Genetic, physiological and environmental factors are implicated in colorectal carcinogenesis. Mutations in the mutL homolog 1 (MLH1) gene, one of the DNA mismatch repair genes, are a main cause of hereditary colon cancer syndromes such as Lynch syndrome. Long-term chronic inflammation is also a key risk factor, responsible for colitis-associated colorectal cancer; radiation exposure is also known to increase colorectal cancer risk. Here, we studied the effects of radiation exposure on inflammation-induced colon carcinogenesis in DNA mismatch repair-proficient and repair-deficient mice. Male an

BACKGROUND: Induction of DNA double strand breaks and alterations in the repair of these breaks is implicated in breast carcinogenesis. Prior studies have demonstrated that peripheral blood mononuclear cells (PBMC) from breast cancer patients exhibit increased numbers of DNA strand breaks after exposure to ionizing radiation, but these studies did not specifically measure DNA double strand breaks and it is not known whether chemical carcinogens produce similar effects.
MATERIALS AND METHODS: PBMC from 32 women undergoing breast surgery were genotyped at nine loci of seven DNA repair genes. DN

We isolated several clones with a wide range of responses to X radiation from an unirradiated human colorectal (HCT 116) tumor cell line. The responses of one of these clones (HCT116-Clone10) and nine other clones to either fractionated or acute (i.e. single, nonfractionated doses) X irradiation in vitro was similar to that of the parental cell line. By contrast, after the same types of treatment, another clone (HCT116-Clone2) manifested a significantly increased survival whereas a third clone (HCT116-CloneK) manifested a significantly decreased survival relative to the parental cell line. BACKGROUND: The role of diet in the etiology of breast carcinoma has been debated for decades. The ecologic approach generally finds that dietary fat is highly associated with breast carcinoma mortality, with fish intake and solar ultraviolet-B (UV-B) radiation, a source of vitamin D, inversely associated. Case-control and cohort studies generally find a variety of chemical, nonfat dietary, environmental, genetic, lifestyle, and reproductive factors to be important.
METHODS: An ecologic study was conducted using breast carcinoma mortality rates (1989-1996), dietary supply data, and latitude (

Recurrent spontaneous abortion (RSA) is usually defined as three or more spontaneous abortions prior to 20-28 weeks gestation. RSA affects approximately 2-5% of all women of childbearing age, and it brings tremendous psychological and psychiatric trauma to the women and also results in economic burden. The causes could be female age, anatomical and chromosomal abnormalities, genetic, endocrinological, placental anomalies, infection, smoking and alcohol consumption, psychological factor, exposure to environmental factors such as heavy metal, environment pollution, and radiation.

Ataxia telangiectasia (AT) is a chromosomal instability syndrome with autosomal recessive inheritance, it is caused by more than 500 mutations of the ATM gene, which is involved in the cellular response to DNA damage. The diagnosis becomes difficult due to the evolution of the disease, their poor knowledge, and limited access to diagnostic tests. Chromosomal damage induced by ionizing radiation (IR) assay is still a sensitive method for early diagnosis, and it is essential for better management and genetic counseling. This paper shows diagnosis and follow-up in four cases with AT.

Familial concentration of Hodgkin's disease has attracted special attention (Razis et al., 1959; De Vore and Doan, 1957; Halazun et al., 1972; Creagan and Fraumeni, 1972) as it could suggest genetical basis. The present report states the unusual occurrence of Hodgkin's disease in four siblings. Three of them descended from the same father and the last one had a different one, as it is shown in the pedigree enclosed. Consanguinity or environmental exposure to radiation has not been detected.

New clinical and pathologic findings in patients with deletion of the long arm of chromosome 13 (13q-) include optic nerve hypoplasia and retinal dysplasia. Fibroblasts derived from patients with a 13q- syndrome with and without retinoblastoma, as well as from familial and sporadic retinoblastoma, are a useful model for the study of genetic susceptibility to the development of spontaneous and radiation-induced cancers. Fibroblasts from patients with hereditary retinoblastoma appear more radiosensitive than fibroblasts from patients with sporadic retinoblastoma or normal control patients.

