Evaluation of a high-yield technique for pancreatic islet isolation from deceased canine donors.

Type 1 diabetes mellitus is one of the most frequently diagnosed endocrinopathies in dogs, and prevalence continues to increase. Pancreatic islet transplantation is a noninvasive and potentially curative treatment for type 1 diabetes mellitus. Institution of this treatment in dogs will require a readily available source of canine islets. We hypothesized that clinically acceptable islet yield and purity could be achieved by using deceased canine donors and standard centrifugation equipment. Pancreata were procured from dogs euthanized for reasons unrelated to this study. Initial anatomic studie

Interactions between the endocrine and exocrine pancreas and their clinical relevance.

In consequence of the close anatomical and functional links between the exocrine and endocrine pancreas, any disease affecting one of these parts will inevitably affect the other. Pancreatic conditions which might cause diabetes mellitus include acute and chronic pancreatitis, pancreatic surgery, cystic fibrosis and pancreatic cancer. The development of diabetes greatly influences the prognosis and quality of life of patients with exocrine pancreatic diseases. It may cause life-threatening complications, such as hypoglycemia, due to the lack of glucagon and the impaired absorption of nutrients

Hyperinsulinemic hypoglycemia due to diffuse nesidioblastosis in adults: a case report.

Persistent hyperinsulinemic hypoglycemia is caused most commonly by an insulinoma in adults or by nesidioblastosis in neonates. In adults, nesidioblastosis is a rare disorder characterized by diffuse or disseminated proliferation of islet cells. We recently encountered a case of nesidioblastosis in an adult. A 71-year-old man was admitted due to intermittent general weakness, abdominal pain, and mild dyspnea. The patient underwent a subtotal gastrectomy for a gastric adenocarcinoma two years ago. After 5 d of admission, the patient showed symptoms of cold sweating, chilling, and hypotension 30

[Tumors of the endocrine pancreas].

The neuroendocrine tumors of the pancreas are rare, but belong to the most common endocrine neoplasms of the abdomen. In most cases they present as solitary tumors, composed of monomorphic cells. Functionally active cells are associated with characteristic hormonal syndromes. Using morphological, immunohistochemical and biological criteria, the classification distinguishes between well differentiated tumors with benign or uncertain behavior, well differentiated (low grade malignant) carcinomas and poorly differentiated (high grade malignant) carcinomas. In recent years the percentage of non-fu

Development of a novel digestion chamber for human and porcine islet isolation.

BACKGROUND: The current technique of human pancreas digestion for islet isolation relies on selective distribution of collagenase delivered via the pancreatic duct to produce digestion and removal of peri-acinar fibrous tissue. However, the collagenase has relatively little effect on the interlobular fibrous tissue, which must therefore be broken down by mechanical means within the digestion chamber so as to release the contained acini and islets. The current way of achieving this in the Ricordi chamber is to place five or six stainless steel balls within the chamber and shake vigorously. The

Development of the islets, exocrine pancreas, and related ducts in the Nile tilapia, Oreochromis niloticus (Pisces: Cichlidae).

Pancreatic development and the relationship of the islets with the pancreatic, hepatic, and bile ducts were studied in the Nile tilapia, Oreochromis niloticus, from hatching to the onset of maturity at 7 months. The number of islets formed during development was counted, using either serial sections or dithizone staining of isolated islets. There was a general increase in islet number with both age and size. Tilapia housed in individual tanks grew more quickly and had more islets than siblings of the same age left in crowded conditions. The pancreas is a compact organ in early development, and

Neural control of islet function by norepinephrine and sympathetic neuropeptides.

It is clear that the sympathoadrenal system has a role in the regulation of endocrine pancreatic function and that the sympathetic nerves of the pancreas can change pancreatic hormone secretion to increase the availability of metabolic fuels. It seems likely that the classical sympathetic neurotransmitter, NE, acts in concert with peptide co-transmitters, such as galanin and NPY. Each is released during the stimulation of pancreatic sympathetic nerves and each is capable of influencing either islet function or pancreatic blood flow. There is considerable indirect evidence that the sympathetic

Exocytotic release of neurotransmitters from nerve fibers in the endocrine pancreas (Brockmann body) of the teleost, Takifugu niphobles.

