Androgens have antiresorptive effects on trabecular disuse osteopenia independent from muscle atrophy.

Aging hypogonadal men are at increased risk of osteoporosis and sarcopenia. Testosterone is a potentially appealing strategy to prevent simu … stored it more efficiently to pre-treatment levels. Conditional AR deletion in late osteoblasts and osteocytes or in the satellite cell lineage increased age-related trabecular bone loss in both hindlimbs without influencing the effect of BTx on trabecular osteopenia. We conclude that androgens have antiresorptive effects on trabecular disuse osteopenia whic …

Rat Osteosarcoma Cells as a Therapeutic Target Model for Osteoregeneration via Sclerostin Knockdown.

There are various conceptually different strategies to improve bone regeneration and to treat osteoporosis, each with distinct inherent adva … the SOST gene, a key regulator of bone formation and remodeling. Sclerostin is expressed by mature osteocytes but also by late osteogenically differentiated cells. Thus, it is difficult and requires long-term cultures to investigate the effects of SOST silencing on the expression of osteogenic markers using primary cells. We, therefore, selected a rat osteo …

In vivo incidence of apoptosis evaluated with the TdT FragEL DNA fragmentation detection kit in cartilage and bone cells of the rat tibia.

Previous studies have shown the occurrence of cell death by apoptosis in cartilage and bone cells, and have suggested a functional relations … counted along the chondro-osseous junction. There were only a few apoptotic osteoblastic cells and osteocytes (0.22 +/- 0.22% and 0.15 +/- 0.16% of total osteoblasts and osteocytes respectively). Histological and electron microprobe studies of mineralisation in aluminium-related osteomalacia.

AIMS: To determine a possible mechanism to explain the presence of aluminium lines within fully calcified bone in aluminium-related osteomal … d and consisted of small focal deposits (less than 40 microns diameter), often (52%) around osteoid osteocytes (probably an underestimate of the association), and larger areas that extended to the aluminium lines at the underlying mineralisation front. Small and large mineralisation nuclei were seen ultrastructurally in the patchy calcification. Quantitative …

New pathogenic insights inform therapeutic target development for renal osteodystrophy.

In an ancillary analysis of cross-sectional observational studies of bone health in end-stage kidney disease (ESKD), Evenepoel et al. reported that subjects with autosomal-dominant polycystic kidney disease (ADPKD) had a unique phenotype in their renal osteodystrophy. ADPKD caused resistance to parathyroid hormone (PTH) producing lower turnover states and preservation of cortical bone mineral density. PTH resistance was probably produced by increased osteocyte sclerostin levels, which is regulated by mechanical loading sensed through primary cilia sensory function affected by mutation in PKD1

Epidemiology, Cause, and Anatomy of Osteonecrosis of the Foot and Ankle.

Osteonecrosis arises throughout the foot and ankle in various forms and due to numerous causes, with a thousand US cases per year estimated for the ankle alone. Although research continues to elucidate specific mechanisms at work, the pathophysiology remains poorly understood. Nevertheless, the various osteonecrosis pathways converge on osteocyte death, and bony lesions follow a pattern of progression. Understanding the specific anatomy and biomechanics associated with common forms of foot and ankle osteonecrosis should help guide diagnosis and interventions, particularly at earlier stages of

Spinal cord injury-induced osteoporosis: pathogenesis and emerging therapies.

Spinal cord injury causes rapid, severe osteoporosis with increased fracture risk. Mechanical unloading after paralysis results in increased osteocyte expression of sclerostin, suppressed bone formation, and indirect stimulation of bone resorption. At this time, there are no clinical guidelines to prevent bone loss after SCI, and fractures are common. More research is required to define the pathophysiology and epidemiology of SCI-induced osteoporosis. This review summarizes emerging therapeutics including anti-sclerostin antibodies, mechanical loading of the lower extremity with electrical sti

[Bone tissue in osteogenesis imperfecta: light and electron microscopic studies].

Osteogenesis imperfecta was studied by light and electron microscopic techniques in 36 cases, of them there were 10 postmortem and 26 bone biopsies. The persons' age was from 0 to 38 years. There were 25 males and 11 females. The microscopic studies indicated a decrease in the basic substance and an increase in the number of osteocytes. The electron microscopic studies revealed a reduction in the granular endoplasmic reticulum, as well as matrix swelling in the mitochondria, their crista degeneration, and the presence of mitochondrial hydroxyappatite; non-uniform mineralization of collagenous …

The cellular basis for extremity bone loss in leprosy.

