Induction of TRIM22 during Clonorchis sinensis infection triggers intracellular ROS accumulation by suppressing the mTOR-mediated Nrf2 signaling pathway.

BACKGROUND: Clonorchis sinensis infection triggers various hepatobiliary complications, including epithelial hyperplasia, chronic inflammation, periductal fibrosis, and cholangiocarcinogenesis through direct contact with worms and their excretory-secretory products (ESPs). We previously profiled differential transcriptome expression from three-dimensional cholangiocyte (H69 cells) spheroids constitutively and repetitively exposed to ESPs using microarrays and RNA-seq analysis. TRIM22 was upregulated in response to ESP exposure. OBJECTIVE: This study aimed to elucidate the pathophysiological m

Long noncoding RNA MALAT1 mediates cardiac fibrosis in experimental postinfarct myocardium mice model.

Cardiac fibrosis is a pathological remodeling response to myocardial infarction (MI) and impairs cardiac contractility. Long noncoding RNA (lncRNA) metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) is increased in patients with MI. However, the functions of MALAT1 in cardiac fibrosis have not been elucidated. This study elucidates the roles of MALAT1 in MI and the underlying mechanisms. The MI model was established by artificial coronary artery occlusion in mice. Western blot analysis and quantitative reverse transcription-polymerase chain reaction were performed to analyze prote

Elk1 and AP-1 sites in the TBP promoter mediate alcohol-induced deregulation of Pol III-dependent genes.

The major risk factors for hepatocellular carcinoma (HCC) are chronic liver diseases that include hepatitis B, hepatitis C, alcoholic liver disease and non-alcoholic steatohepatitis. However, the mechanisms of alcohol-associated HCC remain to be elucidated. The products of RNA Pol III (RNA polymerase III) dependent genes are elevated in both transformation cells and tumor cells. TBP (TATA-box binding protein) is a central transcription factor, which regulates Pol I, Pol II and Pol III gene activity. Our studies have demonstrated that alcohol increases TBP expression and Pol III gene transcript…

Clinical significance of the antibody to the putative core protein of hepatitis C virus in patients with chronic liver disease.

We evaluated the clinical significance of the antibody to hepatitis C core protein (anti-p22) analysing 147 sera from 99 patients; 45 of them had post-transfusion non A non B (NANB) hepatitis, 28 cryptogenic non A non B hepatitis, 12 chronic hepatitis B, 7 chronic hepatitis D, 6 other forms of liver disease (4 primary biliary cirrhosis, 2 autoimmune hepatitis) and 1 rheumatoid arthritis. All sera were tested by commercial 1st and 2nd-generation ELISAs and anti-p22 single antibody ELISA. We found a highly significant correspondence between anti-p22 and commercial assays (p = 0.0001). No loss of genomic imprinting of IGF-II and H19 in placentas of diabetic pregnancies with fetal macrosomia.

OBJECTIVES: Fetal macrosomia is a common complication of maternal diabetes mellitus and is associated with substantial morbidity, but the precise cellular and molecular mechanisms that induce fetal macrosomia are not well understood. The imprinted genes IGF-II and H19 are crucial for placental development and fetal growth. The term placentas from diabetic pregnancies express more insulin-like growth factor II (IGF-II) than those from normal pregnancies. Deregulation of their imprinting status is observed in the macrosomia-associated syndrome, the Beckwith-Wiedemann syndrome. The aim of this st

Targeting CDKs in the RNAPII transcription cycle.

Cyclin-dependent kinases (CDKs) are central regulators of both the cell division cycle and RNA polymerase II (RNAPII)-mediated transcription and have long been pursued as therapeutic targets in cancer and other diseases. Although drug discovery efforts have historically focused on cell-cycle CDKs, transcriptional CDKs have emerged in the past 10-15 years as promising therapeutic candidates. In this Review, we discuss our current understanding of how CDKs regulate gene expression throughout all stages of transcription from initiation and elongation to termination and beyond. We examine how tran…

The Semantics and Mechanisms of Enhancers and Promoters: "What Is True for E. coli Is True for the Elephant, Only More So".

Three fundamental classes of gene regulatory elements were identified in Escherichia coli in the 1960s: operators bound by repressor protein … matin, and they stimulate transcription via directional and short-range interactions with the basic RNA polymerase II machinery at promoters. Enhancer action at a distance is mediated by loops between proximal and distal enhancers that bring activator proteins associated with distal enhancers in proximity to promoters. Thus, Jacques Monod's 1954 conjecture that "wh …

Role of MED15 in Enhancing EMT and Metastasis in Bladder Cancer Through YAP1 Stabilization.

