Ubiquitin-Specific Proteases in Breast Cancer: Unraveling Oncogenic Roles and Therapeutic Opportunities.

The ubiquitin-proteasome system (UPS) is a crucial determinant of protein stability and activity in various aspects of physiological function and disease development. The well-characterized class of regulatory enzymes within the UPS is deubiquitinating enzymes (DUBs), and their effects, particularly those of the ubiquitin-specific proteases (USPs), oppose ubiquitination. All DUB activities can be more or less disrupted in various cancers, including breast cancer, and DUB alterations contribute substantially to tumor initiation, progression, and many forms of therapeutic resistance. In this rev

Endogenous inhibitors of PP2A activate oncogenic and DNA damage response kinases in glioblastoma.

Glioblastomas (GBMs) exhibit constitutive activation of oncogenic kinase signaling pathways, contributing to tumor aggressiveness and resistance to therapy. Kinase inhibitors have demonstrated limited efficacy against GBMs, primarily due to the tumors' ability to adapt to diverse stimuli and effectively rewiring critical downstream signaling networks. This remarkable adaptability underscores the pressing need for novel therapeutic strategies that sustain the inhibition of oncogenic kinase signaling in GBM. This study sought to elucidate the mechanisms by which GBMs maintain the constitutive ac

Molecular Mechanisms of KIT Receptor Dimerization and Oncogenic Activation Revealed by Multiscale Simulations.

The receptor tyrosine kinase KIT is essential for cell proliferation and differentiation, and its aberrant activation contributes to cancers such as acute myeloid leukemia and gastrointestinal stromal tumors. KIT activation normally requires stem cell factor (SCF)-induced dimerization, whereas oncogenic mutations can drive ligand-independent signaling. Here, we employ multiscale molecular dynamics simulations based on the Martini 3 coarse-grained (CG) force field to investigate the dimerization mechanisms of full-length KIT in both wild-type (WT) and oncogenic mutant forms within a realistic m

Human papillomavirus 42: from a low-risk genotype to a potentially oncogenic virus.

Human papillomavirus 42 (HPV42), initially classified as a low-risk genotype isolated from benign vulvar papillomas, has long been associated with benign lesions such as anogenital warts. However, recent studies have uncovered its unexpected oncogenic potential, challenging its traditional classification. HPV42 has been identified as a causative driver of digital papillary adenocarcinoma, a rare and aggressive cutaneous tumour, and is also linked to high-grade squamous intraepithelial lesions in the cervix and anus, as well as frequent infections in the oropharyngeal region. The molecular hall

Oncogenic EGFR Signaling as a Central Regulator of Chemoresistance in Ovarian Cancer: A Mechanistic Review.

Ovarian cancer (OVC) is a leading cause of gynecological cancer mortality due to late-stage diagnosis and chemoresistance. Among the multiple molecular mediators, oncogenic epidermal growth factor receptor (EGFR) signaling has emerged as a key regulator of tumor progression and drug resistance, ultimately governing cancer survival. Therefore, this review focused on the molecular mechanisms of aberrant EGFR signaling to promote chemoresistance in ovarian cancer through multiple interlinking pathways, including the phosphoinositide 3-kinase (PI3K)/protein kinase B (AKT)/mammalian target of the r

Unraveling the pro-oncogenic role of GLIPR2: Its molecular basis in glioma malignancy.

Adult diffuse gliomas remain challenging to treat because of their infiltrative growth and resistance to multimodal therapy. Although GLIPR2 has been implicated in autophagy regulation and tumor-related processes, its role in glioma remains unclear. In this study, transcriptomic and clinical data from TCGA, CGGA, and GTEx were integrated with western blotting and immunohistochemical validation in human glioma specimens. Prognostic significance was evaluated using Kaplan-Meier and ROC analyses. Functional effects of GLIPR2 knockdown were examined in U138 and U251 glioma cells through assays of

Post-translational modifications in prostate cancer: coordinated oncogenic networks and clinical implications.

Prostate cancer (PCa) is one of the most prevalent malignancies affecting male health globally, ranking as the fifth leading cause of cancer-related mortality in men. Post-translational modifications (PTMs), encompassing a diverse array of biochemical alterations such as methylation, acetylation, phosphorylation, ubiquitination, SUMOylation, glycosylation, and lactylation, constitute a critical category of epigenetic regulatory mechanisms that modulate numerous cellular processes in both physiological and pathological contexts. Despite their fundamental importance, the precise functional impli

Ontogeny Dictates Oncogenic Potential, Lineage Hierarchy, and Therapy Response in Pediatric Leukemia.

