Phenotypic Diversity in Cell Wall Lignocellulosic Constituents and Ethanol Yield of USDA Guayule and Mariola Germplasm.

Guayule (Parthenium argentatum A. Gray) is a valuable domestic source for rubber and resin. At its center of origin in the Northern Mexico and Southern Texas deserts, guayule, a perennial shrub, is hybridized with its relative species mariola (Parthenium incanum Kunth). As rubber and resin are the main products derived from guayule, there is interest in using guayule bagasse as a bioenergy feedstock to meet the growing bioenergy and biofuel demands. This study aimed to explore and characterize phenotypic diversity in cell wall constituents (lignin, cellulose, and hemicellulose) and their yield…

Chemical signature of Eurois occulta L. outbreaks in the xylem cell wall of Salix glauca L. in Greenland.

Insect defoliations are a major natural disturbance in high-latitude ecosystems and are expected to increase in frequency and severity due to current climatic change. Defoliations cause severe reductions in biomass and carbon investments that affect the functioning and productivity of tundra ecosystems. Here we combined dendro-anatomical analysis with chemical imaging to investigate the direct and lagged effects of insect outbreaks on carbon investment. We analysed the content of lignin vs. holocellulose, i.e. unspecified carbohydrates in xylem samples of Salix glauca L. collected at Iffiartar…

Evaluation of the fate of wastewater effluent organic matter in receiving water: Effect of sequential photochemical and biological processes.

Effluent organic matter (EfOM) discharged from wastewater treatment plants (WWTPs) carry substantial risks to river ecosystems. The fate and … of EfOM stimulated microbial respiration and growth by producing easily metabolizable less aromatic lignin-type molecules, leading to a 21% increase in biodegradation. Conversely, the irradiation of RNOM removed biodegradable lignin-type molecules, causing a 50% decreased in biodegradation. The second and third irradiation phases of EfOM and RNOM produce …

Successful Production and Ligninolytic Activity of a Bacterial Laccase, Lac51, Made in Nicotiana benthamiana via Transient Expression.

Giant panda could have bamboo as their exclusive diet for about 2 million years because of the contribution of numerous enzymes produced by their gut bacteria, for instance laccases. Laccases are blue multi-copper oxidases that catalyze the oxidation of a broad spectrum of phenolic and aromatic compounds with water as the only byproduct. As a "green enzyme," laccases have potential in industrial applications, for example, when dealing with degradation of recalcitrant biopolymers, such as lignin. In the current study, a bacterial laccase, Lac51, originating from Pseudomonas putida and identifie…

Characterization, expression, and functional analysis of the pathogenesis-related gene PtDIR11 in transgenic poplar.

Lignins and lignans are important for plant resistance to pathogens. Dirigent (DIR) proteins control the regio- and stereo-selectivity of coniferyl alcohol in lignan and lignin biosynthesis. DIR genes have been implicated in defense-related responses in several plant species, but their role in poplar immunity is unclear. We cloned PtDIR11 from Populus trichocarpa; we found that overexpression of PtDIR11 in poplar improved the lignan biosynthesis and enhanced the resistance of poplar to Septotis populiperda. PtDIR11 has a typical DIR domain; it belongs to the DIR-b/d family and is expressed in …

Metagenomic profiling reveals lignocellulose degrading system in a microbial community associated with a wood-feeding beetle.

The Asian longhorned beetle (Anoplophoraglabripennis) is an invasive, wood-boring pest that thrives in the heartwood of deciduous tree species. A large impediment faced by A. glabripennis as it feeds on woody tissue is lignin, a highly recalcitrant biopolymer that reduces access to sugars and other nutrients locked in cellulose and hemicellulose. We previously demonstrated that lignin, cellulose, and hemicellulose are actively deconstructed in the beetle gut and that the gut harbors an assemblage of microbes hypothesized to make significant contributions to these processes. While lignin degrad…

Molecular and physiological mechanisms underlying magnesium-deficiency-induced enlargement, cracking and lignification of Citrus sinensis leaf veins.