Bladder cancer (BLCA) remains a highly prevalent and aggressive malignancy of the urinary tract, characterized by frequent recurrence and metastasis. Mediator complex subunit 15 (MED15) functions as a key transcriptional coactivator bridging transcription factors and RNA polymerase II, yet its role in BLCA progression has not been elucidated. Here, we identify MED15 as markedly upregulated in BLCA tissues. Gene set enrichment analysis (GSEA) revealed that MED15 expression correlates with epithelial-mesenchymal transition (EMT)-related pathways. Functionally, loss- and gain-of-function assays d…

Bacteriocins in Veterinary Medicine: From Antibiotic Limitations to Targeted Solutions.

Antimicrobial resistance (AMR) has emerged as one of the foremost global threats to public health, with the veterinary sector, responsible for nearly three-quarters of global antimicrobial consumption, representing an underappreciated epicenter of this crisis. Despite the extensive literature on bacteriocins as antibiotic alternatives, most reviews focus on human medicine or food preservation, leaving a conspicuous gap in evidence specific to veterinary medicine. The present review addresses this gap by examining the molecular basis of bacteriocin activity (lipid II, bacterial RNA polymerase,

ANP32E drives vulnerability to ATR inhibitors by inducing R-loops-dependent transcription replication conflicts in triple negative breast cancer.

Oncogene-induced replicative stress (RS) drives tumor progression by disrupting genome stability, primarily through transcription-replicatio … increased genomic instability. Genome-wide analyses reveal that ANP32E-driven H2A.Z turnover alters RNA polymerase II processivity, leading to the accumulation of long R-loops at TRC sites. Furthermore, we show that ANP32E overexpression enhances TRC formation and activates an ATR-dependent DDR, predisposing cancer cells to R-loop-mediated genomic fragility. By exp …

COMPASS Ascending: Emerging clues regarding the roles of MLL3/KMT2C and MLL2/KMT2D proteins in cancer.

The KMT2 (lysine methyltransferase) family of histone modifying proteins play essential roles in regulating developmental pathways, and mutations in the genes encoding these proteins have been strongly linked to many blood and solid tumor cancers. The KMT2A-D proteins are histone 3 lysine 4 (H3K4) methyltransferases embedded in large COMPASS-like complexes important for RNA Polymerase II-dependent transcription. KMT2 mutations were initially associated with pediatric Mixed Lineage Leukemias (MLL) and found to be the result of rearrangements of the MLL1/KMT2A gene at 11q23. Over the past severa…

Renal complications and scleroderma renal crisis.

Scleroderma renal crisis (SRC) occurs in 5-10% of SSc patients, who may present with an abrupt onset of hypertension, acute renal failure, headaches, fevers, malaise, hypertensive retinopathy, encephalopathy and pulmonary oedema. Patients at greatest risk of developing SRC are those with diffuse cutaneous or rapidly progressive forms of SSc, and treatment with a recently commenced high dose of corticosteroid. Laboratory tests may demonstrate hypercreatinaemia, microangiopathic haemolytic anaemia (MAHA), thrombocytopaenia and hyperreninaemia. Renal crisis is also linked to a positive ANA speckl

Targeting the DDX3/PAF1 axis enhances chemotherapy efficacy in pancreatic ductal adenocarcinoma.

Pancreatic ductal adenocarcinoma (PDAC) is a lethal malignancy that has a poor survival rate of ∼13 % with limited options for effective therapies. DDX3 is a member of the DEAD-box RNA helicase enzyme family. It acts as an adapter protein that interacts with several transcription factors, enhancing their binding ability to the promoters of genes involved in cancer progression. Previously, we demonstrated that PAF1, a component of the RNA polymerase II-associated factor 1 complex, interacts with DDX3 to promote PDAC stemness. Here, we investigated the therapeutic efficacy of RK-33, a small-mole…

Comparative analysis of pathways induced by TMAO and TNF-α in human microvascular endothelial cells.

Endothelial dysfunction is a systemic disorder that triggers vascular alterations, characterised by a reduction in nitric oxide (NO) synthesis and/or a defective vasodilatory response. Trimethylamine-N-oxide (TMAO) is a gut microbiota-derived dietary metabolite, while tumour necrosis factor alpha (TNF-α) functions as a pro-inflammatory cytokine. Both are known to induce inflammation, metabolic modulations, and endothelial dysfunction, contributing to cardiometabolic diseases. Nevertheless, the comparative effects of TMAO and TNF-α on inflammation and metabolic modulations have yet to be invest
