Evidence for a pathway independent from 2'-deoxyguanosine and reversible by IL-2 by which purine nucleoside phosphorylase inhibitors block T-cell proliferation.

Patients with homozygous deficiency of purine nucleoside phosphorylase (PNP) present with a T-cell selective immune deficiency. To elucidate the potential use of PNP inhibitors in the therapy of cutaneous T-cell lymphomas (CTCLs) the authors studied the effects of CI-1000 (formerly PD141955-2) and CI-972 on a T-cell line MyLa established from a patient with mycosis fungoides. Both PNP inhibitors had significant, dose-dependent, inhibitory effects on the proliferation of the T-cell line. CI-1000 (ED50: 3.7 microM) was approximately six-fold more potent in blocking 3H-thymidine uptake than CI-97

Proteomic profiling of valproic acid-treated zebrafish embryos highlights dysregulation in energy and purine metabolism, and microtubule dynamics: Implications for autism spectrum disorder.

Valproate (VPA), a widely used anticonvulsant, is also employed to establish experimental autism spectrum disorder (ASD) models. This study aims to elucidate mechanisms underlying VPA's effects in ASD by analyzing proteomic profiles and oxidant-antioxidant dynamics in zebrafish embryos, uncovering the cellular pathways driving these changes. Zebrafish embryos were exposed to two concentrations of VPA (10 μM and 25 μM) for 72 h post-fertilization (hpf), and LC-MS/MS analyses were performed. Differentially expressed proteins (DEPs) were subjected to bioinformatic analysis to identify associated

Neurokinin 1 receptor inhibition alleviated mitochondrial dysfunction via restoring purine nucleotide cycle disorder driven by substance P in acute pancreatitis.

Acute pancreatitis (AP) is a life-threatening gastrointestinal disorder for which no effective pharmacological treatments are currently available. One of the pharmacological targets that merits further research is the neurokinin 1 receptor (NK1R), which is found on pancreatic acinar cells and responds to the neuropeptide substance P (SP) that participates in AP. Although a few studies have stated the involvement of SP/NK1R in neurogenic inflammation in AP development, the regulatory mechanism remains unclear. In this study, we found that following activation of NK1R by SP, β-arrestin1, a scaff

Insulin-activated β-cell-liver axis protects against acetaminophen-induced acute liver injury by suppressing oxidative stress and purine catabolism.

β-Cell-hepatocyte communication is vital for the pathogenesis of liver-related diseases, however, its role in acetaminophen (APAP)-induced acute liver injury (ALI) remains poorly understood. This study aimed to investigate the effect of insulin-mediated β-cell-liver crosstalk on APAP-induced ALI and its underlying mechanisms. Streptozotocin-induced β-cell destruction or postprandial insulin stimulation was used to modulate serum insulin levels in mice. As a result, insulin deficiency exacerbated APAP-induced liver injury and inflammation, whereas elevated insulin levels during the postprandial

Multifunctional cellulose-rich bagasse magnetic biochar capable of efficient elimination and separation of purine compounds in beer: DFT calculation and practical system applications.

Purine compounds in beer significantly increase the risk of hyperuricemia and gout, creating a strong demand for low-purine and healthy beer. A multifunctional cellulose-rich bagasse (cellulose content: 40 %-50 %) magnetic porous carbon (MABB) was constructed by employing alkali-modified bagasse and intercalating with Fe3+ to effectively remove three purine nucleosides (NUCs) in beer, namely, guanosine (GUA), adenosine (ADE), and inosine (INO). Characterization results showed that alkalization increased the specific surface area of MABB and iron oxide acted as an active site to facilitate NUC

Effects of corn grain processing and phosphorus content in calf starters on intake, growth performance, nutrient digestibility, blood metabolites, and urinary purine derivatives.

Corn grain with a high phosphorus (P) content (mainly in the form of phytate-P) may need to be processed to improve the digestibility of nutrients for young calves. Processing corn grains can improve the accessibility of phytate-P to the rumen enzymes and increase the bioavailability of P, which benefits the growth and development of calves. The objective of this study was to investigate the effects of feeding starter diets with steam-flaked corn (SFC) compared with ground corn (GC) with 2 P contents of 0.4% and 0.7% DM basis on intake, growth performance, nutrient digestibility, blood metabol

Pharmacophore modeling, molecular docking and molecular dynamics studies on natural products database to discover novel skeleton as non-purine xanthine oxidase inhibitors.

Gout is a common inflammatory arthritis caused by the deposition of urate crystals within joints. It is increasingly in prevalence during the past few decades as shown by the epidemiological survey results. Xanthine oxidase (XO) is a key enzyme to transfer hypoxanthine and xanthine to uric acid, whose overproduction leads to gout. Therefore, inhibiting the activity of xanthine oxidase is an important way to reduce the production of urate. In the study, in order to identify the potential natural products targeting XO, pharmacophore modeling was employed to filter databases. Here, two methods, p

Plasma First Resuscitation Reduces Lactate Acidosis, Enhances Redox Homeostasis, Amino Acid and Purine Catabolism in a Rat Model of Profound Hemorrhagic Shock.

