Iron overload reprogramming lipid metabolism through the IRP1-SCAP axis in fibroblast-like synoviocytes aggravates bone destruction in rheumatoid arthritis.

Rheumatoid arthritis (RA) is a leading cause of disability globally. Although iron accumulation in arthritic lesions has been observed in patients with RA, its specific contribution to disability outcomes remains unclear. Here we demonstrate a comprehensive multiomics approach to elucidate the impact and underlying mechanisms of iron overload in RA. First, clinical radiology in an RA cohort reveals a positive correlation between elevated ferrous iron levels in synovial fluid and joint damage extent. Iron chelator DFO administration significantly alleviates bone destruction in the K/BxN serum-t…

Targeting an IL-17/IL-36 pathogenic hub in hidradenitis suppurativa with a bispecific antibody.

Hidradenitis suppurativa (HS) is a chronic inflammatory skin disorder with a high unmet need, particularly for treatment-resistant tunnels. Integrated multiomics analysis revealed that HS tunnels are spatially organized, self-amplifying inflammatory circuits driven by synergistic IL-17/IL-36γ activation of a unique keratinocyte subset, which activates neutrophils and inflammatory fibroblasts. This mechanistic insight coupled with spatial coenrichment of IL-17A and IL-36R provided the rationale for HB0043, a bispecific antibody that leverages IL-36R binding to locally dual-block both cytokines.…

PWK genetic background reprograms heterochromatin and malignant epithelial states in Neu-driven mammary tumors.

Genetic background can alter the latency and behavior of cancers initiated by the same oncogenic driver, but how inherited variation reshapes the regulatory landscape that permits tumor development remains poorly understood. The PWK genetic background accelerates mammary tumor onset in the NeuT model. We compared PWK NeuT and BALB NeuT tumors using tumor-level RNA-seq, histone ChIP-seq, and single-cell multiome profiling. PWK tumors exhibited broad transcriptional reprogramming characterized by reduced immune-associated expression and increased epithelial differentiation and neuronal-like prog

Phillyrin protects against myocardial ischemia/reperfusion injury by promoting KNL1 K605 acetylation to inhibit the p53/p21 pathway.

Reperfusion therapy is critical for acute myocardial infarction but is often accompanied by myocardial ischemia/reperfusion injury (MIRI). Phillyrin, a natural lignan from Forsythia suspensa, exerts anti-inflammatory and antioxidant effects; however, its role and mechanism in MIRI remain unclear. In this study, HL-1 cardiomyocytes are subjected to oxygen-glucose deprivation/reperfusion (OGD/R). Cell viability, apoptosis, oxidative stress, and inflammation are measured after phillyrin treatment. Multiomics (mRNA-seq, proteomics, and acetylproteomics) is used to identify key targets and pathways…
