Mitochondria and neuroplasticity.

The production of neurons from neural progenitor cells, the growth of axons and dendrites and the formation and reorganization of synapses are examples of neuroplasticity. These processes are regulated by cell-autonomous and intercellular (paracrine and endocrine) programs that mediate responses of neural cells to environmental input. Mitochondria are highly mobile and move within and between subcellular compartments involved in neuroplasticity (synaptic terminals, dendrites, cell body and the axon). By generating energy (ATP and NAD(+)), and regulating subcellular Ca(2+) and redox homoeostasi

[Epidemiology from the Trans Atlantic Inter-Society Consensus Group Guidelines II].

Although there is little direct information or prospective cohort study on the incidence and prevalence of critical limb ischemia (CLI), it can be calculated that the incidence of CLI is approximately 500 to 1,000 per million per year in Western countries and 100 to 200 in Japan. The most important risk factor for the development of CLI is diabetes as well as age, cigarette smoking, lipid abnormalities, and low ankle brachial pressure index. Without pharmacotherapy or interventions, approximately 40% of CLI patients will lose their leg within 6 months, whereas up to 20% will die. As most gener

Separation of microorganisms using electromigration techniques.

Like other colloidal particles bacteria have a surface charge that originates from the ionization of surface molecules and of the adsorption of ions from solution. Bacterial cell wall and membranes containing numerous proteins, lipid molecules, teichoic acids, lipopolisaccharides which give them characteristic charge. Therefore, bacterial cells undergo electrophoresis in a free solution with their own mobility depending on ionic strength and pH of buffer solution. Various electromigration techniques can be used to separate and determine the intact cells. Successful separation of five species o

The roles of estrogen and progesterone in regulating carbohydrate and fat utilization at rest and during exercise.

OBJECTIVE: Compared with men, women use more fat and less carbohydrate to fuel exercise at the same relative intensity. Circulating levels of estrogen and progesterone are likely to play an important role in explaining this gender difference in exercise substrate utilization. METHODS: Studies, mainly using animal models, have shown that estrogen increases fatty acid availability (lipolysis) and decreases carbohydrate availability and uptake. Studies conducted in humans corroborate the reduction in carbohydrate turnover and oxidation in the presence of estrogen, but the impact on fatty acid av

Ultrastructural autoradiographic localization of exogenous arachidonic acid in cultured endothelial and smooth muscle cells.

The uptake and intracellular localization of exogenous arachidonic acid (AA) were investigated in cultured endothelial (EC) and smooth muscle cells (SMC) isolated from bovine aorta. The [14C]AA uptake was assessed biochemically and by light and electron microscopic autoradiography. The highest values of silver grain surface density were associated with the mitochondria, lysosomes, and the Golgi apparatus of the EC. The grain linear density was greater on the nuclear envelope than on plasmalemma. On SMC, the grain density was highest on lipid droplets whereas the linear densities of the nuclear

Prevention of ventricular fibrillation, acute myocardial infarction (myocardial necrosis), heart failure, and mortality by bretylium: is ischemic heart disease primarily adrenergic cardiovascular disease?

It is widely, but mistakenly, believed that ischemic heart disease (IsHD) and its complications are the sole and direct result of reduced co … lipid metabolism that is associated with CAD and IsHD by catecholamine-induced metabolic actions on lipid mobilization by activation of phospholipases. This may also be part of toxic catecholamine hypermetabolic actions by enhancing deleterious cholesterol and lipid actions in damaging coronary vessels by plaque formation as well as inducing obstructive coronary spa …

Adolescent late-onset riboflavin-responsive multiple acyl-CoA dehydrogenase deficiency manifesting with severe multi-organ failure: a case report.