Accumulating evidence links pediatric cancers to prenatal transformation events, yet the influence of the developmental stage on oncogenesis remains elusive. We investigated how hematopoietic stem cell developmental stages affect leukemic transformation, disease progression, and therapy response using a novel, humanized model of NUP98∷NSD1-driven pediatric acute myeloid leukemia, that is particularly aggressive with WT1 co-mutations. Fetal-derived hematopoietic stem cells readily transform into leukemia, and WT1 mutations further enhance stemness and alter lineage hierarchy. In contrast, stem …

Aurora Kinase A: The Prominent Oncogenic Link in Helicobacter pylori-Driven Gastric Carcinogenesis.

Chronic Helicobacter pylori (H. pylori) infection leads to gastric carcinoma (GC), while aurora kinase A (AURKA) is known to be upregulated in several cancers. However, the direct association between AURKA and H. pylori remains largely unexplored. The significance of AURKA in H. pylori infection was investigated using an RNAi-mediated silencing method. The expression of downstream signaling genes and apoptotic markers was analyzed through qRT-PCR and western blot. Cancerous properties were evaluated through scratch wound assay, cell counting through trypan blue, and genomic instability assay.

C-myc Oncogene Numerical Imbalances Analysis in Laryngeal Squamous Cell Carcinoma.

BACKGROUND/AIM: The over-activation of oncogenes is a critical genetic event in the development and progression of solid malignancies. Gene amplification and specific mutations represent the prominent mechanisms that convert proto-oncogenes into their active, oncogenic forms. C-myc oncogene (gene locus: 8q24.21) regulates crucial cell and tissue functions, whereas its deregulation is implicated in carcinogenesis. Our research aimed to investigate the impact C-myc numerical imbalances on a series of laryngeal squamous cell carcinomas (LSCCs) characterized by different clinic-pathological featur…

The Stmn1-lineage contributes to acinar regeneration but not to neoplasia upon oncogenic Kras expression.

BACKGROUND & AIMS: The exocrine pancreas has a limited regenerative capacity, but to what extent all acinar cells are involved in this process is unclear. Nevertheless, the heterogenous nature of acinar cells suggests that cells exhibiting higher plasticity might play a more prominent role in acinar regeneration. In that regard, Stmn1 -expressing acinar cells have been identified as potential facultative progenitor-like cells in the adult pancreas. Here, we studied Stmn1-progeny under physiological conditions, during regeneration, and in the context of Kras G12D expression. METHODS: We follow

CHD8 interacts with BCL11A to induce oncogenic transcription in triple negative breast cancer.

The identification of tumour-specific protein-protein interactions remains a challenge for the development of targeted cancer therapies. In this study we describe our approach for the identification of triple negative breast cancer (TNBC)-specific protein-protein interactions focusing on the oncogene BCL11A. We used a proteomic approach to identify the BCL11A protein networks in TNBC and compared it to its network in B-cells, a cell type in which BCL11A plays crucial roles. This approach identified the chromatin remodeller CHD8 as a TNBC-specific interaction partner of BCL11A. We show that CHD

Denitrogenative Alkylation of K-Ras(G12D) Inhibits Oncogenic Signaling in Cancer Cells.

Pancreatic ductal adenocarcinoma (PDAC) is the most lethal common cancer. More than 90% of PDAC tumors are caused by KRAS mutations, with the majority expressing the K-Ras(G12D) oncoprotein. Despite extensive drug discovery efforts across academia and industry, there are no approved drugs directly targeting K-Ras(G12D) in a mutant-selective manner. We report a series of α-diazoacetamide compounds that form covalent bonds via denitrogenative alkylation of acquired aspartic acid at the mutation site. The lead molecule allosterically inhibits the mitogen-activated protein kinase pathway downstrea

Resolution of oncogene-induced senescence markers in HPV-infected cervical cancer tissue.

BACKGROUND: Oncogene-Induced Senescence (OIS) is a form of senescence that occurs as a consequence of oncogenic overstimulation and possibly infection by oncogenic viruses. Whether senescence plays a role in the pathogenesis of cervical cancer (CC) is not well understood. Moreover, whether cervical epithelial cells that are part of the premalignant cervical intraepithelial neoplasia (CIN), exhibit markers of OIS in Human Papillomavirus (HPV)-infected tissue, has not been investigated. METHODS: We utilized a set of patient-derived premalignant and malignant tissue samples to investigate the pr
