Mechanisms of chromium carcinogenicity and toxicity.

Chromium, like many transition metal elements, is essential to life at low concentrations yet toxic to many systems at higher concentrations. In addition to the overt symptoms of acute chromium toxicity, delayed manifestations of chromium exposure become apparent by subsequent increases in the incidence of various human cancers. Chromium is widely used in numerous industrial processes, and as a result is a contaminant of many environmental systems. Chromium, in its myriad chemical forms and oxidation states, has been well studied in terms of its general chemistry and its interactions with biol

Open-channel block of human TRPV6 by polyamine spermine.

Polyamines are organic cations that are present at sub-millimolar concentrations in the cytoplasm and extracellular fluids and serve as versatile modulators of TRP channels, fine-tuning their functions in physiological and pathological contexts, including pain, inflammation and cancer. Despite extensive functional studies, the structural basis by which polyamines regulate TRP channels remains unclear. Here, we combine calcium imaging, electrophysiology, cryo-electron microscopy, mutagenesis and molecular dynamics simulations to study regulation of human TRPV6 by polyamine spermine. Our functio…

Exploring the Landscape of Aptamers: From Cross-Reactive to Selective to Specific, High-Affinity Receptors for Cocaine.

We reported over 20 years ago MNS-4.1, the first DNA aptamer with a micromolar affinity for cocaine. MNS-4.1 is based on a structural motif … we now report share structural motifs with the previously reported receptor for serotonin. Further mutagenesis studies revealed a palindromic, highly adaptable, broadly cross-reactive hydrophobic motif that could be rebuilt through mutagenesis, expansion of linker regions, and selections into receptors with exceptional affinities and varying specificities.

[Locomotion research with zebrafish].

Recent genetic analysis of behavior was successfully performed in C. elegans and Drosophila. However, since the organization and morphology of their nervous system is different from those of vertebrates, it is unclear how applicable these findings are for vertebrates. Additionally, electrophysiological analysis is difficult in these invertebrates. On the other hand, electrophysiology is well established in vertebrate animals, such as mice and Xenopus, but large-scale mutagenesis is costly and difficult with these vertebrates. I started a genetic analysis of behavior in zebrafish to overcome th…

The response regulator Rrp2 is essential for the expression of major membrane lipoproteins in Borrelia burgdorferi.

Borrelia burgdorferi (Bb), the agent of Lyme disease, exists in nature through a complex enzootic life cycle that involves both ticks and mammals. As Bb transitions between its two diverse niches, profound adaptive changes occur that are reflected in differential patterns of gene expression, particularly involving lipoprotein genes. Using a mutagenesis approach, we show that Rrp2 (gene BB0763), one of the proteins predicted by the Bb genome (www.tigr.org) to be a response regulator of a two-component sensory transduction system, is a pivotal regulator governing the expression of major membrane…

In vitro and in silico characterization of the inhibition of Kir4.1 channels by aminoglycoside antibiotics.

BACKGROUND AND PURPOSE: Aminoglycoside antibiotics are positively charged molecules that are known to inhibit several ion channels. In this study, we have shown that aminoglycosides also inhibit the activity of Kir4.1 channels. Aminoglycosides inhibit Kir4.1 channels by a pore-blocking mechanism, plugging the central vestibule of the channel. EXPERIMENTAL APPROACH: Patch-clamp recordings were made in HEK-293 cells transiently expressing Kir4.1 channels to analyse the effects of gentamicin, neomycin and kanamycin. In silico modelling followed by mutagenesis were realized to identify the residu…

An ENU-induced mutation in AP-2alpha leads to middle ear and ocular defects in Doarad mice.

One of the advantages of N-ethyl- N-nitrosourea (ENU)-induced mutagenesis is that, after randomly causing point mutations, a variety of alleles can be generated in genes leading to diverse phenotypes. For example, transcription factor AP-2alpha ( Tcfap2a) null homozygote mice show a large spectrum of developmental defects, among them missing middle ear bones and tympanic ring. This is the usual occurrence, where mutations causing middle ear anomalies usually coincide with other abnormalities. Using ENU-induced mutagenesis, we discovered a new dominant Tcfap2a mutant named Doarad ( Dor) that ha…

Partial silencing of the NEC1 gene results in early opening of anthers in Petunia hybrida.