BACKGROUND: Multiple acyl-CoA dehydrogenase deficiency (MADD) is a rare autosomal recessive disorder characterized by dysfunctional acyl-CoA dehydrogenases, leading to lipid accumulation in various tissues, including skeletal muscles, liver, and cardiac muscles, etc. Late-onset MADD presents with progressive muscular symptoms (muscle weakness, atrophy, and myalgia) and even multisystem disorders (metabolic encephalopathy, dilated cardiomyopathy, liver failure, acute kidney injury, respiratory failure, and cardiac arrest). Over the past decade, only one case of childhood late-onset MADD with se

Interaction of linear cationic peptides with phospholipid membranes and polymers of sialic acid.

Polysialic acid (PSA) is a natural anionic polymer typically occurring on the outer surface of cell membranes. PSA is involved in cell signaling and intermolecular interactions with proteins and peptides. The antimicrobial potential of peptides is usually evaluated in model membranes consisting of lipid bilayers but devoid of either PSA or its analogs. The goal of this work was to investigate the possible effect of PSA on the structure of melittin (Mlt) and latarcins Ltc1K, Ltc2a, and the activity of these peptides with respect to model membranes. These peptides are linear cationic ones derive

Interleukin-6 induces fat loss in cancer cachexia by promoting white adipose tissue lipolysis and browning.

BACKGROUND: Cancer cachexia is a progressive and multi-factorial metabolic syndrome characterized by loss of adipose tissue and skeletal muscle. White adipose tissue (WAT) lipolysis and white-to-brown transdifferentiation of WAT (WAT browning) are proposed to contribute to WAT atrophy in cancer cachexia. Chronic inflammation, mediated by cytokines such as tumor necrosis factor alpha (TNF-α) and interleukin-6 (IL-6), has been reported to promote cancer cachexia. However, whether chronic inflammation promotes cancer cachexia by regulating WAT metabolism and the underlying mechanism remains uncle

Beneficial effect of hydrogen-rich saline on cerebral vasospasm after experimental subarachnoid hemorrhage in rats.

Cerebral vasospasm (CV) remains a common and devastating complication in patients with subarachnoid hemorrhage (SAH). Despite its clinical significance and extensive research, the underlying pathogenesis and therapeutic perspectives of CV remain incompletely understood. Recently, it has been suggested that molecular hydrogen (H(2)) can selectively reduce levels of hydroxyl radicals (·OH) and ameliorate oxidative and inflammatory injuries to organs in many models. However, whether H(2) can ameliorate CV after SAH is still unknown. This study was designed to evaluate the efficacy of H(2) in prev

Cholesterol Content Regulates the Interaction of αA-, αB-, and α-Crystallin with the Model of Human Lens-Lipid Membranes.

α-Crystallin (αABc) is a major protein comprised of αA-crystallin (αAc) and αB-crystallin (αBc) that is found in the human eye lens and works as a molecular chaperone by preventing the aggregation of proteins and providing tolerance to stress. However, with age and cataract formation, the concentration of αABc in the eye lens cytoplasm decreases, with a corresponding increase in the membrane-bound αABc. This study uses the electron paramagnetic resonance (EPR) spin-labeling method to investigate the role of cholesterol (Chol) and Chol bilayer domains (CBDs) in the binding of αAc, αBc, and αABc

Engineering hydrogel-in-oleogel bigels for reduced-fat and egg-free mayonnaise: relating phase architecture to functional performance.

Hydrogel-in-oleogel (HG-in-OG) bigels have attracted increasing interest as structured lipid systems for fat replacement, yet the relationships between their phase architecture and functionality in complex foods remain insufficiently understood. In this study, food-grade HG-in-OG bigels were prepared by combining phytosterol/γ-oryzanol oleogels with sodium alginate hydrogels at oleogel fractions ranging from 40% to 80%. Increasing the oleogel fraction progressively refined the dispersed hydrogel domains from 4.21 ± 1.57 μm to 1.77 ± 0.69 μm, accompanied by enhanced network rigidity, reduced mo

Tracking biomolecular pathogenesis and recovery in neonatal calf diarrhea via serum infrared spectroscopy.

