Injury to skeletal muscle fibers of mice following lengthening contractions.

We tested the hypothesis that lengthening contractions result in greater injury to skeletal muscle fibers than isometric or shortening contractions. Mice were anesthetized with pentobarbital sodium and secured to a platform maintained at 37 degrees C. The distal tendon of the extensor digitorum longus muscle was attached to a servomotor. A protocol consisting of isometric, shortening, or lengthening contractions was performed. After the contraction protocol the distal tendon was reattached, incisions were closed, and the mice were allowed to recover. The muscles were removed after 1-30 days, a

Androgen cytosol binding during compensatory overload-induced skeletal muscle hypertrophy.

This study was undertaken to evaluate whether the increased androgen cytosol binding is an early or later event in the sequence of skeletal muscle hypertrophy induced by surgical overload. Following removal of the synergistic gastrocnemius and soleus muscles, plantaris muscle weights of overloaded hypophysectomized made rats were heavier than those in the control by 29% at 2 days, 41% at 7 days, 38% at 14 days, and 47% at 35 days. Androgen cytosol receptor binding capacities (femtomoles per milligram protein), determined using a synthetic androgen, [3H]methyltrienolone (R1881), were higher tha

Skeletal muscle actin mRNA. Characterization of the 3' untranslated region.

Plasmids p749, p106, and p150 contain cDNA inserts complementary to rat skeletal muscle actin mRNA. Nucleotide sequence analysis indicates the following sequence relationships: p749 specifies codons 171 to 360; p150 specifies codons 357 to 374 together with 120 nucleotides of the 3'-non-translated region; p106 specifies the last actin amino acid codon, the termination codon and the entire 3' non-translated region. Plasmid p749 hybridized with RNA extracted from rat skeletal muscle, cardiac muscle, smooth (stomach) muscle, and from brain. It also hybridizes well with RNA extracted from skeletal

Distinct subtypes of type I fibers of human skeletal muscle.

We have identified two distinct and consistent type I fiber subtypes in histochemically normal biopsies (vastus lateralis or biceps brachialis) from 12 healthy persons, both sexes, ages 15 to 42. The type I fibers were uniformly light with the regular ATPase (9.4) and uniformly dark with the acid-preincubated reverse ATPase reactions. However, the subtype IA fibers were stained darker than subtype IB fibers with the nicotinamide adenine nucleotide dehydrogenase-tetrazolium reductase, Oil red O, reduced nicotinamide adenine dinucleotide phosphate-tetrazolium reductase, nonspecific esterase, suc

Diabetic Polyneuropathy Patients Exhibit an Altered Skeletal Muscle Proteome Phenotype.

PURPOSE: Diabetic polyneuropathy (DPN) is a debilitating complication of diabetes that affects motor nerves and may impair skeletal muscle health. This study investigated skeletal muscle proteomic alterations associated with DPN compared with type 2 diabetes without DPN (T2D) and healthy controls (CON). EXPERIMENTAL DESIGN: An exploratory proteomic profiling was performed in 15 participants divided into three groups based on clinical and neurophysiological diagnosis (DPN, n = 5; T2D, n = 5; CON, n = 5). Biopsies from the gastrocnemius lateralis muscle were analyzed using LC-MS/MS-based proteo

Miro1 mediates skeletal muscle insulin resistance in type 2 diabetes.

Imbalanced skeletal muscle mitochondrial dynamics contribute to the onset and progression of type 2 diabetes (T2D) by mechanisms that remain incompletely understood. Here, we examined the role of Mitochondrial Rho GTPase 1 (Miro1), an outer mitochondrial membrane enzyme, in regulating skeletal muscle insulin action and glucose homeostasis in type 2 diabetes (T2D). In a cross-sectional clinical study, we show that Miro1 accumulates in the skeletal muscle of patients with obesity and T2D, which was further replicated in mouse models of T2D. This accumulation is explained by inadequate insulin-me

Flow Cytometric Methods for ROS Quantification in Skeletal Muscle Myotubes.

Flow cytometry is a high-throughput technique which can be used to analyze and quantify characteristics of individual cells. Flow cytometry hydrodynamically focuses cells past a series of size and fluorescent detectors to characterise cellular morphology and phenotype. This process of hydrodynamic focusing has previously been shown to be incompatible with myotubes due to their larger size relative to other cell types. Here, we describe the culture and differentiation of myotubes and how myotubes can be analyzed via flow cytometry to quantify changes in cellular and mitochondrial-specific ROS p

Lipidomic Sample Preparation to Detect Phosphatidylcholine Alterations in Skeletal Muscle.