Exocytotic release of neurotransmitters was ultrastructurally investigated in the endocrine pancreas (Brockmann body) of the teleost, Takifugu niphobles. Exocytotic figures of large cored vesicles were found not only on the surface of nerve fibers facing the endocrine cells directly, but also on their surface facing the space of connective tissues including the blood capillaries. The results indicate that some 'neurotransmitters' are released non-synaptically in the endocrine pancreas and that some 'neurotransmitters' may enter the blood stream as 'neurosecretory hormones.'

Immunohistochemistry of opioid peptides in the guinea pig endocrine pancreas.

Previous immunochemical investigations have demonstrated various opioid peptides in the pancreas. However, controversies exist related to the cellular localization of these peptides in the endocrine pancreas. Therefore, the guinea pig endocrine pancreas was immunohistochemically investigated for the presence of opioid peptides derived from pro-dynorphin, pro-enkephalin or pro-opiomelano-cortin. Immunoreactivities were demonstrated on serial semithin sections by the peroxidase anti-peroxidase technique. In routinely immunostained sections, immunoreactivities for dynorphin A and alpha-neo-endorp

Immunocytochemical localization and immunochemical characterization of an insulin-related peptide in the pancreas of the urodele amphibian, Ambystoma mexicanum.

The pancreas of the axolotl, Ambystoma mexicanum, was investigated by immunocytochemical methods for the presence of immunoreactivity to a number of antisera raised against mammalian insulins. All anti-insulin antisera tested revealed substantial amounts of reaction products confined solely to the aldehyde-fuchsinophilic B cells of the endocrine pancreas. The reactive cell population was detected by use of one polyclonal antiserum against bovine insulin and eight different monoclonal antibodies against insulins from various mammalian species. Six of these antibody clones have known specificity

Morphological and functional aspects of pancreatic islet innervation.

Pancreatic islets are collections of 4 functionally-related endocrine cells distributed nonrandomly in the pancreas. Their major physiological actions center about the regulation of metabolic homeostasis. Experimental evidence shows that, in addition to circulating substates, the islets are controlled by outflow from the central nervous system communicated through autonomic nerves. Islet cells also interact with one another via hormonal messengers and, possibly, electrotonic impulses producing a complex--yet well-controlled--system for the integration of numerous types of signals. An ultrastructural description of the cell types in the endocrine pancreas of the pigeon.

We describe completely the electron-microscopical aspects of the endocrine pancreas in the pigeon. The general characteristics of this organ coincides with that found in granivorous birds and consist of D, A, B, IVth type of cells and acinar-D cells. The D cells are electron-dense and contain many large spheric secretory granules with an adherent membrane. The A cells are spheric or columnal, having few cell organelles, a low electron-density and contain spheric or oval secretory granules with a narrow space between their membrane and content. The B cells are elongated, electron-dense, have ma

Uptake of 65Zn in tapetum lucidum.

The uptake of 65Zn (administered as 65ZnCl2) in the tapetum lucidum of ferrets was studied by whole-body autoradiography. The results showed a slow increase in the labelling of the tapetal cells and 7 days after the 65Zn-injection this tissue contained the highest radioactivity in the body. The strong tapetal labelling was registered in both albino and pigmented animals. Previous biochemical and histochemical studies have indicated that there is a specific localization of zinc in the tapetal cells and the results of the present experiments support these reports. There is evidence of a slow tur

Morphological aspects of the endocrine pancreas in a prednisolone-treated infant.

The endocrine pancreas of a corticosteroid-treated infant during the course of nephrotic syndrome was morphologically studied. Islet cell hyperplasia and dispersed endocrine cell aggregates between acinar tissue were observed. A marked numerical increase of B cells was identified by using cytochemical and immunofluorescent methods. Morphometric data support the morphological observations. The findings suggest that corticosteroid treatment induces endocrine cell replication and neogenesis.
