Magnolol inhibits colonic motility through down-regulation of voltage-sensitive L-type Ca2+ channels of colonic smooth muscle cells in rats.

This study aimed to investigate the effect of magnolol (5,5'-diallyl-2,2'-biphenyldiol) on contraction in distal colonic segments of rats and the underlying mechanisms. Colonic segments were mounted in organ baths for isometric force measurement. Whole-cell voltage-sensitive L-type Ca(2+) currents were recorded on isolated single colonic smooth muscle cells using patch-clamp technique. The spontaneous contractions and acetylcholine (ACh)- and Bay K 8644-induced contractions were inhibited by magnolol (3-100 μM). In the presence of Bay K8644 (100 nM), magnolol (10-100 μM) inhibited the contract

[Studying cGMP-dependent mechanisms of vinpocetine effect on smooth muscle cells].

The results of the membrane potential measurements (by the double sucrose gap junction technique) and the smooth muscle tension determination (by the mechanical force measurements) in the rat aorta showed that vinpocetine potentiates the effect of sodium nitroprusside and nitroglycerin on the smooth muscle cells. In the concentration range of 2-20 microM, vinpocetine produced a dose-dependent inhibition of the Ca2+ conductivity of the membrane and decreased the smooth muscle contractility response. At a concentration of 1 microM, the drug acted as an inhibitor of the phosphodiestherase (PDE) a

Endogenous cytosolic Ca(2+) buffering is necessary for TRPM4 activity in cerebral artery smooth muscle cells.

The melastatin transient receptor potential (TRP) channel, TRPM4, is a critical regulator of smooth muscle membrane potential and arterial tone. Activation of the channel is Ca(2+)-dependent, but prolonged exposures to high global Ca(2+) causes rapid inactivation under conventional whole-cell patch clamp conditions. Using amphotericin B perforated whole cell patch clamp electrophysiology, which minimally disrupts cytosolic Ca(2+) dynamics, we recently showed that Ca(2+) released from 1,2,5-triphosphate receptors (IP(3)R) on the sarcoplasmic reticulum (SR) activates TRPM4 channels, producing su

Conditional Loss of MEF2C Expression in Osteoclasts Leads to a Sex-Specific Osteopenic Phenotype.

Myocyte enhancement factor 2C (MEF2C) is a transcription factor studied in the development of skeletal and smooth muscles. Bone resorption studies have exhibited that the reduced expression of MEF2C contributes to osteopetrosis and the dysregulation of pathological bone remodeling. Our current study aims to determine how MEF2C contributes to osteoclast differentiation and to analyze the skeletal phenotype of Mef2c-cKO mice (Cfms-cre; Mef2cfl/fl). qRT-PCR and Western blot demonstrated that Mef2c expression is highest during the early days of osteoclast differentiation. Osteoclast genes, includi

NO-induced vasodilation correlates directly with BP in smooth muscle-Na/Ca exchanger-1-engineered mice: elevated BP does not attenuate endothelial function.

Arterial smooth muscle Na+/Ca2+ exchanger-1 (SM-NCX1) promotes vasoconstriction or vasodilation by mediating, respectively, Ca2+ influx or efflux. In vivo, SM-NCX1 mediates net Ca2+ influx to help maintain myogenic tone (MT) and neuronally activated constriction. SM-NCX1-TG (overexpressing transgenic) mice have increased MT and mean blood pressure (MBP; +13.5 mmHg); SM-NCX1-KO (knockout) mice have reduced MT and MBP (-11.1 mmHg). Endothelium-dependent vasodilation (EDV) is often impaired in hypertension. We tested whether genetically engineered SM-NCX1 expression and consequent BP changes simi

Dietary cholesterol protects against alcohol-induced cerebral artery constriction.

BACKGROUND: Binge drinking represents the major form of excessive alcohol (ethanol [EtOH]) consumption in the United States. Episodic (such as binge) drinking results in blood alcohol levels (BAL) of 18 to 80 mM and leads to alcohol-induced cerebral artery constriction (AICAC). AICAC was shown to arise from EtOH-induced inhibition of large-conductance, calcium/voltage-gated potassium (BK) channels in the vascular smooth muscle. Factors that modulate BK channel-mediated AICAC remain largely unknown. METHODS: Male Sprague Dawley rats were placed on high-cholesterol (2% of cholesterol) diet for

Endothelial tenascin signaling preserves VSMCs contractile phenotype in the human arterial wall in type II diabetes mellitus.

