The Cardiac Neural Crest Cells in Heart Development and Congenital Heart Defects.

The neural crest (NC) is a multipotent and temporarily migratory cell population stemming from the dorsal neural tube during vertebrate embryogenesis. Cardiac neural crest cells (NCCs), a specified subpopulation of the NC, are vital for normal cardiovascular development, as they significantly contribute to the pharyngeal arch arteries, the developing cardiac outflow tract (OFT), cardiac valves, and interventricular septum. Various signaling pathways are shown to orchestrate the proper migration, compaction, and differentiation of cardiac NCCs during cardiovascular development. Any loss or dysr

Development and developmental disorders of the enteric nervous system.

The enteric nervous system (ENS) arises from neural crest-derived cells that migrate into and along the gut, leading to the formation of a complex network of neurons and glial cells that regulates motility, secretion and blood flow. This Review summarizes the progress made in the past 5 years in our understanding of ENS development, including the migratory pathways of neural crest-derived cells as they colonize the gut. The importance of interactions between neural crest-derived cells, between signalling pathways and between developmental processes (such as proliferation and migration) in ensu

Transplanted embryonic neurons integrate into adult neocortical circuits.

The ability of the adult mammalian brain to compensate for neuronal loss caused by injury or disease is very limited. Transplantation aims to replace lost neurons, but the extent to which new neurons can integrate into existing circuits is unknown. Here, using chronic in vivo two-photon imaging, we show that embryonic neurons transplanted into the visual cortex of adult mice mature into bona fide pyramidal cells with selective pruning of basal dendrites, achieving adult-like densities of dendritic spines and axonal boutons within 4-8 weeks. Monosynaptic tracing experiments reveal that grafted

Disruption of Nrxn1α within excitatory forebrain circuits drives value-based dysfunction.

Goal-directed behaviors are essential for normal function and significantly impaired in neuropsychiatric disorders. Despite extensive associations between genetic mutations and these disorders, the molecular contributions to goal-directed dysfunction remain unclear. We examined mice with constitutive and brain region-specific mutations in Neurexin1α, a neuropsychiatric disease-associated synaptic molecule, in value-based choice paradigms. We found Neurexin1α knockouts exhibited reduced selection of beneficial outcomes and impaired avoidance of costlier options. Reinforcement modeling suggested

The human dimension: how the prefrontal cortex modulates the subcortical fear response.

Numerous studies suggest that the amygdala is critical for the acquisition and expression of fear. Conditioned fear in animals has been considered a good model for human anxiety disorders, but animal models of anxiety have several limitations. Conditioned fear in animals can be directed to a specific stressor and is easily extinguished. Furthermore, animals do not seem to be able to develop the capacity to worry excessively about the future. While animal models are useful and can demonstrate psychiatric illnesses, they do not completely mimic the complex cognitive processes that occur in anxio

Complementary cortical and thalamic contributions to cell type-specific striatal activity dynamics during movement.

Coordinated motor behavior emerges from information flow across brain regions. How long-range inputs drive cell type-specific activity within motor circuits remains unclear. The dorsolateral striatum (DLS) contains direct- and indirect-pathway medium spiny neurons (dMSNs and iMSNs) with distinct roles in movement control. In mice performing skilled locomotion, we recorded from dMSNs, iMSNs, and their cortical and thalamic inputs identified by monosynaptic rabies tracing. A recurrent neural network (RNN) classifier and clustering analysis revealed functionally heterogeneous subpopulations in ea

A neurophysiological theory for the pain mechanism of tic douloureux.

In attempting to understand the mechanism of pain production in tic douloureux, one must account for the myelination pathology seen in the primary afferent fibers, the cases where trigger is in a different division than the pain, the frequent lack of a fixed neurologic deficit, the effective trigger stimuli corresponding to large caliber axons which would not seem to involve the small axons usually associated with pain production, and similar puzzling features of the disease. We present a theory which satisfactorily predicts, or is consistent with, most known features of tic; it is based upon

Some electrophysiological properties of neurones of rat locus coeruleus.

1. Electrical activity of neurones of the locus coeruleus (LC) was studied in rats anaesthetized with urethane. By stimulating the dorsal pathway (DP) of axons of LC neurones in the mid=brain and observing field responses in the dorsolateral tegmentum of the pons, micro-electrodes were oriented to record unit discharges of LC neurones. They were evoked by DP stimulation mainly during the negative wave of the field response. 2. In the extraceullar records of spike discharges of LC meurones A and B spikes were distinguished. Very often the third component (C spike) was observed to ride on the de

Possible brain regions involved in parturition in mice.

Parturition is a vital physiological process in the reproduction of female mammals, regulated by neurohumoral mechanisms coordinated by the central nervous system. The uterus is essential for this process; however, the neural pathways connecting the brain to the uterus remain poorly understood. In this study, we combined the pseudorabies virus (PRV) tracing tool with c-Fos immunofluorescence staining to identify brain regions that may regulate uterine muscle activity during parturition. We observed that the paraventricular nucleus (PVN), periaqueductal gray (PAG), and locus coeruleus (LC) were…

Substrate pathways demonstrated by transplanted Mauthner axons.

