Reframing cancer therapy through tumor plasticity.

A major barrier to cancer therapy is the emergence of drug-tolerant states that allow tumor cells to bypass the targeted pathway. These refractory states arise through reversible cellular plasticity, a process that also occurs during development and regeneration, giving rise to new lineages or phenotypes and restoring drug sensitivity upon treatment withdrawal. Over time, genetic mutations may accumulate, leading to permanent resistance. This paradigm expands the focus from targeting genetic mechanisms alone to also exploiting nongenetic vulnerabilities, driving clinical strategies that preven

Financial Toxicity in Plastic Surgery: A Scoping Review.

INTRODUCTION: Financial toxicity (FT) describes the physical, psychosocial, and financial burden of health care-related costs on patients' well-being. Plastic surgery patients may be susceptible to FT due to high procedure and adjunct therapy costs. Despite these risks, FT resulting from plastic and reconstructive surgery remains unclear. This scoping review assessed the prevalence and impact of FT on plastic surgery patients. METHODS: MEDLINE, Embase, Web of Science Core Collection, Scopus, and the Cumulative Index to Nursing and Allied Health Literature were searched from inception through

Artificial Photoreforming Technology for Plastic Waste Valorization.

The escalating depletion of fossil fuels, rising greenhouse gas emissions, and pervasive plastic pollution pose unprecedented threats to the environment and human well-being, underscoring an urgent need for sustainable and clean resource recycling technologies. Solar-driven photoreforming of plastic waste simultaneously generates value-added chemicals, fuels, and carbon materials. Conceptually, this process resembles an artificial photosynthesis system, wherein solar energy serves as the driving force, photocatalysts function as active media, and waste plastics act as the feedstocks. However, …

Neurotransmitter-activated GPCR signaling in myelin plasticity.

Myelination is increasingly recognized as a dynamic and adaptive process regulated by oligodendrocytes throughout life. Beyond providing electrical insulation, myelin supports axonal metabolism and may serve as an energy reservoir under metabolic stress, highlighting the importance of physiological myelin turnover. Dysregulation of myelin dynamics contributes to a wide spectrum of neurological disorders, including demyelinating, neurodegenerative, and neuropsychiatric diseases. Growing evidence indicates that neurotransmitter signaling through G protein-coupled receptors (GPCRs) expressed by o

Metabolic plasticity in the pancreatic cancer metastatic cascade.

Most deaths from pancreatic ductal adenocarcinoma (PDAC) result from metastatic organ failure rather than primary tumor burden. Across the metastatic cascade, tumor cells encounter hypoxia, nutrient limitation, oxidative stress, immune pressure, and changing stromal conditions, and only those capable of dynamic metabolic adaptation successfully disseminate and colonize distant sites. In this review, we summarize how metabolic plasticity shapes PDAC dissemination, organ-specific colonization, and metastatic outgrowth. We further discuss how this framework may shift therapeutic strategy from bro

The Critical Period Microbiota Shape Brain Plasticity.

The gut microbiota is increasingly recognized as a regulator of brain function, yet its role in experience-dependent plasticity during postnatal development remains largely unknown. Here, we show that disrupting the gut microbiota with antibiotics during critical periods of visual cortex development impairs ocular dominance plasticity (ODP) in juvenile mice. Antibiotic treatment induces marked changes in microbial community composition and is accompanied by extensive transcriptional remodeling of the visual cortex, including pathways involved in extracellular matrix organization, blood-brain b

State of the Art of Artificial Intelligence in Plastic Surgery.

Introduction: Artificial intelligence (AI) raises a fundamental question for modern plastic surgery: will it enable personalized, patient-centered, efficient care, or lead to a dystopian, automated care that diminishes human autonomy, which is dominated by generic "AI slop" that eventually deteriorates the doctor-patient relationship? Methods: This study examines current applications of large language models, generative AI, and neural networks in plastic surgery, assessing AI's roles in surgical planning, automation of repetitive tasks, outcome evaluation, education, and patient communication,

Synaptic Plasticity-Intrinsic Excitability and Antidepressant Discovery.

Major depressive disorder remains a leading cause of disability, and decades of monoamine-centered pharmacology have yielded delayed and often incomplete relief. Rapid-acting antidepressants reshaped the field by linking swift symptom improvement to glutamatergic plasticity, yet durable benefit depends on how newly reconfigured circuits are stabilized and tuned. This review synthesizes evidence that antidepressant efficacy arises from the coordinated engagement of synaptic plasticity, spanning induction and consolidation, and intrinsic excitability, which provides gain control, and proposes an

Developmentally constrained plasticity as a therapeutic vulnerability in cancer.

Cancer cells can exploit developmental lineage programs to generate phenotypic heterogeneity under therapeutic pressure. Although a subset of resistant tumors preserves its founding lineage, receptor, or oncogenic dependency, accumulating evidence shows that others enter reversible persister states or stabilize alternative lineage programs. Here we frame malignant plasticity as developmentally constrained, not limitless: cell of origin, lineage history, injury memory, genetic gates, chromatin state, transcription-factor circuits, and tumor-ecosystem feedback help delimit and probabilistically

Adapting to scarcity: Plasticity in rural healthcare practice.