The NEC1 gene, previously isolated from Petunia hybrida, is expressed at high levels in nectaries, and in a very localized fashion in stamens, particularly in the anther stomium cells and the upper part of the filament. To elucidate the function of the NEC1 gene, co-suppression was employed for down-regulation of NEC1 expression, and transposon insertion mutagenesis was used to knock out the NEC1 function. Among the transgenic plants and plants carrying dTph1 inserted in the NEC1 gene, an "early open anther" phenotype was observed. In this mutant phenotype, the anthers already open in young fl…

The charybdotoxin receptor of a Shaker K+ channel: peptide and channel residues mediating molecular recognition.

Charybdotoxin (CTX) is a peptide of known structure that inhibits Shaker K+ channels by a pore-blocking mechanism. Point mutagenesis of all 30 solvent-exposed residues identified the part of the CTX molecular surface making contact with the receptor in the K+ channel. All close-contact residues are clustered in a well-defined interaction surface; the shape of this surface implies that the outer opening of the Shaker channel conduction pore abruptly widens to a 25 x 35 A plateau. A mutagenic scan of the S5-S6 linker sequence of the Shaker K+ channel identified those channel residues influencing…

Breakage in breakage-fusion-bridge cycle: an 80-year-old mystery.

2021 marked the 80th anniversary of Barbara McClintock's pioneering article on the breakage-fusion-bridge (BFB) cycle. Of the three steps of the BFB cycle, breakage remains the least understood despite its major contribution to mutagenesis. We discuss recent findings shedding light on how chromatin bridges break in yeast and animal cells.

Is tetrachloroethylene genotoxic or not?

A recent study published in Mutagenesis, in which the ability of tetrachloroethylene to induce DNA damage, detected by the alkaline comet assay, in mouse tissues (liver and kidney) was examined, has resulted in different interpretations of the data for liver as either positive or negative for genotoxicity. Here, I discuss the statistical approaches used and comment on the different conclusions reached.

Improving the Activity of Phi29 DNA Polymerase by SaProt Prediction.

Phi29 DNA polymerase has broad application value due to its excellent functions such as strand displacement. However, the catalytic efficiency of the natural Phi29 DNA polymerase is low, which greatly limits its application scope. This study attempts to modify Phi29 DNA polymerase using the deep prediction model SaProt to improve its catalytic efficiency. First, the 3D structure of Phi29 DNA polymerase was predicted by AlphaFold, and the reliability of the model was verified by tools such as PROCHECK. Subsequently, SaProt was used to perform saturation mutagenesis calculations on the entire se…

Wall incorporation of the β-1,3-glucan cross-linking protein Pir1 in the human pathogen Candida albicans is facilitated by the presence of two or more Pir repeat units.

The Pir1 protein in the prevalent pathogenic yeast Candidaalbicans has been hypothesized to be important for cellular integrity by crosslinking cell wall β-1,3-glucans. However, recent studies with deletion mutants have reported contrasting results concerning its actual importance for wall integrity. Here, we present functional characterization of the two members of the Pir family (Pir1 and Pir32) as well as protein structure modeling and mutagenesis studies to elucidate how Pir1, the most important family member, is incorporated into the cell wall. Our data show that Pir1 indeed is involved i…

Porin A and α/β-hydrolase are necessary and sufficient for hemolysis induced by Bartonella bacilliformis.

Carrion's disease is endemic to the South American Andes and is characterized by fatal hemolytic anemia. This neglected tropical disease is caused by Bartonella bacilliformis, a fastidious and slow-growing pathogen difficult in genetic manipulation. In this study, we determine that porin A and α/β-hydrolase are both necessary and sufficient for hemolysis induced by B. bacilliformis. These genes are identified through a screen of a Tn5 transposon mutant library. Using markerless deletion mutagenesis, porin A and α/β-hydrolase deletion mutants are generated and functionally analyzed by hemolysis…
