Genetically engineered mouse models have been generated to recapitulate major signaling pathways deregulated in melanoma. Although these models are invaluable to delineate the relationship between gene mutations and targeted therapeutics, no spontaneously occurring melanomas are available in laboratory mice, which might be used to discover novel disrupted pathways, other than the widely studied MAPK, PI3-AKT and CDK4-INK4A-RB1. Different subpopulations of cells from two different murine melanomas (K-1735 and B16) and a fibrosarcoma (UV-2237) were injected into the internal carotid artery of anesthetized syngeneic mice. Eight cases of choriocarcinoma were induced in WKA female rats by simple fetectomy without viral inoculation or administration of chemical carcinogens. Among them, one line has been successfully subcutaneously transplanted over 10 generations in syngeneic rats and this line was designated as RCHO. The histological appearance of RCHO consisted of trophoblastic giant cells and small basophilic tumor cells, which resembled normal rat placenta. RCHO possessed marked mammotropic and lactogenic activities in female recipients and induced atrophy of the testes in male recipients.

Malignant ileo-caecal immunocytomata, which secrete immunoglobulin, frequently arise spontaneously in LOU/C/Wsl rats. The tumors are confined to the ileo-caecal region and could originate either in the wall of the caecum or in the ileo-caecal lymph nodes. After excision of the ileo-caecal lymph nodes, the incidence of the type of tumor was found to be reduced significantly. The results strongly suggest that the first cells to undergo malignant transformation are located in the ileo-caecal nodes.

Mice heterozygous for a null p53 allele were administered three well-characterized carcinogens to learn more about mechanisms of carcinogenesis and to evaluate the p53-deficient mouse as a tool for identifying potential human carcinogens. Twenty-two of 39 rabbits inoculated with Herpesvirus saimiri developed malignant lymphoma and either died or were killed between 17 and 165 days after inoculation.