Osteoclasts and osteolytic osteocytes have been observed in the majority of 60 samples of bone taken from five patients with lepromatous or tuberculoid leprosy. These results are interpreted to mean that bone loss in patients with leprosy is an acceleration of a normal cellular process and not the result of avascular necrosis. The acceleration of bone resorption could be due to local release of products from M. leprae or host cells, a hypothesis testable by organ culture methods. The presence of lymphocytes and mononuclear cells in bone samples in this and previous studies is discussed with re…

Mechanical loading under estrogen deficiency enhances mineralization and osteoblast maturation in an advanced 3D vascularized bone organoid driven by hypertrophy and apoptosis.

Bone is a highly mechanosensitive tissue that responds to mechanical cues and adapts to maintain a homeostatic balance between bone formation and resorption, thereby preserving structural integrity and strength. Estrogen is the key modulator of skeletal homeostasis that regulates the mechanosensitivity and survival of osteocytes, promotes osteoblast activity and reduces osteoclastogenesis. The decline in circulating estrogen after menopause leads to increased bone resorption, widely associated with postmenopausal osteoporosis, but is also associated with vascular dysregulation, mineral heterog…

Evaluating the antimicrobial and anti-inflammatory mechanisms of dexmedetomidine in managing bone infection: a laboratory perspective.

BACKGROUND: Osteomyelitis involves bone destruction, impaired bone formation, and systemic inflammation. Dexmedetomidine (DXMT) possesses antioxidant, anti-inflammatory, and anti-apoptotic properties alongside sedative and analgesic effects. This study evaluates DXMT's effects on markers of infection and bone healing using osteocyte-like cells infected by Staphylococcus aureus (S. aureus). METHODS: Human osteosarcoma-derived SAOS-2 cells were differentiated to an osteocyte-like phenotype over 28 days using potassium dihydrogen phosphate. Differentiation was verified via qPCR for osteogenic ma

Loss of the Ubiquitin-Associated Domain of sqstm1/p62 in Zebrafish Causes a Phenotype Resembling Paget's Disease of Bone.

The ubiquitin-binding protein p62, encoded by Sequestosome 1 (SQSTM1), is an essential molecular adaptor for selective autophagy. Heterozygous mutations deleting or disrupting the ubiquitin-associated (UBA) domain of p62 have been reported as the major genetic cause for Paget's disease of bone (PDB), the second most common skeletal disease, characterized by hyperactive osteoclasts and focal increases of bone turnover. In this study, we aimed to determine the impact of a similar sqstm1/p62 mutation on the skeleton of zebrafish. We successfully established a sqstm1tmΔUBA zebrafish line with prem

HERC1 deficiency causes osteopenia through transcriptional program dysregulation during bone remodeling.

Bone remodeling is a continuous process between bone-forming osteoblasts and bone-resorbing osteoclasts, with any imbalance resulting in met … ith the changes associated with reductions in testosterone and dihydrotestosterone levels. Finally, osteocytes isolated from knockout mice showed a higher expression of osteocytic genes and an increase in the Rankl/Opg ratio, indicating a relevant cell-autonomous role of HERC1 when regulating the transcriptional program of bone formation. Overall, these find …

Micropetrosis in hemodialysis patients.

Micropetrosis develops as a result of stagnation of calcium, phosphorus and bone fluid, which appears as highly mineralized bone area in the osteocytic perilacunar/canalicular system regardless of bone turnover of the patients. And microcracks are predisposed to increase in these areas, which leads to increased bone fragility. However, micropetrosis of hemodialysis (HD) patients has not been discussed at all. Micropetrosis area per bone area (Mp.Ar/B·Ar) and osteocyte number per micropetrosis area (Ot.N/Mp.Ar) were measured in nine HD patients with renal hyperparathyroidism (Group I), twelve p

CKD-induced wingless/integration1 inhibitors and phosphorus cause the CKD-mineral and bone disorder.

In chronic kidney disease, vascular calcification, renal osteodystrophy, and phosphate contribute substantially to cardiovascular risk and are components of CKD-mineral and bone disorder (CKD-MBD). The cause of this syndrome is unknown. Additionally, no therapy addresses cardiovascular risk in CKD. In its inception, CKD-MBD is characterized by osteodystrophy, vascular calcification, and stimulation of osteocyte secretion. We tested the hypothesis that increased production of circulating factors by diseased kidneys causes the CKD-MBD in diabetic mice subjected to renal injury to induce stage 2