Altered phospholipid compositions in skeletal muscle occur in muscular dystrophy and other pathological conditions; however, origins and relationships to the disease course are unclear. Liquid chromatography-mass spectrometry (LC-MS) technologies allow the determination of phospholipid compositions and alteration patterns in disease states. Here, we describe a basic protocol to prepare methanolic extracts from skeletal muscle, which may be analyzed by lipidomic LC-MS in order to detect compositional alterations in phosphatidylcholine, a major phospholipid species of skeletal muscle, as well as

Vitamin B12 Improves Skeletal Muscle Mitochondrial Biology in Aged Mice.

Age-related skeletal muscle deterioration is a commonly reported disability among older adults, attributed to several factors including mitochondrial dysfunction, a major hallmark of aging. Therapies to attenuate or reverse mitochondrial decline are limited. Despite identified positive relationships between vitamin B12 (B12) and mitochondrial biology, the impact of B12 supplementation on skeletal muscle mitochondria, in advanced aged, has not been examined. Thus, the impact of B12 supplementation on skeletal muscle mitochondrial biology was examined in (i) aged female mice, given 12 weeks of B

Hypoxia and hypercapnia elicit overlapping but distinct skeletal muscle toxicities.

Skeletal muscle dysfunction is strongly associated with elevated mortality in acute and chronic pulmonary diseases. Hypoxaemia and hypercapnia, which are central hallmarks of respiratory failure, represent critical cellular signals regulating muscle loss, causing disruption of skeletal muscle mass, myofibre metabolic profile and oxidative capacity, and regenerative potential after injury. Both hypoxaemia and hypercapnia elicit alterations in protein synthesis and degradation, myogenesis and autophagy - key cellular processes that significantly impact skeletal muscle integrity. Recent data have

A Method for Fabricating Long Decellularized Scaffolds from Skeletal Muscle.

Skeletal muscle tissue engineering (SMTE) is a rapidly evolving field with applications spanning regenerative medicine, disease modeling, drug screening, and biohybrid robotics. Effective SMTE requires scaffolds that reproduce the anisotropic architecture and mechanical properties of native muscle while supporting macroscale tissue formation. Decellularized tissues are strong candidates; however, existing approaches face key limitations. Whole-muscle decellularization requires complex perfusion systems and often fails to fully clear the tissue core. In contrast, minced-tissue processing comple

Reference Methods for Measuring Skeletal Muscle Mass: A Critical Perspective.

Skeletal muscle (SM) is an integral organ component in the pathophysiology of many acute and chronic diseases. But is there a 'gold' standard or accepted reference method for quantifying the amount and composition of human SM mass? Exploring that question led us to recognize the existence of a SM measurement paradigm that divides methods into two broad categories, in vitro and in vivo. In vitro methods quantify SM mass, weighing intact muscles as part of whole cadaver evaluations, only 51 of which are reported in medical literature with no recent additions. In vivo methods are used to evaluate

Cellular and molecular pathways linking obesity to skeletal muscle dysfunction.

Obesity is increasingly recognized as a condition that directly impairs skeletal muscle structure, metabolism, and endocrine function through complex molecular and cellular mechanisms extending beyond the classical concept of sarcopenic obesity. This narrative review aimed to synthesize current evidence regarding the intracellular signaling pathways, metabolic alterations, and endocrine interactions involved in obesity-induced skeletal muscle dysfunction independent of overt sarcopenia. Relevant literature from experimental, clinical, and review studies was identified through searches of PubMe

Sex-divergent skeletal muscle remodeling emerges early in cancer cachexia.

Cancer cachexia is characterized by progressive skeletal muscle wasting and dysfunction, yet the early events that precede overt muscle wasting remain poorly defined. Here, we utilized a time-coursed C26 carcinoma model in male and female mice to define the precachectic transcriptomic, proteostatic, immune, and metabolic adaptations between sexes. Males progressed more rapidly to cachexia and exhibited earlier impairments in body composition and grip strength, with shifts in muscle fiber size distribution detectable before changes in muscle mass. Transcriptomic analysis of the tibialis anterio
