Species composition and relative abundance of sand flies of the genus Lutzomyia (Diptera: Psychodidae) at an endemic focus of visceral leishmaniasis in Colombia.

Ecological studies on the sand fly Lutzomyia longipalpis (Lutz & Neiva) were conducted during 1990-1993 at a small rural community in Colombia where American visceral leishmaniasis is endemic. Weekly sand fly collections were made from pigpens, houses, and natural resting sites, using hand-held aspirators, sticky (oiled) paper traps, and opossum-baited Disney traps. In total, 263,094 sand flies were collected; L. longipalpis predominated (86.1%), followed by L. trinidadensis (11.0%), L. cayennensis (2.7%), and 8 other Lutzomyia species. The species composition and sex ratio of these sand flies…

Effects of sand viper (Cerastes cerastes) venom on isolated smooth muscle and heart and on haematological and cardiovascular parameters in the guinea-pig.

The effects of the venom of the sand viper (Cerastes cerastes) on haematological and cardiovascular parameters and on isolated ileum, trachea, pulmonary artery and atrium from the guinea-pig were studied. In concentrations from 0.2 micrograms/ml to 0.6 mg/ml, snake venom caused concentration-dependent relaxation of the longitudinal ileal segments and the epinephrine-precontracted pulmonary artery rings, increased the tone of the tracheal rings and increased the rate of the spontaneously beating atrium but depressed the amplitude of its contraction. Low doses (0.04-0.4 mg) of the venom caused t…

Joint analysis of microsatellites and flanking sequences shows shared demographic response of coastal sand dune plant communities to past environmental changes.

Understanding recent change in effective population size is crucial to evaluate how specie respond to environmental modifications and their ability to adapt to global changes. However, the impact of past environmental change on the demography history of species communities has rarely been studied across multiple species, members of the same community sharing the same habitat. In this study, we compare historical change in effective population size across eight species of coastal sand dune plant communities along the coastal dune in Southwest France. We developed and sequenced species-specific …

Sand fly (Diptera: Psychodidae: Phlebotominae) species composition in an area with a recent American Tegumentary Leishmaniasis outbreak in Minas Gerais, Brazil.

American tegumentary leishmaniasis (ATL) constitutes an infectious, non-contagious pathology that manifests as ulcerative lesions on the integumentary system and mucosal surfaces. The causative agents are parasites belonging to the genus Leishmania (Protozoa: Kinetoplastida: Trypanosomatidae). In Brazil, the principal ATL agents include Leishmania (Leishmania) amazonensis, Leishmania (Viannia) guyanensis, and Leishmania (Viannia) braziliensis. ATL transmission to humans occurs via the hematophagous activity of phlebotomine sand flies (Diptera: Psychodidae: Phlebotominae). Despite the endemic s…

Comparative quantification of Leishmania infantum in experimental phlebotomine sand fly infections using kDNA and single-copy Meta-1 gene qPCR assays.

BACKGROUND: Leishmaniasis is a parasitic disease transmitted by female sand flies and caused by protozoan parasites of the genus Leishmania. Accurate quantification of parasite load within vectors is essential for understanding transmission dynamics and vector competence. This study compares two quantitative polymerase chain reaction (qPCR) methods for detecting and quantifying Leishmania infantum in three experimentally infected sand fly species (Phlebotomus perniciosus, Phlebotomus argentipes, and Phlebotomus orientalis). METHODS: One method targets kinetoplast minicircle DNA, which offers …

Phlebotomine sand fly survey, blood meal source identification, and description of Sergentomyia imihra n. sp. in the central Sahara of Algeria.

BACKGROUND: Phlebotomine sand flies (Diptera: Psychodidae) are important vectors of various pathogens, mainly Leishmania parasites. In the Old World, the most important genus in term of pathogens transmission is the genus Phlebotomus, which includes many proven or suspected vectors of several Leishmania species, while the genus Sergentomyia remains so far unproven as a vector of human pathogens. Algeria is one of the most affected countries by human leishmaniasis. METHODS: In the present study, an entomological survey was carried out in two provinces, Ghardaïa and Illizi, located in the north…

Influence of filter backwashing on iron, manganese, and ammonium removal in dual-media rapid sand filters used for drinking water production.

Iron (Fe), manganese (Mn), and ammonium (NH4+) removal from groundwater using rapid sand filtration is a widely employed method in drinking water production. Over time, Fe and Mn oxides accumulate in the filter, which necessitates frequent backwashing to avoid clogging. In this study, we investigated the impact of backwashing on the microbial community and filter chemistry in a dual-media filter comprising anthracite and sand layers. Specifically, we focused on the removal of Fe, Mn, and NH4+ over the runtime of the filter. With increasing runtime, depth profiles of dissolved and particulate F…

Influence of filter age on Fe, Mn and NH4+ removal in dual media rapid sand filters used for drinking water production.

Rapid sand filtration is a common method for removal of iron (Fe), manganese (Mn) and ammonium (NH4+) from anoxic groundwaters used for drinking water production. In this study, we combine geochemical and microbiological data to assess how filter age influences Fe, Mn and NH4+ removal in dual media filters, consisting of anthracite overlying quartz sand, that have been in operation for between ∼2 months and ∼11 years. We show that the depth where dissolved Fe and Mn removal occurs is reflected in the filter medium coatings, with ferrihydrite forming in the anthracite in the top of the filters …

Biting rhythms and infection rates of anthropophilic sand fly species (Diptera: Phlebotominae) in sites with different land use in southern Mexico.