BACKGROUND: Phenotypic switching of vascular smooth muscle cells (VSMCs) is a hallmark of coronary artery disease (CAD). How type 2 diabetes mellitus (T2DM), a major risk factor for cardiovascular complications, reshapes these VSMCs states remains poorly defined. METHODS: Single-cell RNA sequencing (scRNA-seq), spatial transcriptomics, high-dimensional weighted gene coexpression network analysis (hdWGCNA), and cell‒cell interaction analysis were used to profile human arterial tissue from CAD patients with or without T2DM who underwent coronary artery bypass graft surgery. Spot-level cell-type

Incorporation of Smooth Muscle Cells Derived from Human Adipose Stem Cells on Poly(Lactic-co-Glycolic Acid) Scaffold for the Reconstruction of Subtotally Resected Urinary Bladder in Athymic Rats.

BACKGROUND: The urinary tract can be affected by both congenital abnormalities as well as acquired disorders, such as cancer, trauma, infection, inflammation, and iatrogenic injuries, all of which may lead to organ damage requiring eventual reconstruction. As a gold standard, gastrointestinal segment is used for urinary bladder reconstruction. However, one major problem is that while bladder tissue prevents reabsorption of specific solutes, gastrointestinal tissue actually absorbs them. Therefore, tissue engineering approach had been attempted to provide an alternative tissue graft for urinary

Differential distribution and functional impact of BK channel beta1 subunits across mesenteric, coronary, and different cerebral arteries of the rat.

Large conductance, Ca2+i- and voltage-gated K+ (BK) channels regulate myogenic tone and, thus, arterial diameter. In smooth muscle (SM), BK channels include channel-forming α and auxiliary β1 subunits. BK β1 increases the channel's Ca2+ sensitivity, allowing BK channels to negatively feedback on depolarization-induced Ca2+ entry, oppose SM contraction and favor vasodilation. Thus, endothelial-independent vasodilation can be evoked though targeting of SM BK β1 by endogenous ligands, including lithocholate (LCA). Here, we investigated the expression of BK β1 across arteries of the cerebral and p

Mechanical stretch regulates hypertrophic phenotype of the myometrium during pregnancy.

The adaptive growth of the uterus is a critical event that involves changes in cellular phenotypes throughout pregnancy. In early pregnancy, uterine growth is due to hyperplasia of uterine smooth muscle cells (SMCs) within the myometrium; however, the major component of myometrial growth occurs after mid-gestation. This study sought to test the hypothesis that increase in myometrial growth seen during late pregnancy is due to SMC hypertrophy caused by mechanical stretch of uterine tissue by a growing fetus(es) by providing direct measurements of individual SMC size. We employed a stereological

Vascular Sphingosine Kinase 1 Regulates Angiotensin II-Induced Hypertension.

BACKGROUND: S1P (sphingosine-1-phosphate) signaling plays a key role in regulating the cardiovascular system. In experimental essential hypertension (HTN), expression of Sphk1 (sphingosine kinase 1), a key S1P-producing enzyme, is upregulated in the vasculature. Mice with global Sphk1 deletion exhibit alleviated AngII (angiotensin II)-induced HTN. However, cellular mechanisms underlying this protective effect remain unclear due to pleiotropic S1P actions. METHODS: Mice with targeted deletion of Sphk1 in smooth muscle cells (SMCs) or endothelial cells were generated using the Cre-loxP system.

Smooth muscle cells transiently acquire a CD34+ progenitor state upon injury to drive pathological vascular remodeling.

Neointimal hyperplasia, a key pathological feature of many cardiovascular diseases, is driven by vascular smooth muscle cells (SMCs), yet the role of specific SMC subtypes remains unclear. This study identifies a smooth muscle-derived transient progenitor cell (STPC) population, marked by CD34 expression, which emerges after artery injury. These STPCs exhibit high proliferative capacity and generate the majority of neointimal SMCs. Genetic ablation of STPCs significantly reduces neointimal SMC accumulation and attenuates hyperplasia. Mechanistically, SMC-specific knockdown of DCLK1 markedly su

Zinc-binding protein metallothionein 3 protects vascular smooth muscle cells from ferroptosis via blocking lysosomal degradation of GPX4.