The use of aggressive crystalloid resuscitation to treat hypoxemia, hypovolemia, and nutrient deprivation promoted by massive blood loss may lead to the development of the blood vicious cycle of acidosis, hypothermia, and coagulopathy and, utterly, death. Metabolic acidosis is one of the many metabolic derangements triggered by severe trauma/hemorrhagic shock, also including enhanced proteolysis, lipid mobilization, as well as traumatic diabetes. Appreciation of the metabolic benefit of plasma first resuscitation is an important concept. Plasma resuscitation has been shown to correct hyperfibr

Human polymerase alpha inhibitors for skin tumors. Part 2. Modeling, synthesis and influence on normal and transformed keratinocytes of new thymidine and purine derivatives.

Recently, the three-dimensional structure of the active site of human DNA polymerase alpha (pol alpha) was proposed based on the application of molecular modeling methods and molecular dynamic simulations. The modeled structure of the enzyme was used for docking selective inhibitors (nucleotide analogs and the non-nucleoside inhibitor aphidicolin) in its active site in order to design new drugs for actinic keratosis and squamous cell carcinoma (SCC). The resulting complexes explained the geometrical and physicochemical interactions of the inhibitors with the amino acid residues involved in bin

New bioinformatics approach to analyze gene expressions and signaling pathways reveals unique purine gene dysregulation profiles that distinguish between CD and UC.

BACKGROUND: Expression of purine genes is modulated by inflammation or experimental colitis and altered expression leads to disrupted gut function. We studied purine gene dysregulation profiles in inflammatory bowel disease (IBD) and determined whether they can distinguish between Crohn's disease (CD) and ulcerative colitis (UC) using Pathway Analysis and a new Comparative Analysis of Gene Expression and Selection (CAGES) method. METHODS: Raw datasets for 22 purine genes and 36 probe-sets from National Center for Biotechnology Information (NCBI) GEO (Gene Expression Omnibus) (http://www.ncbi.

Adenosine and purine nucleosides protect rat primary astrocytes from peroxynitrite-potentiated, glucose deprivation-induced death: preservation of intracellular ATP level.

Previously we have reported that immunostimulated astrocytes became highly vulnerable to glucose deprivation. In the present study we examined the effect of various kinds of nucleosides on the augmented death of glucose-deprived immunostimulated astrocytes. Preincubation with interferon-gamma (100 U/ml) and lipopolysaccharide (1 microg/ml) for 48 h and continuous exposure to glucose deprivation (4 h) significantly induced the lactate dehydrogenase (LDH) release, as a marker of cell injury or death, from astrocytes. The glucose deprivation-induced augmented cell death in immunostimulated astroc

Purine nucleotides induce regulated secretion of von Willebrand factor: involvement of cytosolic Ca2+ and cyclic adenosine monophosphate-dependent signaling in endothelial exocytosis.

von Willebrand factor (vWF) is stored and released from endothelial secretory granules called Weibel-Palade (WP) bodies. Acute release can be induced by thrombin, histamine, and other mediators of thrombosis or inflammation. Their effect is thought to be mediated by an increase in intracellular free calcium ([Ca2+]i). Purine nucleotides such as adenosine triphosphate (ATP) and adenosine diphosphate (ADP) are released from platelet dense granules and from ischemic tissues and are important regulators of platelet function and vascular tone. In the present study, we investigated whether they coul

DFT and TD-DFT insights into the conformational, electronic, and spectral properties of purine-based glycol nucleic acid (GNA) monophosphates and their Zn(II) coordination effects.

The structurally simpler Glycol nucleic acids (GNAs) have been known to be chemically robust alternatives to canonical nucleic acids, but their conformational and electronic properties remain underexplored. In this work, density functional theory (DFT), time-dependent DFT (TD-DFT), and molecular docking were applied to purine-based GNAs and their Zn(II) complexes. Conformational analysis reveals alternative low-energy conformers with opposite nucleobase orientation relative to their crystal geometries. The Zn(II) coordination leads to reduction of orbital energy gap and increases dipole moment

Synthesis and biological evaluation of 6,9-disubstituted purine analogues inducing autophagy-mediated apoptosis and potentiating carboplatin response in high-grade serous ovarian cancer.

High-grade serous ovarian carcinoma (HGSOC) is the most lethal ovarian cancer subtype, characterized by late diagnosis, high recurrence, and resistance to platinum-based chemotherapy and PARP inhibitors. TP53 inactivation and defects in homologous recombination repair, including BRCA1/2 loss, necessitate new therapeutic strategies that engage alternative cell death pathways. Among candidate scaffolds, purine analogues have attracted attention due to their antimetabolite and kinase-modulating properties. Herein, we report the synthesis and biological evaluation of novel purine analogues, highli