Rural healthcare practitioners face persistent challenges such as limited staffing, geographic isolation, and broad scopes of responsibility. These conditions require constant adaptation to ensure care delivery. This study introduces plasticity as a conceptual lens for understanding how rural healthcare teams collectively navigate scarcity through flexible role and task shifting. Drawing on constructivist grounded theory, we conducted 19 interviews with nurses and physicians in emergency departments in two rural communities in Ontario, Canada, to explore how plasticity functions in rural healt

Optimizing phage elution for biopanning of plastic materials.

Biopanning of phage-displayed peptides is a useful method for peptide probe discovery and has been applied to multiple plastic targets. The most widely used elution method on plastic targets, acidic glycine buffer, is well-established as a general binding disrupter for protein targets, but a comparative study of elution buffers for plastic targets has never been published. Here we demonstrate how elution from plastics with mild surfactant C12E8 or ethanol increases phage yield up to 1000-fold compared to the commonly used acidic elution while also yielding more diverse phage populations. This …

Plastic Surgery Involvement Reduces Cranioplasty Complications.

BACKGROUND: Cranioplasty is commonly performed after decompressive craniectomy to provide structural protection of the cerebrum and restore cosmesis. However, procedural complications are not insignificant and can result in prolonged recovery. Although cranioplasties may be performed solely by neurosurgeons, plastic and reconstructive surgeons (PRS) are occasionally consulted for skull reconstruction and wound closure. This study analyzes the impact of PRS involvement in cranioplasty outcomes, including primary and secondary cranioplasty cases. METHODS: A retrospective chart review was conduc

MYRF restrains mesothelial plasticity to protect the lung.

In this issue of Developmental Cell, Luna et al.1 identify myelin regulatory factor (MYRF) as a critical determinant of lung mesothelial specification and quiescence. Their study demonstrates that MYRF loss leads to congenital diaphragmatic hernia or a pleuroparenchymal fibroelastosis-like phenotype, revealing the mesothelium as an actively restrained progenitor population.

Non-Synaptic Mechanism of Ocular Dominance Plasticity.

Classic experiments showing that monocular visual disruption alters synaptic connections to binocular neurons established the fundamental concept of synaptic plasticity. Synaptic inputs that are activated coincidently with postsynaptic action potential firing are strengthened, and inputs from cells firing before or after the postsynaptic action potential are weakened. An implicit assumption, however, is that the speed of impulse transmission is not altered by visual deprivation. If so, spike time arrival at binocular neurons would be affected, thereby inducing synaptic plasticity. This possibi

Critical period plasticity enables credit assignment.

Synaptic plasticity is often guided by instructive inputs to neural circuits, but learning only succeeds when these instructions reach neurons that mediate relevant outputs. This creates the credit assignment problem: how does a neuron receive instructions suited to its own behavioral function? Here we report a developmental mechanism that enables credit assignment by using instructive inputs to organize the downstream architecture through which learning is expressed. In the olivocerebellar learning system, the inferior olive provides instructive inputs that guide plasticity within the cerebel

Cellular Plasticity in Malignant Transformation: Mesothelial Cells.

BACKGROUND: Mesothelial proliferations range from reactive lesions to benign and malignant tumors, bearing witness to the plasticity of these cells. Their diagnosis often requires combination of morphological assessment, immunohistochemistry, and molecular testing. SUMMARY: Considerable progress has been made in recent years in all the above aspects. The separation of reactive from neoplastic mesothelial proliferations has become more robust. Mesothelioma in situ is now recognized as precursor of invasive mesothelioma and better tools exist for its differentiation from benign mimics. Differen

Transcriptomic Plasticity Is a Hallmark of Metastatic Pancreatic Cancer.

UNLABELLED: Metastasis is the leading cause of cancer deaths. To develop strategies for intercepting metastatic progression, a better understanding of how tumor cells adapt to vastly different organ contexts is needed. To investigate this question, a single-cell transcriptomic atlas of primary tumors and diverse metastatic samples (liver, omentum, peritoneum, stomach wall, lymph node, and diaphragm) from a patient with pancreatic ductal adenocarcinoma who underwent rapid autopsy was generated. Using unsupervised archetype analysis, both shared and site-specific gene programs were identified, i

Metabolic regulation of synaptic plasticity in anorexia nervosa.

Anorexia nervosa (AN) is increasingly understood as a metabo-psychiatric disorder in which metabolic biology and neural circuit function are intrinsically intertwined. Genetic studies reveal that AN is associated with heritable metabolic traits suggesting that metabolic vulnerability contributes to the disorder. The metabolic profile of AN further shapes brain responses; endocrine signals such as insulin, leptin, ghrelin, and adiponectin elicit atypical neural responses in circuits regulating appetite, reward, interoception, and cognitive control. This altered signaling is accompanied by circu
