; and some frontotemporal dementias), alpha-synuclein (Dementia with Lewy bodies) and huntingtin (Huntington's

Localization of ISG15 and conjugated proteins in bovine endometrium using immunohistochemistry and electron microscopy.

The interferon-stimulated gene ISG15, a ubiquitin homolog, becomes conjugated to and regulates uterine proteins in response to conceptus-derived interferon-tau on d 18 of pregnancy. It was hypothesized here that cellular localization of ISG15 within endometrial cells might provide insight regarding function. Uteri were collected from cows (approximately 21-d estrous cycle) on d 17-21/0 of the estrous cycle and pregnancy and d 23, 45, and 50 of pregnancy. Intracellular ISG15 and its conjugates were present on d 17 of pregnancy, peaked to highest levels from d 18 to 23 and then declined to low b