Little is known about the physiological and molecular mechanisms underlying magnesium (Mg)-deficiency-induced enlargement, cracking and lign … iency veins of lower leaves (MDVLL) than those in Mg-sufficiency veins of lower leaves (MSVLL), but lignin concentration was higher in MDVLL than that in MSVLL. However, all four parameters were similar between Mg-deficiency veins of upper leaves (MDVUL) and Mg-sufficiency veins of upper leaves (MSVUL). Using label-free, liquid chromatography-tandem mass …

Short-term facilitation of microbial litter decomposition by ultraviolet radiation.

Solar radiation plays an important role in carbon cycling by increasing the decomposition rates of plant litter and soil organic matter (i.e … bial respiration in an incubation experiment. We also found that UV exposure had limited effects on lignin and other cell wall structures, but one month of microbial decomposition (in absence of UV exposure) significantly reduced lignin β-aryl ether inter-unit linkages and acetylated xylans. These results indicate that abiotic photodegradation alone was …

Plant Tissue Cultures.

Plant tissue cultures are an efficient system to study cell wall biosynthesis in living cells in vivo. Tissue cultures also provide cells and culture medium from which enzymes and cell wall polymers can easily be separated for further studies. Tissue cultures with tracheary element differentiation or extracellular lignin formation have provided useful information related to several aspects of xylem and lignin formation. In this chapter, methods for nutrient medium preparation and callus culture initiation and its maintenance as well as those for protoplast isolation and viability observation a…

One-pot quantitative hydrolysis of lignocelluloses mediated by black liquor.

Black liquor from the kraft process facilitates quantitative biomass hydrolysis converting cellulose and hemicellulose into organic acids such as lactic acid (~50%), and lignin into small molecular aromatics, without gasification and black tar formation. Oxygen transfer between lignin and carbohydrates may be the mechanism. With this method, three tons of lignocellulosic biomass can potentially produce up to one ton of lactic acid, and one ton of small molecular aromatics. This novel usage of black liquor is environmentally viable because it is accompanied by significant emission reduction of …

Catabolism of acetosyringone and co-metabolic transformation of 2,4,6-trichlorophenol by a novel FAD-dependent monooxygenase.

Acetosyringone (AS), a prototypical syringyl-type monomer of lignin, functions as a model compound for the study of microbial catabolism of S-lignin-derived aromatics. In this study, we present the discovery of a novel metabolic pathway for AS catabolism, initiated by a previously uncharacterized FAD-dependent oxidoreductase, designated AsdA. In contrast to the sole previously documented AS funneling route, which entails side chain modification and conversion to syringic acid, AsdA catalyzes direct hydroxylation of the aromatic core. This represents a mechanistically distinct entry into centra…

A Library of Versatile Lignocellulose-Derived Monomers From 5-(Chloromethyl)furfural.

The valorization of biomass-derived platform chemicals into functional monomers is a promising strategy for developing more sustainable polymeric materials. Herein, we report a versatile synthetic approach for the preparation of a library of lignocellulosic (LC) monomers by coupling 5-(chloromethyl)furfural (CMF), once considered a waste product of furan production, with selected lignin-derived monomers. The reaction was optimized using vanillin, affording a bis aldehyde monomer (Fur-Van) in 93% yield after only 2 h under mild conditions using K2CO3 as the base. The methodology was successfull…

Strong and flexible lignocellulosic film fabricated via a feasible molecular remodeling strategy.

Biomass-derived sustainable film is a promising alternative to synthetic plastic, but hampered by strength, toughness and flexibility trade-off predicament. Here, a feasible and scalable strategy was proposed to fabricate strong and flexible lignocellulosic film through molecular reconstruction of cellulose and lignin. In this strategy, polyphenol lignin was absorbed and wrapped on the surface of cellulose fiber, forming strong interfacial adhesion and cohesion via intramolecular and intermolecular hydrogen bonding. Further, covalent ether bond was generated between the hydroxyl groups of lign…

An Aqueous Fungal-Enzymatic Strategy for the Surface Engineering of Bamboo toward Sustainable High-Performance Biocomposites.

Bamboo biocomposites hold significant potential for applications in the construction, automotive, and logistics sectors. However, their widespread adoption is hindered by inherently weak interfacial bonding, primarily due to the poor surface wettability and unfavorable surface characteristics of natural bamboo. This study presents an innovative fungal-enzymatic pretreatment strategy by inducing the white-rot fungus Trametes versicolor (T. versicolor) to secrete highly active laccase, which achieves partial depolymerization and modification of lignin, thereby regulating and controlling bamboo s…