Aortic dissection (AD) is a life-threatening vascular emergency characterized by medial degeneration and vascular smooth muscle cell (VSMC) loss. Although disruption of zinc homeostasis has been reported in patients with AD, how zinc ions and their regulatory proteins influence VSMC survival and disease progression remains unknown. In this study, single-cell analyses revealed that ferroptosis and zinc-related pathways were significantly enriched in VSMCs from patients with AD, showing a strong correlation between the two processes, and zinc levels were markedly elevated in dissected aortas. ATOX1-driven ECM degradation and vascular smooth muscle cell apoptosis accelerate aortic dissection progression.

BACKGROUND: The development of aortic dissection (AD) is closely associated with extracellular matrix degradation and the apoptosis of vascular smooth muscle cells (VSMCs). Antioxidant-1 (ATOX1), a copper-binding protein, the precise mechanisms by which it contributes to extracellular matrix (ECM) degradation, VSMC apoptosis, and the onset of AD remain to be further elucidated. METHODS AND RESULTS: Through high-throughput sequencing, we identified a significant increase in the expression of ATOX1 in patients with AD. Further validation using tissue staining, RT-PCR and Western blot revealed t

GSDME-dependent pyroptosis drives abdominal aortic aneurysm via promoting vascular senescence.

Senescence contributes to the pathology of abdominal aortic aneurysm (AAA); however, the regulation of senescence in AAA remains unclear. Here, we sought to determine the role of gasdermin-E (GSDME)-dependent non-canonical pyroptosis in AAA. GSDME-dependent non-canonical pyroptosis is activated in the lesioned vascular walls of mouse models and patients with AAA. GSDME deficiency inhibits vascular senescence and AAA progression. Combined analyses of single-cell RNA sequencing (scRNA-seq), bulk RNA-seq, and multiplex flow cytometry demonstrate that GSDME is essential for the reprogramming of va

Inhibition of long interspersed nuclear element-1 by nucleoside reverse transcriptase inhibitors attenuates vascular calcification.

Vascular calcification (VC) is a critical vascular pathological event, contributing to the rise in both the prevalence and fatality of cardiovascular diseases. However, the lack of effective therapeutic strategies for VC is attributed primarily to the incomplete understanding of its underlying molecular mechanisms. In this study, we discovered that long interspersed nuclear element 1 (LINE1) was significantly upregulated in the calcified arteries of both human individuals and mouse models. Mechanistically, silencing LINE1 expression or inhibiting its activity with adding nucleoside reverse tra

lincRNA-p21 drives apoptosis and calcification of vascular smooth muscle cell via small extracellular vesicles under hyperphosphatemic conditions in chronic kidney disease.

Vascular calcification, a major contributor to cardiovascular morbidity, involves pathological osteogenic transdifferentiation of vascular smooth muscle cell (VSMC) under hyperphosphatemic conditions in chronic kidney disease. This study investigates the role of lincRNA-p21 and small extracellular vesicles in phosphate-induced mouse aortic smooth muscle cell (MASMC) calcification. Exposure to phosphate (2.6 mmol/L) triggered time-dependent calcification, characterized by enhanced calcium deposition, endoplasmic reticulum remodeling, and intracellular calcium accumulation (p < 0.05 vs. 24/48-h

Smooth Muscle LRRC8A Knockout Preserves Vascular Function in Ang II Hypertension.

BACKGROUND: Ang II (angiotensin II) causes hypertension and vascular inflammation both directly and indirectly via cytokines, including TNFα (tumor necrosis factor-α). In vascular smooth muscle cells (VSMCs), Ang II and TNFα activate Nox1 (NADPH oxidase 1) to produce superoxide. TNFα receptors associate with Nox1 and LRRC8A (leucine-rich repeat containing 8A) anion channels to modulate inflammation and contractility in a RhoA-dependent manner. VSMC-specific LRRC8A knockout mesenteric arteries are protected from TNFα-induced injury, and vasodilation is preserved. We hypothesized that LRRC8A kno

Enhanced Yoda1-induced vasoconstriction in pulmonary arterial smooth muscle from monocrotaline-induced pulmonary hypertensive rats.

Mechanical forces such as shear stress, stretch, and pressure play critical roles in regulating vascular function. Mechanosensitive ion channels convert these mechanical stimuli into cytosolic Ca2+ signals that modulate vascular tone. The Piezo family of mechanosensitive channels, comprising Piezo1 and Piezo2, consists of Ca2+-permeable ion channels activated by mechanical stress. Piezo1 is predominantly expressed in non-neuronal cells, whereas Piezo2 is mainly localized to sensory neurons and mechanosensory tissues. The present study investigated the contribution of Piezo1 channels to vasocon