Neonatal calf diarrhea (NCD) is an important health and economic problem in dairy cattle production because of its association with morbidity, mortality, and production losses. Moving beyond basic diagnostic classification, this study aims to track the biomolecular pathogenesis and recovery in NCD by evaluating host-level molecular damage using serum ATR-FTIR spectroscopy. Serum samples obtained from 1 to 20-day-old Holstein calves were analyzed in the mid-infrared spectral range, and the integral areas, bandwidths, and spectrochemical indices of critical bands were calculated. During the acti

Hyperspectral imaging as an objective diagnostic tool for secondary lymphedema in breast cancer patients.

BACKGROUND: Secondary lymphedema is a common complication after breast cancer treatment. Traditional diagnostic techniques often lack objectivity, are prone to operator bias, and show limited reproducibility. There is a need for diagnostic methods that are reliable, noninvasive, and suitable for monitoring disease progression. This study (German Clinical Trials Register, DRKS-ID: DRKS00027366) aimed to evaluate hyperspectral imaging as a tool for detecting and classifying secondary lymphedema. METHODS: We conducted a prospective multicenter study including 58 women with unilateral secondary l

Effects of dietary camelina oil intake on skeletal muscle metabolism and sarcopenia in older mice.

The age-related decrease in skeletal muscle mass and function, mostly known as sarcopenia, increases the risk of mobility impairments, chronic disease and early mortality. Physical activity and targeted dietary approaches are the most effective intervention to prevent or limit sarcopenia. Omega-3 polyunsaturated fatty acids (PUFA) could alleviate some aspects of age-related diseases. We investigated the effect of a chronic intake of a diet containing a high content of omega-3 PUFA in old mice exposed to a normocaloric or an obesogenic diet. Female C57BL/6J mice received a low-fat or a high-fat

Substitution of bridge carbons for sulphur in calix[4]arene-bis-α-hydroxymethylphosphonic acid transformed mobile carrier into ionic channel accompanied with evoked muscle contraction and impaired neurotransmission powered by membrane action of resulting thiocalix[4]arene-bis-α-hydroxymethylphosphonic acid.

The action of calix[4]arenes C-424, C-425 and C-1193 has been investigated on suspended cholesterol/egg phosphatidylcholine lipid bilayer in a voltage-clamp mode. Comparative analysis with the membrane action by calix[4]arene-bis-α-hydroxymethylphosphonic acid (C-99) has shown that the substitution of bridge carbons for sulphur and addition of another methyl group to two alkyl tales in the lower rim of former dipropoxycalix[4]arene C-99 transformed mobile carrier that C-99 created in lipid bilayer (Shatursky et al., 2014) into a transmembrane pore as exposure of the bilayer membrane to sulphur

Diet-induced induction of hepatic serine/threonine kinase STK38 triggers proinflammation and hepatic lipid accumulation.

Nonalcoholic fatty liver disease (NAFLD) is one of the most common liver diseases worldwide. Although the involvement of chronic overnutrition, systemic inflammation, and insulin resistance in the development of NAFLD is well-established, however, the associations among these remain to be elucidated. Several studies have reported that chronic overnutrition, such as excessive consumption of fats (high-fat diet, HFD), can cause insulin resistance and inflammation. However, the mechanisms by which HFD exerts inflammation and thereby promotes insulin resistance and intrahepatic fat accumulation re

Dapagliflozin ameliorates diabetic renal injury through suppressing the self-perpetuating cycle of inflammation mediated by HMGB1 feedback signaling in the kidney.

Dapagliflozin, the Sodium-glucose cotransporter 2 (SGLT2) inhibitor class of glucose-lowering agents, has shown the significantly nephroprotective effects to reduce the risk of kidney failure in diabetes. However, the underlying mechanisms are incompletely understood to explain the beneficial effects of dapagliflozin on kidney function. Here, we demonstrated that the administered of dapagliflozin for 12 weeks improved the proteinuria, histomorphology damage, oxidative stress, and macrophage infiltrations in the kidney of streptozotocin (STZ)-induced diabetic mice. Meanwhile, dapagliflozin atte