A substrate pathway is a set of aligned guidance cues. (Such cues may be either cells or molecules.) CNS substrate pathways can be demonstrated by transplanting axons to different starting locations. The stereotyped routes of transplanted axons will then demonstrate the locations of effective substrate pathways. To map CNS substrate pathways, Mauthner axons were transplanted to various unnatural locations along the CNS of Xenopus embryos. The routes of 24 experimental Mauthner axons were traced. Twenty-one of these axons grew along parts of a stereotyped route extending in the ventral marginal

Xenopus ADAM 13 is a metalloprotease required for cranial neural crest-cell migration.

BACKGROUND: Cranial neural-crest (CNC) cells originate from the lateral edge of the anterior neuroepithelium and migrate to form parts of the peripheral nervous system, muscles, cartilage, and bones of the face. Neural crest-cell migration involves the loss of adhesion from the surrounding neuroepithelium and a corresponding increase in cell adhesion to the extracellular matrix (ECM) present in migratory pathways. While proteolytic activity is likely to contribute to the regulation of neural crest-cell adhesion and migration, the role of a neural crest-specific protease in these processes has

Advances in ontogeny of the enteric nervous system.

The neurons and glia that comprise the enteric nervous system (ENS), the intrinsic innervation of the gastrointestinal tract, are derived from vagal and sacral regions of the neural crest. In order to form the ENS, neural crest-derived precursors undergo a number of processes including survival, migration and proliferation, prior to differentiation into neuronal subtypes, some of which form functional connections with the gut smooth muscle. Investigation of the developmental processes that underlie ENS formation has progressed dramatically in recent years, in no small part due to the attention

Modulation of spinal circuits following phase-dependent electrical stimulation of afferent pathways.

Objective.Peripheral electrical stimulation (PES) of afferent pathways is a tool commonly used to induce neural adaptations in some neural disorders such as pathological tremor or stroke. However, the neuromodulatory effects of stimulation interventions synchronized with physiological activity (closed-loop strategies) have been scarcely researched in the upper-limb. Here, the short-term spinal effects of a 20-minute stimulation protocol where afferent pathways were stimulated with a closed-loop strategy named selective and adaptive timely stimulation (SATS) were explored in 11 healthy subjects

PACAP/Receptor System in Urinary Bladder Dysfunction and Pelvic Pain Following Urinary Bladder Inflammation or Stress.

Complex organization of CNS and PNS pathways is necessary for the coordinated and reciprocal functions of the urinary bladder, urethra and urethral sphincters. Injury, inflammation, psychogenic stress or diseases that affect these nerve pathways and target organs can produce lower urinary tract (LUT) dysfunction. Numerous neuropeptide/receptor systems are expressed in the neural pathways of the LUT and non-neural components of the LUT (e.g., urothelium) also express peptides. One such neuropeptide receptor system, pituitary adenylate cyclase-activating polypeptide (PACAP; Adcyap1) and its cogn…

Mapping Projectome Heterogeneity of Subiculum Neuron Cell Types.

The subiculum (SUB) is the main output structure of the hippocampus, influencing diverse behaviors through its widespread cortical and subcortical connections. Our previous work creating the mouse Hippocampus Gene Expression Atlas (HGEA) identified four genetically distinct cellular layers across five columnar domains in the SUB, with gene expression boundaries corresponding to distinct connectivity patterns and brain-wide networks involved in spatial navigation, social behavior, and neuroendocrine regulation. Using the Digital Brain Mouse Projectome Atlas (MPA) tool, we conducted 'virtual tra

Neural selection in corticothalamocortical loops: Heterogeneity as a computational resource.

Neuroscience has long relied on (typically binary) categorisations to manage the complexity of brain organisation. Yet growing anatomical and physiological evidence reveals that these dichotomies artificially partition systems that are inherently continuous and heterogeneous. In the thalamus, neurons display mixed afferent-efferent motifs and graded physiological properties that defy rigid categorisation. In the cerebral cortex, laminar somatic labels obscure the contribution of multilayer active dendritic nonlinearities, such as burst spiking, which radically reshapes modes of neural communic

Multi-brain network functional connectivity in major depressive disorder: a fMRI systematic review of mechanisms and clinical translation.

Major Depressive Disorder (MDD) poses significant health risks, yet diagnosis lacks objective biomarkers. This systematic review synthesizes functional Magnetic Resonance Imaging (fMRI) studies (2020-2025, n = 52) on functional connectivity (FC) in MDD. We found robust FC alterations within and between core networks (Default Mode, Salience, Central Executive), linked to rumination, emotion dysregulation, and cognitive deficits. These alterations varied with suicidal ideation, comorbidities, childhood trauma, and biological sex. Treatments (antidepressants, rTMS, ECT) demonstrated distinct norm