Radiation-induced glioma (RIG) is a highly aggressive brain cancer arising as a consequence of radiation therapy. We report a case of RIG that arose in the brain stem following treatment for paediatric medulloblastoma, and the development and characterisation of a matched orthotopic patient-derived xenograft (PDX) model (TK-RIG915). Patient and PDX tumours were analysed using DNA methylation profiling, whole genome sequencing (WGS) and RNA sequencing. While initially thought to be a diffuse intrinsic pontine glioma (DIPG) based on disease location, results from methylation profiling and WGS we

While radiation therapy (RT) can improve cancer outcomes, it can lead to radiation-induced heart dysfunction (RIHD) in patients with thoracic tumors. This study examines the role of adaptive immune cells in RIHD. In Salt-Sensitive (SS) rats, image-guided whole-heart RT increased cardiac T-cell infiltration. We analyzed the functional requirement for these cells in RIHD using a genetic model of T- and B-cell deficiency (interleukin-2 receptor gamma chain knockout (IL2RG-/-)) and observed a complex role for these cells. Surprisingly, while IL2RG deficiency conferred protection from cardiac hyper

Porokeratosis encompass a group of acquired and familial, preneoplastic, keratinization disorders, clinically characterized by atrophic macules or patches with a peripheral keratotic rim, the cornoid lamella. Genetic background is recognized as crucial in its pathophysiology, while immunosuppression and ultraviolet radiation represent triggering factors. We report the case of a woman who developed disseminate superficial actinic porokeratosis following the intake of hydroxyurea for a polycythaemia vera. Clinical, dermoscopic and histopathology data are showed, and the role of drug as a second-

Neurofibromatosis type 2 (NF2) is a dominantly inherited genetic condition. Bilateral vestibular schwannoma, which are benign tumors, composed of neoplastic Schwann cells that arise from the eighth cranial nerve, are the hallmark of NF2. Standard approaches for treatment of growing vestibular schwannoma include observation, surgical removal and radiation therapy. Molecular targeted therapies also present great prosperity in recent years. In this review, we summarize the latest progresses on the treatment of NF2-associated vestibular schwannoma.

Patients receiving radiotherapy often experience toxicity of the skin and mucous membranes. While radiotherapy is a mainstay of myeloablative conditioning for allogeneic hematopoietic stem cell transplantation (ASCT), no risk factors for radiotoxicity have been identified in this setting. Here, we report on a patient with excessive radiation-induced toxicity after ASCT who carried a heterozygous mutation in the Ataxia telangiectasia mutated (ATM) gene. This is the first case to suggest a genetic basis for increased radiotoxicity after myeloablative ASCT.

Welding involves exposure to fumes, gases, radiation, electricity, noise, and heat. Herein, we describe 2 welders presenting with lung infiltrates and elevated liver enzyme levels. Both of these patients had pulmonary siderosis ("welder's lung") on lung biopsy along with evidence of systemic iron overload. Evaluation for genetic hemochromatosis and other known causes of iron overload was unrevealing. Welding with chronic inhalation of iron particles maybe an under-recognized source of systemic iron overload.

Hereditary Hemorrhagic Telangiectasia (HHT) is a genetic disorder of blood vessels, which affects about 1 in 10,000 people. The disorder is also sometimes referred to as Osler-Weber-Rendu Disease after the doctors who studied HHT. The disorder affects males and females from all racial and ethnic groups, and the most commonly affected organs are the nose, lungs, gastrointestinal tract, and brain--in that order. Radiation therapy should be considered for intractable bleeding from the upper airway or alimentary tract.

A review of the factors involved in treating the cancer patient during pregnancy is presented with respect to the fetus and the mother. The fetus may be adversely affected by diagnostic procedures, radiation therapy, cytotoxic chemotherapy or by the mother's metastatic disease or diminished health. Prenatal consideration must be given to the mother's additional nutritional requirements, susceptibility to infection, the effect of cancer therapies on blood counts and likelihood of hemorrhage. Postpartum and postnatal treatment should include cancer assessment, contraceptive and genetic counselli

Evolutionary innovation is a key driver of the colonization of new environments and the adaptive radiations of major groups. Novel traits typically evolve through the modification of pre-existing characters but the genetic paths underlying their origin have been challenging to trace, and the general requirements for and relative order of different kinds of gene mutations have been difficult to assess. Here, we trace the genomic origins of four procoagulant venom toxins (factor X, factor V, group I phospholipase A2, and Kunitz-type toxins) that collectively underlie a novel, especially potent b

PURPOSE: Radiation-induced brain injury (RIBI) poses significant clinical challenges, underscoring the limited mechanistic understanding in this field. This study systematically investigates both the genetic and metabolic alterations induced by RIBI and their differential regional impacts across brain structures.
METHODS: Mice received cranial irradiation with a single 30 Gy X-ray dose. Behavioral assessments, including the open field test (OFT), elevated plus maze test (EPM), and Morris water maze test (MWM), were conducted to evaluate the impact of RIBI on mouse behavior. Hippocampal and co