Could tropical forest conversion shape sand fly (Diptera: Phlebotominae) biting rhythms and Leishmania infection rates? Using a Shannon trap, we estimated the bite rate and infection prevalence among anthropophilic sand flies at sites with different land use in southern Mexico. We estimated the expected monthly infection rate of the Leishmania parasite along the gradient and generated information on the biting rhythm of sand flies in a poorly characterized cutaneous leishmaniasis endemic region. We used generalized mixed linear and mixed additives models to evaluate differences in the biting r…

Co-transport and co-release of Eu(III) with bentonite colloids in saturated porous sand columns: Controlling factors and governing mechanisms.

Accurate prediction of the colloid-driven transport of radionuclides in porous media is critical for the long-term safety assessment of radioactive waste disposal repository. However, the co-transport and corelease process of radionuclides with colloids have not been well documented, the intrinsic mechanisms for colloids-driven retention/transport of radionuclides are still pending for further discussion. Thus the controlling factors and governing mechanisms of co-transport and co-release behavior of Eu(III) with bentonite colloids (BC) were discussed and quantified by combining laboratory-sca

Detection of Specific Antibodies against Toscana Virus among Blood Donors in Northeastern Italy and Correlation with Sand Fly Abundance in 2014.

Toscana virus (TOSV) is a Phlebovirus transmitted by phlebotomine sand flies and is an important etiological agent of summer meningitis in the Mediterranean basin. Since TOSV infection is often asymptomatic, we evaluated the seroprevalence in blood donors (BDs) in the Bologna and Ferrara provinces (Northeastern Italy)-the areas with the highest and lowest numbers of TOSV neuroinvasive cases in the region, respectively. A total of 1208 serum samples from BDs were collected in April-June 2014 and evaluated for the presence of specific TOSV-IgG by ELISA. The IgG-reactive samples were confirmed by…

Aerobic midgut microbiota of sand fly vectors of zoonotic visceral leishmaniasis from northern Iran, a step toward finding potential paratransgenic candidates.

BACKGROUND: Leishmaniasis is caused by Leishmania parasites and is transmitted to humans through the bite of infected sand flies. Development of Leishmania to infective metacyclic promastigotes occurs within the sand fly gut where the gut microbiota influences development of the parasite. Paratransgenesis is a new control method in which symbiotic bacteria are isolated, transformed and reintroduced into the gut through their diet to express anti-parasitic molecules. In the present study, the midgut microbiota of three sand fly species from a steppe and a mountainous region of northern Iran, wh…

Establishment of a method for Lutzomyia longipalpis sand fly embryo microinjection: The first step towards potential novel control strategies for leishmaniasis.

Leishmaniasis is a vector-borne parasitic disease transmitted by sand flies that affects 1.3 million people across 98 countries, with limited control strategies due to the lack of an available vaccine and the emergence of insecticide resistance.  Novel control strategies that are being explored for mosquito-borne diseases, such as Wolbachia bacterial inhibition of pathogens and genetically modified insects (e.g. using CRISPR-Cas9 editing), rely on the ability to consistently inject embryos of the target species.  Here we present a novel method to obtain and inject preblastoderm sand fly embryo…

Transmission blocking sugar baits for the control of Leishmania development inside sand flies using environmentally friendly beta-glycosides and their aglycones.

BACKGROUND: The sand fly Lutzomyia longipalpis is the main vector of American visceral leishmaniasis, a disease caused by parasites of the genus Leishmania. Adults of this insect feed on blood (females only) or sugar from plant sources, but their digestion of carbohydrates is poorly studied. Beta-glycosides as esculin and amygdalin are plant compounds and release toxic compounds as esculetin and mandelonitrile when hydrolyzed. Beta-glucosidase and trehalase are essential enzymes in sand fly metabolism and participate in sugar digestion. It is therefore possible that the toxic portions of these…

Species composition of sand flies and bionomics of Phlebotomus papatasi and P. sergenti (Diptera: Psychodidae) in cutaneous leishmaniasis endemic foci, Morocco.

BACKGROUND: Cutaneous Leishmaniasis (CL) is one of the most neglected tropical diseases in Morocco. Leishmania major and L. tropica are the main culprits identified in all endemic foci across the country. These two etiological agents are transmitted by Phlebotomus papatasi and P. sergenti, the two most prevalent sand fly species in Morocco. Previous studies reflected gaps of knowledge regarding the environmental fingerprints that affect the distribution of these two potential vectors across Morocco. METHODS: The sand flies were collected from 48 districts across Morocco using sticky paper tra…

Environmental Niche Modelling of Phlebotomine Sand Flies and Cutaneous Leishmaniasis Identifies Lutzomyia intermedia as the Main Vector Species in Southeastern Brazil.

Cutaneous leishmaniasis (CL) is caused by a protozoan of the genus Leishmania and is transmitted by sand flies. The state of Espírito Santo (ES), an endemic area in southeast Brazil, has shown a considerably high prevalence in recent decades. Environmental niche modelling (ENM) is a useful tool for predicting potential disease risk. In this study, ENM was applied to sand fly species and CL cases in ES to identify the principal vector and risk areas of the disease. Sand flies were collected in 466 rural localities between 1997 and 2013 using active and passive capture. Insects were identified t…
